RESUMO

This paper proposes an analytical framework for the analysis of organizational cognition that borrows from distributed and ecological cognition. In so doing, we take a case study featuring a decision on the topic of agreeing on a set point in the agenda of a meeting. It is through the analysis of a few minutes of video-recording used in the case that enables us to demonstrate the power of applying distributed and ecological cognition to organizing processes. Cognitive mechanism, resources, and processes are identified within this combined framework. Mechanisms are described as "socio-material" (CM1)-where "people" and "artifacts" are the related cognitive resources-and as "conceptual" (CM2)-with "group" identity, "topic" understanding, meaning of "procedures," and perception of "time" as resources. Processes are defined as "coupling," "de-coupling," and "un-coupled" depending on the type of relation in place. Finally, the paper presents an agent-based computational simulation to demonstrate the potentials of operationalizing this approach.

While a substantial body of literature suggests that lasting community mentoring relationships can have a range of positive effects on youths, little is known about these effects in the Nordic welfare context, where community mentees may have lower risk profiles compared to many previous samples. This study explores how the duration (length) of child mentoring relationships predicts parental perceptions of child well-being among 197 children served by Denmark's most extensive community-based youth mentoring program. We find that children who have had a mentor for at least one year are perceived to have significantly higher well-being. In contrast, we find no significant differences in well-being between children who had mentors for less than one year and children on a waiting list. Previous research, conducted in primarily North American contexts, finds that longer mentoring relationships substantially improve school behavior and reduce risk taking. Our results add to the literature by indicating that a minimum mentoring relationship duration of one year appears to be similarly important in promoting well-being for youths involved in community-based mentoring programs in a Nordic welfare context.

Expression of recombinant proteins traditionally require a cellular system to transcribe and translate foreign DNA to a desired protein. The process requires special knowledge of the specific cellular metabolism in use and is often time consuming and labour intensive. A cell free expression system provides an opportunity to express recombinant proteins without consideration of the living cell. Instead, a cell free system relies on either a cellular lysate or recombinant proteins to carry out protein synthesis, increasing overall production speed and ease of handling. The one-pot cell free setup is commonly known as an in vitro transcription/translation reaction (IVTT). Here we focused on a PURE (Protein synthesis Using Recombinant Elements) IVTT system based on recombinant proteins from Escherichia coli. We evaluated the cell free system's ability to express functional insulin analogues compared to Saccharomyces cerevisiae, a well-established system for large scale production of recombinant human insulin and insulin analogues. Significantly, it was found that correct insulin expression and folding was governed by the inherent properties of the primary amino acids sequence of insulin, whereas the eukaryotic features of the expression system apparently play a minor role. The IVTT system successfully produced insulin analogues identical in structure and with similar insulin receptor affinity to those produced by yeast. In conclusion we demonstrate that the PURE IVTT system is highly suited for expressing soluble molecules with higher order features and multiple disulphide bridges.

Septin9 (Sept9) is a member of the filament-forming septin family of structural proteins and is associated with a variety of cancers and with hereditary neuralgic amyotrophy. We have generated mice with constitutive and conditional Sept9 knockout alleles. Homozygous deletion of Sept9 results in embryonic lethality around day 10 of gestation whereas mice homozygous for the conditional allele develop normally. Here we report the consequences of homozygous loss of Sept9 in immortalized murine embryonic fibroblasts. Proliferation rate was not changed but cells without Sept9 had an altered morphology compared to normal cells, particularly under low serum stress. Abnormal, fragmented, and multiple nuclei were more frequent in cells without Sept9. Cell migration, as measured by gap-filling and filter-invasion assays, was impaired, but individual cells did not move less than wild-type cells. Sept9 knockout cells showed a reduced resistance to hypo-osmotic stress. Stress fiber and vinculin staining at focal adhesion points was less prominent. Long septin filaments stained for Sept7 disappeared. Instead, staining was found in short, often curved filaments and rings. Furthermore, Sept7 was no longer localized to the mitotic spindle. Together, these data reveal the importance of Sept9 for septin filament formation and general cell stability.

Abortion in dairy cows in well-managed dairies is not common but differences have been reported probably due to variation in animal health, nutrition and management, as well as difficulties in observing the aborted material. A 38-month study of 507 large Danish dairy herds revealed 3354 late-term abortions and 224,419 calvings were recorded. During the study period, a total of 3717 submissions were made to the Danish Institute for Food and Veterinary Research (DFVF). A broad spectrum of abortive agents was isolated but none were found to be statistically associated with abortions. The number of abortions in a month on a dairy was significantly (P<0.001) associated with the number of cows in the third trimester but explained only 11% (R(adj)(2)=0.114) of the variability of the reported abortions. A total of 23 herds (4.5%) reported 531 abortions (15.8%). Although a marginally significant (P=0.11) risk of increased abortions was found to exist in bovine viral diarrhea virus- (BVDV) infected herds, it could be at least partially explained by additional calvings in those herds. Temporal correlation between inseminations and abortions was statistically significant (P<0.001) with the highest correlation (r=0.47-0.51) after lagging abortions on insemination by 6-8 months. No indication of spatial clustering was detected for either specific-abortogenic pathogens or high aborting dairies using either Cuzick-Edwards' (P>0.17) or spatial scan tests (P>0.23). Ederer-Myers-Mantel test was applied to 3 years of data on the highest aborting dairies and showed that July had nearly double the expected number of maximum monthly abortions (P<0.001). These findings provide further insight into the reported abortion pattern in Danish dairies and may facilitate planning future control programs.

From the pK(a) values of the conjugate acids of a large series of hydroxylated piperidines and hexahydropyridazines, a consistent difference in basicity was found between stereoisomers having an axial or equatorial hydroxyl (OH) group either beta or gamma to the amine. Compounds with an equatorial OH group in the 3-position were 0.8 pH units more acidic than otherwise identical compounds with an axial OH group, whilst compounds with an equatorial OH group in the 4-position relative to the amine were 0.4 pH units more acidic than the corresponding compound with an axial OH. A similar effect was observed for the COOMe substituent. The difference in electron-withdrawing power of axial and equatorial substituents was explained by a difference in charge-dipole interactions in the two systems. Since this stereoelectronic substituent effect causes differences in basicity in different conformers, certain piperidines and hexahydropyridazines were found to change conformation upon protonation. A method for predicting the pK(a) of piperidines which takes stereochemistry into account is described.
