RESUMO

Wrinkling is prominent manifestation of aging skin. A mutant phenotype characterized by systemic wrinkles and thickened skin was discovered in Xiang pig populations with incidence about 1-3%. The feature in histological structure was epidermal hyperplasia and thickening, collagen fibers disorder. To uncover genetic mechanisms for the mutant phenotype of Xiang pigs with systemic wrinkle (WXP), a genome-wide of structural variations (SVs) in WXP was described by next generation resequencing, taking Xiang pigs (XP) and European pigs (EUP) as compares. Total of 32,308 SVs were detected from three pig groups and 965 SVs were identified specifically from WXP, involving 481 protein-coding genes. These genes were mainly enriched in nuclear structure, ECM components and immunomodulatory pathways. According to gene function and enrichment analysis, we found that 65 candidate SVs in 59 protein genes were probably related with the systemic wrinkle of WXP. Of these, several genes are reported to be associate with aging, such as EIF4G2, NOLC1, XYLT1, FUT8, MDM2 and so on. The insertion/deletion and duplication variations of SVs in these genes resulted in the loss of stop-codon or frameshift mutation, and aberrant alternative splicing of transcripts. These genes are involved in cell lamin filament, intermediate filament cytoskeleton, supramolecular complex, cell differentiation and regulation of macromolecule metabolic process etc. Our study suggested that the loss of function or aberrant expression of these genes lead to structural disorder of nuclear and the extracellular matrix (ECM) in skin cells, which probably was the genetic mechanisms for the mutant phenotype of systemic skin wrinkle of Xiang pig.

Purpose To investigate 18F-FDG PET/CT imaging characteristics of multiple myeloma (MM) and bone metastatic tumor,and evaluate the diagnostic value of 18F-FDG PET/CT in the identification of MM and bone metastatic tumor.Materials and Methods Thirty patients who were definitely diagnosed as MM and another 30 cases with bone metastatic tumor confirmed by through pathology in the First Affiliated Hospital of Harbin Medical University from September 2010 to February 2017 were chosen to receive 18F-FDG PET/CT imaging.Focal distribution,type of bone destruction,maximum standardized uptake value and metabolic homogeneity in the two groups were compared.In addition,18F-FDG metabolic profile was also compared with that of the 30 controls with healthy bone.Results MM and bone metastatic tumor were mostly seen on spine,pelvis and chest bone,followed by limbs.Focal occurrence rate of the spine,pelvis and limbs had no statistical difference (P>0.05).MM would often involve skull while bone metastatic tumor involved skull less often and differences among patients in the two groups were of statistical significance (P<0.05).Uptaking abilities of MM and bone metastatic tumor on 18F-FDG were higher than that of healthy bones and the difference was of statistical importance (P<0.05).MM on 18F-FDG was mostly represented as diffuse slight uptake and bone metastatic tumor was more often represented as imbalanced uptake.Among MM focal in this group,osteolytic bone destruction occupied 96.7% and was mostly represented as "chisel-like" or "insect-bite-like".In addition,the bone was in expansive change,which was obvious in ribs and osteoblastic bone change was rare (3.3%).Among bone metastatic tumor focal,bone destruction was 76.7%,mostly represented as focal lesions and osteoblastic change was about 36.7%.As bone destruction occurred in MM and bone metastatic tumor,soft tissues mass was formed.Difference in the two groups had no statistical significance (x2=0.07,P>0.05).Conclusion 18F-FDG PET/CT examination can obtain anatomical,metabolic and other imaging features and is of higher value for the identification and diagnosis of MM and bone metastatic tumor.

RESUMO

HEV is a positive strand RNA virus containing 3 open reading frames(ORFs), while the function of its binding protein pORF3 is poorly understood. In our study, the interactive protein of the pORF3 from human liver cDNA library was screened by the SOS recruitment system, and the construction of the pSOS bait-ORF3 plasmid was identified. The expression of the pORF3 was analyzed by Western blotting, and the detection result of the self activation of bait protein was obtained as expected. Totally, 8 putative interacting proteins had been successively screened. Some of these cellular binding proteins of pORF3, especially the important immunological pathway kinase p110δ, may advance our understanding of the role in ORF3 protein of HEV, and the possible implications of these candidate proteins for the further research are discussed.

AIM: To investigate the ameliorative effect of water extract of propolis (WEP) preconditioning on mesenteric microcirculation of rats subjected to small intestinal ischemia reperfusion (I/R). METHODS: Thirty-two male Wistar rats were randomly divided into sham, I/R and WEP (100, 200 mg/kg) preconditioning groups. Model of small intestinal I/R injury was made by clamping super mesenteric artery for 45 min followed by 120 min of reperfusion in rats. Mesenteric microcirculation was detected at the end of reperfusion. The degree of small intestinal injury was The content of soluble intercellular adhesion molecule-1 (sICAM-1) in plasma and myeloperoxidase (MPO) activity in intestinal tissue were detected using enzyme linked-immuno-sorbent assay (ELISA) and spectrophotometer, respectively. RESULTS: (1) WEP preconditioning alleviated significantly the pathologic lesion in small intestine, and wet/dry ratio, compared to those in I/R group (P<0.01). (2) The disturbance of the blood flow in microvessel induced by I/R was improved significantly by WEP. In addition, WEP preconditioning alleviated significantly the decrease in diameters of microvessels and microcirculatory blood velocity (P<0.05 vs I/R group) and inhibited the adherence of leukocytes to venule (P<0.01 vs I/R group) in a dose-dependent manner. SICAM-1 content in plasma and MPO activity in intestinal tissue were decreased in WEP preconditioning group, compared to those in I/R group (P<0.05 or P<0.01). CONCLUSION: WEP preconditioning ameliorates mesenteric microcirculation of rats subjected to small intestinal I/R through suppressing the activation of polymorphonuclear neutrophils mediated by ICAM-1.

BACKGROUND: It has been proved that matrix metalloproteinase-9 (MMP-9) has a correlation with stability of atherosclerotic plaque, and propolis flavone plays a role in anti-atherosclerosis. OBJECTIVE: To investigate the effect of propolis flavone on expression of MMP-9 in human umbilical vein endothelial cell (HUVEC). DESIGN, TIME AND SETTING: The comparative observation was performed at the Institute of Life Science, Taishan Medical University from January to August 2008. MATERIALS: The propolis flavone was extracted by the Institute of Life Science, Taishan Medical University. The neonatal umbilical cord with 1 hour was supplied by Tai’an Central Hospital, and the informed consent was obtained from the puerperant. METHODS: HUVEC was cultured and identified in vitro, followed by treated with propolis flavoneat with different concentrations for 24 hours. The blank was used as the control group. MAIN OUTCOME MEASURES: The effect of propolis flavone on expression of MMP-9 in HUVEC was detected by cytoimmunochemical staining and ELISA. RESULTS: Propolis flavonecan with all concentrations can restrain the expression of MMP-9 after co-culture with the cells for 24 hours, which showed in a dose-dependent manner. CONCLUSION: The propolis flavone can obvious depress the expression of matrix metalloproteinase-9 in HUVEC.

Tissue culture, biochemical techniques and radioimmunoassay were used to study the effects of Compound Lian Zhu Capsule on micrangium lesions in diabetic rats. The results indicated that blood sugar, glycosylated hemoglobin (GHb), urinary protein and malondialdehyde (MDA) contents, aldose reductase (AR) activity and 3H-TdR incorporation rate in the vascular smooth muscle cell (VSMC) were significantly higher, and plasma NO content in the diabetes mellitus (DM) group were significantly lower than those in the normal control group (both P < 0.05). The above indexes in the Chinese medicine (TCM) treatment group were improved significantly as compared with the DM group, with no significantly differences, except urine volume and urinary protein, as compared with the normal control group. It is suggested that Compound Lian Zhu Capsules cansignificantly alleviate the complicated lesions of the micrangium in diabetic rats.

ObjectiveTo observe the effect of dehydroepiandrostrone (DHEA) on retinal capillary pericytes’ proliferation under high glucose and research the prevention and treatment of diabetic retinopathy (DR).MethodsRetinas were dissected from eyes of fresh killed cattle and retinal capillary pericytes were separated and cultured.The effect of DHEA (10、50、100、500、1 000nmol/L) on pericytes’ proliferetion was evaluated by 3H-thymidine incorporation and li-quid scintillation techniques.ResultsThere was significant difference between DHEA (50、100、500、1 000nmol/L) and control culturing for 48h.The increasing rate was 42.67% in DHEA of 500nmol/L.ConclusionDHEA could promote pericytes’ proliferation under high glucose.This provided reliable evidence for selecting effective drugs for prevention and treatment of DR clinically.

Objective To observe the effect of high glucose on retinal capillary pericytes in vitro and research the pathogenesis of diabetic retinopathy (DR). Methods Retinas dissected from eyes of fresh killed cattle were separated and retinal capillary pericytes were cultured. The effect of high glucose on pericytes was evaluated by measurement of lactate dehydrogenase(LDH),3H-thymidine incorporation and liquid scintillation techniques.Morphological changes were observed under microscope and electric microscope.Results There was no significant difference in LDH activity in the supernatants between high glucose(10、20、40mmol*L-1) and normal glucose(5mmol*L-1)cultured for 48h or 96h. High glucose had an effective inhibition on pericytes' proliferation cultured for 48h,96h and 144h, and the inhibition rate increased when the glucose concentration rised and time passed by. The inhibition rate of pericytes was 78.09% after cultured in high glucose (40mmol*L-1) for 144h. Conclusion High glucose has an effect inhibition on pericytes' proliferation, and the inhibition rate increase when the glucose concentration rises and time passes by.

To provide necessary evidence for studying the pharmacological and clinical effectiveness of Styela clava,the quantities of 21 chemical elements in Styela clava were analysised.It was found that the quantities of several mineral elements in it were higher than that in sea water,which indicated Styela clava had a relatively high ability to concentrate mineral elements.

AIM:To investigate the ameliorative effect of ischemic postconditioning (I-postC) on pia mater microcirculation in rats subjected to cerebral ischemia reperfusion (I/R) and its mechanisms. METHODS:Thirty-two male Wistar rats were randomly divided into sham,I/R,I-postC,and ischemic preconditioning (IPC) groups. The global cerebral I/R injury was induced by shunting carotid artery in rats. Pia mater microcirculation and cerebral microcirculatory perfusion were measured after reperfusion. The content of soluble intercellular adhesion molecule-1 (sICAM-1) in plasma was detected using enzyme linked-immunosorbent assay (ELISA). Myeloperoxidase (MPO),malondialdehyde (MDA),and superoxide dismutase (SOD) in cerebral tissue were detected. The expressions of vascular endothelial cell cadherin (VE-cadherin) and NF-?B p65 in cerebral tissue were assayed by Western blotting. RESUMO

This paper deals with three cases of long jumpers suffering from fracture at middle one-third of the femoral shaft. This kind of fracture is transverse and produced by indirect force at the landing moment. This is a characteristic fracture which we call a jumping femoral fracture.Physical examination reveals swelling with tenderness and deformity of the injured thigh. X-ray shows a transverse fracture and angulating deformity at middle one-third femoral shaft in all cases.Two cases were performed by open reduction and internal fixation with K?ntschcr or medullary nail, and one case was performed by traction through the intercondylus of the lower femur. All patients got good result after one year, but open reduction was better than skeletal traction.According to clinical observations, CT examination of the femur in normal person and experiment of the femoral shaft with fresh cadaver, the mechanism is based on the imbalance of the body rising high into the air, which produces both scissors stress and rotational force at landing. In the meanwhile the posterior group muscles of the thigh were more likely to be relaxed reflectively, leaving the femoral shaft unprotected.
