RESUMO

Hierarchical self-assembly of synthetic polymers in solution represents one of the sophisticated strategies to replicate the natural superstructures which lay the basis for their superb functions. However, it is still quite challenging to increase the degree of complexity of the as-prepared assemblies, especially in a large scale. Liquid-liquid phase separation (LLPS) widely exists in cells and is assumed to be responsible for the formation of many cellular organelles without membranes. Herein, through integrating LLPS with the polymerization-induced self-assembly (PISA), a coacervate-assisted PISA (CAPISA) methodology to realize the one-pot and scalable preparation of hierarchical bishell capsules (BCs) from nanosheets with ultrathin lamellae phase (sub-5 nm), microflakes, unishell capsules to final BCs in a bottom-up sequence is presented. Both the self-assembled structure and the dynamic formation process of BCs have been disclosed. Since CAPISA has combined the advantages of coacervates, click chemistry, interfacial reaction and PISA, it is believed that it will become a promising option to fabricate biomimetic polymer materials with higher structural complexity and more sophisticated functions.

Among the few available mRNA delivery vehicles, lipid nanoparticles (LNPs) are the most clinically advanced but they require cumbersome four components and suffer from inflammation-related side effects that should be minimized for safety. Yet, a certain level of proinflammatory responses and innate immune activation are required to evoke T-cell immunity for mRNA cancer vaccination. To address these issues and develop potent yet low-inflammatory mRNA cancer vaccine vectors, a series of alternating copolymers "PHTA" featured with ortho-hydroxy tertiary amine (HTA) repeating units for mRNA delivery is synthesized, which can play triple roles of condensing mRNA, enhancing the polymeric nanoparticle (PNP) stability, and prolonging circulation time. Unlike LNPs exhibiting high levels of inflammation, the PHTA-based PNPs show negligible inflammatory side effects in vivo. Importantly, the top candidate PHTA-C18 enables successful mRNA cancer vaccine delivery in vivo and leads to a robust CD8+ T cell mediated antitumor cellular immunity. Such PHTA-based integrated PNP provides a potential approach for establishing mRNA cancer vaccines with good inflammatory safety profiles.

Rechargeable aqueous zinc-ion batteries (ZIBs) have attracted extensive attention due to their low cost and high safety. However, the critical issues of dendrite growth and side reactions on the Zn metal anode hinder the commercialization of ZIBs. Herein, we demonstrated that the formation of Zn4SO4(OH)6·5H2O byproducts is closely relevant to the direct contact between the Zn electrode and SO42-/H2O. On the basis of this finding, we developed a cation-exchange membrane of perfluorosulfonic acid (PFSA) coated on the Zn surface to regulate the Zn plating/stripping behavior. Importantly, the PFSA film with abundant sulfonic acid groups could simultaneously block the access of SO42- and H2O, accelerate the Zn2+ ion transport kinetics, and uniformize the electrical and Zn2+ ion concentration field on the Zn surface, thus achieving a highly reversible Zn plating/stripping process with corrosion-free and dendrite-free behavior. Consequently, the PFSA-modified Zn anode exhibits high reversibility with 99.5% Coulombic efficiency and excellent plating/stripping stability (over 1500 h), subsequently enabling a highly rechargeable Zn-MnO2 full cell. The strategy of the cation-exchange membrane proposed in this work provides a simple but efficient method for suppression of side reactions.
