RESUMO

The binding of SARS-CoV-2 nucleocapsid (N) protein to both the 5'- and 3'-ends of genomic RNA has different implications arising from its binding to the central region during virion assembly. However, the mechanism underlying selective binding remains unknown. Herein, we performed the high-throughput RNA-SELEX (HTR-SELEX) to determine the RNA-binding specificity of the N proteins of various SARS-CoV-2 variants as well as other {beta}-coronaviruses and showed that N proteins could bind two unrelated sequences, both of which were highly conserved across all variants and species. Interestingly, both these sequence motifs are virtually absent from the human transcriptome; however, they exhibit a highly enriched, mutually complementary distribution in the coronavirus genome, highlighting their varied functions in genome packaging. Our results provide mechanistic insights into viral genome packaging, thereby increasing the feasibility of developing drugs with broad-spectrum anti-coronavirus activity by targeting RNA binding by N proteins.

Neuroinflammation is an important part of the development of neurodegenerative diseases such as Alzheimer's disease (AD), Parkinson's and amyotrophic lateral sclerosis. Inflammatory factors destroy the balance of the microenvironment, which results in changes in neural stem cell differentiation and proliferation behaviour. However, the mechanism underlying inflammatory factor-induced NSC behavioural changes is not clear. Resistin is a proinflammatory and adipogenic factor and is involved in several human pathology processes. The neural stem cell microenvironment changes when the concentration of resistin in the brain during an inflammatory response disease increases. In the present study, we explored the effect and mechanism of resistin on the proliferation and differentiation of neural stem cells. We found that intracerebroventricular injection of resistin induced a decrease in GFAP-positive cells in mice by influencing NSC differentiation. Resistin significantly decreased TEER and increased permeability in an in vitro blood-brain barrier model, which is consistent with the results of an HBMEC-astrocyte coculture system. Resistin-inhibited astrocyte differentiation is mediated through TLR4 on neural stem cells. To our knowledge, this is the first study reporting the effect of resistin on neural stem cells. Our findings shed light on resistin-involved neural stem cell degeneration mechanisms.

Objective:To study the expression level of peripheral blood mircoRNA-155 and the variation characteristics of CD4+T cell percentage and to expound the clinical significance of mircoRNA-155 quantitative test and CD 4+T cell percentage cytological test.Methods:Fluorescent quantitative PCR had been used to detect the expression level of mircoRNA -155 in the peripheral blood.Flow cytometry had been used to detect the cell percentage of the peripheral blood T lymphocyte subset CD 3+T cell,CD4+T cell and CD8+T cell.The mircoRNA-155 expression levels and the cell percentages of CD 4+T were tested respectively in the selected 40 patients with atopic dermatitis ,30 patients with skin eczema and 30 healthy controls.Finally,the differences of the above groups were counted and analyzed .Results:The relative expression levels of mircoRNA-155 of the atopic dermatitis group were higher than those of the eczema group and the normal control group ( P<0.05 ).The CD4+T cell percentage of the atopic dermatitis group was higher than those of the eczema group and the normal control group ( P<0.05 ) .Conclusion: The expression level of the peripheral blood mircoRNA-155 in the patients with atopic dermatitis is increased significantly and CD 4+T cell percentage in the patients is raised as well.Consequently ,the clinical detection of miRNA-155 and CD4+T cell shall be of great significance .

Objective:To investigate the correlation between single nucleotide polymorphism ( SNP ) of rs7517847 and rs10489629 on IL-23R gene and susceptibility of ankylosing spondylitis in Jilin.Methods:IL-23R gene polymorphisms of 188 cases on ankylosing spondylitis were detected by polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP ) , compared with those of 100 cases of healthy control group.Results:The frequency distribution differences ,between AS group and comparison group of separate genotype and the allele on two SNPs (rs7517847 and rs10489629) both showed statistical significance (P<0.05).Meanwhile, by the assumed genetic way , compared homozygous mutant GG of rs 7517847 with TG+TT, compared homozygous mutant AA of rs10489629 with GA+GG,such frequency distribution difference between the above two groups also showed statistical significance ( P<0.05).Conclusion:Polymorphisms about IL-23R gene of the rs7517847 and rs10489629 are both associated with susceptibility of AS in Jilin.The risk of AS will be increased if the person both with G/A allele and GG/AA genotype , which may be one of susceptive factors by AS.

<p><b>OBJECTIVE</b>To establish a convenient and accurate method of DNA molecular marker for the identification of corium stomachium galli.</p><p><b>METHOD</b>Cytb mtDNA sequences of Gallus gallus domestica and three other species of poultry were downloaded from Genbank. Species-specific PCR primers were designed according to the differential DNA fragments of the cytb genes. PCR tests were performed with the DNAs extracted from G. gallus domestica, three other poultry species and domestic mammal animals.</p><p><b>RESULT</b>The specific primers of G. gallus domestica could only amplify the cytb mtDNA of G. gallus domestica.</p><p><b>CONCLUSION</b>The primers are specific to G. gallus domestica mtDNA and can used to discern Corium stomachium from the false medicine.</p>

RESUMO

Objective To analyze the trend of cervical cancer, age structure and influencing factors. Methods Retrospective study and survey. The 1563 patients with cervical cancer and gynecological medical examination of 600 non-cervical cancer patients who took part in Physical examination in the obstetrics and gynecology department received the enclosed analysis questionnaire. Results Cervical cancer is becoming a trend of cervical cancer patients being younger, high risk period at the age of 36 to 55, and the downward trend towards the menopausal women. The number of being pregnant, ma]-sexuality (the first sexual intercourse at a younger age, or many sexual partners), and smoking are the causes which lead to cervical cancer. Conclusion Give great importance to a trend of cervical cancer patients being younger; screening, publicity and education about cervical cancer;, missionaries and strengthen anti-cancer awareness, promote a healthy lifestyle, which is the key to prevention and treatment of cervical cancer.

RESUMO

Objective To establish a method for quantitative determination of triptolide and triptonide in human plasma samples by HPLC.Methods The samples were extracted with Oasis HLB solid phase extraction cartridge and analyzed with reserved phase HPLC coupled with diode array detection.Results Recoveries rates obtained from spiked plasma for the two diterpenoid were higher than 80%.The linearity ranged from 10 ng/ml to 1 000 ng/ml.Detection limits of the method were 3.0ng/ml for triptolide and 4.5ng/ml for triptonide.Conclusion The method is fast and accurate,which is applicable to determination of diterpenoid in cases of suspected poisoning of Triptergium wilfordii Hook.f. in forensic practice.
