RESUMO

Triploids are rare in nature because of difficulties in meiotic and gametogenic processes, especially in vertebrates. The Carassius complex of cyprinid teleosts contains sexual tetraploid crucian carp/goldfish (C. auratus) and unisexual hexaploid gibel carp/Prussian carp (C. gibelio) lineages, providing a valuable model for studying the evolution and maintenance mechanism of unisexual polyploids in vertebrates. Here we sequence the genomes of the two species and assemble their haplotypes, which contain two subgenomes (A and B), to the chromosome level. Sequencing coverage analysis reveals that C. gibelio is an amphitriploid (AAABBB) with two triploid sets of chromosomes; each set is derived from a different ancestor. Resequencing data from different strains of C. gibelio show that unisexual reproduction has been maintained for over 0.82 million years. Comparative genomics show intensive expansion and alterations of meiotic cell cycle-related genes and an oocyte-specific histone variant. Cytological assays indicate that C. gibelio produces unreduced oocytes by an alternative ameiotic pathway; however, sporadic homologous recombination and a high rate of gene conversion also exist in C. gibelio. These genomic changes might have facilitated purging deleterious mutations and maintaining genome stability in this unisexual amphitriploid fish. Overall, the current results provide novel insights into the evolutionary mechanisms of the reproductive success in unisexual polyploid vertebrates.

ETHNOPHARMACOLOGICAL RELEVANCE: Toxicodendron vernicifluum (Stokes) F.A. Barkley (syn. Rhus verniciflua or vernicifera Stokes, Anacardiaceae) (RVS), the lacquer tree, also known as sumac, has been used in China, Japan and South Korea for thousands of years as a highly durable coating material and a traditional herbal medicine, which contains medicinal ingredients with anti-tumor, anti-inflammatory, antiviral, and anti-rheumatic activities. AIM OF THIS REVIEW: This review intends to provide a comprehensive and critical appraisal of RVS, including its phytochemical data, botanical and pharmacological literature that support its therapeutic potential in treatment on human diseases, with emphasis on the isolation of natural occurring compounds and detailed pharmacological investigations. MATERIALS AND METHODS: Specific information of RVS was collected by using the key words "Toxicodendron vernicifluum", "Rhus verniciflua Stokes", "Rhus vernicifera Stokes" and "Lacquer tree" through published scientific materials (including PubMed, ScienceDirect, Wiley, ACS, CNKI, Scifinder, Springer, Web of Science, Google Scholar, and Baidu Scholar) and other literature sources. RESULTS: The major phytoconstituents, 175 of which are presented in this review, including flavonoids, urushiols, terpenes, phenolic acids and other types of compounds, of which flavonoids and urushiols are main components. The extracts and isolates purified from RVS showed a wide range of in vitro and in vivo pharmacological effects, such as anti-cancer, anti-oxidation, anti-inflammatory, antimicrobial, tyrosinase inhibition and so on. CONCLUSION: The modern pharmacological research of RVS mainly focus on the pharmacological effects of crude extract or active constituents, of which the flavonoids are widely studied. However, there are few reports on the relationship between pharmacological effects and their structures. And at present, there is still a lack of researches that are of both effective and in-depth. Meanwhile, there is little research on quality control. Apart from the wood and lacquer, other botanical parts also need to be explored further. In addition to phenolic compounds, the study on other types of components in T. vernicifluum would start more sparks for the discovery of new bioactive principles.

To analyze the efficacy and safety of Shensong Yangxin Capsules in treatment of bradycardia combined with premature beat. Databases, such as CNKI, VIP, WanFang, SinoMed, PubMed, Cochrane Library, ClinicalTrials were retrieved by computers for relevant randomized controlled trials of Shensong Yangxin Capsules in treatment of bradycardia combined with premature beat. Two researchers independently screened out the literatures, extracted data according to the inclusion criteria, and applied the Risk of Bias assessment tool in assessing the methodological quality. The Cochrane systematic evaluation software RevMan 5.3 was used for data analysis. Totally 9 randomized controlled trials including 706 subjects were included. The intervention measure was the single administration with Shensong Yangxin Capsules, and the control measure was the blank control. The results showed that Shensong Yangxin Capsules had an obvious effect on average heart rate(MD=6.59, 95%CI[3.87, 9.31], I~2=90%), premature beat efficacy(RR=1.72, 95%CI[1.53, 1.93], I~2=0%), heart rate efficacy(RR=1.74, 95%CI[1.40, 2.17], I~2=47%), and objective efficacy(RR=1.50, 95%CI[1.31, 1.70], I~2=31%). Eight studies reported safety events, with no significant adverse reaction. In conclusion, the single administration with Shensong Yangxin Capsules may have a certain effect in improving heart rate, controlling premature beats and alleviating clinical symptoms in patients with bradycardia combined with premature beat, with no obvious adverse reaction. Shensong Yangxin Capsules can be used in clinic. This potential conclusion needs to be confirmed in future trials using rigorous methodology.

To systemically analyze the efficacy and safety of Babaodan Capsules in treatment of viral hepatitis. Databases such as CNKI,Wan Fang Date,VIP,Sino Med,PubMed,and Cochrane Library were electronically searched for relevant randomized controlled trials about Babaodan Capsules in the treatment of viral hepatitis,from database establishment to November 11,2018. Two researchers independently screened the literature and extracted data according to the inclusion criteria. GRADE system was used to evaluate evidence quality,and we used the Cochrane Rev Man 5. 3 software for Meta-analysis. Six randomized controlled trials including 520 subjects were included. Babaodan Capsules combined with conventional treatment were used as intervention measures,and the conventional treatment was used as the control measures. The results showed Babaodan Capsules combined with conventional treatment had better efficacy on reducing the total bilirubin( MD =-16. 25,95% CI[-19. 86,-12. 63]),alanine aminotransferase( MD =-26. 62,95% CI[-41. 18,-12. 06]),total bile acid( MD=-46. 02,95%CI[-49. 18,-42. 85]) and improving clinical efficiency( RR = 1. 34,95%CI[1. 13,1. 59]) than conventional treatment alone. In addition,Babaodan Capsules combined with conventional treatment can delay the progression of liver fibrosis to some extent. Qualitative analysis showed that the combined treatment regimen was more effective in relieving clinical symptoms. There was no significant difference between the two regimens in increasing albumin and prothrombin activity. Babaodan Capsules combined with conventional treatment showed no adverse reactions. In summary,for patients with viral hepatitis,the combination of Babaodan Capsules and conventional treatment has more advantages in reducing total bilirubin,alanine aminotransferase and total bile acid and is more effective in improving clinical symptoms as compared with conventional Western medicine,with no serious adverse reactions. Its clinical application with syndrome differentiation method can be considered. However,due to the limited number and quality of the original researches,more multi-center,high-quality randomized controlled trials are needed for further verification.

In order to test the hypothesis that the expression levels of GH/IGF axis genes would be negatively related to the expression of the myostatin genes in fish species, we cloned six growth regulating genes including growth hormone receptors-1/-2 (GHRs), insulin-like growth factors-I/-II (IGFs) and myostatins-a/-b (MSTNs) from blunt snout bream (Megalobrama amblycephala). The contents of mRNA transcripts for the six genes were determined in the different tissues of adult and developmental stages for the embryonic and larval periods. The results of quantitative real-time PCR showed that GHRs, IGFs and MSTNs were widely expressed in the tissues we tested, with the relatively lower expression levels in mesonephros, gonad and spleen for the six genes. The analysis of expression correlation coefficients among these six genes showed that GHR 1, GHR 2 and MSTN b were correlated with each other in adult tissues (P<0.01). For the developmental stages, GHR 1 had a similar expression pattern to GHR 2 during the examined periods, both with the highest expression levels at 160 hpf (hours post-fertilization) (P<0.05). IGF-II had higher expression levels than that of IGF-I before 400 hpf (P<0.05), while IGF-I was active after 52 hpf. The maximum of MSTN a and MSTN b mRNA levels were at 24 hpf and 400 hpf, respectively. The analysis of expression correlation coefficients showed that GHR 1, GHR 2, IGF-I, IGF-II and MSTN b were positively correlated with each other during embryonic development (P<0.01). The results from this study suggested that the relationship between GH/IGF axis genes and MSTNs was complex and not absolutely negative correlated in fish species.

A new "dual-mode" chromogenic and fluorescent turn-on probe (2) for the selective sensing of biological thiols is reported. In MeOH-H(2)O cosolvent at physiological pH 7.40 (MeOH-H(2)O = 3:7), biological thiols cleave the 2,4-dinitrobenzenesulfonyl group to release the chromo- and fluorophore merocyanine (3).

RESUMO

A irreversible Hg2+ selective ratiometric fluorescence probe FR, a fluorescein fluorophore linked to a rhodamine B hydrazide by a thiourea spacer, was designed and synthesized. The developed probe FR exhibited great ratiometric fluorescence enhancement and remarkable yellow-magenta color change toward Hg2+ with excellent selectivity in aqueous acetone solution, and the ratiometric fluorescence response to Hg2+ was not interfered by other metal cations including Fe3+, Co2+, Ni2+, Cr3+, Zn2+, Pb2+, Cd2+, Ca2+, Mg2+, Ba2+ and Mn2+. The linear range and the detection limit of this supposed ratiometric fluorescence method for Hg2+ were 0.0-10.0x10(-6) and 5x10(-8) M, respectively.

A new highly sensitive and selective colorimetric method for fluoride determination in water is described. The novel reagent used is a hemicyanine dye (HC), which forms a stable complex with Zr-EDTA, the fluoride-binding site, through the hydroxyl groups. The hemicyanine-chelating Zr-EDTA complex (HC-Zr-EDTA) is ready to react with fluoride ion to release HC. This result in remarkable color change of the sensing solutions from red (lambda(max)=513nm) to yellow (lambda(max)=427nm) at pH 4.40. When applied to the colorimetric determination of fluoride ions, a linear range from 3.0x10(-6) to 5.0x10(-5)mol/L and a detection limit of 2.8x10(-6)mol/L with a correlation coefficient of 0.9993 can be achieved under the optimized conditions. Because of the specific affinity of fluorides for the [Zr-EDTA], there is little interference by other ions. This method has been successfully applied to the determination of fluorides in toothpaste samples.

Seventeen microsatellite markers of Aristichthys nobilis previously discovered by our lab were selected to analyze the genetic diversity and characteristics of two populations of Aristichthys nobilis from Jiangxi and Sichuan provinces. The following parameters were calculated: heterozygosity, polymorphism information content (PIC), valid allele number, allele frequency, genetic distance, genetic similarity coefficient, Hardy-Weinberg balance deflection index and so on. Results show that there are 4 monomorphic and 13 polymorphic markers among the 17 selected microsatellite markers. The average of allele number in each microsatellite locus of the Jiangxi population and Sichuan populations is 3.325 and 3.882, respectively; the average valid allele number is 3.531 and 2.676, respectively; and the number of total alleles of these 17 microsatellite loci is 71. The PIC of polymorphic loci varies between 0.077-0.960, and the average PIC is 0.417. The average observed heterozygosity (Ho) of two populations is 0.385 and 0.360, respectively and the average expected heterzygosity (He) is 0.452 and 0.422, respectively. The genetic similarity coefficient of two populations of Aristichthys nobilis is 0.897 and the genetic distance of these populations is 0.109.

RESUMO

A new chemosensor based on rhodamine B thiohydrazide is described. Chemosensor B was found to show a reversible dual chromo- and fluorogenic response toward Hg2+ in aqueous solution in a highly selective and sensitive manner. This was suggested to result from the coordination of Hg2+ at the N, S binding sites in B to open its spiro ring.

A fluorescence immunoassay for human IgG (Ag) was developed using a pH-sensitive polymer prepared by thermal initiation or redox initiation polymerization as a carrier. In the competitive immunoassay, appropriate quantity of Ag was immobilized on the polymer and the standard Ag (or sample) solution, and a constant amount of fluorescein isothiocyanate labeled goat anti-human IgG antibody (Ab-FITC) was added. Immobilized Ag and the standard (or sample) Ag competed for binding to the Ab-FITC in 37 degrees C in homogeneous format. After changing the pH to separate the polymer-immune complex precipitate, it was re-dissolved and determined by fluorescence method. The results showed that the immobilization efficiency, immunological reaction activities of immobilized Ag and phase transition pH range were improved as Ag was immobilized by thermal initiation instead of redox initiation polymerization. Under optimum conditions, the calibration graphs for the Ag in both methods, thermal initiation and redox initiation, were linear over the concentration range of 0.0-1000ngmL(-1), with detection limits 8 (thermal initiation) and 12ngmL(-1) (redox initiation), respectively. Moreover, some pH-sensitive polymer prepared only in organic solvent or under high temperature could also be used as an immunoreaction carrier by thermal initiation polymerization. Thermal initiation polymerization was a better immobilization mode.

The preparation and utilization of a novel composite silica-hemin nanoparticles (CSHNs) as mimetic peroxidase are reported in this article. Experimental results showed that the composite nanoparticles had unique advantages over free hemin molecules in good stability and highly catalytic activity. By employing these nanoparticles as biocatalyst, we developed a new spectrofluorometric method for the determination of trace level glucose. The calibration graph for glucose was linear over the range 4.0 x 10(-7) mol/L - 7.0 x 10(-5) mol/L, with a detection limit of 1.0 x 10(-7) mol/L. The proposed method has been successfully applied to the determination of glucose in serum samples and offers the advantages of being rapid, stable, sensitive, and renewable.

A hybrid cluster complex, formed by chelating a chromogenic ligand to a [2Fe-2S] cluster, sensitively exhibited differential colorimetric responses towards Hg2+, Cd2+, Cr3+, Pb2, Sn2+, Cu2+, Zn2+, Fe3+ and Co2+ in water at physiological pH. Speciation of some of these metal elements, such as Cr(III) and Sn(IV), was also studied by UV/Vis absorption.
