RESUMO

OBJECTIVETo observe the efficacy of Guchong Zhixue Decoction (GCZXD) in treating dysfunctional uterine bleeding of anovulatory type with Qi-deficiency and blood stasis syndrome, and to systematically explore its hemostasis mechanism.

METHODSSixty patients were randomly divided into the treated group (n = 30, treated by GCZXD taken orally) and the control group (n=30, treated by Yigong Zhixue Oral Liquid, taken orally). Hemostatic effect of treatment, improvements of clinical symptoms, thickness of endometrium, prothrombin time (PT) and activated partial prothrombin time (APTT), serum level of Ca2+, endocrine hormone, levels of PGE2 and PGF2alpha in menstrual blood serum, and contents of endometrial estrogen receptor (ER) and progestin receptor (PR) before and after treatment were observed. Results The total effective rate of GCZXD in hemostasis was 93.3% in the treated group, which was significantly better than that in the control group (70.0%, P < 0.05). Significant difference was shown in improving clinical symptoms, thickness of endometrium, PT and APTT, serum level of Ca2+, endocrine hormone, levels of PGE2 and PGF2alpha in menstrual blood serum, and contents of endometrial ER and PR in the treated group when compared with before treatment (P < 0.05, P < 0.01) also when compared with the control group after treatment (P < 0.05).

CONCLUSIONGCZXD has favorable hemostatic effect in treating dysfunctional uterine bleeding of Qi deficiency and blood stasis syndrome, and it also con improve the accompanied symptoms significantly.

RESUMO

OBJECTIVESTo study the changes of cellular immunity function in patients with hepatocellular carcinoma (HCC) and its correlation with disease severity.

METHODST lymphocyte subsets and CD28 co-stimulation molecule in CD8+ T cells in 22 HCC patients were detected using three-color flow cytometry. Serum interleukin-2 (IL-2), transforming growth factor beta 1 (TGFbeta1), and interleukin-6 (IL-6) were determined by ELISA and radioimmunoassay. A group of 30 patients with chronic hepatitis B (CHB), liver cirrhosis (LC), or normal adults (NC) served as controls.

RESULTSCompared with NC, the number of CD8+CD28- T cells increased and CD8+CD28+ T cells decreased in patients with HCC. The number of CD4+ T cells, CD4+/CD8+ ratios, IL-2 level all decreased and CD8+ T cells, IL-6, TGFbeta1 levels all increased in patients with HCC, LC and CHB. The CD4+ T cells, CD4+/CD8+ ratios and IL-2 level in patients with HCC were lower than those with CHB. Serum IL-6 and TGFbeta1 in patients with HCC were higher than those with LC and CHB. The levels of IL-6 and TGFbeta1 correlated with the stages of the tumors.

CONCLUSIONSHCC patients have a cellular immunity dysfunction. Rectifying the imbalanced function could be a potential way for treating HCC. Measurement of these factors would be useful for early diagnosis and evaluating the prognoses of these patients.

RESUMO

OBJECTIVETo detect the hepatic tissue and serum level of TGFbeta1 in patients with viral hepatitis, in order to clarify their relationship of the starting, developing of hepatic fibrosis.

METHODSThis study included 92 patients with viral hepatitis. Liver puncture was performed in 31 patients. Hepatic collagen staining (Masson's three colors) and TGFbeta1 immunohistochemistry staining of the liver tissue specimens were performed, morphometric quantitative measurements of hepatic histological collagen and TGFbeta1 were made. The serum level of TGFFbeta1 was detected by ELISA.

RESULTSThe surface density of hepatic TGFbeta1 increased linearly with the elevation of fibrosis stage (P < 0.05), there were no significant differences between every two groups of G1, G2, G3 and G4 (P > 0.005). There was a closely positive correlation between the levels of TGFbeta1 in hepatic tissue and serum, the coefficient was 0.896 (P < 0.01). The levels of TGFbeta1 in tissue and serum both had positive correlation with hepatic collagen, coefficients were 0.863 and 0.667 (P < 0.001). The level of TGFbeta1 in tissue and serum both had positive correlation with serum levels of PCIII, HA, LN, CIV (P < 0.001).

CONCLUSIONSThere was a closely relationship between the levels of TGFbeta1 in hepatic tissue and serum and liver fibrosis. The detection of TGFbeta1 in liver and serum are more sensitive than HA, LN, CIV in early period of hepatic fibrosis.
