Understanding how natural aging impacts rodent performance in translational behavior tests is critical to teasing apart impairments due to age-related decline from neurodegenerative disorder modeling. Reduced neuropilin and tolloid-like 1 (NETO1), an accessory protein of ionotropic glutamate receptors involved in synaptic plasticity, was associated with Alzheimer's disease, yet aging effects on Neto1 remain unclear. For these reasons, our goal was to characterize how Neto1 expression corresponded with social, repetitive, and spatial learning behaviors and stress response across the C57BL/6J mouse lifespan. We measured social preferences in three-chamber tests, and motor stereotypies by marble burying. Cognitive flexibility is typically assessed in the Morris water maze (MWM), wherein C57BL/6J mice exhibit deficits with age. However, fatigue or locomotor impairment may confound interpretation of MWM performance. Therefore, we used a less arduous water T-maze (WTM) to compare spatial learning flexibility in 2, 9-15, and 24-month-old male and female mice to test the hypothesis that deficits would emerge with age. In both sexes, 9-15-month-olds made more chamber entries during social preference tests, while 2-month-olds did less social sniffing than aged mice. No age or sex differences emerged in marble burying or serum corticosterone measurements. In 24-month-olds hippocampal Neto1was increased relative to 2-month-olds, and male cognitive flexibility was strong, while spatial learning and reversal learning of 24-month-old females was impaired in WTM irrespective of Neto1 expression. The WTM is a useful alternative assessment for cognitive flexibility deficits in aged mice, and the role of hippocampal Neto1 in promoting social sniffing is of interest.

Gelatin, as a common hemostatic agent, has been processed into a variety of forms for clinical applications. To enhance wound healing and reduce postoperative complications after liver trauma or surgery, naturally-derived materials can be incorporated into gelatin to improve its physical and biological properties. In this study, we developed an extracellular matrix (ECM) enriched gelatin (EG) sponge through combining liver ECM digestion and gelatin at different proportions. By increasing the gelatin concentration, the EG sponge exhibited reduced porous structure, lower water absorption rate, superior degradation resistant, and higher elastic modulus, whereas, by increasing the ECM concentration, the porous structure and swelling ratio of the EG sponge were significantly improved. We tested the in vivo response of EG sponge for liver parenchyma wound repair as compared with the ECM-only or gelatin-only sponges. Liver wound repaired with the gelatin-only sponge exhibited a severe inflammation and tissue adhesion. In contrast, both ECM-only and EG sponges repaired liver wound showed desired biocompatibility as evidenced by a smooth liver surface, reduced wound size, earlier material absorption, and accelerated liver regeneration. In conclusion, the properly designed EG sponge is a more effective and safer topical hemostatic agent than the traditional gelatin sponge for repairing liver injury.

RESUMO

A multimodal nursing pedagogy utilizing simulation was incorporated into annual nurse competencies at a 465-bed academic medical center to translate into practice the required surveillance of a potentially septic patient and the necessary clinical decision-making. Pretest/posttest was used to determine knowledge retention from simulation. Nurses' sepsis knowledge following simulation was improved. Simulation can provide long-term knowledge retention of sepsis for application into clinical practice and be a viable option for competency assessment for professional nurses.

RESUMO

Limited research has been conducted on how nurses define or perceive "quality nursing care." We conducted focus groups to identify nurses' perceptions of quality care at a Midwestern academic medical center. Transcripts of the focus group sessions were analyzed using thematic analysis techniques, and 11 themes emerged: Leadership, Staffing, Resources, Timeliness, Effective Communication/Collaboration, Professionalism, Relationship-Based Care, Environment/Culture, Simplicity, Outcomes, and Patient Experience.

Preventing hospital-acquired infections, including ventilator-associated pneumonia (VAP), has become a priority for Wyoming Medical Center. Despite the implementation of a standardized care bundle protocol to reduce the facility's high rate of VAP, rates of VAP remained higher than the national benchmark. A recent breakthrough in care happened when the center's intensive care unit created a multidisciplinary VAP committee with the goal of reducing the incidence of potentially deadly VAP. The VAP committee implemented a VAP bundle protocol and adopted the use of a polyurethane endotracheal tube on the basis of literature review. These interventions, in addition to comprehensive education and the synergism between physicians, nurses, and respiratory therapists, have not only reduced the center's VAP rate but also prevented the occurrences of VAP during 33 consecutive months.
