RESUMO

The high rate of deaths around the world from noncommunicable diseases (NCDs) (70%) is a consequence of a poor diet lacking in nutrients and is linked to lifestyle and environmental conditions that together trigger predisposing factors. NCDs have increased 9.8% of public health spending worldwide, which has been increasing since 2000. Hence, international organizations such as the WHO, the Pan American Health Organization, and the Food and Agriculture Organization of the United Nations have been developing strategic plans to implement government and economic policies to strengthen programs in favor of food security and nutrition. A systematic review is presented to document an analysis of the origin and characteristics of obesity, cardiovascular disease, chronic respiratory diseases, diabetes, and cancers affecting a large part of the world's population. This review proposes a scientifically based report of functional foods including fruits, vegetables, grains, and plants, and how their bioactive compounds called nutraceuticals-when consumed as part of a diet-benefit in the prevention and treatment of NCDs from an early age. Multifactorial aspects of NCDs, such as culture and eating habits, are limitations to consider from the clinical, nutritional, and biochemical points of view of everyone who suffers from them.

Cancer induced by a viral infection is among the leading causes of cancer. Hepatitis C Virus (HCV) is a hepatotropic oncogenic positive-sense RNA virus that leads to chronic infection, exposing the liver to a continuous process of damage and regeneration and promoting hepatocarcinogenesis. The virus promotes the development of carcinogenesis through indirect and direct molecular mechanisms such as chronic inflammation, oxidative stress, steatosis, genetic alterations, epithelial-mesenchymal transition, proliferation, and apoptosis, among others. Recently, direct-acting antivirals (DAAs) showed sustained virologic response in 95% of cases. Nevertheless, patients treated with DAAs have reported an unexpected increase in the early incidence of Hepatocellular carcinoma (HCC). Studies suggest that HCV induces epigenetic regulation through non-coding RNAs, DNA methylation, and chromatin remodeling, which modify gene expressions and induce genomic instability related to HCC development that persists with the infection's clearance. The need for a better understanding of the molecular mechanisms associated with the development of carcinogenesis is evident. The aim of this review was to unravel the molecular pathways involved in the development of carcinogenesis before, during, and after the viral infection's resolution, and how these pathways were regulated by the virus, to find control points that can be used as potential therapeutic targets.

Flavivirus detection in humans and mosquito reservoirs has been an important issue since it can cause a variety of illnesses and could represent a health problem in geographical zones where the vector is endemic. In this work, we designed and characterized a biosensor based on gold nanoparticles (AuNPs) and antibody 4G2 for the detection of dengue virus (DENV) in vitro, obtaining different conjugates (with different antibody concentrations). The AuNP-4G2 conjugates at concentrations of 1, 3, and 6 µg/mL presented an increase in the average hydrodynamic diameter compared to the naked AuNPs. Also, as part of the characterization, differences in the UV-Vis absorbance spectrum and electrophoretic migration were observed between the conjugated AuNPs (with BSA or antibody) and naked AuNPs. Additionally, we used this biosensor (AuNP-4G2 conjugate with 3 µg/mL antibody) in the assembly of a competitive lateral flow assay (LFA) for the development of an alternative test to detect the flavivirus envelope protein in isolated DENV samples as a future tool for dengue detection (and other flaviviruses) in the mosquito vector (Aedesaegypti) for the identification of epidemic risk regions. Functionality tests were performed using Dengue virus 2 isolated solution (TCID50/mL = 4.58 × 103) as a positive sample and PBS buffer as a negative control. The results showed that it is possible to detect Dengue virus in vitro with this gold nanoparticle-based lateral flow assay with an estimated detection limit of 5.12 × 102 PFU. We suggest that this biosensor could be used as an additional detection tool by coupling it to different point-of-care tests (POCT) for the easy detection of other flaviviruses.

An HPLC-UV/FLD method for simultaneous detection of ten antibiotics in surface waters was developed. Antibiotics were extracted from water using solid phase extraction. An Atlantis T3 column was used with acetonitrile and 0.05% trifluoroacetic acid as a mobile phase for separation, with a total running time of 45 min. Signal detection was performed at 280 nm; fluoroquinolones were additionally quantified by fluorescence detection. Validation parameters such as linearity, recovery and precision were evaluated. The limits of detection (LOD) in river waters were in the range 0.1-1.3 µg/L for antibiotics detected by UV, and 0.039 and 0.073 µg/Lfor fluoroquinolones detected by FLD. LOD are sufficiently low to consider this method as a first alternative for HPLC-MS methods that will allow alerting for the presence of antibiotics in surface waters. This screening method is rapid, sensitive, reproducible and economical.

Osteoporosis is a chronic disease in adult women caused by menopause and some other factors, which entails deficiency of calcium in diet. Natural products are the best source of nutriments to reduce the risk of chronic diseases. Nopal (Opuntia ficus-indica) is a plant characterized by its nutritional components and benefits to health. Its calcium content increases with maturation process that could be beneficial for consumers. Nopal powder (NP) was elaborated from nopal harvested within 16-24 weeks of maturation, and the nutritional content was determined. An experimental clinical trial was performed to evaluate the effect of NP. A total of 69 women between 40 and 60 years old participated in the study. During 24 weeks, experimental group (n = 56) consumed a daily dose of 5 g of NP and control group (n = 13) continue with habitual diet. Changes in bone mineral density (BMD), body mass index (BMI), body fat percentage and serum calcium were assessed. Between baseline and after 24 weeks of consumption, no significant changes were found in BMD P = .885 experimental group and P = .970 control group, BMI P = .865 experimental group and P = .984 control group, body fat P = .744 experimental group and P = .740 control group and serum calcium P = .282 experimental group and P = .959 control group. These results indicate that advanced maturation NP does not have influence in bone health, BMI, and body composition in adult women.

There is evidence that support the clinical usage of glycemic index (GI) and glycemic load (GL) in the prevention of chronic disease. Objective: To determine the GI and GL of the Opuntia ficus-indica fruit. Methodology: An analytic, transversal study was made involving 25 healthy volunteers accepted by an informed consent with a normal body mass index, glucose, glycoside hemoglobin, cholesterol and serum triglycerides. The homogeneity of the population was evaluated with anthropometrical and biochemical data using principal component analysis (PCA). The equivalent of 50 g of carbohydrates test food (tuna) and 50 g of dextrose as food standard was provided for the measure of the glucose curve. The GI was determined by calculating the area under the curve by the triangulation method. The CG was reported as the product of IG by carbohydrate loading provided. Results and Conclusions: The IG of the tuna was 48.01 ± 17.4, classified as low, while the CG was 24.0 ± 8.7 rated as high. The chemometric analysis by PCA showed that the selection of the normal population for determining the IG, it is important to consider the values of cholesterol and triglycerides.


Existe evidencia que apoya la utilidad clínica de que el índice glucémico (IG) y la carga glucémica (CG) podrían ser impor- tantes en la prevención de enfermedades crónicas. Objetivo: Determinar el IG y la CG del fruto de Opuntia ficus-indica (tuna). Metodología: Se realizó un estudio analítico, transversal, con 25 voluntarios sanos con valores normales de índice de masa corporal, glucosa, hemoglobina glucosilada, colesterol y triglicéridos séricos, que mediante consentimiento informado acepta- ron participar. Con los datos antropométricos y bioquímicos se evaluó la homogeneidad de la población usando análisis de componente principal (PCA). Se proporcionó el equivalente a 50 g de hidratos de carbono de alimento de prueba (tuna) y 50 g de dextrosa como alimento estándar para medir la curva de glucosa. El IG se determinó calculando el área bajo la curva por el método de triangulación. La CG se reportó como el producto del IG por la carga de hidratos de carbono propor- cionada. Resultados y conclusiones: El IG de la tuna fue de 48.01 ± 17.4, clasificado como bajo, mientras que la CG fue de 24.0 ± 8.7, clasificada como alta. El análisis quimiométrico por PCA demostró que, en la selección de la población normal para la determinación del IG, es importante considerar los valores de colesterol y triglicéridos en sangre.

La composición química, minerales, azúcares neutros, compuestos fenólicos y capacidad antioxidante fueron analizados en la cascarilla de garbanzo. La cascarilla de garbanzo presentó 72% de fibra dietaria, del cual el 24.4% fue celulosa. Los compuestos fenólicos y actividad antioxidante de de la cascarilla de garbanzo fueron evidentes, predominando los taninos condensados totales con 13.28 mgEC/g, donde la fracción soluble fue mayor respecto a la fracción ligada. El contenido de proteína y grasa fue de 4.5 y 0.4%. En conclusión, la cascarilla de garbanzo tiene propiedades nutricionales y funcionales que pueden ser consideradas en el diseño de nuevos productos alimenticios para mejorar la salud de los consumidores(AU)


The chemical composition, minerals, neutral sugars, phenolic compounds and antioxidant capacity were analyzed in the chickpea husk. Chickpea husk presented 72% of dietary fiber of which 24.4% was cellulose. The content of phenolic compounds and antioxidant capacity of chickpea husk was evident, predominating condensed tannins with a total content of 13.28 mgECat/g, of which soluble fraction was the higher than bound fraction. The content of protein and fat was 4.5 and 0.4 %, respectively. In conclusion, chickpea husk has nutritional and functional properties that can be considered in the design of new food products to improve the health of consumers(AU)

Background and aim. Acetylsalicylic acid (ASA) has been shown to downregulate HCV expression; however, the involved mechanisms are unknown. We used proteomic analysis to compare protein expression profiles between human hepatocarcinoma cells (Huh7) and Huh7-HCV cells harboring expression of non-structural HCV proteins, to elucidate the mechanism(s) involved in ASA-mediated downregulation of HCV replication. Material and methods. Both cell lines were treated or untreated with 4 mM ASA and harvested at 0, 24, 48 and 72 h to isolate total proteins, which were resolved by two-dimensional gel electrophoresis (2DE) to separate them by isoelectric point (pI), followed by fractionation by molecular weight (MW). Gels were scanned and analyzed with PD-Quest software V8.0.1, and proteins were elucidated by the specific pI and MW using TAGIDENT software. Statistics analysis included the t-test. esults and Discussion. Different protein patterns among hepatocytes expressing HCV-proteins in ASA treated and untreated cells were found. Among proteins differentially expressed in Huh7-HCV cells, we found proteins related to cell proliferation (MTMR6, FAM22, HDGF and HCF-1) after 24 h of ASA treatment; and upregulation of angiostatin, PI4KA and STAT-1 after 48 h of treatment. Finally, at 72 h of ASA exposure, we identified overexpression of adenylsuccinate synthase, 2'-3'-di-deoxyadenosine, ubiquitin-protein-ligase E6A, adenylosuccinate-lyase and nibrin (NBN). Conclusion. We found that ASA induces different protein patterns in Huh7-HCV cells promoting activation of proteins involved in cell progression, repair of double strand breaks, proliferation, inhibition of apoptosis and growth stimulation at the same time that it decreased HCV expression.

The determination of the antioxidant activity of Turnera diffusa using partial least squares regression (PLSR) on chromatographic data is presented. The chromatograms were recorded with a diode array detector and, for each sample, an enhanced fingerprint was constructed by compiling into a single data vector the chromatograms at four wavelengths (216, 238, 254 and 345 nm). The wavelengths were selected from a contour plot, in order to obtain the greater number of peaks at each of the wavelengths. A further pretreatment of the data that included baseline correction, scaling and correlation optimized warping was performed. Optimal values of the parameters used in the warping were found by means of simplex optimization. A PLSR model with four latent variables (LV) explained 52.5% of X variance and 98.4% of Y, with a root mean square error for cross validation of 6.02. To evaluate its reliability, it was applied to an external prediction set, retrieving a relative standard error for prediction of 7.8%. The study of the most important variables for the regression indicated the chromatographic peaks related to antioxidant activity at the used wavelengths.

In the present contribution, the partial least squares (PLS) method was used to establish a correlation between the antioxidant activity (obtained by DPPH assay) and chromatographic profiles of TURNERA DIFFUSA extracts. Chromatograms were obtained using HPLC-DAD. A model was constructed using 40 samples with 2550 X variables corresponding to the responses obtained at different times; the Y variables consisted of experimental values of antioxidant activity of each extract (measured as EC50). Prior to this analysis, alignment of chromatograms was performed based on consideration of seven high-intensity signals present in all samples. The PLS1 model was validated by cross-validations; its capacity was evaluated using correlation parameters R², root mean square error of calibration (RMSEC), and root mean square error of prediction (RMSEP). The best results were achieved with zero order chromatograms using five-point smoothing (R² = 0.96, RMSEC = 3.31, and RMSEP = 7.86). Under these conditions, the optimal number of components was five. The model was applied to the prediction of antioxidant activity of commercial products; no significant differences were found between the experimental and predicted antioxidant activities for 83 % of them.

Damiana is a shrub used as a dietary supplement and as an additive in several foods. This report presents the first validated analytical method for QC of products made from damiana. Through bioassay-guided fractionation, the compound showing the best antioxidant activity, determined using 2,2-diphenyl-1-picryl-hydrazyl assay in TLC, was purified and identified as 8-C-beta-[6-deoxy-2-O-(alpha-1-rhamnopyranosyl)-xylo- hexopyranos-3-uloside] (Cl50 5.56 g/mL). This is the only natural source from which this compound has been reported, and has been found in all native damiana samples analyzed. Therefore, it was used as a biomarker to develop an HPLC-diode-array detection analytical procedure using the first-order derivative of the chromatograms. Separation was achieved using an AccQ Tag C18 150 x 3.9 mm (4 microm) column, with a gradient of methanol and 0.1% trifluoroacetic acid in water. The method was shown to be accurate, sensitive, and reproducible, providing a useful quantitative means to apply QC to damiana products.
