This study examined the changes in the number of visitors to regions during periods of high particulate matter (PM) concentrations in Seoul and analyzed the regional differences of these changes. Further, it examined the spatial characteristics that affect these regional differences. This study mapped the regional differences by conducting a spatial cluster analysis using GIS and examined factors affecting the regional differences using logistic regression analysis. The visiting population data used in this study were obtained from the Big Data on the de facto population measured every hour at mobile base stations, and all analyses were conducted in terms of weekdays and weekends. The results indicated that the visiting population decreases significantly on weekdays when there are high PM concentrations; however, visits increase on weekends, even during periods of high PM concentrations. Moreover, there was a huge regional gap in visiting population changes. Regions with more commercial use, higher bus accessibility, and better pedestrian environment (pedestrian paths, Walk Score) were more likely to be hotspots, whereas regions with high residential and industrial use were more likely to be cold spots. These results can be used as the basic data for PM policies based on regional characteristics.

While incineration is among the most commonly used technologies for waste disposal, there is ongoing public concern regarding the adverse health impact. The aim of this study is thus to use health statistics to assess the relative risk of asthma-related hospitalization for those living in close proximity to incineration facilities. We also examine differences in asthma risk related to age demographics. The spatial relationship between incineration facilities and asthma-related hospital admissions in Seoul is analyzed for the period of 2009-2011 using the Rapid Inquiry Facility (RIF) and SaTScan software. The relative risk of asthma-related hospitalization decreased with increasing distance from incinerators, but increased among those living within a 2-km radius. The relative risks of asthma-related hospitalization were 1.13 (95% confidence interval (CI): 1.10-1.17), 1.12 (95% CI: 1.08-1.17), and 1.18 (95% CI: 1.10-1.27) for all ages, those aged below 15 years, and those aged 65 years and older, respectively. This study is the first to observe an increased risk of asthma-related hospitalization in relation to a person's distance from an incinerator in Seoul, Korea. It is clear that asthma should be considered an adverse health outcome during health impact assessments of incineration plants.

Hypoxia-inducible factor HIF-1 is responsible for radiation resistance and poor prognosis in cancer therapy. As part of our drug discovery program, a novel HIF inhibitor, LW6, was identified as a small compound that inhibits the accumulation of HIF-1alpha. We found that LW6 decreased HIF-1alpha protein expression without affecting HIF-1beta expression. MG132, a proteasome inhibitor, protected HIF-1alpha from LW6-induced proteasomal degradation, indicating that LW6 affects the stability of the HIF-1alpha protein. We found that LW6 promoted the degradation of wild type HIF-1alpha, but not of a DM-HIF-1alpha with modifications of P402A and P564A, at hydroxylation sites in the oxygen-dependent degradation domain (ODDD). LW6 did not affect the activity of prolyl hydroxylase (PHD), but induced the expression of von Hippel-Lindau (VHL), which interacts with prolyl-hydroxylated HIF-1alpha for proteasomal degradation. In the presence of LW6, knockdown of VHL did not abolish HIF-1alpha protein accumulation, indicating that LW6 degraded HIF-1alpha via regulation of VHL expression. In mice carrying xenografts of human colon cancer HCT116 cells, LW6 demonstrated strong anti-tumor efficacy in vivo and caused a decrease in HIF-1alpha expression in frozen-tissue immunohistochemical staining. These data suggest that LW6 may be valuable in the development of a HIF-1alpha inhibitor for cancer treatment.

Previously, we reported that overexpression of Opa (Neisseria gonorrhoeae opacity-associated)-interacting protein 5 (OIP5) caused multi-septa formation and growth defects, both of which are considered cancer-related phenotypes. To evaluate OIP5 as a possible cancer therapeutic target, we examined its expression level in 66 colorectal cancer patients. OIP5 was upregulated about 3.7-fold in tumors and over 2-fold in 58 out of 66 colorectal cancer patients. Knockdown of OIP5 expression by small interfering RNA specific to OIP5 (siOIP5) resulted in growth inhibition of colorectal and gastric cancer cell lines. Growth inhibition of SNU638 by siOIP5 caused an increase in sub-G1 DNA content, as measured by flow cytometry, as well as an apoptotic gene expression profile. These results indicate that knockdown of OIP5 may induce apoptosis in cancer cells. Therefore, we suggest that OIP5 might be a potential cancer therapeutic target, although the mechanisms of OIP5-induced carcinogenesis should be elucidated.
