RESUMO

The effect of mannan polysaccharide on the haemopoiesis recovery in irradiated mice has been investigated. Mannan has been shown to exert both the protective and the stimulatory effect: it accelerates restoration of femur bone marrow cellularity and nucleate cell number in the peripheral blood and causes a larger initial yield and subsequent more rapid postirradiation dynamics of pluripotent haemopoietic stem cells and precursor cells of granulocytes and macrophages.

RESUMO

Anabol is a biopolymer from the complex of components extracted from the surface layer of the Lactobacillus bulgaricus cell wall. Its effect on certain indices of the functional activity of mononuclear phagocytes was studied. Administration of the preparation in a dose of 20 mg/kg 24 hours before the investigation resulted in lowering of the stain half-life in the general blood flow and increasing of the fibronectin levels in blood plasma of rats. Activation of the metabolic activity estimated by nitroblue tetrazolium test as well as resident cells and induced macrophages was observed. 24 hours after the anabol administration there was noted a marked increase in the pool of the precursors of granulocytes and macrophages, the stimulating effect being preserved for up to 3 days. The results showed that anabol had an effect on various elements of the system of mononuclear phagocytes. It may be useful in complex therapy aimed at increasing the host resistance to diverse toxic substances and bacterial infection.

In this investigation the influence of anabol, a biopolymer preparation incorporated into a complex of extracted components obtained from the surface layer of Lactobacillus bulgaricus cell wall, on some functional activity characteristics of mononuclear phagocytes has been studied. The injection of the preparation in a dose of 20 mg/kg of body weight 24 hours prior to the experiment leads to a decrease in the time necessary for the elimination of one-half of the total amount of ink from the blood stream and to an increase in the content of fibronectin in the blood plasma of rats. The activation of the metabolic activity of resident and induced macrophages, evaluated by the nitro blue tetrazolium reduction test, is observed. On the next day after the injection of anabol the sharp increase of the pool of granulocyte and microphage precursors is observed; the stimulating effect may be retained as long as 3 days. The results thus obtained indicate that anabol acts on different elements of the system of mononuclear phagocytes and can be effective in the complex therapy aimed at the increase of the body resistance to any toxic substances and bacterial infection.

RESUMO

The recovery of a population of granulopoietic-monocytic colony-forming units of mouse bone marrow was only slightly inhibited during the first two weeks after exposure to a mixture of radiation (4 Gy) and heat (burn of 10 per cent of the skin). In the interval between days 14 and 28 following the effect, their number reached the control level, then it decreased again.

RESUMO

CFU-DC in the bone marrow of CBA and BALB/c mice, which are contrast in total radiosensitivity, have close characteristics: D0 is 1.35 and 1.32 Gy, respectively. The proliferation rate of CFU-DC after single exposure to a non-lethal dose of 4 Gy is higher in CBA than in BALB/c mice. The time of doubling the CFU-DC population during the period of exponential growth after irradiation is 40 and 72 h for CBA and BALB/c mice, respectively.

RESUMO

The ability of CFUdc to repair radiation-induced lesions in the irradiated body was studied by the methods of dose fractionation and dose-rate reduction. With the dose-rate decreased from 1-0.5 Gy/min to 0.02 Gy/min, a mean lethal dose per cell increased from 1.35 up to 1.93 Gy. With fractionation of the dose, the known picture of repair of sublethal radiation lesions was obtained the second survival peak being insignificant. The authors discuss the possible causes of the distinctions in the repair parameters of CFUdc obtained by the two methods.

RESUMO

After exposure in vitro and in situ CFUdc of CBA mouse bone marrow and spleen were characterized by the comparable parameters of radiosensitivity and oxygen-dependent modification: the values of D0 for bone marrow and spleen cells were 1.31 and 1.35 Gy (in vitro) and 1.36 and 1.37 Gy (in situ), and the values of the oxygen effect were 2.3 and 2.5, respectively.

RESUMO

Pretreatment of mouse bone marrow cells with rabbit antiserum to mouse brain (RAMB) decreases the capacity of these cells to form granulocyte-macrophage colonies in agar culture in vivo (CFU-DC) and does not affect the formation of colonies in agar culture in vitro (CFU-C). Mouse bone marrow CFU-DC are analogous to the pluripotent hemopoietic stem cells (CFU-C) forming spleen colonies in lethally irradiated mice as regards the ability for being inactivated with RAMB but differ from CFU-C. CFU-DC were found in the mouse thymocyte population.
