RESUMO

Owing to a complex application of topical analysis and tracer technique, it is possible to carry out a light optic and electron microscopic investigation of newly formed capillaries growing in the rabbit cornea after its chemical burn. The ultrastructural analysis demonstrates certain polymorphism of morphological organization of endotheliocyte in the newly formed capillaries. There is a rather elevated amount of free ribosomes, mitochondria, microtubules and microfilaments in cytoplasm. The granular endoplasmic reticulum and Golgi complex are hypertrophied. Weibel--Palade bodies appear. Taking into account certain morpho-functional peculiarities of endothelial cells along the course of the growing capillaries, on the 8th day of growth three zone are distinguished: 1--area of nondifferentiated endothelium (apex of the capillary), 2--transitional zone, 3--zone of relatively differentiated endothelium situating in the place where the capillary gets off the parental vessel. According to the zones distinguished, the ways of trans-endothelial transport of molecules are investigated. In formation of the capillary barrier-transport function an important role belongs to polymorphism of the endothelial cells along the course of the growing capillary which is determined by differentiation degree of these cells depending on their participation in permeability.

RESUMO

Methodical aspects of scanning electron microscopy of corrosion preparations (SEMCP) are described on the basis of the authors' own data and those from the literature. A critical analysis of the currently used injection masses and the results obtained by SEMCP is given. New data are emphasized, introduced into pathological morphology after the adoption of SEMCP. Relief formations of casts of the blood and lymph vessels in health and disease are classified. The prospects of and main approaches to the use of SEMCP in pathological morphology are discussed.

RESUMO

The physiological saline was injected into the excretory duct of the cat parotid gland under the pressure of 30, 70 and 120 cm H2O. It was found with the aid of transmission electronic microscopy, morphometry and statistical analysis that the liquid injection into the gland produces compression of the blood capillaries weaving the acinus. The compression of the capillary tubes is shown by a significant reduction in space given to the lumen of the capillaries and their endothelial layer. The compression of the capillary tubes is combined with a two-fold lessening suggest that the additional blood volume entering the gland in response to its perfusion by the liquid does not reach the blood capillaries and is thrown off into the vein vessels through the shunt communications and that regulation of the blood volume getting into the cat parotid gland capillaries is likely to depend on the hydraulic and osmotic state in the interstitial space of glandular lobes.

RESUMO

A method for obtaining injection replications of microvessels for scanning electron microscopy using nonsaturated polyether resin PH-8 is described. Possible applications of the method in question to study microcirculatory bed are discussed. With resine PH-8 it is possible to obtain complete and detailed replications which give information on three-dimensional organization both of the microcirculatory bed and of the vascular microrelief peculiarities.

RESUMO

Regularities in structural rearrangement of epithelium of blood capillaries in the rat parotid salivary gland have been studied under various functional conditions of the organ. The data obtained demonstrate that secretion is accompanied by dilatation of anuclear segments of capillary endothelium, decreasing thickness of endotheliocytes, rise of hematocrit number by adding the total area of the capillary fenestrated surface, the form factor of intraendothelial contacts and decreasing length of the contacting cytolemmas. Since extreme values of the metrical parameters studied have been revealed during the phase of the secrete excretion, a suggestion is made that increasing hydraulic permeability of the metabolic vessels with subsequent transport of the fluid to the terminal parts of the gland create the conditions promoting to reach the threshold level of pressure in acini that is necessary to overcome high resistance of nerrow intercalated sections for the secretion to reach the ducts.

Topological approach, suggested in the work, is a further development of methodology in studying microcirculation, especially its morphological basis. Owing to this approach, it is possible to consider spacial-time (coordinative) interrelations of different transport communications together with their histotopography. Hemato-lymphatic interrelations in the peritoneum can serve as an excellent example. Regularly repeating ansambles of transport communications (vascular and others) are interpreted as systems invariable for structural-functional units of the organ as to transport of fluid in tissues. Such complexes (modules) possess certain anatomy in hemodynamical and metabolic sense. Analysis of hemato-lymphatic interactions mediated by the interstitial space makes it possible to unite, within the module, processes of transmural, interstitial and axial (intervascular) transport. Movement of blood, interstitial fluid and lymph in tissues must be of oscillatory (discrete) character. As their trigger mechanism regulating rhythm and sequence of transport processes can serve the activity of metabolic lymphatic microvessels. Regulation of blood volume in capillaries depends on hydraulic and osmotic conditions in the interstitial space and on intensity of transmural transport. Topological approach can be effective in analyzing transport interrelations in tissues of different organs.
