Plant growth-promoting bacteria (PGPB) are known to increase plant tolerance to several abiotic stresses, specifically those from dry and salty environments. In this study, we examined the endophyte bacterial community of five plant species growing in the Thar desert of Pakistan. Among a total of 368 culturable isolates, 58 Bacillus strains were identified from which the 16 most divergent strains were characterized for salt and heat stress resilience as well as antimicrobial and plant growth-promoting (PGP) activities. When the 16 Bacillus strains were tested on the non-host plant Arabidopsis thaliana, B. cereus PK6-15, B. subtilis PK5-26 and B. circulans PK3-109 significantly enhanced plant growth under salt stress conditions, doubling fresh weight levels when compared to uninoculated plants. B. circulans PK3-15 and PK3-109 did not promote plant growth under normal conditions, but increased plant fresh weight by more than 50% when compared to uninoculated plants under salt stress conditions, suggesting that these salt tolerant Bacillus strains exhibit PGP traits only in the presence of salt. Our data indicate that the collection of 58 plant endophytic Bacillus strains represents an important genomic resource to decipher plant growth promotion at the molecular level.

The pot experiment was conducted to select appropriate integrated weed management method in rice under different salinity levels (0, 4 and 8 dS m(-1)). All the parameters including rice and weed measured were significantly influenced by weed control treatments at all salinity levels. Treatments including weed-free condition, Pretilachlor @0.375 kg ai ha(-1) + hand weeding, Propanil + Thiobencarb @ 0.9 kg ai ha(-1) and 1.8 kg ai ha(-1)+ hand weeding performed better under all salinity levels. Pretilachlor @ 0.375 kg ai ha(-1) with one round of hand weeding and propanil + thiobencarb 0.9 kg ai ha(-1) + 1.8 kg ai ha(-1) with one round of hand weeding were comparable to weed-free yields, and were superior to other treatments under salinity condition. Considering all the parameters, pretilachlor @ 0.375 kg ai ha(-1) + one round of hand weeding (at 65 DAT), propanil + thiobencarb 0.9 kg ai ha(-1) +1.8 kg ai ha(-1) + one round of hand weeding (at 65 DAT) gave the most effective control of weeds in rice under saline environments.

OBJECTIVE: This study was carried out to evaluate the drug prescribing, utilization pattern and adverse drug reactions recording associated with drugs prescribed to glaucoma patients. MATERIALS AND METHODS: A total of 50 glaucoma patients were included in the study, based on inclusion and exclusion criteria. All the observations were recorded in drug utilization and ADR recording documentation form. RESULTS: Out of 50 patients suffering from glaucoma, 38 patients (76%) were diagnosed open angle glaucoma, 4 patients (8%) closed angle glaucoma and 8 patients (16%) post-operative respectively. There were 19 patients (38%) males and 31 patients (62%) were females. The age range between 41-50 years had the maximum number of patients 15 (30%). A total of 17 patients (34%) had family history of glaucoma. Timolol was prescribed to 34 patients (68%), followed by dorzolamide 18 patients (36%) and acetazolamide 14 patients (28%). A total of 32 patients (64%) were prescribed single drug therapy whereas 18 patients (36%) were on multiple drug therapy. A total of 25 patients (50%) reported ADR. In the present study, latanoprost was associated with maximum number of ADRs 9 patients (18%) followed by acetazolamide 7 patients (14%), dorzolamide 4 patients (8%), then timolol 3 patients (6%) and pilocarpine 2 patients (4%). According to Naranjo scale, in 6 patients (24%) the ADR were unlikely, 12 patients (48%) were given possible score, 3 patients (12%) were given probable score, and 4 patients (16%) were given definite scores. CONCLUSION: In the present study, the maximum patients were in the age group of 41-50 years. The most commonly prescribed drugs were timolol followed by dorzolamide, acetazolamide. Latanoprost was associated with maximum number of ADRs.

Oil palm tissues are rich in polyphenols, polysaccharides and secondary metabolites; these can co-precipitate with RNA, causing problems for downstream applications. We compared two different methods (one conventional and a kit-based method - Easy-Blue(TM) Total RNA Extraction Kit) to isolate total RNA from leaves, roots and shoot apical meristems of tissue culture derived truncated leaf syndrome somaclonal oil palm seedlings. The quality and quantity of total RNA were compared through spectrophotometry and formaldehyde gel electrophoresis. The specificity and applicability of the protocols were evaluated for downstream applications, including cDNA synthesis and RT-PCR analysis. We found that the conventional method gave higher yields of RNA but took longer, and it was contaminated with genomic DNA. This method required extra genomic DNA removal steps that further reduced the RNA yield. The kit-based method, on the other hand, produced good yields as well as well as good quality RNA, within a very short period of time from a small amount of starting material. Moreover, the RNA from the kit-based method was more suitable for synthesizing cDNA and RT-PCR amplification than the conventional method. Therefore, we conclude that the Easy-BlueTM Total RNA Extraction Kit method is suitable and superior for isolation of total RNA from oil palm leaf, root and shoot apical meristem.

Rice straw is produced as a by-product from rice cultivation, which is composed largely of lignocellulosic materials amenable to general biodegradation. Lignocellulolytic actinobacteria can be used as a potential agent for rapid composting of bulky rice straw. Twenty-five actinobacteria isolates were isolated from various in situ and in vitro rice straw compost sources. Isolates A2, A4, A7, A9 and A24 were selected through enzymatic degradation of starch, cellulose and lignin followed by the screening for their adaptability on rice straw powder amended media. The best adapted isolate (A7) was identified as Micromonospora carbonacea. It was able to degrade cellulose, hemicelluloses and carbon significantly (P ≤ 0.05) over the control. C/N ratio was reduced to 18.1 from an initial value of 29.3 in 6 weeks of composting thus having the potential to be used in large scale composting of rice straw.

Cosmos caudatus Kunth. (Asteraceae), commonly known as ulam raja, is widely grown as an herbal aromatic shrub. In Malaysia, its young leaves are popularly eaten raw as salad with other greens and have been reported to possess extremely high antioxidant properties, which may be partly responsible for some of its believed medicinal functions. In early 2010, a suspected powdery mildew was observed on ulam raja plants at the Agricultural Park of Universiti Putra Malaysia. Initially, individual, white, superficial colonies were small and almost circular. Later, they enlarged and coalesced to cover the whole abaxial leaf surface. With development of the disease, all green parts (leaves, stems, and petioles) became covered with a continuous mat of mildew, giving a dusty appearance. Newly emerged leaves rapidly became infected. Diseased leaves ultimately senesced and dried up, making them aesthetically unattractive and unmarketable. The pathogen produced conidia in short chains (four to six conidia) on erect conidiophores. Conidiophores were unbranched, cylindrical, 125 to 240 µm long, with a slightly swollen foot cell. Individual conidia were hyaline, ellipsoid, and 25 to 30 (27.5) × 15 to 20 (17.5) µm with fibrosin inclusions. Morphological descriptions were consistent with those described for Sphaerotheca fuliginea or S. fusca, which has lately been reclassified as Podosphaera fusca (1). From extracted genomic DNA of P. fusca UPM UR1, the internal transcribed spacer (ITS) region was amplified with ITS1 (5'-TCCGTAGGTGAACCTGCGG-3') and ITS4 (5'-TCCTCCGCTTATTGATATGC-3'). A BLAST search of GenBank with an ITS rDNA sequence of this fungus (GenBank Accession No. HQ589357) showed a maximum identity of 98% to the sequences of two P. fusca isolates (GenBank Accession Nos. AB525915.1 and AB525914.1). To satisfy Koch's postulates, the pathogenicity of fungal strain UPM UR1 was verified on 4-week-old plants. Inoculation was carried out by gently rubbing infected leaves onto healthy plants of C. caudatus. Ten pots of inoculated plants were kept under a plastic humid chamber and 10 pots of noninoculated plants, placed under another chamber, served as controls. After 48 h, the plants were then placed under natural conditions (25 to 28°C). Powdery mildew symptoms, similar to those on diseased field plants, appeared after 7 days on all inoculated plants. The white, superficial colonies enlarged and merged to cover large areas within 2 weeks. The infected leaf tissues became necrotic 6 to 8 days after the appearance of the first symptoms. Sporulation of P. fusca was observed on all infected leaves and stems. No symptoms were seen on the control plants. To our knowledge, this is the first report of P. fusca causing powdery mildew on C. caudatus in Malaysia. This pathogen has also been reported previously to be economically important on a number of other hosts. This study was designed to investigate the hypothesis that the inhibition of ferrochelatase will cause in situ build up of high concentrations of protoporphyrin-IX which may act as a putative agent for photodestruction of cancer cells. The parenteral administration of lead acetate, a known inhibitor of ferrochelatase, to mice bearing cutaneous tumors (papillomas and carcinomas) caused a six-fold enhancement in the concentration of protoporphyrin-IX in tumors within a period of one month. Forty-eight hours after the second injection of lead, mice were exposed to visible light, at a light dose of about nine kilo lux for a period of one hour (in four sittings of fifteen minutes each keeping a gap of ten minutes between two exposures). A significant reduction in tumor size was observed starting as early as day one following the treatment. Continuous treatment for six consecutive days resulted in almost complete ablation of the tumor mass in most of the animals. Complete regression of the tumors was observed at two to three days following the first exposure. Our observations on in situ accumulation of protoporphyrin-IX by heme-biosynthesis inhibition represent a novel method for photodynamic therapy of cancer cells. It is important to emphasize that lead is a fairly toxic agent and developing a non-toxic agent is one of our future goals.
