RESUMO

An immunocytochemical analysis of hormonal status of transgenic rats containing human growth hormone gene has been done. The enhanced expression of the endogenous growth hormone gene was demonstrated with poly- and monoclonal antibodies inside somatatropes of pituitary. No activity of the heterologous growth hormone gene was revealed in kidney, pancreas or liver as it might be expected according to specificity of MT1 and TAT promotors. Transgenic animals of F0, F1 and F2 generation exhibited disturbance of functional morphology of glucagon and insulin producing cells. Lymphocyte infiltration was found in pancreatic islets. The transgenic rabbits and swine with the gene of releasing factor of human growth hormone did not reveal any severe disturbance. Although one swine demonstrated alterations in glucagon producing cells and one rabbit revealed a disturbed morphology of the stomach tissues. The data are discussed in relation to general problems of transgene activity and interaction with endogenous homolog.

By means of covalent chromatography on thiopropyl-sepharose 6B the N-terminal, as well as other tryptic cysteine-containing peptides of the bovine tryptophanyl-tRNA-synthetase (EC 6.1.1.2) were purified and characterized, their structures being determined by a combination of plasma desorption mass spectrometry and peptide sequencing. In total, six different peptides containing seven cysteine residues were analysed. The N-terminal amino acid (presumably, alanine) was shown to be acetylated in the nature enzyme amino acid sequences of some cysteine-containing peptides proved to differ from those deduced from the cDNA structure, thus indicating the presence of the enzyme's isoforms. The purification does not affect the peptides' sulfhydryl groups. The number of cysteine residues in the peptides could be determined with a high accuracy by measuring their masses before and after alkylation with 4-vinylpyridine.

RESUMO

Revealing of enterochromaffine and enterochromaffine-like cells of the rat stomach was undertaken under the effect of bombesin and alpha- and beta-adrenoblocking agents. Fibers containing gastrin-releasing-peptide were immunohistochemically revealed in neural plexus of the rat digestive tract. Spontaneous rhythmic release of the stomach monoamine cells' secret was shown. The effect of bombesin on the enterochromaffine-like cells of the rat stomach fundus containing histamine, was shown to be actualized through beta-adrenoreceptors. Serotonin excretion from the enterochromaffine cells of the stomach antrum depends neither on bombesin nor on the adrenergic effects.

RESUMO

Using antisera against serotonin (5-OT) and neurotensin (NT) with immunofluorescence, 5-OT-immunoreactive and NT-immunoreactive nerve fibers have been detected in the walls of the femoral and pial arteries and intragastric arterioles. With electron microscopy, adrenergic and cholinergic axons were found in close anatomical proximity to some intragastric capillaries and arterioles. A possible role of the detected nerves is being discussed.

RESUMO

Localization of serotonin (5-OT)- and neurotensin (NT)-containing cells in the tunica mucosa of the gastric antral part and the duodenum of the rat and chick has been revealed by means of the immunofluorescent method and the method with application of the peroxidase-antiperoxidase complex. For immune-histochemical revealing of biologically active substances in endocrine cells Sovient antisera are used for the first time. Experiments are performed on injection of antiserotonin serum of various dosage into the rat blood bed. The reaction of serotonin-containing cells of the gastric antral part tunical mucosa and those of the duodenum to changes in serotonin concentration in blood serum is demonstrated.

RESUMO

Ability of homologous antistreptococcal sera obtained in various periods after immunization (7, 14, 21, 30 and 60 days) to alter the testis following intravenous injection was studied in guinea-pigs. Only 30-day immune sera containing maximum antibodies to testicular antigens (fluorescent and spermacytotoxic) were able to induce alteration of the testis after preliminary (10 days before) intracutaneous administration of complete Freund's adjuvant. Within the first two days after the alteration induction, fixation of globulin from the immune serum on the structures of spermatozoas from the testis was determined by the immunofluorescent technique. Atrophy of the spermatogenic epithelium developed by the 7th day after the alteration induction. IgM containing spermatocytotoxic antibodies are responsible for the testis impairment induced by the spermatocytotoxic antibodies.

RESUMO

A substance with insulin-like activity was obtained from visceral organs (excluding foot, gills and mantle) of the freshwater bivalve molluscs Unio pictorum and Anodonta cygnea. Injecting this substance to guinea pigs, antiserum was also obtained. Using the obtained antiserum in immunostaining experiments, the cells which produce insulin-like substance were identified in the intestinal epithelium of the midgut in A. cygnea.

RESUMO

Insulin-like immunoreactivity can be localized in the cells of the mid-gut of Unio pictorum Lam. The cells containing the substance immunoreactive to mammalian anti-insulin serum, can be stained with paraldehyde-fuchsin or alcian blue and give positive PAS reaction. Intramuscular glucose administration produces degranulation of the cytoplasm in these cells: the quantity of cells stainable with paraldehyde-fuchsin significantly decreased 24 hours after glucose injection.
