The kinetics of cortisol was investigated in 26 healthy subjects and in 9 patients with rheumatoid arthritis induced by exogenous glucocorticosteroids. The cortisol concentration was determined in blood serum by using a high-efficiency liquid chromatography method. The cortisol kinetic parameters were found by empirically constructed formulae. An inadequate synthesis of cortisol was found in patients with rheumatoid arthritis after prednisolone administration. The results can be used for evaluating the pharmacodynamic properties of glucocorticosteroids.

Serum hydrocortisone concentrations were measured by high performance liquid chromatography (HPLC) and radioimmunoassay (RIA) in 24 children with different diseases and 16 healthy adults. HPLC and RIA proved to be equally sensitive. However the authors recommend HPLC as a reliable, rapid, and ecologically pure method, which is, in addition, selective and economic.

The volume of retention of corticosterone, hydrocortisone, aldosterone, dexamethasone, cortisone, methylprednisolone, and prednisolone during their separation by high-performance liquid chromatography was evaluated. The studies were performed on a Milichrome microcolumn chromatographer. The order of glucocorticoid release and volume of their retention were under study. Measurement of endogenous and exogenous plasma glucocorticoids is practically important in clinical medicine. Steroid concentrations indicate the pharmacodynamics and pharmacokinetics of exogenous glucocorticoids and their effects on the production of endogenous steroids and reflect the biorhythms of corticosterone and hydrocortisone.
