The building industry is responsible for a large amount of waste, and the measurement and modelling of this waste could be used to develop better waste management plans. Several theoretical models explain the relationships between waste and building characteristics, but local practices may result in different behaviours. This study aimed to measure and analyse the waste generated through construction. It was based on the analysis of 18 building sites located in the region of Porto Alegre, Brazil. Waste was measured at these sites, and the results showed an average waste generation rate of 0.151 m3 m-2. A regression analysis of the collected data presented a satisfactory performance in two models. The first model was developed to explain total waste generation, including the effects of certain attributes, with an R2 = 0.81. The changes in waste generated during construction were estimated. The second model considered time schedules and examined the effect of the construction stage on waste generation, and reached an R2 = 0.91. The model with time indicated an S-shaped relationship. The models presented satisfactory statistical parameters and could be used to produce better waste management plans in the preconstruction stage.

In Brazil, the use of wood in construction is primarily temporary, and it can represent a great percentage of construction waste. It is typically discarded with minimal reuse or recycling. As landfill wood disposal could result in methane emissions and/or leaching of hazardous constituents polluting water or soil, the implementation of temporary wood waste reduction strategies must be a critical issue for local construction companies. To manage and control wood waste generation, including setting some reduction goals, it is necessary to identify the influencing factors and ways to quantitatively predict their relative contributions. This study uses a multiple regression statistical model to estimate the amount of temporary wood waste generated in the construction of high-rise buildings by considering the influencing factors related to the design/construction as well as site and safety installations. The case study includes 22 high-rise residential buildings. The regression model predicted approximately 89% of the factors involved in the generation of wood waste in similar constructions. The dependent variables that had an influence on the amount of wood waste are related to design features ('number of floors'), structural construction system ('in situ moulded concrete volume'), and site and safety installations ('site wood fence' and 'wood use rate'). Ways of minimising wood waste generation and the contributions of each type of temporary wood use were discussed.

Reducing construction waste is becoming a key environmental issue in the construction industry. The quantification of waste generation rates in the construction sector is an invaluable management tool in supporting mitigation actions. However, the quantification of waste can be a difficult process because of the specific characteristics and the wide range of materials used in different construction projects. Large variations are observed in the methods used to predict the amount of waste generated because of the range of variables involved in construction processes and the different contexts in which these methods are employed. This paper proposes a statistical model to determine the amount of waste generated in the construction of high-rise buildings by assessing the influence of design process and production system, often mentioned as the major culprits behind the generation of waste in construction. Multiple regression was used to conduct a case study based on multiple sources of data of eighteen residential buildings. The resulting statistical model produced dependent (i.e. amount of waste generated) and independent variables associated with the design and the production system used. The best regression model obtained from the sample data resulted in an adjusted R(2) value of 0.694, which means that it predicts approximately 69% of the factors involved in the generation of waste in similar constructions. Most independent variables showed a low determination coefficient when assessed in isolation, which emphasizes the importance of assessing their joint influence on the response (dependent) variable.

OBJECTIVES: To determine the antibiotic resistance and fingerprint profiles of methicillin-resistant coagulase-negative staphylococci (MRCoNS) from animal infections among different practices and examine the history of antibiotic treatment. METHODS: Isolates were identified by mass spectrometry and tested for antimicrobial resistance by broth dilution, microarrays and sequence analysis of the topoisomerases. Diversity was assessed by PFGE, icaA PCR and staphylococcal cassette chromosome mec (SCCmec), arginine catabolic mobile element (ACME) and multilocus sequence typing. Clinical records were examined retrospectively. RESULTS: MRCoNS were identified as Staphylococcus epidermidis (n=20), Staphylococcus haemolyticus (n=17), Staphylococcus hominis (n=3), Staphylococcus capitis (n=1), Staphylococcus cohnii (n=1) and Staphylococcus warneri (n=1). PFGE identified one clonal lineage in S. hominis isolates and several in S. haemolyticus and S. epidermidis. Fourteen sequence types were identified in S. epidermidis, with sequence type 2 (ST2) and ST5 being predominant. Ten isolates contained SCCmec IV, seven contained SCCmec V and the others were non-typeable. ACMEs were detected in 11 S. epidermidis isolates. One S. hominis and 10 S. epidermidis isolates were icaA positive. In addition to mecA-mediated ß-lactam resistance, the most frequent resistance was to gentamicin/kanamycin [aac(6')-Ie-aph(2')-Ia, aph(3')-III] (n=34), macrolides/lincosamides [erm(C), erm(A), msr, lnu(A)] (n=31), tetracycline [tet(K)] (n=22), streptomycin [str, ant(6)-Ia] (n=20), trimethoprim [dfr(A), dfr(G)] (n=17), sulfamethoxazole (n = 34) and fluoroquinolones [amino acid substitutions in GyrA and GrlA] (n=30). Clinical data suggest selection through multiple antibiotic courses and emphasize the importance of accurate diagnosis and antibiograms. CONCLUSIONS: MRCoNS from animal infection sites are genetically heterogeneous multidrug-resistant strains that represent a new challenge in the prevention and therapy of infections in veterinary clinics.

A high-resolution multi-proxy analysis was conducted on a 1.5-m-long core of Tortonian age (~ 10.5 Ma; Late Miocene) from Austria (Europe). The lake sediments were studied with a 1-cm resolution to detect all small-scale variations based on palynomorphs (pollen and dinoflagellate cysts), ostracod abundance, geochemistry (carbon and sulfur) and geophysics (magnetic susceptibility and natural gamma radiation). Based on an already established age model for a longer interval of the same core, this sequence can be limited to approx. two millennia of Late Miocene time with a resolution of ~ 13.7 years per sample. The previous study documented the presence of solar forcing, which was verified within various proxies on this 1.5-m core by a combination of REDFIT spectra and Gaussian filters. Significant repetitive signals ranged in two discrete intervals corresponding roughly to 55-82 and 110-123 years, fitting well within the lower and upper Gleissberg cycle ranges.Based on these results, the environmental changes along the 2000-year Late Miocene sequence are discussed. No major ecological turnovers are expected in this very short interval. Nonetheless, even within this brief time span, dinoflagellates document rapid changes between oligotrophic and eutrophic conditions, which are frequently coupled with lake stratification and dysoxic bottom waters. These phases prevented ostracods and molluscs from settling and promoted the activity of sulfur bacteria. The pollen record indicates rather stable wetland vegetation with a forested hinterland. Shifts in the pollen spectra can be mainly attributed to variations in transport mechanisms. These are represented by a few phases of fluvial input but mainly by changes in wind intensity and probably also wind direction. Such influence is most likely caused by solar cycles, leading to a change in source area for the input into the lake.Furthermore, these solar-induced variations seem to be modulated by longer solar cycles. The filtered data display comparable patterns and modulations, which seem to be forced by the 1000-year and 1500-year cycles. The 1000-year cycle modulated especially the lake surface proxies, whereas the 1500-year cycle is mainly reflected in hinterland proxies, indicating strong influence on transport mechanisms.

Precipitation over India is driven by the Indian monsoon. Although changes in this atmospheric circulation are caused by the differential seasonal diabatic heating of Asia and the Indo-Pacific Ocean, it is so far unknown how global warming influences the monsoon rainfalls regionally. Herein, we present a Miocene pollen flora as the first direct proxy for monsoon over southern India during the Middle Miocene Climate Optimum. To identify climatic key parameters, such as mean annual temperature, warmest month temperature, coldest month temperature, mean annual precipitation, mean precipitation during the driest month, mean precipitation during the wettest month and mean precipitation during the warmest month the Coexistence Approach is applied. Irrespective of a ~ 3-4 °C higher global temperature during the Middle Miocene Climate Optimum, the results indicate a modern-like monsoonal precipitation pattern contrasting marine proxies which point to a strong decline of Indian monsoon in the Himalaya at this time. Therefore, the strength of monsoon rainfall in tropical India appears neither to be related to global warming nor to be linked with the atmospheric conditions over the Tibetan Plateau. For the future it implies that increased global warming does not necessarily entail changes in the South Indian monsoon rainfall.

BACKGROUND: Epidural Anesthesia (EA) is a well-established procedure. The aim of the present study was to evaluate the incidence of immediate complications following epidural puncture, such as sanguineous puncture, accidental dural perforation, unsuccessful catheter placement or insufficient analgesia and to identify patient and maneuver related risk factors. METHODS: A total of 7958 non-obstetrical EA were analyzed. The risk of each complication was calculated according to the preconditions and the level of puncture. For probabilistic evaluation we used a logistic regression model with forward selection. RESULTS: The risk of sanguineous puncture (n = 247, 3.1%) increases with both the patient's age (P = 0.013) and the more caudal the approach (P < 0.01). Dural perforation (n = 123, 1.6%) was found to be influenced only by advanced age (P = 0.019). Unsuccessful catheter placement (n = 68, 0.94%) occurred more often in smaller individuals (P < 0.001) and at lower lumbar sites (P < 0.01). Amongst all cases with successful catheter placement a (partial) insufficient analgesia was found in 692 cases (8.8%). This risk of insufficient analgesia decreased with patient's age (P <0 .01), being least likely for punctures of the lower thoracic spine (P < 0.001). CONCLUSIONS: Compared to more cranial levels, EA of the lower spine is associated with an increased risk of sanguineous and unsuccessful puncture. Insufficient analgesia more often accompanies high thoracic and low lumbar approaches. The risk of a sanguineous puncture increases in elderly patients. Gender, weight and body mass index seem to have no influence on the investigated complications.

High resolution pollen and dinoflagellate analyses were performed on a continuous 98-cm-long core from Tortonian deposits of Lake Pannon in the Styrian Basin in Austria. The sample distance of 1-cm corresponds to a resolution of roughly one decade, allowing insights into environmental and climatic changes over a millennium of Late Miocene time. Shifts in lake level, surface water productivity on a decadal- to centennial-scale can be explained by variations of rainfall during the Tortonian climatic optimum. Related to negative fine scale shifts of mean annual precipitation, shoreline vegetation belts reacted in an immediate replacement of Poaceae by Cyperaceae as dominant grasses in the marshes fringing the lake. In contrast to such near-synchronous ecosystem-responses to precipitation, a delayed lake level rise of 4-6 decades is evident in the hydrological budget of Lake Pannon. This transgression, caused by a precipitation increase up to > 1200 mm/yr, resulted in a complete dieback of marshes. Simultaneously, "open-water" dinoflagellates, such as Impagidinium, took over in the brackish lagoon and fresh water dinoflagellates disappeared. As soon as the rainfall switched back to moderate levels of ~ 1100-1200 mm/yr, the rise of the lake level slowed down, the marsh plants could keep up again and the former vegetation belts became re-established. Thus, mean annual precipitation, more than temperature, was the main driving force for high-frequency fluctuations in the Tortonian wetlands and surface water conditions of Lake Pannon. Such high resolution studies focusing on Tortonian decadal to centennial climate change will be crucial to test climate models which try to compare the Tortonian models with predictions for future climate change.

Pollen analyses have been proven to possess the possibility to decipher rapid vegetational and climate shifts in Neogene sedimentary records. Herein, a c. 21-kyr-long transgression-regression cycle from the Lower Austrian locality Stetten is analysed in detail to evaluate climatic benchmarks for the early phase of the Middle Miocene Climate Optimum and to estimate the pace of environmental change.Based on the Coexistence Approach, a very clear signal of seasonality can be reconstructed. A warm and wet summer season with c. 204-236 mm precipitation during the wettest month was opposed by a rather dry winter season with precipitation of c. 9-24 mm during the driest month. The mean annual temperature ranged between 15.7 and 20.8 °C, with about 9.6-13.3 °C during the cold season and 24.7-27.9 °C during the warmest month. In contrast, today's climate of this area, with an annual temperature of 9.8 °C and 660 mm rainfall, is characterized by the winter season (mean temperature: -1.4 °C, mean precipitation: 39 mm) and a summer mean temperature of 19.9 °C (mean precipitation: 84 mm).Different modes of environmental shifts shaped the composition of the vegetation. Within few millennia, marshes and salt marshes with abundant Cyperaceae rapidly graded into Taxodiaceae swamps. This quick but gradual process was interrupted by swift marine ingressions which took place on a decadal to centennial scale. The transgression is accompanied by blooms of dinoflagellates and of the green alga Prasinophyta and an increase in Abies and Picea. Afterwards, the retreat of the sea and the progradation of estuarine and wetland settings were a gradual progress again.Despite a clear sedimentological cyclicity, which is related to the 21-kyr precessional forcing, the climate data show little variation. This missing pattern might be due to the buffering of the precessional-related climate signal by the subtropical vegetation. Another explanation could be the method-inherent broad range of climate-parameter estimates that could cover small scale climatic changes.

BACKGROUND: Cervical cancer is the second most frequent cancer among woman worldwide and is considered to be caused by infection with high-risk papilloma viruses. Genetic immunization using recombinant adeno-associated virus (rAAV) vectors has shown great promise for vaccination against human papillomavirus (HPV) infections. METHODS: rAAV5, -8 and -9 vectors expressing an HPV16 L1/E7 fusion gene were generated and applied intranasally for combined prophylactic and therapeutic vaccination of mice. RESULTS: The rAAV5 and the rAAV9 vectors showed efficient induction of both humoral and cellular immune responses, whereas rAAV8 failed to immunize mice by the intranasal route. The L1-specific immune response evoked by expression of the L1/E7 fusion gene, however, was lower than that evoked by expression of the L1 antigen alone. This deficiency could be compensated by application of Escherichia coli heat-labile enterotoxin or monophsphoryl lipid as adjuvant upon vaccination with rAAV5-L1/E7. Coimmunization of rAAV9-L1/E7 with rAAV5-L1 or boosting of rAAV9-L1/E7 with rAAV5-L1 strongly increased L1-specific neutralizing antibody titres to levels above those achieved by vaccination with vectors expressing L1 alone. Both vectors elicited a vibrant cytotoxic T-lymphocyte response against L1 or E7. Nasal immunization with rAAV5 or rAAV9 was superior to vaccination with HPV16-L1 virus-like particles (VLPs) or HPV16-L1/E7 CVLPs with respect to humoral and cellular immune responses. Vaccination with the rAAV vectors led to a significant protection of animals against a challenge with different HPV tumour cell lines. CONCLUSIONS: Our results show that rAAV5 and rAAV9 vectors are promising candidates for a non-invasive nasal vaccination strategy.

A detailed ultra-high-resolution analysis of a 37-cm-long core of Upper Miocene lake sediments of the long-lived Lake Pannon has been performed. Despite a general stable climate at c. 11-9 Ma, several high-frequency oscillations of the paleoenvironments and depositional environments are revealed by the analysis over a short time span of less than 1000 years. Shifts of the lake level, associated with one major 3rd order flooding are reflected by all organisms by a cascade of environmental changes on a decadal scale. Within a few decades, the pollen record documents shifting vegetation zones due to the landward migration of the coast; the dinoflagellate assemblages switch towards "offshore-type" due to the increasing distance to the shore; the benthos is affected by low oxygen conditions due to the deepening. This general trend is interrupted by smaller scale cycles, which lack this tight interconnection. Especially, the pollen data document a clear cyclicity that is expressed by iterative low pollen concentration events. These "negative" cycles are partly reflected by dinoflagellate blooms suggesting a common trigger-mechanism and a connection between terrestrial environments and surface waters of Lake Pannon. The benthic fauna of the core, however, does not reflect these surface water cycles. This forcing mechanism is not understood yet but periodic climatic fluctuations are favoured as hypothesis instead of further lake level changes. Short phases of low precipitation, reducing pollen production and suppressing effective transport by local streams, might be a plausible mechanism. This study is the first hint towards solar activity related high-frequency climate changes during the Vallesian (Late Miocene) around Lake Pannon and should encourage further ultra-high-resolution analyses in the area.

Adeno-associated viruses (AAV) have been developed and evaluated as recombinant vectors for gene therapy. More recently, due to the advantages they offer for gene transfer, several AAV serotypes have gained increasing interest. However, monoclonal antibodies for the characterization and quantitation of vectors derived from different serotypes are at present not available. Serotype-specific monoclonal antibodies (mAbs) against the capsids of the serotypes 1/6, 4 and 5 are described. These antibodies, designated as ADK1a and b, ADK4 or ADK5a and b, reacted specifically with the indicated serotype capsids in cell lysates. ADK 1a and b cross-reacted with its highly related AAV6 serotype, but not with the other serotypes tested. The new antibodies recognized exclusively assembled capsids and neither free nor denatured capsid proteins as shown by fractionation experiments. In immunofluorescence experiments, the mAbs stained only distinct intranuclear foci in cells expressing the capsid protein. The development of capture ELISAs for quantitation of AAV1 and 6, AAV4 or AAV5 capsids illustrates that these novel monoclonal antibodies provide valuable tools for characterization of vector stocks.

Adeno-associated viruses (AAV) have been developed and evaluated as recombinant vectors for gene therapy in many preclinical studies, as well as in clinical trials. However, only a few approaches have used recombinant AAV (rAAV) to deliver vaccine antigens. We generated an rAAV encoding the major capsid protein L1 (L1h) from the human papillomavirus type 16 (HPV16), aiming to develop a prophylactic vaccine against HPV16 infections, which are the major cause of cervical cancer in women worldwide. A single dose of rAAV5 L1h administered intranasally was sufficient to induce high titers of L1-specific serum antibodies, as well as mucosal antibodies in vaginal washes. Seroconversion was maintained for at least 1 year. In addition, a cellular immune response was still detectable 60 weeks after immunization. Furthermore, lyophilized rAAV5 L1h successfully evoked a systemic and mucosal immune response in mice. These data clearly show the efficacy of a single-dose intranasal immunization against HPV16 based on the recombinant rAAV5L1h vector without the need of an adjuvant.

Infections by human papillomaviruses (HPV) are the major cause of uterine cancer in women worldwide. Aiming to develop a combined prophylactic and therapeutic vaccine we have previously demonstrated immunogenicity of chimeric virus-like particles consisting of a C-terminally truncated HPV 16 L1 capsid protein fused to an E7 portion. Here we show that genetic vaccination with a corresponding DNA was inefficient in the induction of a L1-specific prophylactic immune response. DNA immunization with C-terminally truncated HPV 16 L1 genes of different lengths revealed that only short deletions (L1(1-498)) were tolerated for eliciting a humoral immune response against viral capsids. This correlates with the observation that the C-terminal sequences are critical for nuclear localization, capsomere and capsid assembly. However, only the ability of L1 protein to form capsomeres or capsids showed a direct influence on the outcome of the immune response. C-terminal insertion of 60 amino acids of E7 was tolerated in fusion constructs, whereas insertion of full-length E7(1-98) or shuffled E7 (149 aa) completely abolished the humoral immune response. The L1(1-498)/E7(1-60) fusion construct not only induced L1-specific antibodies but also L1- and E7-specific CTL responses after DNA vaccination.
