RESUMO

Fish adapt to changing environments by maintaining homeostasis or making energy trade-offs that impact fitness. We investigated the effect of Zn on the fitness and physiology of Barbus meridionalis, a native cyprinid fish species, under two exposure scenarios. The Osor stream's mine-effluent reach represented long-term (chronic) exposure, while the upstream reach served as a control/acute exposure. Acute exposure involved exposing B. meridionalis to 1mg/L Zn for 96 h in the laboratory. We examined physiological traits (Standard metabolic rate SMR, Maximum metabolic rate MMR, Absolute Aerobic scope AAS, Critical swimming capacity Ucrit) and antioxidant system, AS (Superoxide dismutase, SOD; Catalase, CAT; Glutathione peroxidase, GPX; Glutathione-S-transferase, GST; Glutathione, GSH; Thiobarbaturic acid reactive substances, TBARS) biomarkers. The results indicated that Zn had no significant effect on osmoregulatory cost (SMR) in either exposure scenario but impaired energetically costly exercise (low MMR). AAS reduction in both exposure groups suggested compromised energy allocation for life-history traits, evidenced by decreased locomotor performance (Ucrit) after acute exposure. Tissue-specific and time-dependent responses were observed for AS biomarkers. The fish exhibited ineffective control of oxidative damage, as evidenced by high TBARS levels in the liver and gills, despite increased CAT and GSH in the liver under acute conditions. Our findings demonstrate differential responses at the subcellular level between the two exposure scenarios, while trait-based endpoints followed a similar pattern. This highlights the utility of a trait-based approach as a supplementary endpoint in biomonitoring studies, which provides insights into impacts on individual fitness and population demography.

The green-collar strategies for nanomaterial synthesis with novel structural competencies have received significant attention in nanotechnology owing to their potential benefits. The utilization of silica nanoparticles for wastewater treatment through heavy metal ions remediation is the focal point of the present study. With this intent, silica was extracted from bagasse ash by the sol-gel method and modified using chitosan. Chemical and physical characteristics of silica(S), silica/Chitosan (SCs), were reckoned through X-ray Powder Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), and Scanning Electron Microscopy (SEM) and the efficiency of synthesized biomaterials for removing heavy metal ions. Cadmium and Lead from wastewater was evaluated by conducting closed batch experiments. Isotherm and kinetics models were applied to understand the adsorption mechanism. Results of heavy metal ions removal showed that the S possesses the highest removal efficiency of 88% for cadmium. Equilibrium was established within 56 min following a Langmuir isotherm model and pseudo-second-order reaction. The synthesized biomaterials were also tested against the fungal (Aspergillus Niger) and bacterial strains (Escherichia coli and Staphylococcus aureus) to determine their antimicrobial properties Maximum inhibition of 26 mm was shown by SCs for E.coli. Synthesized samples were not so effective for A.niger. The high adsorption potential of silica nanoparticles reveals their potential to treat wastewater containing inorganic pollutants like calcium and lead released from the sugar industry firsthand, thereby building a circular economy by controlling the pollution from source to sink. The synthesized silica nanoparticles and silica/chitosan biomaterials demonstrated high adsorption potential for heavy metal ions, making them promising candidates for integration into Algal Membrane Bioreactors to enhance wastewater treatment efficiency and remove toxic pollutants. Their multifunctional properties, including antimicrobial activity, also offer potential for improving microbial control within AMBRs, ensuring a more effective and sustainable wastewater treatment process.

Access to clean water is the mandatory requirement for every living being to sustain life. So, membrane-based integrated approach of adsorption and separation technology has recently been preferred by scientists over other conventional techniques, for wastewater treatment. Current research focused on the synthesis of novel imidazolium (A1) based IL, which was further functionalized with hydroxyapatite (HAp; extracted from Labeo rohita scales), to create possible solutions towards environmental remediation. Cellulose acetate (CA) was used for the fabrication of three different ionic liquid membranes. All the synthesized products were characterized by FTIR, XRD and TGA. Two dyes of different nature, i.e., congo red (anionic) and crystal violet (cationic) were selected because of their highly toxic and carcinogenic effects, for batch adsorption experiments. Antibacterial activity of the synthesized membranes was also evaluated against S. aureus. Results of the study revealed that CA-HA1 1:2 acted as the best adsorbent towards the removal of crystal violet, exhibiting removal efficiency of 98% with the contact time of 24 h while in case of congo red adsorption, CA-HA1 (1:2) proved as prime adsorbent with the removal efficiency of 96% for the same preceding contact time. Considering the antibacterial character of the synthesized membranes, CA-A1 (1:1) emerged as very efficient antibacterial agent with the inhibition zone of 50 mm after 48 h. The overall behavior of monolayer and multilayer adsorption was witnessed for both dyes while kinetic studies favored the pseudo-second order reaction for all adsorbents.

Wetland is an important feature of the landscape that provides beneficial services. However, with the ever-increasing heavy metal load, the quality of wetlands is deteriorating. Dongzhangwu Wetland in Hebei, China, was taken as our study site. It provides breeding and foraging grounds to migratory water birds such as Little Egret (Egretta garzetta), Great Egret (Ardea alba), and Grey Heron (Ardea cinerea). The current study aimed to quantify heavy metals exposure hazard and risk to the water migratory birds by employing a non-destructive approach. Oral intake was considered the main exposure route to calculate total exposure via multiple phases. The concentrations of Cr, Zn, Cu, Pb, As, Ni, Mn, and Cd were investigated in water, soil and food compartments of three different habitat components (Longhe River, Natural Pond and Fish Pond). The results showed: (1) The trend of potential daily dose (PDD) was Mn > Zn > Cr > Pb > Ni > Cu > As > Cd while for hazard quotient (HQ) was Cr > Pb > Cu > Zn > As > Ni > Mn > Cd, making Cr, Pb, Cu, Zn, and As the priority pollutants in all habitats, with Natural Pond eliciting the highest exposure. (2) The cumulative heavy metal exposure, explained by the integrated nemerow risk index, characterized all three habitats for all the birds in high exposure risk category. (3) The exposure frequency index identified that all the birds are frequently exposed to heavy metals from multiple phases in all three habitats. (4) Little Egret is at the highest pedagogy of exposure from single or multiple heavy metal(s) in all three habitats. A rigorous management plan for identified priority pollutants is required to improve wetland functioning and ecological services. Decision-makers could use the developed tissue residue objectives for protecting Egret species in Dongzhangwu Wetland as benchmarks.

Dyes contaminated water has caused various environmental and health impacts in developing countries especially Pakistan due to different industrial activities. This issue has been addressed in present study by fabricating biocompatible ionic liquid (IL) membranes for the remediation of Crystal violet (CV) dye from contaminated water. Novel ammonium-based IL such as Triethyl dimethyl ammonium sulfate ([C3A][C2H6]SO4); (A2) was synthesized and further functionalized with hydroxyapatite (HAp; extracted from refused fish scales) resulting in the formation of HA2. Furthermore, A2 and HA2 were then used to fabricate the cellulose acetate (CA) based membranes with different volume ratios. The physicochemical properties of membranes-based composite materials were investigated using FTIR, XRD, and TGA and used for the adsorption of CV in the closed batch study. In results, CA-HA2 (1:2) showed higher efficiency of 98% for CV reduction, after the contact time of 90 min. Kinetic studies showed that the adsorption of CV followed the pseudo-second-order kinetic model for all adsorbents. The antibacterial properties of the synthesized membrane were investigated against gram-positive strain, S. aureus and CA-A2 (1:1) showed better antibacterial properties against S. aureus. The developed membrane is sustainable to be used for the adsorption of CV and against bacteria.

BACKGROUND: Pakistan is home to 4.6 million children who have been orphaned. Limited data on caregiving in orphanages suggests that caregivers do not have specialized training and experience heavy workloads and high-stress levels. Supporting these caregivers to provide responsive and consistent caregiving can improve their well-being along with the psychological and physical development of children who have been orphaned. This research explored the main caregiving-related challenges faced by caregivers in orphanage settings, their professional and personal needs and perceived emotional and behavioural problems manifesting in children under their care. METHOD: This research adopted a qualitative research design with a thematic analysis approach. Semi-structured interviews were conducted with 14 caregivers who were currently caring for four to 12-year-old children in Pakistani orphanages. RESULTS: Five main themes: (1) religiosity, (2) economic relief, (3) caregivers' needs and well-being, (4) caring for children who have been orphaned and (5) need for context specific training, emerged from the data, which included several subthemes. Findings revealed the presence of positive religious views regarding the upbringing and care of children who have been orphaned. Work-provided accommodation was an important economic relief. Caregivers' psychological, physiological and personal lives were affected by job-related stress and demands. Many helpful and unhelpful parenting practices were documented, and challenges such as children's verbal and physical aggression, stealing, non-cooperation and poor social skills were reported. The main professional issues included low salary, high numbers of children in care and lack of context specific professional caregiving training provided. CONCLUSION: This study established the need for a tailored programme that suits the context specific caregiving needs in Pakistani orphanages to support the training and professional growth of caregivers and promote their wellbeing along with positive developmental outcomes in the children under their care.

BACKGROUND: Individuals with a prior diagnosis of chronic lymphocytic leukaemia (CLL) have a higher risk of developing melanoma and exhibit poorer outcomes than patients without CLL. However, there are limited data reporting the clinicopathological features of melanoma diagnosed in patients with CLL. AIMS: To review clinicopathological characteristics of patients with coexisting diagnoses of melanoma and CLL. METHODS: A retrospective review was undertaken for patients with coexisting diagnoses of melanoma and CLL between 2005 and 2015 in 11 centres in the UK and Ireland. RESULTS: Overall, 46 cutaneous melanomas identified in 45 patients were included. In 28 (62.2%) patients, melanoma was diagnosed after an existing diagnosis of CLL. In this group, mean Breslow thickness was 2.7 mm (range 0.2-25 mm). Ten patients (35.7%) developed locoregional recurrence and 8 (28.6%) developed distant metastases. Melanoma-specific mortality was 5 of 28 (17.9%) and all-cause mortality was 13 of 28 (46.4%). In 17 patients, melanoma was diagnosed before CLL. In this group, mean BT was 2.9 mm (range 0.4-14 mm); five patients (29.4%) developed locoregional recurrence and three (17.6%) developed distant metastases. Melanoma-specific mortality was 1 of 17 (5.8%) and all-cause mortality was 5 of 17 (29.4%) in this group. CONCLUSIONS: To our knowledge, this is the first and largest cohort study to report clinicopathological data of coexisting melanoma and CLL in the UK and Ireland. Although the thickness of primary melanoma was not different before or after a CLL diagnosis, melanoma recurrence and melanoma-specific mortality appear to be more common in patients with a prior diagnosis of CLL.

Gastric glomus tumors (GGTs) are rare mesenchymal neoplasms that arise from cells of the glomus body. These occur in the submucosa of the gastric wall and are usually benign in nature. However, it is difficult to predict tumor behavior due to the lack of reliable histological features. Diagnosis can be challenging due to the lack of specific clinical features, and radiologic and endoscopic findings. Computed tomography (CT) scan, esophagogastroduodenoscopy (EGD), and endoscopic ultrasound (EUS) are key diagnostic modalities. However, the final diagnosis depends on the postoperative immunohistochemical and pathological analysis. Most GGTs can be cured by surgical or endoscopic resection. We report a case of GGT in a middle-aged woman who presented with new-onset anemia and was found to have a gastric mass that was later diagnosed as GGT after immunohistochemical staining.

OBJECTIVE: To assess musculoskeletal disorders among male cricketers in an urban centre. METHODS: The cross-sectional study was conducted from October to November 2020 in Lahore, Pakistan, and comprised male cricket players aged 10-25 years playing in four domestic clubs. Data was collected about musculoskeletal disorders experienced during the preceding 12 months using the Extended Nordic Musculoskeletal Questionnaire. Data was analysed using SPSS 22. RESULTS: Of the 89 players with a mean age of 19.24±3.12 years, 35(39.3%) were bowlers, 26(29.2%) were batsmen, 17(19.1%) were all-rounders, and 11(12.4%) were wicketkeepers. The anatomical distribution of disorder was lower-back 68(76.4%), shoulder 40(44.9%), neck 39(43.8%), upper-back 37(41.6%), knees 31(34.8%), ankle/feet 29(32.6%), thighs 27(30.3%), wrist/hands 18(20.2%), and elbows 17(19.1%). There were 22(24.7%) players who had at any time seen a doctor or a physiotherapist, while 24(27%) players had a history of taking sick leave. CONCLUSIONS: The most affected anatomical segments were lower-back, shoulder, knee, ankle and upper-back.

Biloma is an intrahepatic or extrahepatic collection of bile within the abdominal cavity. It can occur spontaneously, or as a result of trauma to the biliary tree. The clinical presentation can be variable and non-specific. Early diagnosis is crucial given the high mortality rate. Diagnostic modalities include abdominal ultrasound, hepatobiliary scintigraphy, computerized tomography (CT), and magnetic resonance imaging (MRI). Treatment options include interventional radiology (IR)-guided drainage, endoscopic drainage, or surgical drainage with a bile leak repair. We report a case of a middle-aged non-cirrhotic patient who presented with abdominal pain and was noted to have extensive portal vein thrombosis. She underwent transjugular intrahepatic portosystemic shunt (TIPS) with thrombectomy and the hospital course was complicated by elevated liver enzymes and found to have intrahepatic biloma requiring IR-guided drainage.

It is rare for patients to have hemorrhagic complications after abdominal paracentesis. Abdominal wall hematomas and hemoperitoneum are the most common hemorrhagic complications of paracentesis. The incidence rate of hemorrhage-related complications is unknown. The risk of hemorrhage-related complications can be elevated in patients with underlying kidney disease and those who are thrombocytopenic or coagulopathic. However, there is no correlation between the degree of thrombocytopenia or coagulopathy and the risk of bleeding. It is important to identify the high-risk patients to prevent these hemorrhage-related complications. In rare instances, secondary complications can develop from hemoperitoneum. We present a case of a cirrhotic patient who underwent a diagnostic paracentesis leading to subsequent intra-abdominal hematoma followed by small bowel obstruction (SBO) due to large abdominal hematoma compressing small bowel loops.

Colonic ischemia is the most common form of gastrointestinal ischemia, which frequently affects the elderly population. The diagnosis and treatment can be challenging since it is prevalent in patients who are debilitated and have multiple comorbidities. However, most cases remain undetected until further complications emerge. Some of these patients will develop prolonged complications like chronic ischemic colitis or stricture requiring surgical intervention. Here we present a case of a colonic stricture secondary to recurrent ischemic colitis in an elderly female patient with multiple medical problems.

The substitution of petrochemical plastics by bio-based and biodegradable plastics are in need of an evaluation for the potential toxic impacts that they can have on marine wildlife. This study aims to assess the toxicological effects of polylactic acid microparticles at two concentrations, 10 and 100 µg/L, during 8 days on the blue mussel, Mytilus edulis. No significant oxidative stress (catalase, glutathione-S-transferase and superoxide dismutase activities), neurotoxicity (acetylcholinesterase), or immunotoxicity (lysosomal membrane stability and acid phosphatase activity) were detectable. The multivariate analysis of metabolomic data allowed us to differentiate the individuals according to the exposure. From the loading plot of OPLS-DA, 48 ions down-regulated in the individuals exposed to microplastics. They were identified based on HRMS data as glycerophospholipids.
