The present study evaluates the effect of nickel oxide nanoparticles on some biochemical parameters and midgut tissues in the ground beetle Blaps polychresta as an indicator organism for nanotoxicity. Serial doses of the NiO-NPs colloid (0.01, 0.02, 0.03, 0.04, 0.05, and 0.06 mg/g) were prepared for injecting into the adult beetles. Insect survival was reported daily for 30 days, and the sublethal dose of 0.02 mg/g NiO-NPs was selected for the tested parameters. After the treatment, nickel was detected in the midgut tissues by X-ray microanalysis. The treated group demonstrated a significant increase in aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities when compared to the untreated group. However, the treated group demonstrated a significant decrease in ascorbate peroxidase (APOX) activity when compared to the untreated group. Histological and ultrastructural changes in the midgut tissues of treated and untreated beetles were also observed. The current findings provide a precedent for describing the physiological and histological changes caused by NiO-NPs in the ground beetle B. polychresta.

The present research aims to evaluate the impact of industrial processes and anthropogenic activities on the beetle Pimelia latreillei inhabiting the polluted site at Zawya Abd El- Qader, Alexandria, Egypt. Beetles were collected from the vicinity of five factories. The genotoxic effects of environmental exposures to industrial heavy metals were monitored using a broad range of assays, including energy-dispersive X ray microanalysis and X-ray diffraction (SEM and EDX)), qRT-PCR gene expression assay, micronuclei formation, and transmission electron microscope (TEM). Energy dispersive X-ray microanalysis for the soil and testicular tissues of beetles collected from the polluted site revealed a higher percentage of heavy metals than the beetles collected from the reference site (Sidi Kirier, Alexandria, Egypt). To analyze/monitor genotoxicity in P. latreillei sampled from the polluted site, the transcription levels of levels of heat shock proteins (Hsps) and accessory gland seminal fluid protein (AcPC01) in testicular tissues were recorded. The incidence of micronuclei (MN) formation in the testicular cells was also observed. Quantitative RT-PCR (RT-qPCR) analysis was carried out to detect the changes in the gene expression of the aforementioned proteins. Genes encoding heat shock proteins (Hsp60, Hsp70, and Hsp90) were significantly overexpressed (> 2-fold) in specimens sampled from the polluted site; however, AcPC01 gene expression was under-expressed (<1.5-folds). The incidence of MN was significantly increased in specimens sampled from the polluted site. Ultrastructure anomalies (nuclear and cytoplasmic disruption) were also observed in the testicular cells of the beetles sampled from the polluted site compared to those sampled from the unpolluted site. Our results, therefore, advocate a need for adequate measures to reduce increasing environmental pollution in the urban-industrial areas.

The expression levels of Esterase Isoenzyme and total soluble protein fractionation were studied in two coleopteran insects Blaps polycresta and Trachyderma hispida to evaluate the possible hazards from ceramic and plastic factories in the Khorshed Region, East of Alexandria, Egypt. Two insect collection sites were selected. The first site was the garden of the Faculty of Science, Moharram Bek, Alexandria University, which is considered a non-polluted site, and Khorshed district, considered as the polluted site. Percentages of heavy metals were estimated using SEM-X-ray microanalysis of soft tissues of both sexes of the two coleopteran insects. Esterase Isoenzymes were found to be overexpressed in B. Polycresta but not T. hispida. Female B. polycresta from the polluted site exhibited overexpression of the second and third loci. Furthermore, the females were found to be more affected than males, which only showed the overexpression of the second loci. T. hispida (females and males) collected from the reference site were found to have increased esterase activity compared with those sampled from the polluted site. The Snake-Skin™ Dialysis tubing technique, used for optimizing the protein extraction method, reflected the highest quantified proteins compared to other, traditional methods. SDS polyacrylamide gel electrophoresis (SDS-PAGE) analysis of the whole-body protein reflected definite variations between T. hispida and B. polycresta in fraction number and activity at the two sites. Varied expression levels for metallothionein (MT) heavy metal resistance proteins for B. polycresta and T. hispida were also detected in the study. Based on these results, we suggest that biochemical biomarkers could infer environmental hazards, B. polycresta and T. hispida are successful biomarkers for heavy metal pollution.

The aim of this study was to analyze the impact of organophosphorus (OP) pollutants on oxidative stress and ultrastructural biomarkers in the midgut of the honeybee Apis mellifera collected from three locations that differ in their extent of spraying load with OP insecticides: a weakly anthropised rural site, Bolin which is considered as a reference site; moderately spraying site, El Kaza; and a strongly anthropised urban site, Tiba with a long history of pesticide use. Results showed that high concentrations of chlorpyrifos, malathion, diazinon, chlorpyrifos-methyl, and pirimiphos-methyl were detected in midgut at locations with extensive pesticide spraying. Reduced glutathione content, superoxide dismutase, catalase, and glutathione peroxidase displayed lowest activities in the heavily sprayed location (Tiba). Lipid peroxidation level in the midgut of honeybees in the sprayed locations was found to be significantly higher compared to the reference values. Meanwhile, various ultrastructural abnormalities were observed in the epithelial cells of midgut of honeybees collected from El Kaza and Tiba, included confluent and disorganized microvilli and destruction of their brush border, the cytoplasm with large vacuoles and alteration of cytoplasmic organelles including the presence of swollen mitochondria with lysis of matrices, disruption of limiting membranes, and disintegration of cristae. The nuclei with indented nuclear envelope and disorganized chromatin were observed. These investigated biomarkers indicated that the surveyed honeybees are being under stressful environmental conditions. So, we suggest using those biomarkers in the assessment of environmental quality using honeybees in future monitoring of ecotoxicological studies.
