RESUMO

Commercial system of multimicrotests for biochemical identification of enterobacteria MMT E2 and routine tube tests were compared in a controlled epidemiologic experiment with 205 enterobacterial strains. The system has been developed at the Allergen Research and Production Amalgamation. System MMT E2 fairly well met the requirements to diagnostic preparations of this kind; its results were compatible and well reproducible. This system in complex with commercial MMT E1 system may be used for species identification of enterobacteria. Practical application of both the systems will considerably simplify the process of identification of these microorganisms.

RESUMO

Comparative evaluation of a system of multimicrotests for biochemical identification of enterobacteria (MMT E1) developed at the ALLERGEN Research and Production Amalgamation and of tube tests for the same purpose in a controlled epidemiologic experiment has shown that the suggested system meets the requirements to such diagnostic preparations by all the major qualitative parameters (compatibility and reproducibility of the test results). Practical use of MMT E1 system will simplify and facilitate the process of enterobacteria identification.

RESUMO

The first Soviet kits for the serological identification of streptococci, groups A, B, and C, on the basis of the coagglutination test were developed. Each kit was intended for 35-40 determinations. The optimum concentration of streptococci during their identification by means of the reagents making up the kit was about 1.6 x 10(9) cells/ml. The specificity of the reagents in comparison with the results of the identification of streptococci by reference methods was 97.3 +/- 0.9%. The reagents making up the kits can be presumably used for solving a number of practical problems in the epidemiological surveillance of streptococcal infection.

When grown on meat-peptone agar with heated blood, different Haemophilus species (H. influenzae, H. parahaemolyticus, H. parasuis, H. pleuropneumoniae), including different H. influenzae serovars (a, b, c, d, e, f), and Pasteurella multocida have identical fatty acid composition, characterized by the prevalence of fatty acids with 16 carbon atoms, constituting about 70% and more of the total number of fatty acids, and a low level of fatty acids with 18 carbon atoms. P. multocida strains cultivated on meat-peptone agar with unheated blood have a greatly increased content of fatty acids with 18 carbon atoms, while the content of fatty acids with 16 carbon atoms is much lower. The identity of fatty acid composition under similar cultivation conditions, together with their similarity in other phenetic signs, is indicative of close phylogenic relationship between bacteria belonging to the genus Haemophilus and P. multocida.

RESUMO

Biochemical plates for the differentiation between Enterobacteriaceae have been employed for studies of the biochemical activities of hemophilic microorganisms. Thirty-seven hemophilic strains have been examined with the use of this test system. A wide set of diagnostic tests used with these plates permits not only biochemical identification of the hemophils but their biotyping as well.

RESUMO

Cultures isolated from human and animal blood have been examined in tests with 3% KOH solution; their sensitivities to various concentrations of sulfanol have been tested in order to specify the tinctorial characteristics of these cultures; antibiotic sensitivity of this group of microorganisms has been studied with the use of discs. The findings evidence that gram-variable nonspore-carrying aerobic bacilli isolated from human and animal blood should be referred to gram-positive microorganisms; their antibiotic sensitivity has been detected. The tests employed in the study may be rationally used for the identification of polychromic bacteria isolated from the blood.

The study of 26 B. bronchiseptica strains with typical morphological and biochemical properties resulted in the detection of 8 strains having the main specific agglutinogens of 3 Bordetella species (serovars) in different combinations. The presence of the agglutinogens was confirmed in the agglutination test and the agglutinin adsorption test with the use of monospecific antisera to the main agglutinogens. The comparison of natural B. bronchiseptica serovars and artificial convertants (resulting from the conversion of B. parapertussis by B. pertussis phages) revealed their identical biochemical activity, their capacity for causing necrosis when injected intradermally into rabbits and for the formation of two types of colonies, differing in size and serological activity. In contrast to B. parapertussis convertants, B. bronchiseptica serovars had no lysogenic properties and were sensitive to B. pertussis and B. bronchiseptica phages.

Bacteria of the species B. parapertussis and B. bronchiseptica have proved to be identical in their fatty-acid composition with a high level (35.7-39%) of methylene-hexadecanoic acid, found to be absent in B. pertussis in experimental conditions. At the same time the total content of methylene-hexadecanoic acid and its biosynthetic precursor, hexadecenoic acid, in the first two Bordetella species is similar to the content of hexadecenoic acid in B. pertussis, which, along with the presence of common characteristics in the sign under consideration (the low level of C18:1), indicates the close relationship of these three Bordetella species. Bacteria of the species H. influenzae, H. parainfluenzae, H. aegyptius, H. aphrophilus have similar fatty-acid composition with the prevalence of hexadecanoic and hexadecenoic acids and the low level of fatty acids with 18 carbon atoms. The data on fatty-acid composition may suggest the presence of philogenetic links between the genera Bordetella and Haemophilus.

RESUMO

The saccharolytic capacity in respect to 14 carbohydrates, the lipolytic activity, the presence of urease, catalase, galactosidase, the formation of hydrogen sulphide and indol and also serological properties with the species agglutinating sera were studied in the representatives of the genus of hemoglobinophilic microbes: H. influenzae, H. parainfluenzae, H. aegiptius, H. haemolyticus, H. aphrophilus. There were revealed differences in the individual representatives of the genus by the enzymatic activity and serological properties. Thus, representatives of H. influenzae possessed urease activity, but all of them lacked galactosidase. H. aegiptius possessed urease and galactosidase, and H. parainfluenzae, H. haemolyticus and H. aphrophilus--galactosidase of high activity, but no urease. Representatives of each of the species were agglutinated by homologous sera only.

RESUMO

Strains (29) of Corynebacterium genus were studied with the use of the method of numerical taxonomy. 48 characteristics (including morphological, tinctorial, cultural and biochemical ones) were analyzed. All the strains tested proved to belong to the same taxon. The present method permitted to reveal various biotypes within the group studied.

Type-specific agglutinating sera to capsular antigenas of H. influenzae were recovered (from 1:3200 to 1:25600). With the given immunization scheme there formed mostly IgG immunoglobulins, and only insignificant amount of IgM (separation of G-200 Sephadex gel). IgG were found to be responsible for the type-specificity of the sera to bacteria, whereas the cross reactions within the H. influenzae genus were connected connected with the IgM. Use of the sera heated at various temperature regimens offered a possibility of obtaining highly specific agglutinating sera with the aid of which express serodiagnosis of the isolated H. influenzae strains was possible.
