RESUMO

The functional components of leaves and stalks from 14 sweet potato cultivars were investigated by determining lutein, ß-carotene, chlorophyll, tannin and phenolic acid contents. It was found that the contents of the functional components in different cultivars differ significantly (p<0.05). Lutein, ß-carotene and total chlorophyll contents were high in leaves and ranged from 19.01-28.85, 35.21-52.01 and 440.9-712.2 mg/100 g, respectively. The tannin and total phenolic acid contents of sweet potato leaves ranged from 2,280-4,460 and 2,640.2-4,200.9 mg/100 g, respectively. Significant correlations have been observed among cultivar, lutein, ß-carotene, chlorophyll, and other antioxidants. The leaves of Healthymi cultivar contained the highest level of lutein, ß-carotene and total chlorophyll, and Geonpungmi cultivar contained the highest level of the other antioxidant, among the all cultivars examined. Sweet potato leaves and stalks contain abundant functional components that make them potentially useful as fresh vegetables or processed foods.

This study was performed to evaluate and compare the antioxidant substance content and antioxidant activities of white (Superior) and colored (Hongyoung, Jayoung, Jasim, Seohong, and Jaseo) potatoes. The potatoes were extracted with 80% ethanol and were evaluated for the total polyphenol, flavonoid, and anthocyanin contents and for 1,1-diphenyl- 2-picrylhydrazyl (DPPH)/2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity, reducing power, and ferrous metal ion chelating effect. The total polyphenol, flavonoid, and anthocyanin contents of Hongyoung and Jayoung were higher than white and other colored potatoes. All colored potato extracts, except for Jaseo and Seohong, showed higher ABTS radical scavenging activities than the general white potato extract. Hongyoung and Jayoung had the highest ABTS and DPPH radical scavenging activities. Optical density values for the reducing power of Jayoung and Jaseo at concentration of 2 mg/mL were 0.148 and 0.090, respectively. All colored potato extracts had lower ferrous metal ion chelating effect than the white potato. A significant (P<0.05) positive correlation was observed between total polyphenol content and total flavonoid content (r=0.919), anthocyanin content (r=0.992), and ABTS radical scavenging activity (r=0.897). Based on these results, this research may be useful in developing the Hongyoung and Jayoung cultivars with high antioxidant activities.

We performed de novo transcriptome sequencing for Panax ginseng and Panax quinquefolius accessions using the 454 GS FLX Titanium System and discovered annotation-based genome-wide single-nucleotide polymorphism (SNPs) using next-generation ginseng transcriptome data without reference genome sequence. The comprehensive transcriptome characterization with the mature roots of four ginseng accessions generated 297,170 reads for 'Cheonryang' cultivar, 305,673 reads for 'Yunpoong' cultivar, 311,861 reads for the G03080 breeding line, and 308,313 reads for P. quinquefolius. In transcriptome assembly, the lengths of the sample read were 156.42 Mb for 'Cheonryang', 161.95 Mb for 'Yunpoong', 165.07 Mb for G03080 breeding line, and 166.48 Mb for P. quinquefolius. A total of 97 primer pairs were designed with the homozygous SNP presented in all four accessions. SNP genotyping using high-resolution melting (HRM) analysis was performed to validate the putative SNP markers of 97 primer pairs. Out of the 73 primer pairs, 73 primer pairs amplified the target sequence and 34 primer pairs showed polymorphic melting curves in samples from 11 P. ginseng cultivars and one P. quinquefolius accession. Among the 34 polymorphic HRM-SNP primers, four primers were useful to distinguish ginseng cultivars. In the present study, we demonstrated that de novo transcriptome assembly and mapping analyses are useful in providing four HRM-SNP primer pairs that reliably show a high degree of polymorphism among ginseng cultivars.

Flooding of fields due to heavy and/or continuous rainfall influences soybean production. To identify soybean varieties with flooding tolerance at the seedling emergence stage, 128 soybean varieties were evaluated using a flooding tolerance index, which is based on plant survival rates, the lack of apparent damage and lateral root development, and post-flooding radicle elongation rate. The soybean varieties were ranked according to their flooding tolerance index, and it was found that the tolerance levels of soybean varieties exhibit a continuum of differences between varieties. Subsequently, tolerant, moderately tolerant and sensitive varieties were selected and subjected to comparative proteomic analysis to clarify the tolerance mechanism. Proteomic analysis of the radicles, combined with correlation analysis, showed that the ratios of RNA binding/processing related proteins and flooding stress indicator proteins were significantly correlated with flooding tolerance index. The RNA binding/processing related proteins were positively correlated in untreated soybeans, whereas flooding stress indicator proteins were negatively correlated in flooded soybeans. These results suggest that flooding tolerance is regulated by mechanisms through multiple factors and is associated with abundance levels of the identified proteins.

We experimentally analyze the stimulated Raman scattering characteristics of a high-birefringence fiber (HBF), which presents an extraordinary level of spectral broadening incurred by the strong nonlinear interaction between the pump and Stokes pulses via the polarization-mode dispersion and group-velocity dispersion of the fiber. We also investigate the impact of the inter-pulse time-delay on the additional spectra broadening when dual-wavelength pump pulses are used. Exploiting these unique SRS properties of the HBF, we develop a novel Raman continuum source based on an all-fiber dual-wavelength master-oscillator power amplifier that can generate a dip-free spectrum in the 1200-1400-nm spectral range. We finally obtain a broadband continuum having an average power of ~840 mW and a 3-dB bandwidth of ~240 nm centered at 1200-1400 nm, which also represents a good spectral flatness and conversion efficiency. This type of source is very useful and important for optical coherence tomography applications, for example.

Despite the intensive soybean [Glycine max (L.) Merrill] genome studies, the high chromosome number (20) of the soybean plant relative to many other major crops has hindered the development of a high-resolution genomewide genetic map derived from a single population. Here, we report such a map, which was constructed in an F15 population derived from a cross between G. max and G. soja lines using indel polymorphisms detected via a G. soja genome resequencing. By targeting novel indel markers to marker-poor regions, all marker intervals were reduced to under 6 cM on a genome scale. Comparison of the Williams 82 soybean reference genome sequence and our genetic map indicated that marker orders of 26 regions were discrepant with each other. In addition, our comparison showed seven misplaced and two absent markers in the current Williams 82 assembly and six markers placed on the scaffolds that were not incorporated into the pseudomolecules. Then, we showed that, by determining the missing sequences located at the presumed beginning points of the five major discordant segments, these observed discordant regions are mostly errors in the Williams 82 assembly. Distributions of the recombination rates along the chromosomes were similar to those of other organisms. Genotyping of indel markers and genome resequencing of the two parental lines suggested that some marker-poor chromosomal regions may represent introgression regions, which appear to be prevalent in soybean. Given the even and dense distribution of markers, our genetic map can serve as a bridge between genomics research and breeding programs.

Narrow leaflet soybean (Glycine max) varieties tend to have more seeds per pod than broad leaflet varieties. Narrow leaflet in soybean is conferred by a single recessive gene, ln. Here, we show that the transition from broad (Ln) to narrow leaflet (ln) is associated with an amino acid substitution in the EAR motif encoded by a gene (designated Gm-JAGGED1) homologous to Arabidopsis JAGGED (JAG) that regulates lateral organ development and the variant exerts a pleiotropic effect on fruit patterning. The genomic region that regulates both the traits was mapped to a 12.6-kb region containing only one gene, Gm-JAG1. Introducing the Gm-JAG1 allele into a loss-of-function Arabidopsis jagged mutant partially restored the wild-type JAG phenotypes, including leaf shape, flower opening, and fruit shape, but the Gm-jag1 (ln) and EAR-deleted Gm-JAG1 alleles in the jagged mutant did not result in an apparent phenotypic change. These observations indicate that despite some degree of functional change of Gm-JAG1 due to the divergence from Arabidopsis JAG, Gm-JAG1 complemented the functions of JAG in Arabidopsis thaliana. However, the Gm-JAG1 homoeolog, Gm-JAG2, appears to be sub- or neofunctionalized, as revealed by the differential expression of the two genes in multiple plant tissues, a complementation test, and an allelic analysis at both loci.

This study investigated the functional compounds, including isoflavones, phenolics, soyasapogenols, and tocopherols, that were detected in 204 soybean [ Glycine max (L.) Merrill] germplasms. The soybean samples were divided into three groups according to origin: America, China, and Korea. The soybean samples were also classified into three groups on the basis of 100-seed weight: small (<13 g), medium (13-24 g), and large (>24 g). Among the soybean germplasms, CSRV121 (Bosukkong) had the highest level of isoflavone content (4778.1 µg g(-1)), whereas CS01316 had the lowest isoflavone content (682.4 µg g(-1)). Of the soybeans from the three different countries of origin, those from Korea showed the highest average concentration of total isoflavones (2252.6 µg g(-1)). The small seeds had the highest average total isoflavone concentration (2520.0 µg g(-1)) of the three different seed sizes. Among the 204 soybean germplasms, CS01405 had the highest content of total phenolics (5219.6 µg g(-1)), and CSRV017 (Hwangkeumkong) had the lowest phenolic content (654.6 µg g(-1)). The mean concentrations of total phenolic compounds were 2729.1 µg g(-1) in American soybean seeds, 1680.4 µg g(-1) in Chinese soybean seeds, and 1977.6 µg g(-1) in Korean soybean seeds. Of the soybean seeds from the three different countries of origin, American soybean seeds had the highest average concentration of total phenolic compounds, and Korean varieties showed the second highest value. Small soybean seeds had the highest average content of total phenolic compounds (2241.7 µg g(-1)), whereas medium-sized (1926.8 µg g(-1)) and large (1949.9 µg g(-1)) soybeans had lower concentrations of phenolic compounds. In whole soybean germplasms, the level of total soyasapogenols was higher in CS01173 (1802.3 µg g(-1)) and CS01346 (1736.8 µg g(-1)) than in the other types of soybeans. The mean concentrations of total soyasapogenol were 1234.0 µg g(-1) in American, 1294.5 µg g(-1) in Chinese, and 1241.5 µg g(-1) in Korean soybean varieties. Chinese soybean varieties showed the highest mean concentration of total soyasapogenol, and Korean soybean seeds showed the second highest level. The medium-seed group had the highest soyasapogenol content (1269.3 µg g(-1)) of the seeds that were grouped by size. A larger amount of soyasapogenol B than soyasapogenol A was detected. In whole soybeans, CS01202 showed the highest level of total tocopherols (330.5 µg g(-1)), whereas CSRV056 (Pungsannamulkong) had the lowest content (153.3 µg g(-1)). Chinese soybeans had the highest average concentration of total tocopherols (255.1 µg g(-1)). By comparison, the medium-sized Chinese soybean group had the highest (256.1 µg g(-1)) average total tocopherol content.

In order to develop a novel system for the discrimination of five ginseng cultivars (Panax ginseng Meyer), single nucleotide polymorphism (SNP) genotyping assays with real-time polymerase chain reaction were conducted. Nucleotide substitution in gDNA library clones of P. ginseng cv. Yunpoong was targeted for the SNP genotyping assay. From these SNP sites, a set of modified SNP specific fluorescence probes (PGP74, PGP110, and PGP130) and novel primer sets have been developed to distinguish among five ginseng cultivars. The combination of the SNP type of the five cultivars, Chungpoong, Yunpoong, Gopoong, Kumpoong, and Sunpoong, was identified as 'ATA', 'GCC', 'GTA', 'GCA', and 'ACC', respectively. This study represents the first report of the identification of ginseng cultivars by fluorescence probes. An SNP genotyping assay using fluorescence probes could prove useful for the identification of ginseng cultivars and ginseng seed management systems and guarantee the purity of ginseng seed.

Narrow leaflet cultivars tend to have more seeds per pod than broad leaflet cultivars in soybean [Glycine max (L.) Merr.], which suggests that the leaflet-shape trait locus is tightly linked to or cosegregates with the trait locus controlling the number of seeds per pod (NSPP). Here, we attempted to further elucidate the relationship between leaflet shape and NSPP. A BC(3)F(2) population from a cross between the 'Sowon' (narrow leaflets and high NSPP) and 'V94-5152' (broad leaflets and low NSPP) variants was used. The results of the molecular genetic analyses indicated that, although the NSPP characteristic, in particular, the occurrence of 4-seeded pods, is governed by additional modifying genes that are likely present in Sowon, the two traits cosegregate in the BC(3)F(2) population. The mapping results generated using public markers demonstrated that the narrow leaflet-determining gene in Sowon is an allele of the previously highly studied ln gene on chromosome 20. A high-resolution map delimited the genomic region controlling both the leaflet shape and NSPP traits to a sequence length of 66 kb, corresponding to 0.7 cM. Among the three genes annotated in this 66 kb region, Glyma20g25000.1 appeared to be a good candidate for the Ln-encoding gene, owing to its 47.8% homology with the protein encoding for the JAGGED gene that regulates lateral organ development in Arabidopsis. Taken together, our results suggested that phenotypic variations for narrow leaflet and NSPP are predominantly from the pleiotropic effects of the ln gene. Thus, our results should provide a molecular framework for soybean breeding programs with the objective of improving soybean yield.

The Rsv4 gene confers resistance to all the known strain groups of soybean mosaic virus in soybean (Glycine max (L.) Merr.). To construct a fine genetic map near Rsv4 in soybean, we employed a comparative genomics approach that used genome sequence information of the model legume Lotus japonicus. Sequences of the soybean expressed sequence tags (ESTs) AI856415 and BF070293 mapping to one side of the Rsv4 gene showed high similarity with gene sequences of the transformation-competent artificial chromosome (TAC) clone LjT32P24 of Lotus. LjT32P24 is tightly linked to another sequenced TAC clone, LjT26I01, in Lotus. A new marker, AW307114A, developed from soybean EST AW307114, which is homologous to a Lotus gene within LjT26I01, was mapped to the other side of the Rsv4 gene. The identification of the microsyntenic relationship facilitated the development of additional 2 EST markers between BF070293-S and AW307114A bracketing the Rsv4 gene. Several other markers developed in this study were mapped to putative homoeologous or duplicated chromosomal regions in soybean. Alignment between the soybean maps indicated that Rsv4 is located near a local chromosomal rearrangement. This targeted comparative mapping serves to provide a foundation for marker-assisted selection and cloning of the Rsv4 gene.

The water content of stratum corneum (SC) has been reported to vary depending on the anatomical sites. Each anatomical site respectively have their own proper water content of SC which reflects the status of skin health. Also, this anatomical region can be divided into relatively wet and dry area according to the degree of the water content of SC. In this study, we tried to investigate time series trend of the water content. Skin hydration was measured on the different anatomical sites for the first four seconds using single frequency-susceptance measuring method. It is observed that the slope of the graph for the water content during the first four seconds is different depending on the anatomical sites and can be divided into two groups by the degree of the slope. These grouped areas are well aligned with traditional wet and dry area. It is presumed that the difference of the gradient could be caused by the thickness of SC. In this study, the different gradient of time series trend of water content of SC on different anatomical sites was found, and it could be conveniently used to distinguish the wet and dry area of the skin.

A compact and easy-to-use home health monitor was developed. A palm-size health monitor contained a finger probe as sensor unit. In the finger probe, light from a light emitting diode (LED) array was illuminated on a finger nail bed, and transmitted light was measured to obtain photoplethysmography (PPG) signals. Hematocrit, pulse, respiration rate, and saturated oxygen in arterial blood (SpO(2)) were measured simultaneously from PPGs using five different wavelengths: 569, 660, 805, 904, and 975 nm. To predict hematocrit, a dedicated algorithm was used based on scattering theory of red blood cells using these wavelengths. Preliminary clinical tests showed that the achieved percent errors were +/- 8.2% for hematocrit when tested with 549 persons (N = 549). Digital filtering techniques were used to extract respiratory information from a single PPG signal. SpO(2) was predicted on the basis of the ratio of the wavelengths 660 nm and 940 nm. The accuracies were within clinically acceptable errors. In addition, the compact home health monitor included a blood pressure monitoring unit. For convenient and simultaneous measurement with the other previously mentioned signals, blood pressure was measured on a finger. An air cuff was installed on the same finger where PPGs were measured. Achieved mean differences were +/- 3.8 mmHg for systole and +/- 5.1 mmHg for diastole. One can use the palm-size monitor simply by inserting a finger into the home health monitor that is suitable for telemedicine.
