RESUMO

Pseudomonas stutzeri KC can rapidly degrade carbon tetrachloride (CCl4) to CO2 by a fortuitous reaction with pyridine-2,6-bis(thiocarboxylic acid), a metal chelator encoded by pdt genes. These genes were first identified after a spontaneous mutant, strain CTN1, lost the ability to degrade CCl4. Here we report the complete genome of strain KC and show that these pdt genes are located on an integrative and conjugative element (ICE), designated ICEPsstKC. Comparative genome analyses revealed homologues of pdt genes in genomes of members of other gammaproteobacterial orders. Discrepancies between the tree topologies of the deduced pdt gene products and the host phylogeny based on 16S rRNA provided evidence for horizontal gene transfer (HGT) in several sequenced strains of these orders. In addition to ICEPsstKC, HGT may be have been facilitated by other mobile genetic elements, as indicated by the location of the pdt gene cluster adjacent to fragments of other ICEs and prophages in several genome assemblies. We could here show that the majority of cells from the culture collection DSMZ had lost the ICE. The presence of the pdt gene cluster on mobile genetic elements has important implications for the bioremediation of CCl4 for bioremediation of CCl4 and needs consideration when selecting suitable strains.

Wastewater-based epidemiology (WBE) expanded rapidly in response to the COVID-19 pandemic. As the public health emergency has ended, researchers and practitioners are looking to shift the focus of existing wastewater surveillance programs to other targets, including bacteria. Bacterial targets may pose some unique challenges for WBE applications. To explore the current state of the field, the National Science Foundation-funded Research Coordination Network (RCN) on Wastewater Based Epidemiology for SARS-CoV-2 and Emerging Public Health Threats held a workshop in April 2023 to discuss the challenges and needs for wastewater bacterial surveillance. The targets and methods used in existing programs were diverse, with twelve different targets and nine different methods listed. Discussions during the workshop highlighted the challenges in adapting existing programs and identified research gaps in four key areas: choosing new targets, relating bacterial wastewater data to human disease incidence and prevalence, developing methods, and normalizing results. To help with these challenges and research gaps, the authors identified steps the larger community can take to improve bacteria wastewater surveillance. This includes developing data reporting standards and method optimization and validation for bacterial programs. Additionally, more work is needed to understand shedding patterns for potential bacterial targets to better relate wastewater data to human infections. Wastewater surveillance for bacteria can help provide insight into the underlying prevalence in communities, but much work is needed to establish these methods.IMPORTANCEWastewater surveillance was a useful tool to elucidate the burden and spread of SARS-CoV-2 during the pandemic. Public health officials and researchers are interested in expanding these surveillance programs to include bacterial targets, but many questions remain. The NSF-funded Research Coordination Network for Wastewater Surveillance of SARS-CoV-2 and Emerging Public Health Threats held a workshop to identify barriers and research gaps to implementing bacterial wastewater surveillance programs.

Background: Wastewater measurements of SARS-CoV-2 RNA have been extensively used to supplement clinical data on COVID-19. Most examples in the literature that describe wastewater monitoring for SARS-CoV-2 RNA use samples from wastewater treatment plants and individual buildings that serve as the primary residence of community members. However, wastewater surveillance can be an attractive supplement to clinical testing in K-12 schools where individuals only spend a portion of their time but interact with others in close proximity, increasing risk of potential transmission of disease. Methods: Wastewater samples were collected from two K-12 schools in California and divided into solid and liquid fractions to be processed for detection of SARS-CoV-2. The resulting detection rate in each wastewater fraction was compared to each other and the detection rate in pooled clinical specimens. Results: Most wastewater samples were positive for SARS-CoV-2 RNA when clinical testing was positive (75% for solid samples and 100% for liquid samples). Wastewater samples continued to test positive for SARS-CoV-2 RNA when clinical testing was negative or in absence of clinical testing (83% for both solid and liquid samples), indicating presence of infected individuals in the schools. Wastewater solids had a higher concentration of SARS-CoV-2 than wastewater liquids on an equivalent mass basis by three orders of magnitude.

Recovery research has amply documented the benefits of employee recovery for their well-being and performance. However, most research to date has mainly focused on intraindividual benefits, so that we know relatively little about how focal employees' recovery activities might affect others at work. Bridging the gaps in the recovery and leadership literatures, this study focuses on leader-follower dyads and examines the interpersonal effects of leaders' recovery activities on followers' daily performance quality. In particular, our proposed model-which is informed by the conservation of resource and emotional contagion theories-explains the spillover-crossover process by which leaders' pleasurable recovery activity at home influences their positive affect (PA) displays at work, their followers' PA, and ultimately followers' task performance and creative behavior. We conducted two experience sampling method studies that relied on leader-member dyads of full-time working adults who responded to multiple online surveys for five workdays (n1 = 511 daily observations) and 10 workdays (n2 = 576 daily observations). In both studies, multilevel path analyses showed that leaders' previous-night pleasurable recovery activity leads to increased morning PA displays at work (observed by followers), which in turn crosses over to followers' midday PA and positive work outcomes. In particular, the affective crossover from leaders to followers was stronger when followers were in an inadequate morning recovery state and, therefore, more in need of resource replenishment. Rare cells, such as circulating tumor cells or circulating fetal cells, provide important information for the diagnosis and prognosis of cancer and prenatal diagnosis. Since undercounting only a few cells can lead to significant misdiagnosis and incorrect decisions in subsequent treatment, it is crucial to minimize cell loss, particularly for rare cells. Moreover, the morphological and genetic information on cells should be preserved as intact as possible for downstream analysis. The conventional immunocytochemistry (ICC), however, fails to meet these requirements, causing unexpected cell loss and deformation of the cell organelles which may mislead the classification of benign and malignant cells. In this study, a novel ICC technique for preparing lossless cellular specimens was developed to improve the diagnostic accuracy of rare cell analysis and analyze intact cellular morphology. To this end, a robust and reproducible porous hydrogel pellicle was developed. This hydrogel encapsulates cells to minimize cell loss from the repeated exchange of reagents and prevent cell deformation. The soft hydrogel pellicle allows stable and intact cell picking for further downstream analysis, which is difficult with conventional ICC methods that permanently immobilize cells. The lossless ICC platform will pave the way for robust and precise rare cell analysis toward clinical practice.

Wastewater-based epidemiology has gained attention throughout the world for detection of SARS-CoV-2 RNA in wastewater to supplement clinical testing. Raw wastewater consists of small particles, or solids, suspended in liquid. Methods have been developed to measure SARS-CoV-2 RNA in the liquid and the solid fraction of wastewater, with some studies reporting higher concentrations in the solid fraction. To investigate this relationship further, six laboratories collaborated to conduct a study across five publicly owned treatment works (POTWs) where both primary settled solids obtained from primary clarifiers and raw wastewater influent samples were collected and quantified for SARS-CoV-2 RNA. Settled solids and influent samples were processed by participating laboratories using their respective methods and retrospectively paired based on date of collection. SARS-CoV-2 RNA concentrations, on a mass equivalent basis, were higher in settled solids than in influent by approximately three orders of magnitude. Concentrations in matched settled solids and influent were positively and significantly correlated at all five POTWs. RNA concentrations in both settled solids and influent were correlated to COVID-19 incidence rates in the sewersheds and thus representative of disease occurrence; the settled solids methods appeared to produce a comparable relationship between SARS-CoV-2 RNA concentration measurements and incidence rates across all POTWs. Settled solids and influent methods showed comparable sensitivity, N gene detection frequency, and calculated empirical incidence rate lower limits. Analysis of settled solids for SARS-CoV-2 RNA has the advantage of using less sample volume to achieve similar sensitivity to influent methods.

Monitoring wastewater for SARS-CoV-2 from populations smaller than those served by wastewater treatment plants may help identify small spatial areas (subsewersheds) where COVID-19 infections are present. We sampled wastewater from three nested locations with different sized populations within the same sewer network at a university campus and quantified SARS-CoV-2 RNA using reverse transcriptase droplet digital polymerase chain reaction (PCR). SARS-CoV-2 RNA concentrations and/or concentrations normalized by PMMoV were positively associated with laboratory-confirmed COVID-19 cases for both the sewershed level and the subsewershed level. We also used an antigen-based assay to detect the nucleocapsid (N) antigen from SARS-CoV-2 in wastewater samples at the sewershed level. The N antigen was regularly detected at the sewershed level, but the results were not associated with either laboratory-confirmed COVID-19 cases or SARS-CoV-2 RNA concentrations. The results of this study indicate that wastewater monitoring based on quantification of SARS-CoV-2 RNA using PCR-based methods is associated with COVID-19 cases at multiple geographic scales within the subsewershed level and can serve to aid the public health response.

Grounded in self-regulatory resources and conservation of resources theories, the current research examines poor self-regulatory capacity as a precursor to microbreaks and their possible outcomes at work. Full-time employees completed multiple online surveys for 10 (n1 = 779 daily observations) and 5 workdays (n2 = 1,024 daily observations). In Study 1, multilevel path analysis results showed that on days when employees had poorer recovery at home (i.e., poor sleep quality), they experienced higher fatigue in the next morning (low self-regulatory capacity) and thus took microbreaks more frequently at work. In turn, their engagement in microbreaks was related to higher work engagement during the day and lower end-of-work fatigue. Furthermore, perceived health climate was found to moderate the path from morning fatigue to microbreaks. In Study 2, we replicated and confirmed the serial mediation paths found in Study 1 (poor sleep quality → morning fatigue → microbreaks → work engagement and end-of-work fatigue). Building on Study 1, Study 2 also identified microbreak autonomy as a mechanism by which perceived health climate moderates the path between morning fatigue and microbreaks (i.e., mediated moderation effect). Exploratory analyses discovered intriguing patterns of socialization microbreaks versus other microbreaks, providing further implications for the theoretical perspective. Overall, the findings support the theoretical resource perspective of microbreaks as an effective energy management strategy while at work. With abundant health-related information, the modern workforce is advised to engage in health-promoting behaviors such as good sleep, physical activities, and a healthy diet to stay productive at work. However, no study has provided a theoretical framework or empirical evidence on the association between employees' unhealthy eating behavior and the quality of their performance. Drawing from the stress and coping literature, the current study proposes a moderated mediation model to investigate the day-specific roles of (un)healthy lifestyle in regard to personal well-being and performance at work. We used daily diary data collected from 97 full-time employees and employed an experience sampling method (ESM) to examine this within-person phenomenon for 2 weeks. Our multilevel path analysis reveals that employees' unhealthy eating behavior in the evening led to emotional strain (e.g., guilt) as well as physical strain (e.g., stomachache, diarrhea) on the next morning; the emotional and physical strains experienced in the morning served as key mediators resulting in decreased quality of performance (i.e., less helping and more withdrawal behaviors) in the afternoon. Furthermore, emotional stability was found to moderate the relationship between unhealthy eating behavior and morning strains, such that employees with higher emotional stability tended to experience less negative emotions and fewer physical symptoms. The theoretical and practical implications of these findings are discussed, along with suggestions for future studies on health-related behaviors. This study offers an alternative approach to address on-going concerns about burnout of healthcare employees. Departing from the existing job-demand based approach proposing that healthcare employees' burnout can be resolved by reducing demands, we theorize that patient-centered prosocial behavior, even if it often increases job demands, could serve as potential job resources that fuel positive energy to vitalize nurses at work. We further theorize that this possibility could be more pronounced among a group of nurses with a strong sense of ethical membership regarding their hospital (i.e., moral identification). To test our hypotheses, we used a sample of 202 nurses from 104 South Korean hospitals. We found that, even controlling for workloads as an indicator of job demand, nurses who engage in patient-centered prosocial behavior (i.e., relational caring) are likely to feel vitalized, and this pattern is more salient among a group of nurses with high moral identification. Results indicate that prosocial behavior could be an alternative job resource that helps nurses flourish at work.

Published and unpublished reports show that SARS-CoV-2 RNA in publicly owned treatment work (POTW) wastewater influent and solids is associated with new COVID-19 cases or incidence in associated sewersheds, but methods for comparing data collected from diverse POTWs to infer information about the relative incidence of laboratory-confirmed COVID-19 cases, and scaling to allow such comparisons, have not been previously established. Here, we show that SARS-CoV-2 N1 and N2 concentrations in solids normalized by concentrations of PMMoV RNA in solids can be used to compare incidence of laboratory confirmed new COVID-19 cases across POTWs. Using data collected at seven POTWs along the United States West Coast, Midwest, and East Coast serving ∼3% of the U.S. population (9 million people), we show that a 1 log change in N gene/PMMoV is associated with a 0.24 (range 0.19 to 0.29) log10 change in incidence of laboratory confirmed COVID-19. Scaling of N1 and N2 by PMMoV is consistent, conceptually, with a mass balance model relating SARS-CoV-2 RNA to the number of infected individuals shedding virus in their stool. This information should support the application of wastewater-based epidemiology to inform the response to the COVID-19 pandemic and potentially future viral pandemics.

Wastewater-based epidemiology may be useful for informing public health response to viral diseases like COVID-19 caused by SARS-CoV-2. We quantified SARS-CoV-2 RNA in wastewater influent and primary settled solids in two wastewater treatment plants to inform the preanalytical and analytical approaches and to assess whether influent or solids harbored more viral targets. The primary settled solids samples resulted in higher SARS-CoV-2 detection frequencies than the corresponding influent samples. Likewise, SARS-CoV-2 RNA was more readily detected in solids using one-step digital droplet (dd)RT-PCR than with two-step RT-QPCR and two-step ddRT-PCR, likely owing to reduced inhibition with the one-step ddRT-PCR assay. We subsequently analyzed a longitudinal time series of 89 settled solids samples from a single plant for SARS-CoV-2 RNA as well as coronavirus recovery (bovine coronavirus) and fecal strength (pepper mild mottle virus) controls. SARS-CoV-2 RNA targets N1 and N2 concentrations correlated positively and significantly with COVID-19 clinically confirmed case counts in the sewershed. Together, the results demonstrate that measuring SARS-CoV-2 RNA concentrations in settled solids may be a more sensitive approach than measuring SARS-CoV-2 in influent.

Growing concerns about intensive information and communication technology (ICT) use led to abundant research on its debilitating effects on employees' abilities to meet family demands. Drawing on the stressor-strain model, we conducted a daily diary study to investigate how different types of daily ICT demands experienced during work hours and after work influence work-family conflict (WFC) in the evening. We collected data from 98 full-time employees (793 day-level observations) for 10 consecutive workdays to understand employees' work-nonwork interface experiences, namely, negative spillover and role conflict. First, we examined a multilevel mediation model to test the negative spillover effect of on-the-job ICT demands on WFC in the evening via negative affect (NA) at the end of the workday. Second, we investigated the effects of off-the-job ICT demands on WFC to provide evidence of role conflict in the nonwork domain. Further, we tested the protective role of boundary control in these phenomena. The multilevel analysis results revealed that different types of ICT demands experienced at work have idiosyncratic impacts on WFC. Also, while extended availability after work hours yields greater WFC, this link was weaker for the employees who perceive high boundary control.

Customer mistreatment is becoming an important topic for work stress researchers and practitioners given the rise of service industry. Taking stressor-emotion-control perspectives, the authors examine day-level relationships between call center workers' customer mistreatment experiences and their impaired recovery outcomes mediated by end-of-work negative affect. Furthermore, control concepts in the job and personal domains are tested as cross-level moderators. Specifically, job control and recovery self-efficacy are identified to reduce the within-person process of customer mistreatment affecting recovery outcomes. Seventy-one call center employees completed a registration survey and then two daily surveys over 2 consecutive weeks, yielding 481 time-lagged day-level observations. Multilevel path analyses show that customer mistreatment is positively associated with increased negative affect, which, in turn, predicts poor sleep quality at night and poor recovery state the next morning. Job control attenuates the day-level effect of customer mistreatment on negative affect, and recovery self-efficacy alleviates the day-level effects of negative affect on sleep quality and next-morning recovery state. Despite the growing research on work recovery and its well-being outcomes, surprisingly little attention has been paid to at-work recovery and its job performance outcomes. The current study extends the work recovery literature by examining day-level relationships between prototypical microbreaks and job performance as mediated by state positive affect. Furthermore, general work engagement is tested as a cross-level moderator weakening the indirect effects of microbreaks on job performance via positive affect. Using multisource experience sampling method, the authors collected two daily surveys from 71 call center employees and obtained objective records of daily sales performance for two consecutive weeks (n = 632). Multilevel path analysis results showed that relaxation, socialization, and cognitive microbreaks were related to increased positive affect at work which, in turn, predicted greater sales performance. However, breaks for nutrition-intake (having snacks and drinks) did not show significant effects. Importantly, microbreaks had significant indirect effects on job performance via positive affect only for workers who had lower general work engagement, whereas the indirect effects did not exist for workers who had higher general work engagement. Furthermore, Bayesian multilevel analyses confirmed the results. Theoretical and practical implications, limitations, and future research directions are discussed. (PsycINFO Database Record
