RESUMO

Systematic condition and analytical surveys were carried out on Zoe Leonard's (b. 1961) Mouth Open, Teeth Showing 2000, an installation artwork in Tate's collection consisting of 162 children's dolls. The dolls were manufactured at various points within the 20th century and encompass several potentially problematic synthetic polymers found in modern and contemporary museum collections. To explore the doll materials and conservation condition, a multi-analytical approach was used to identify key synthetic polymer types and additives present, including portable and bench analytical techniques. Challenging degradation phenomena associated with different types of doll have been discussed and related to their material composition, which has helped our understanding of the conservation challenges inherent to this contemporary artwork.

Trait impulsivity and callous-unemotional traits are associated with behavioural problems among high-risk adolescents. While both are known to influence behaviour, the nature of their expression in high-risk behaviours, particularly those related to inhibitory control, is not well understood. In the current, preliminary study, we examined whether and how these traits predicted deficits in behaviour driven by bottom-up, automatic versus deliberate, top-down inhibitory processes among high-risk adolescents. Two go/no-go task variants, emotional and non-emotional, were used to assess reactive response inhibition, and the Balloon Analogue Risk Task was used to assess the ability to resist deliberate risky choices. The results showed that the two types of self-reported trait measures were differentially associated with performance on the two types of behavioural inhibition tasks. Trait impulsivity predicted non-emotional inhibitory control whereas callous-unemotional traits predicted risky choices. The results also showed that the emotions task elicited slower reaction times and higher false alarm rates than did the letters task, and that participants had greater difficulty inhibiting responses to negatively than to positively valenced no-go stimuli. While preliminary, the results suggest that the interplay between trait impulsivity and callous-unemotional traits is an important determinant of inhibitory behaviour in this high-risk adolescents.

How do individuals rank body shapes? Does this relate to the body part one is most dissatisfied with? Our study investigates whether one can generalize regarding how women represent the body. Three BMI-calibrated images from the Photographic Figure Rating Scale, representative of thin (BMI = 14.72), medium (BMI = 20.33), and large (BMI = 29.26) shapes, were divided into torso, legs and arms, and saved as individual images on a black background. Of 27 possible composite images, 8 were chosen based on a Torso (thin vs. large) × Leg (thin/large vs. medium) × Arm (thin vs. large) design. Our 44 female participants ordered these from thinnest to largest. This was first according to torso, then leg, and finally arm: 41 individuals agreed on the thinnest image (thin torso, thin legs, thin arms), followed by a second image (thin torso, thin legs, large arms, n = 26; or thin torso, medium legs, thin arms, n = 10). One participant differed markedly in her choice of the first image (large torso, medium legs, thin arms). Interestingly, she scored 10 on the EDI-2 scale of Bulimic Tendencies, revealing high risk for bulimia, suggesting that our task might be useful for studying eating disorders. Our juxtaposition of two analytic procedures-partial order scalogram analysis (POSAC) and cluster analysis-enables one to uncover such outliers in a data set. Importantly, the 2D POSAC space clearly reveals the hierarchical structure of the body image.

OBJECT: High-grade gliomas are the most common form of adult brain cancer, and patients have a dismal survival rate despite aggressive therapeutic measures. Intratumoral hypoxia is thought to be a main contributor to tumorigenesis and angiogenesis of these tumors. Because hypoxia-inducible factor 1α (HIF-1α) is the major mediator of hypoxia-regulated cellular control, inhibition of this transcription factor may reduce glioblastoma growth. METHODS: Using an orthotopic mouse model with U87-LucNeo cells, the authors used RNA interference to knock down HIF-1α in vivo. The small interfering RNA (siRNA) was packaged using a novel multifunctional surfactant, 1-(aminoethyl) iminobis[N-(oleicylcysteinylhistinyl-1-aminoethyl)propionamide] (EHCO), a nucleic acid carrier that facilitates cellular uptake and intracellular release of siRNA. Stereotactic injection was used to deliver siRNA locally through a guide-screw system, and delivery/uptake was verified by imaging of fluorescently labeled siRNA. Osmotic pumps were used for extended siRNA delivery to model a commonly used human intracranial drug-delivery technique, convection-enhanced delivery. RESULTS: Mice receiving daily siRNA injections targeting HIF-1α had a 79% lower tumor volume after 50 days of treatment than the controls. Levels of the HIF-1 transcriptional targets vascular endothelial growth factor (VEGF), glucose transporter 1 (GLUT-1), c-MET, and carbonic anhydrase-IX (CA-IX) and markers for cell growth (MIB-1 and microvascular density) were also significantly lower. Altering the carrier EHCO by adding polyethylene glycol significantly increased the efficacy of drug delivery and subsequent survival. CONCLUSIONS: Treating glioblastoma with siRNA targeting HIF-1α in vivo can significantly reduce tumor growth and increase survival in an intracranial mouse model, a finding that has direct clinical implications.

Social information gathering by infants 6 and 12 months old was examined as a foundation for later social learning that may be uniquely human. Infant performance on a contingency/extinction task was studied following a caregiver demonstration of the contingency on varied reinforcement schedules. Infants who observed caregivers receive any reinforcing stimulation in pretraining decreased responding over their own acquisition period, possibly because they began to habituate to the reinforcer. In extinction, infants who observed caregivers receive partial reinforcement in pretraining were more persistent in responding than others. This suggested that direct experience with partial reinforcement is not needed for greater persistence in extinction. These studies revealed details of social learning in the 1st year.

Malarial anemia is associated with a shift in iron distribution from functional to storage compartments. This suggests a relative deficit in erythropoietin production or action similar to that observed in other infections. Our study in Kenyan children with asymptomatic malaria aimed at investigating whether malaria causes increased erythropoiesis, and whether the erythropoietic response appeared appropriate for the degree of resulting anemia. Longitudinal and baseline data were used from a trial with a 2 x 2 factorial design, in which 328 anemic Kenyan children were randomly assigned to receive either iron or placebo, and sulfadoxine-pyrimethamine or placebo. Erythropoiesis was evaluated by serum concentrations of erythropoietin and soluble transferrin receptor. Prospectively collected data showed that malarial infection resulted in decreased hemoglobin concentrations, and increased serum concentrations of erythropoietin and transferrin receptor. Conversely, disappearance of malarial antigenemia resulted in increased hemoglobin concentrations, and decreased concentrations of these serum indicators. Additionally, our baseline data showed that current or recent malarial infection is associated with increased serum concentrations of erythropoietin and transferrin receptor, and that these were as high as or perhaps even higher than values of children without malarial infection and without inflammation. Our findings indicate that in asymptomatic malaria, the erythropoietic response is adequate for the degree of anemia, and that inflammation probably plays no or only a minor role in the pathogenesis of the resulting anemia. Further research is needed to demonstrate the role of deficient erythropoietin production or action in the pathogenesis of the anemia of symptomatic malaria.
