RESUMO

This study investigated the expression dynamics of bone morphogenetic protein 4 (BMP4) and its receptors (BMPR1A, BMPR1B, and BMPR2) in bovine endometrium and examined the physiological function and regulatory mechanism of BMP4 expression. The messenger RNA (mRNA) expression of BMP4 and its receptors was detected in bovine endometrium of both ipsilateral (corpus luteum [CL]-side) and contralateral (non-CL-side) uterine horns during the estrous cycle and early pregnancy. BMP4 protein levels were higher in the endometrial tissues obtained from those cows in early pregnancy than in the estrous cycle. Immunohistochemical analysis showed that BMP4 and its receptors were localized in endometrial epithelial cells. The addition of BMP4 to cultured endometrial epithelial cells did not affect caspase-3/-8 mRNA expression, whereas it significantly inhibited cell proliferation. Both prostaglandin (PG) E2 and PGF2α concentrations in the culture supernatant were decreased when stimulated by BMP4. Furthermore, BMP4 mRNA expression was increased by stimulation with tumor necrosis factor-α (TNF) and interferon-γ (IFNG). In conclusion, BMP4 is produced in bovine endometrial epithelial cells and may contribute to the regulation of cell proliferation and suppression of PG secretion through autocrine or paracrine mechanisms. BMP4 expression in the bovine endometrium may be regulated by TNF and IFNG.

RESUMO

Concomitant prescriptions of psychotropic drugs such as sleeping pills, antidepressants, and anti-anxiety medications are common. The relationship between the number of psychotropic drug prescriptions and the incidence of drug overdose has not been reported. However, efforts have been made to reduce the number of concomitant prescriptions hoping that fewer prescriptions of multiple drugs will lower the incidence of drug overdoses. Furthermore, among sleeping pills, prescriptions of barbiturates have been gradually decreasing due to the risk of severe side effects and addiction. This report features a case of an overdose of pentobarbital tablets that caused the classic medical triad (impaired consciousness, hypotension, and hypothermia) of barbiturate intoxication under the characteristics of borderline personality disorder.

RESUMO

Here, we designed a surface-grafted hydrogel (SG gel) that exhibits thermoresponsive changes in surface properties. Quantitative measurements using a self-made device showed that the adhesive strength between the SG gel surface and a Bakelite plate due to hydrophobic interaction changed significantly with temperature.

We performed a molecular phylogenetic analysis for the ground skink populations of the genus Scincella Mittleman, 1950 on Yonagunijima Island, Southern Ryukyus and representative samples of Scincella species known from other islands of the East Asian Archipelago. Partial mitochondrial cytochrome c oxidase subunit I and cytochrome b sequence data were used. Additionally, we examined the nuclear genetic variation between the Yonagunijima samples and those of closely related species. The genetic distances were largely equivalent between the Yonagunijima population and the two closest species, S. boettgeri from other southern Ryukyu Islands and S. formosensis from Taiwan. Moreover, there were nuclear haplotypes unique to the Yonagunijima population. We thus recognize the Yonagunijima population as a distinct species and describe it as Scincella dunan sp. nov. Morphologically, S. dunan sp. nov. is similar to S. boettgeri, but differs in some morphometric and meristic characters, and dorsolateral stripe pattern. These findings further support the biogeographically unique status of Yonagunijima Island among the southern Ryukyus islands.

Recent studies by our group have suggested that the vitamin D3 decomposition product VDP1 [(1R,3aR,7aR)-1-[(1R)-1,5-dimethylhexyl]octahydro-7a-methyl-4H-inden-4-one] confers the potent bactericidal action to Helicobacter pylori by targeting the membranal dimyristoyl-phosphatidylethanolamine (di-14:0 PE). In this study we synthesized a new VDP1 derivative to advance further investigation as for the correlative relationship between VDP1 structure and anti-H. pylori activity or PE vesicle collapse induction activity. The derivative VD3-7 [(1R,7aR)-4-fluoro-7a-methyl-1-((R)-6-methylheptan-2-yl)octahydro-1H-indene] retained a fluorine atom in place of the oxygen atom of VDP1. The fluorination of the carbonyl portion of VDP1 forfeited the effective anti-H. pylori activity. We, therefore, prepared Coomassie brilliant blue (CBB)-containing unilamellar vesicles consisting of various PE molecular species, and examined the vesicle collapse induction activity of either VDP1 or VD3-7 by detecting the CBB eluted from the PE unilamellar vesicles. VDP1 strongly induced CBB elution from the unilamellar vesicles of rectus-PE retaining the same two fatty acid side-chains shorter than carbon numbers 14, indicating that VDP1 specifically disrupted the vesicular conformation of those PE unilamellar vesicles. Meanwhile, VD3-7 had no influence on the structural stability of any PE unilamellar vesicles. This study obtained additional evidence that VDP1 acts as a bactericidal agent on H. pylori by targeting the membranal di-14:0 PE.

BACKGROUND: Major BCR-ABL1 mRNA in patients with chronic myeloid leukemia (CML) has generally been analysed by real-time polymerase chain reaction (PCR). Application of the international scale (IS) for the quantification of major BCR-ABL1 mRNA has been recommended in several sets of guidelines, including those of the European LeukemiaNet. The aim of this study was to clarify the efficacy of digital PCR technology for the IS of BCR-ABL1 mRNA in the patients with CML by comparing with real-time PCR. METHODS: The analysis of BCR-ABL1 mRNA was carried out by the Ipsogen® BCR-ABL1 Mbcr IS-MMR DX Kit (Qiagen), and the QuantStudio 3D Digital PCR System (Thermo Fisher Scientific ) using 20 peripheral blood samples obtained from the 9 patients with CML at Sapporo Medical University Hospital. RESULTS: The correlation between the data obtained by digital PCR and by real-time PCR was really high at R = 0.96. The detection limit of digital PCR was up to 0.003% and was equal to IS with 0.01% or less in comparison with real-time PCR. CONCLUSIONS: Digital PCR technology is promising for predicting the IS value with similar efficacy to real-time PCR and should be useful for simple monitoring of the effects of tyrosine kinase inhibitor (TKI) treatments.

RESUMO

The migration of metal ions into a nanoporous electrode was controlled using an electrophoretic effect under the influence of an external electric field based on the concept of bipolar electrochemistry. Robust, uniform and well-aligned arrays of Co and Pt nanorods were observed to form vertically against the electrode.

Alternating current (ac) bipolar electropolymerization of 3,4-ethylenedioxythiophene (EDOT) was performed in the presence of hexachloroplatinate ([PtCl6]2-) or poly(styrenesulfonate) (PSS). We demonstrated that both [PtCl6]2- and PSS were successfully incorporated into electrogenerated poly(3,4-ethylenedioxythiophene) (PEDOT) as dopants to offer hybrid fibers composed of (i) PEDOT and platinum nanoparticles (PtNPs) (PEDOT-Pt hybrid fibers) and (ii) PEDOT and PSS (PEDOT-PSS hybrid fibers), respectively, in one step, grown from the very edges of Au wires used as bipolar electrodes (BPEs).

Wilms' tumor 1 (WT1) is overexpressed in various hematopoietic tumors and widely used as a marker of minimal residual disease. WT1 mRNA has been analyzed using quantitative real-time polymerase chain reaction (real-time PCR). In the present study, we analyzed 40 peripheral blood and bone marrow samples obtained from cases of acute myeloid leukemia, acute lymphoblastic leukemia, and myelodysplastic syndrome at Sapporo Medical University Hospital from April 2012 to January 2015. We performed quantification of WT1 was performed using QuantStudio 3D Digital PCR System (Thermo Fisher Scientific ) and compared the results between digital PCR and real-time PCR technology. The correlation between digital PCR and real-time PCR was very strong (R = 0.99), and the detection limits of the two methods were equivalent. Digital PCR was able to accurately detect lower WT levels compared with real-time PCR. Digital PCR technology can thus be utilized to predict WT1/ABL1 expression level accurately and should thus be useful for diagnosis or the evaluation of drug efficiency in patients with leukemia.

A novel bubble-induced ultrafast floating and sinking of micromotors based on the difference between buoyant force and gravity is proposed. Asymmetric micromotors were prepared by modification with Au and Pt layers for the two faces of glassy carbon beads (GCBs) by the bipolar electrodeposition technique. After the accumulation of enough oxygen bubbles by the decomposition of H2O2 at the Pt layer, the upward net force acting on the micromotor drove its movement to the air/solution interface. In order to reverse the direction of net force for the sinking of the micromotors, sodium dodecyl sulfate (SDS) was added into the fuel solution, which could facilitate the release of bubbles and decrease the diameter of the bubbles. However, the lifetime of the bubbles was increased significantly. After the addition of a small amount of salt, the lifetime of the bubbles was obviously reduced. As a consequence, the breakup of bubbles on the micromotor changed the direction of the net force from up to down which pulled the micromotor down to the bottom of the solution. The velocity of the micromotor was dependent on the net force exerted on the micromotor, leading to an ultrafast motion of the micromotor. It still reached 1.2 cm s-1 after 3 h. Moreover, the simple asymmetric deposition technique showed great promise for the further application of the micromotors in bioanalysis and environmental remediation.

Alternating current (AC) bipolar electropolymerization of 3,4-ethylenedioxythiophene (EDOT) using a gold (Au) wire as a bipolar electrode (BPE) on a substrate surface resulted in gradual growth of the corresponding poly(3,4-ethylenedioxythiophene) (PEDOT) thin film from the terminals of the Au wire on the substrate. Studies to clarify the polymerization behavior were conducted under various electrolytic conditions, including monomer concentration, applied frequency, monomer structure, and substrate material. This method could be used to draw conducting polymer films on a nonconductive substrate, guided by an applied external electric field, and thus has potential for circuit patterning in organic electronic devices.

The technique of jaw tracking can be used in volumetric modulated arc therapy (VMAT) and intensity modulated radiation therapy (IMRT). In this technique, jaw tracks apertures of the multi leaf collimator (MLC) during irradiation. It is reported that dose variation is occurred by the changing accuracy of MLC position in VMAT and IMRT. Though jaw moves dynamically in the irradiation by using this technique, the influence of the jaw position accuracy on dose variation is not examined. The purpose of this study is to verify the influence of jaw position accuracy on dose variation in VMAT using jaw tracking. We appended intentional errors on jaw position in digital image communication in medicine-radiation therapy (DICOM-RT) plans created using jaw tracking technique. These plans were irradiated on the phantom that was inserted ion chamber, and we measured dose variation by changing the intentional error. The dose variation in planning target volume (PTV) was enlarged by increasing the error, and agreed with the variation of a collimator scatter factor within 0.03%. In clinical case of oropharyngeal cancer, the maximum dose variations in parotid gland were 0.179% and 1.23% when the errors were 1 mm and 10 mm, respectively. Dose variation in parotid gland was larger than the variation in PTV and spinal cord because of increasing MLC transmission. The dose variation caused by jaw position error was smaller than it caused by MLC position error. So, we can keep the dose error slightly that is related to jaw position error in VMAT by maintaining jaw position accuracy correctly.

Conducting polymers can be easily obtained by electrochemical oxidation of aromatic monomers on an electrode surface as a film state. To prepare conducting polymer fibres by electropolymerization, templates such as porous membranes are necessary in the conventional methods. Here we report the electropolymerization of 3,4-ethylenedioxythiophene and its derivatives by alternating current (AC)-bipolar electrolysis. Poly(3,4-ethylenedioxythiophene) (PEDOT) derivatives were found to propagate as a fibre form from the ends of Au wires used as bipolar electrodes (BPEs) parallel to an external electric field, without the use of templates. The effects of applied frequency and of the solvent on the morphology, growth rate and degree of branching of these PEDOT fibres were investigated. In addition, a chain-growth model for the formation of conductive material networks was also demonstrated.

BACKGROUND: Determining reliable evolutionary rates of molecular markers is essential in illustrating historical episodes with phylogenetic inferences. Although emerging evidence has suggested a high evolutionary rate for intraspecific genetic variation, it is unclear how long such high evolutionary rates persist because a recent calibration point is rarely available. Other than using fossil evidence, it is possible to estimate evolutionary rates by relying on the well-established temporal framework of the Quaternary glacial cycles that would likely have promoted both rapid expansion events and interisland dispersal events. RESULTS: We examined mitochondrial cytochrome b (Cytb) and control region (CR) gene sequences in two Japanese wood mouse species, Apodemus argenteus and A. speciosus, of temperate origin and found signs of rapid expansion in the population from Hokkaido, the northern island of Japan. Assuming that global warming after the last glacial period 7-10 thousand years before present (kyr BP) was associated with the expansion, the evolutionary rates (sites per million years, myr) of Cytb and CR were estimated as 11-16 % and 22-32 %, respectively, for A. argenteus, and 12-17 % and 17-24 %, respectively, for A. speciosus. Additionally, the significant signature of rapid expansion detected in the mtDNA sequences of A. speciosus from the remaining southern main islands, Honshu, Shikoku, and Kyushu, provided an estimated Cytb evolutionary rate of 3.1 %/site/myr under the assumption of a postglacial population expansion event long ago, most probably at 130 kyr BP. Bayesian analyses using the higher evolutionary rate of 11-17 %/site/myr for Cytb supported the recent demographic or divergence events associated with the Last Glacial Maximum. However, the slower evolutionary rate of 3.1 %/site/myr would be reasonable for several divergence events that were associated with glacial periods older than 130 kyr BP. CONCLUSIONS: The faster and slower evolutionary rates of Cytb can account for divergences associated with the last and earlier glacial maxima, respectively, in the phylogenetic inference of murine rodents. The elevated evolutionary rate seemed to decline within 100,000 years.

We report the first ever use of electrochemically mediated atom transfer radical polymerization (eATRP) employing a bipolar electrochemical method for the fabrication of both gradient and patterned polymer brushes. A potential gradient generated on a bipolar electrode allowed the formation of a concentration gradient of a Cu(I) polymerization catalyst through the one-electron reduction of Cu(II) , resulting in the gradient growth of poly(NIPAM) brushes from an initiator-modified substrate surface set close to a bipolar electrode. These polymer brushes could be fabricated in three-dimensional gradient shapes with control over thickness, steepness, and modified area by varying the electrolytic conditions. Moreover, by site-selective application of potential during bipolar electrolysis, a polymer brush with a circular pattern was successfully formed. Polymerization was achieved using both a polar monomer (NIPAM) and a nonpolar monomer (MMA) with the eATRP system.

Scincella boettgeri and S. formosensis are respectively small-bodied, morphologically similar skinks, endemic to the Southern Ryukyus and Taiwan. To estimate the phylogeography of both species, we performed phylogenetic analyses using the mitochondrial cytochrome b (cyt b) gene sequences based on 102 individuals of S. boettgeri from 12 Southern-Ryukyu islands and 33 S. formosensis from six localities in Taiwan. A total of 67 haplotypes were recognized for S. boettgeri and 21 for S. formosensis. The phylogenetic analyses revealed that the populations of Scincella spp. in the Southern Ryukyus and Taiwan are composed of three major clades, the Yonagunijima clade from Yonagunijima Island, the Southern-Ryukyu clade from the Southern Ryukyus exclusive of Yonagunijima Island, and the Taiwan clade from Taiwan. These clades showed high levels of genetic divergence, suggesting that the species have been isolated since the Early Pliocene. The Southern-Ryukyu and Taiwan clades were further divided into three and four subclades, respectively. Two of the three Southern-Ryukyu subclades are partially sympatric on two islets of the Yaeyama Group, suggesting this population represents a secondary contact subsequent to their allopatric differentiations.

RESUMO

We demonstrate a marked performance improvement in a 512 QAM transmission by employing frequency-domain equalization (FDE) instead of an FIR filter. FDE enables us to compensate for distortions due to hardware imperfections in the transmitter with higher precision, which successfully reduced the power penalty by 4 dB in a 54 Gbit/s (3 Gsymbol/s)-160 km transmission. FDE also allows the transmission distance to be extended up to 240 km.

We demonstrate substantial performance improvements in 256 QAM transmission in terms of both data rate and distance that we realized by using a digital back-propagation (DBP) method. 160 Gbit/s-160 km and 64 Gbit/s-560 km transmissions were successfully achieved with a polarization-multiplexed 256 QAM signal, in which the symbol rate and transmission distance were greatly increased by compensating for the interplay between dispersion and nonlinearity, which is responsible for the transmission impairment especially for a higher symbol rate and longer distance.
