RESUMO

Recent studies have shown that insects harbor numerous viruses of various taxa and that viral infections are often latent without noticeable symptoms. The red firebug Pyrrhocoris apterus, a true flightless bug from the family Pyrrhocoridae, is widely used for physiological studies on insect metabolism, endocrinology, and digestion. While exploring the transcriptome of P. apterus salivary glands, a nearly complete genomic sequence of a novel RNA virus was reconstructed. The virus, provisionally named Pyrrhocoris apterus virus 1 (PaV1), possesses eight potential open reading frames (ORFs) encoding for an array of proteins, some of which are involved in virus replication while others ensure success of the virus in multiple ways, including evasion of the host immune response. In addition to the information obtained from sequence analyses, we documented virus transmission, virus-induced mortality, host response upon persistent PaV1 infection, virion morphology, and putative virus-induced structures in salivary gland cells in a laboratory culture of red firebug. We propose that PaV1 belongs to a novel viral species of a new, yet-to-be established family.

RESUMO

The complete genomic sequence of a new virus from cherry trees was determined. Its genome is 5857 nt long and resembles that of members of the genus Luteovirus in its genomic organization and nucleotide sequence. Based on the species demarcation criteria for luteoviruses, the virus represents a new luteovirus species. Furthermore, a 47-nt-long inverted repeat was found at the 3' end of its genome. The virus has been provisionally named cherry-associated luteovirus (ChALV) and is the fourth member of the family Luteoviridae reported to naturally infect woody plants.

RESUMO

Cauliflower mosaic virus (CaMV) - a plant pararetrovirus that naturally causes diseases in Brassicaceae and Solanaceae plant hosts worldwide - has been detected by PCR for the first time in herbarium samples of Usnea sp. lichens. The virus's presence in these lichens did not result in any micro- or macromorphological changes, and the herbarium records were classified as representative for the distinct species. Sequence analyses classified all the detected viruses into one lineage of CaMV isolates. We have shown here that herbarium samples could be a good source for virus study, especially where a longer time span is involved.
