RESUMO

The heart of the fruit fly, Drosophila melanogaster, is a particularly suitable model for cardiac studies. Optical coherence microscopy (OCM) captures in vivo cross-sectional videos of the beating Drosophila heart for cardiac function quantification. To analyze those large-size multi-frame OCM recordings, human labelling has been employed, leading to low efficiency and poor reproducibility. Here, we introduce a robust and accurate automated Drosophila heart segmentation algorithm, called FlyNet 2.0+, which utilizes a long short-term memory (LSTM) convolutional neural network to leverage time series information in the videos, ensuring consistent, high-quality segmentation. We present a dataset of 213 Drosophila heart videos, equivalent to 604,000 cross-sectional images, containing all developmental stages and a wide range of beating patterns, including faster and slower than normal beating, arrhythmic beating, and periods of heart stop to capture these heart dynamics. Each video contains a corresponding ground truth mask. We expect this unique large dataset of the beating Drosophila heart in vivo will enable new deep learning approaches to efficiently characterize heart function to advance cardiac research.

RESUMO

Polarization properties of a soliton generated in a fiber laser of zero dispersion are investigated. Similar to the solitons generated in a fiber laser of all anomalous dispersion, the polarization ellipse of the soliton rotated during pulse evolution inside the cavity. The number of rotations relies on the cavity averaged birefringence with nonlinear bias. The larger the cavity averaged birefringence is, the bigger the bias is. When the period multiplying of solitons appears, the number of rotations depends on both multiplying periods and the cavity averaged birefringence. Multiple polarization states can be observed at a fixed position in the cavity depending on the multiplying period. When the cavity length is equal to n times of the averaged beat length, the polarization ellipse of the soliton rotates n∗m times at a fixed position, where m is equal to the multiplying period.

RESUMO

WHO has declared the outbreak of monkeypox as a public health emergency of international concern. In less than three months, monkeypox was detected in more than 30 000 people and spread to more than 80 countries around the world. It is believed that the immunity formed to smallpox vaccine can protect from monkeypox infection with high efficiency. The widespread use of Vaccinia virus has not been carried out since the 1980s, which raises the question of the level of residual immunity among the population and the identification of groups requiring priority vaccination. We conducted a cross-sectional serological study of remaining immunity among Moscow residents. To do this, a collection of blood serum samples of age group over 30 years old was formed, an in-house ELISA test system was developed, and a virus neutralization protocol was set up. Serum samples were examined for the presence of IgG antibodies against Vaccinia virus (n=2908), as well as for the ability to neutralize plaque formation with a Vaccinia virus MNIIVP-10 strain (n=299). The results indicate the presence of neutralizing antibody titer of 1/20 or more in 33.3 to 53.2% of people older than 45 years. Among people 30-45 years old who probably have not been vaccinated, the proportion with virus neutralizing antibodies ranged from 3.2 to 6.7%. Despite the higher level of antibodies in age group older than 66 years, the proportion of positive samples in this group was slightly lower than in people aged 46-65 years. The results indicate the priority of vaccination in groups younger than 45, and possibly older than 66 years to ensure the protection of the population in case of spread of monkeypox among Moscow residents. The herd immunity level needed to stop the circulation of the virus should be at least 50.25 - 65.28%.

Transition from a gain-guided soliton (GGS) to a fully developed noise-like pulse (NLP) is numerically demonstrated in fiber lasers operated in the normal dispersion regime, which explains well the experimental observation of spectrum evolution that the bottom of the averaged spectrum gradually broadens with pump power increasing. Numerical results suggest that the transition could also happen under the condition of cavity linear phase delay bias change with fixed pump power. It is demonstrated that the peak power clamping effect and the normal dispersion are the key factors leading to the spectrum evolution. In addition, intermittent meta-stable states between GGS and NLP can be obtained when the cavity dispersion is chosen at small normal dispersion.

Using numerical simulation, we have investigated the generation of color solitons consisting of two radiation fragments with different carrier frequencies in a dual-wavelength laser. The proposed mechanism for the formation of such solitons involves nonlinear losses that increase with increasing intensity, the dispersion of the refractive index, spectral gain inhomogeneity, and the generation of a doublet radiation spectrum, owing to the corresponding spectral-dependent losses in the laser. The proposed theory explains the main features of the experimentally observed formation of color domains in fiber lasers and has the potential for further development of methods for controlling the nonlinear dynamics of laser radiation.

Vaccination against COVID-19 has occurred in Russia for more than two years. According to the Russian official clinical guidelines to maintain tense immunity in the conditions of the ongoing COVID-19 pandemic, it is necessary to use booster immunization six months after primary vaccination or a previous COVID-19 contraction. It is especially important to ensure the maintenance of protective immunity in the elderly, who are at risk of severe courses of COVID-19. Meanwhile, the immunological effectiveness of the booster doses has not been sufficiently substantiated. To investigate the immunogenicity of Sputnik V within the recommended revaccination regimen and evaluate the effectiveness of booster doses, we conducted this study on 3983 samples obtained from individuals previously vaccinated with Sputnik V in Moscow. We analyzed the level of antibodies in BAU/mL three times: (i) six months after primary immunization immediately before the booster (RV), (ii) 3 weeks after the introduction of the first component of the booster (RV1), and (iii) 3 weeks after the introduction of the second component of the booster (RV2). Six months after the primary vaccination with Sputnik V, 95.5% of patients maintained a positive level of IgG antibodies to the receptor-binding domain (RBD) of SARS-CoV-2. The degree of increase in the specific virus-neutralizing antibodies level after revaccination increased with a decrease in their initial level just before the booster dose application. In the group of people with the level of antibodies up to 100 BAU/mL six months after the vaccination, a more than eightfold increase (p < 0.001, Wilcoxon criterion with Bonferroni adjustment) in the level of specific antibodies was observed (Me = 8.84 (IQR: 3.63−30.61)). A significant increase in the IgG level after receiving both the first and the second booster doses occurred at the initial titer level up to 300 BAU/ mL (p < 0.001) in those who did not contract COVID-19 in the past and up to 100 BAU/mL (p < 0.001) in those who were previously infected with SARS-CoV-2. A significant increase in the antibody level after the first dose of the booster was noted for people who had up to 500 BAU/mL (p < 0.05), regardless of the previous COVID-19 infection. Thus, revaccination is most effective in individuals with an antibody level below 500 BAU/mL, regardless of the vaccinee age and COVID-19 contraction. For the first time, it has been shown that a single booster dose of the Sputnik vaccine is sufficient to form a protective immunity in most vaccinees regardless of age and preexisting antibody level.

Due to the weak birefringence of single mode fibers, solitons generated in fiber lasers are indeed vector pulses and exhibit periodic parameter change including polarization evolution even when there is a polarizer inside the cavity. Period doubling eigenstates of solitons generated in a fiber laser mode-locked by the nonlinear polarization rotation, i.e., period doubling of polarization components of the soliton, are numerically explored in detail. We found that, apart from the synchronous evolution between the two polarization components, there exists asynchronous development depending on the detailed operation conditions. In addition, period doubling of one polarization component together with period-one of another polarization component can be achieved. When the period tripling window is obtained, much complexed dynamics on the two polarization components could be observed.

We report on experimental generation and evolution of circumstance-susceptible, narrow-bandwidth, h-shaped pulse in a thulium-doped fiber (TDF) laser. With typical mode-locking technique based on nonlinear amplifying loop mirror, a type of h-shaped pulse is generated in a net normal dispersion regime for the first time to our best knowledge. Different from pulses with similar profiles achieved in typical anomalous dispersion regime, the h-shaped pulse here exhibits extremely narrow spectral bandwidth and meanwhile becomes highly circumstance-susceptible. Not alike the well-preserved h-shaped profile with anomalous dispersion, here the h-shaped pulse can easily evolve into various other pulse patterns with circumstance variations, including peak-depressed profiles, burst-like emission, multiple h-shaped pulses, and even some highly complex temporal cases. Despite that, the h-shaped pulse broadens as the pump power increasing, being a typical pump-related characteristic dominated by the peak-power-clamping effect. Moreover, it is observed that the h-shaped pulse profile can be re-shaped by incorporating a piece of unpumped TDF into the cavity, i.e., introducing some reabsorption. Our results substantiate the experimental revelation of such a type of particular-profile pulse in the normal dispersion regime, demonstrating some new evolution features facilitated by the dispersion-relevant circumstance-susceptibility.

RESUMO

Raman-scattering-assisted noise-like pulse (NLP) generation was achieved by using an appropriate segment of high nonlinearity fiber in an erbium-doped fiber laser. Broadband spectrum with 203 nm 3-dB bandwidth was obtained, which, to the best of our knowledge, is the broadest bandwidth achieved for NLPs. The broadband operation is the result of tailored cavity design, which optimizes various effects including the Raman scattering effect to maximize the bandwidth of NLPs. Further broadening the NLP spectrum up to 294 nm was achieved by using spectral filtering outside the cavity with a polarization beam splitter.

We report on the experimental observation of a color domain (CD) phenomenon in a dissipative soliton fiber laser. The CDs are constituted by a wavelength-dependent condensed phase of pulses in motion. Single CD, dual CD, and tricolor domain that occupy all the cavity space are observed. The CDs have flexible tunability of duty cycle, amplitude, wavelength, and wavelength spacing. The CDs derive from the wavelength-dependent slow evolution of gain, and the formation mechanism of multiple CDs can be explained by cross gain saturation.

We report experimental observation of passive harmonic mode locking (HML) in which the basic pattern is a soliton crystal. Several crystal states were generated from an initial large bound state by increasing the pump power. The soliton crystals are identical and progressively span along the cavity to finally take a regular spacing leading to HML of solitons crystal.

On the basis of numerical simulation results, we put forward a way to realize harmonic passive mode locking of fiber lasers with an ultrahigh-repetition-rate pulse train. The equidistant distribution of ultrashort pulses filling the total laser cavity is due to bound-soliton mechanisms. In the case of large bound energy, such long soliton trains are very stable and have the ideal periodic structure as a soliton crystal.

On the basis of numerical simulation of fiber laser passive mode locking with anomalous dispersion we have found the dissipative solitons with powerful pedestals having oscillating structure. The pedestal structure causes a complex structural spectrum. These solitons can be multistable: with the same laser parameters the pedestals can have different structures. For some nonlinear-dispersion parameters there exist solitons with asymmetric structural pedestals moving relatively solitons with symmetric ones.

We report the observation of bound states of 350 pulses in a ring fiber laser mode locked by nonlinear rotation of the polarization. The phenomenon is described theoretically using a multiscale approach to the gain dynamics; the fast evolution of a small excess of gain is responsible for the stabilization of a periodic pattern, while the slow evolution of the mean value of gain explains the finite length of the quasiperiodic soliton train.
