RESUMO

Experiments on rabbits were made to examine excretory-absorption liver function and the blood kallikrein-kinin system after thermal burn and treatment with gamma-globulin immune to burn skin toxin. Immunotherapy leads to a more rapid and effective recovery of the functional indicators in the burnt animals than in burns without or with the treatment by nonimmune preparations. It was demonstrated that specific detoxification plays an important role in multiple modality treatment of burn disease.

RESUMO

Experiments on rats were made to study the action of toxic glycoprotein isolated from burn skin on the lysosomal apparatus of hepatocytes and morphological structure of the liver. It was shown that 2-2.5 h after intraperitoneal injection to normal rats of toxic preparations of varying purity, the animals developed morphological alterations similar to those seen in the liver of the burn animals. At the same time there was an appreciable increase in the percentage of unsedimentable activity of catepsine D of the total one, thereby indicating the toxin-induced injury to the lysosomal apparatus of hepatocytes.

RESUMO

In vivo and in vitro experiments have demonstrated the toxic effect of high-molecular protein isolated from burnt skin extract (BSE) by immunosorption. The toxin preparations cause the death of mice with RES blockade and inhibit the leukocyte migration capacity in culture. It was shown in control experiments that BSE after passage through the immunosorbent fully loses its toxic properties because of toxin binding to insoluble-base-fixed antibodies.

RESUMO

The toxic properties of serum were studied in rats with thermal burns by bioassays (in hemocultures) and in mice with the blocked reticuloendothelial system. Both methods have proved the biphasic character of the serum toxic effect time course in burnt animals. The first phase of toxemia was observed during the first 24 h and the second one days 5-7 after the burn. A mechanism of the phenomenon seen is suggested. The data obtained by the two different methods are comparable. This indicates the identity of the substances that produce the toxic action on the hemocultures and on the mice with the blocked reticuloendothelial system. Both methods can be recommended for testing toxemia in burns.

Experiments were conducted on rats. The influence of the serum of a burn convalescent on the toxic properties, the level of proteolytic enzymes activity, and morphological changes following burn were studied. After the burn the blood serum and the extracts of the organs acquired toxic properties; there was an increase in the activity of proteolytic enzymes, and marked morphological changes developed Administration of the serum of aburn convalescent promoted detoxication, diminished the activity of proteolytic enzymes distinctly, and decreased the morphological disturbances.

Experiments were conducted on germ-free (gnotobiotic) rats. A study was made of the toxic properties of the organs and the activity of proteolytic enzymes in them after thermal burns. Despite the absence of pathogenic microbial flora the germ-free rats developed the state of toxemia with an increase of the level of proteolytic enzymes. The extent of the toxic and catabolic reaction depended on the severity of the burn. Intoxication and an increase in proteolysis in the gnotobiotic rats were similar, and, in a number of cases greater than in usual animals. A conclusion was drawn on the important role of the tissue sources of intoxication in burns. A correlation between the toxemia and proteolysis was established.

RESUMO

Toxic and hemolytic properties of the sera of dogs which received massive homologus blood transfusions were studied in experiments on 20 dogs and 432 mice. The toxicity of the recipient's sera was evaluated in vivo on mice with blocked reticulo-endothelial system and by the method of hemocultures. Red cells labeled with radioactive chromium were used to study the hemolytic activity. Investigations carried out demonstrated the acute blood loss wilth the subsequent massive transfusion of the homologous blood to result in the production in the recipient's serum of toxigenic products. These toxigenic properties were determined by two biotesting methods. The maximal toxigenic action was noted in 2 hours and in the course of the 1st day after the substitution. No toxic effect of the sera was found on the 17th day. A parallel study of hemolytic properties of the sera revealed a marked increase of its activity; it was the greatest during the first 24 hours. In three days the hemolytic properties of the serum were no different from the initial one.

RESUMO

Experiments were conducted on sterile and contaminated guinea pigs. The toxicity of the serum and the water-salt extracts of the internal organs after the burn was studied by the action on the leukocyte migration in a culture. Despite the complete absence of the microbial flora, the toxic properties of the serum and of the extracts of the internal organs of the sterile animals were the same as in the contaminated organs. The data obtained pointed to the important role of the histiogenic factors in the development of burn intoxication.
