RESUMO

HIV infection is accompanied by activation of lipid peroxidation, oxidative modification of lipoprotein complexes, and a decrease in activity of antioxidant enzymes (superoxide dismutase (SOD), catalase and glutathione peroxidase (GP)) in blood serum. A significant increase of glutathione reductase observed under these conditions is considered as a function of the antioxidant defence. These changes were already seen at early (symptomless) stages of this disease, however, at manifested forms they were more pronounced.

RESUMO

In the article general conformities to law and features of violation of the thioldisulfid an redox system of whey of blood are described for patients with the without a symptom transmitter of HIV and manifestly forms AIDS. The role of her components is grounded - general, protein and non protein -SH, -S-S- groups in the mechanism of forming of oxidative stress and distress at HIV of infection. The clinic and laboratory criterions of expressed of peroxides processes are specified on the indexes of violation of redox transformations of -SH, -S-S- groups, neutralization and utilization of MDA and stability of LPC in the whey of blood at HIV of infection. Found out intercommunication between expressed of violation of the studied indexes, oxidative stress and distress and features of clinical flow of HIV of infection, allowed authors to attribute this pathology to "free radical diseases". In this connection, indicated analytic it is suggested to use as additional biochemical markers of oxidative stress and distress, and also for the ground of setting of antioxidants and their combinations in complex prophylactic or therapeutic application at HIV of infection.

RESUMO

The interaction of blood serum immunoglobulins of M, G, and A classes of the donors with monospecific serums (MSS)-anti-IgM, anti-IgG and anti-IgG was established to be associated by Ag(+)-sensitive-SH containing non protein compounds release. This phenomenon formation should be related to a parallel running associated reaction mediated by conformational and/or some other changes of immunoglobulins macrostructure under highly specific intermolecular interaction with adequate MSS in the reactive mixtures. As a rule these processes are associated by the break and reduction of mixed disulphide bounds between thiol containing nonprotein compounds and proteins. HIV antigen glycoproteins and morphine preliminary introduced into the analogic reactive mixtures were found to block this phenomenon. If in these reactive mixtures the serums including three serotypes hepatitis B virus antigen is introduced this phenomenon is preserved. This effect of HIV antigen glycoproteins and morphine could be explained by their direct and/or mediated influence on the immunoglobulins macrostructure. As a result of the latter the immunoglobulins structure-functional status is infringed, being indirectly evidenced by absence of the associated reaction of release Ag(+)-sensitive-SH containing non protein compounds in the reactive mixtures. The processes presented are capable to play an essential role in formation of polyclonal gammapathy under HIV-infection.

RESUMO

The phenomenon of release of Ag(+)-sensitive SH containing protein compounds during reaction of serum antibodies to HIV with HIV antigen glycoproteins (AgHIV) (both reagents taken from diagnostic kits) is described. These compounds were detected in nonprotein fractions of serum anti-HIV + AgHIV reaction media and disappeared after dialysis of nonprotein fractions. On the other hand, this phenomenon was not detected during addition of AgHIV to the serum containing no AgHIV. Presumably, the phenomenon should be referred to secondary concomitant reaction, which seems to be mediated by conformational and/or other processes only during highly specific antigen-antibody intermolecular interactions. That is why this phenomenon seems to characterize the biochemical phase of immune reactions, which can be associated with the denaturing processes as well, running in serum anti-HIV + AgHIV reaction media, and it can be used as an additional test in evaluating anti-HIV antibodies.

RESUMO

The author demonstrates that patients with ischemic heart disease have phenomenological peculiarities of adaptation by stability of thiol-dependent mechanism of structural-functional condition of blood serum proteins; describes the phenomenon of emergence of free nonprotein SH-groups in protein-free reaction mixtures of the serum varying in adrenaline concentrations in unstable angina pectoris (this phenomenon indicates structural-functional destabilisation of serum proteins and adequate immunopathological process); suggests an alternative method and pathogenetic diagnosis of IHD clinical forms in which thiol-containing analytes serve as molecular markers.

For the donors and for the patients with inflammatory processes is thiol-dependent the gear of immune responses in vitro an antigen--antibody on dynamics(changes) of change (+/- delta) of the contents SH- and S-S-group reaction mixtures. Thus, is conducted the analysis of interplay of proteins of an acute phase (CRP, orosomucoid and transferin) serums of a blood of the donors and patients with serology by related diagnostic (complementary) monospecific serums (MSS) against CRP (Anti-CRP), against Oroso (Anti-Oroso) i against Transf (Anti-Transf). Is established, that as against the donors, for the patients with inflammatory processes these reacting are accompanied by a phenomenon of a liberation of energy of Ag(+)-sensing non proteins SH-groups and they occur in supernatants of deprotheinized of reaction mixtures. At the same time, both for the donors, and for the patients, these reacting are accompanied modification by changes kept in repair (+/- delta) proteins SH- and S-S-rpy[symbol: see text], in integral reaction mixtures (in which one protein did not deposit). Such data testify, that the inflammatory process, apparently, can be accompanied by such rearrangement of a structurally functional condition of proteins of an acute phase, that under operating MSS in reaction mixtures descends labelised blended disulfide of communications between them and low molecular weight thiols. As a result of it there is a liberation of energy of Ag(+)-sensing non proteins SH-groups. This parameter can be used for an estimation of functional activity of proteins of an acute phase.

RESUMO

A two-staged test for immune status by methods described by L. A. Kozhemyakin et al. (1987), as well as AIDS immunodiagnosis were conducted with the help of automated working place (AWP). Application of AWP will make it possible to obtain up to 150 indexes. The principle of AWP functional algorithm is based upon menu bar selection or initial data input information, and contributes to well-founded methods of clinical immunology.
