RESUMO

In former experiments it could be shown, that new bone is formed around ashwood after implantation in the tibia of rabbits, in spite of a foreign-body tissue reaction. Equally prepared ashwood half-cylinders were now implanted in the soft tissue of rats and evaluated 2, 6, 12 and 30 weeks after implantation. Around the implants a foreign-body tissue reaction developed and demonstrates the hitherto insufficient preparation of the implants.

RESUMO

The model of an experimental posttraumatic osteomyelitis is described. Acute and chronic osteomyelitis was produced by inoculation of 10(5) Staph. aureus or E. coli to fractured femora--stabilised or not--with intramedullar nailing. The results are discussed. The model seems to be useful to further studies on posttraumatic osteomyelitis.

RESUMO

The influence of a biological tissue glue--based on fibrinogen clotting--on bone healing has been investigated in animal experiments. Standardized cortical defects in the tibiae of rabbits were closed with a plug of homologous or heterologous fibrin. The course of restoration has been studied using histological techniques, x-ray examinations and isotopes (Ci 99 TC). Heterologous fibrin delays bone formation, due to a local immune response. Homologous fibrin on the other hand cubances and accelerates sprouting of capillary vessels and connective tissue cells which results in rapid new formation of bone.

Three groups of rabbits were injected intra-articularly into the knee joints with preformed complexes of hydroxamated collagen and antiserum to denatured collagen respectively, with hydroxamated collagen and normal serum, hydroxamated collagen, anticollagen serum or saline as controls. From twelve rabbits, receiving only one injection and investigated 8 h later, only the joint tissues of rabbits which received immune complexes showed acute arthritis with severe infiltration of polymorphonuclears, leucocyte thrombi in vessels and haemorrhages. From six rabbits, receiving two consecutive injections (day 0 and day 2) and investigated 12 h after the last injection, only the joint tissue of rabbits which received immune complexes showed subacute-chronic arthritis. From sixteen rabbits receiving four consecutive injections day 0, day 2, day 7 and day 14) and investigated 7 days after the last injection, only the joint tissue of rabbits which received immune complexes showed severe chronic arthritis with infiltration of plasma cells, lymphocytes and nodule-like accumulation of lymphocytes whilst their joint fluid showed additionally a distinctly increased number of polymorphonuclears (3-15 x 10(6)) which differed with P less than 0-01 statistically from all controls.

RESUMO

We report on investigations describing the use of a biological adhesive system for tissue adhesion and local haemostasis. This system consists of highly concentrated native fibrinogen and thrombin. Clotting factor XIII is admitted for reinforcement of the fibrin network. In experimental partial kidney resection this system was used with excellent results. This experience led to successful application of this method in 15 patients who underwent partial nephrectomy for various diseases.

The immunological reactions of heterologous fibrin were investigated in a sensitive experimental system. In 10 rabbits autologous full-thickness skin grafts were glued with bovine fibrinogen cryoprecipitate. 5 of these animals had been immunized against this adhesive before transplantation. In a control group the graft was fixed with homologous fibrinogen cryoprecipitate. Examination of the sera of the animals for antibodies against fibrinogen was performed before immunization, 1 day prior to and 14 days after skin grafting. 14 days after skin adhesion all grafted areas were examined by histological methods. In the group without preoperative immunization antibodies were found against fibrinogen. The skin graft healed in this series in the same manner as in the sham group. In the series with preoperative immunization the glued skin graft was discharged early and the healing procedure of the defect was retarded.

Thirty rabbits were investigated in 3 groups: a) one consisting of non-immunized animsls which were injected intraarticularly with human soluble collagen, human gammaglobulin or saline, b) the other consisting of human gammaglobulin-immunized animals which were injected with human soluble collagen and human gammaglobulin into the knee joints, and c) the third consisting of human collagen-immunized rabbits which were injected with human soluble collagen and human gammaglobulin into the knee joints. Eighteen hours after injection animals were sacrificed, joint fluid collected, cell number in fluid determined and synovial tissue prepared for histologic investigation. Distinct acute synovitis was observed in collagen injected knees of collagen immunized rabbits, as shown by distinctly increased cell number in fluids and histologic investigation. In comparison to collagen- or gammaglobulin injected knees of non-immunized animals, increase in cell number of collagen injected knees of collagen injected animals was statistically significant with P less than 0.005 respectively with P less than 0.01, and with P less than 0.025 in comparison to gammaglobulin injected contralateral knee. Distinct acute inflammation was observed morphologically in synovial tissue of collagen injected knees of collagen immunized animals only. Acute synovitis was also observed in positive controls, i.e. gammaglobulin injected knees of gammaglobulin immunized animals. These observations show, that appearance of collagen in molecular disposable form can induce acute inflammation of joints in state of collagen immunization.
