RESUMO

The article presents the results for the study of lipid correction capacity and safety of concomitant use of policosanol and rosuvastatin compared with rosuvastatin monotherapy in patients with stable coronary artery disease and moderate hepatic dysfunction. Fifty-seven subjects aged 37 to 72 years (mean age 54.4 years +/- 6.5 years) have been enrolled into the study with the following inclusion criteria: therapy with statins for more than 8 weeks, failure to achieve target LDL cholesterol levels and moderately elevated liver enzymes. The following laboratory tests were performed at baseline and after 12 weeks of follow-up: blood lipid profile (total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), very low density lipoprotein cholesterol (VLDL-C), high density lipoprotein cholesterol (HDL-C) and triglycerides (TG), lipid peroxidation (ma- Ionic dialdehyde (MDA), glycosylated hemoglobin HbAlc (%) and liver function tests (gamma-glutamyl transpeptidase-gamma-GTP) and alanine aminotransferase (ALT). Concomitant use of policosanol with rosuvastatin was superior to rosuvastatin monotherapy in terms of reduction of pro-aterogenicity of lipid metabolism by decreasing serum TC, LDL-C and TG, increasing serum HDL-C and decreasing the pro-oxidative activity (MDA) with simultaneous substantial improvement of hepatic function. Concomitant use of policosanol at the dose of 20 mg/day and rosuvastatin at 10-20 mg/day was favorably tolerated. None of the subjects had any discontinuations of therapy due to adverse events.

RESUMO

The sterol composition of Palaemon adspersus is studied. It is established that the nonsaponifiable fraction of Palaemon adspersus contains cholesterol, 7-dehydrocholesterol, desmosterol, substance "240" and inconsiderable amounts of other still nonidentified sterols.

RESUMO

The ergosterol and lecithin absorption IR-spectra were studied in a nonpolar anhydrous medium. The thermodynamical and spectral characteristics of dimeric associates and the enthalpy value for trimeric associates of this sterol are determined. Thermodynamical and spectral parameters of ergosterol intermolecular associates with lecithin in a nonpolar anhydrous medium are found. It is established that the intermolecular interaction of lecithin with ergosterol occurs according to the mechanism of hydrogen bond. A conclusion is drawn that the presence of binary bonds and methyl groups in the cyclic and aliphatic parts of the sterol molecule affects greatly the structure of the model membrane and its strength. It is shown that under conditions of the experiment the oxygen of the phosphate group contributes to formation of the molecular associates of lecithin with sterols and not that of the carbonyl group. The obtained experimental data may be at use when studying structural disturbances of native membranes in norm and with different pathologies.

RESUMO

A method is developed for isolating desmosterol fro the brain of some animals. Sterols with Rf 0.37 and 0.51 are identified. Desmosterol is found in the brain unsaponifiable fraction of calves, piggies, chickens and rats. Its content in the mentioned animals brain is compared. Desmosterol content is shown to rise sharply in the rat brain during the first 12 days after birth. Interrelation between the level of sterols in the brain and formation of the nerve tissue myelin sheath is discussed.

RESUMO

Composition of sterols of the guinea pigs spinal cord unsaponifiables fraction was studied with experimental allergic encephalomyelitis (EAE). It is established that under conditions of EAE the cholesterol content is decreased by 48% as compared to normal. The amount of latosterol lowers as well and is 2.64 mg/g under normal conditions and 1.61 mg/g with EAE. A steroid compound which is more polar than cholesterol is found in the spinal cord. It is supposed to be cholesterol ether.

By thin-layer chromatography compounds of sterol origin were isolated from the nonsaponified fraction of bone tissue of rats. The occurrence of squalene, lanosterol, cholesterol and incompletely identified Cdelta2427-sterol in the organic matrix of the bone was demonstrated by means of UV- and IR-spectrometry. Bone tissue incubation with 1,2-C14-aceticacid yielded C14-labeled lanosterol, C28, 29-sterols, cholesterol and squalene. The rate of cholesterol synthesis in the bone tissue was lower than that in the liver and intestine of rats.
