Respiratory muscle training plays a significant role in reducing blood lactate concentration (bLa) and attenuating negative physiological stress reactions. Therefore, we investigated if voluntary isocapnic hyperpnoea (VIH) performed after a maximum anaerobic effort influences bLa and perceived fatigue level in well-trained speedskaters. 39 elite short-track speedskaters participated in a trial with two parallel groups: experimental and control. All the participants performed the Wingate Anaerobic Test (WAnT). The experimental group performed a VIH-based recovery protocol 20 min after exercise, the control group used passive recovery only. Blood samples were taken 3 and 30 min after the WAnT to measure bLa. Fatigue was self-appraised on a 0-10 perceived rating-of-fatigue (ROF) scale 3 and 30 min after the WAnT. Noteworthy, but not statistically significant changes between the experimental and control groups were observed for changes in bLa (p = 0.101). However, statistically significant changes between the groups were found for ROF (p = 0.003, ηp2 = 0.211, ω2 = 0.106). Moreover, statistically significant interactions between post-exercise bLa clearance and VO2max (p = 0.028) and inspiratory muscle strength (p = 0.040) were observed. Our findings provided preliminary insight that VIH may be an efficient recovery protocol after anaerobic exercise performed by elite athletes. The association between VO2max and post-exercise bLa clearance indicates the vital role of aerobic fitness in repeated-efforts ability in short-track speedskaters. The study was registered at ClinicalTrials.gov as NCT05994092 on 15th August 2023.

BACKGROUND: Patients underwent cyclists and triathletes test, train, and race both indoors and outdoors. However, the differences between indoor and outdoor performance remain understudied. This study aimed to analyze the relationship between indoor and outdoor performance in cycling. We investigated the influence of training environment history (indoor vs. outdoor) and application of e-sport platform on the aforementioned relationship. METHODS: Forty-three well-trained triathletes performed indoor and outdoor field tests to establish maximum sprint power (MSP) and functional threshold power (FTP). The main effects for FTP and MSP were assessed by repeated-measures ANOVA to analyze the differences between power output obtained indoors and outdoors. Multiple covariates, including training environment history and application of ZWIFT e-sport platform, were applied. Multiple linear regression was performed to investigate outdoor FTP prediction based on indoor testing. RESULTS: No significant differences were found for MSP and FTP obtained indoors and outdoors. However, the high individual variability in the disparity between indoor and outdoor power output (PO) in triathletes was observed. There was a statistically significant interaction between FTP and training environment history (P<0.001 for FTP expressed in W*kg-1 and W) and FTP and BMI (P=0.042 and P=0.034 for FTP expressed in W*kg-1 and W, respectively). The prediction formulas to establish outdoor FTP based on indoor testing had high accuracy (R2 0.80 and 0.68 for full and simple model, respectively). CONCLUSIONS: The study underlines the crucial role of the appropriate testing environment, corresponding to the training environment and racing demands. A high individual variability in the disparity between indoor and outdoor PO at FTP are associated with training environment history and BMI. No such interactions were found for MSP.

Background: The ratio of oxygen uptake (VO2) to minute ventilation (VE) is described as the oxygen uptake efficiency slope (OUES). OUES has been suggested as a valuable submaximal cardiorespiratory index; however, its characteristics in endurance athletes remain unknown. In this study, we a) investigated OUES between different time intervals, b) assessed their prediction power for VO2peak, and c) derived new prediction equations for OUES tailored for well-trained individuals. Materials and Methods: A total of 77 male (age = 21.4 ± 4.8 yrs; BMI = 22.1 ± 1.6 kg·m-2; peak oxygen uptake = 4.40 ± 0.64 L·min-1) and 63 female individuals (age = 23.4 ± 4.3 yrs; BMI = 23.1 ± 1.6 kg·m-2; peak oxygen uptake = 3.21 ± 0.48 L·min-1) underwent the cycling cardiopulmonary exercise test. OUES was measured at 75%, 90%, and 100% of exercise duration. Prediction power and new models were derived with the multiple linear regression method. Results: In male subjects, OUES [mL·min-1/L·min-1] from 75% = 4.53 ± 0.90, from 90% = 4.52 ± 0.91, and from 100% = 4.41 ± 0.87. In female subjects, OUES [mL·min-1/L·min-1] from 75% = 3.50 ± 0.65, from 90% = 3.49 ± 0.62, and from 100% = 3.41 ± 0.58. OUES did not differ between time intervals in male (p = 0.65) and female individuals (p = 0.69). OUES strongly predicts peak VO2 independently from the measuring interval (ß = 0.71-0.80; R 2 = 0.50-0.63). The prediction model designed for elite athletes was OUES [mL·min-1/L·min-1] = -1.54 + 2.99; BSA [m2]-0.0014; (age [in years]; sex [1 = male, 2 = female]) (R 2 = 0.36). Conclusion: OUES enables an accurate prediction of peak cardiorespiratory fitness in elite endurance athletes. OUES is a feasible alternative to maximal exercise testing. A new prediction equation should be used for highly trained individuals. Physicians should understand OUES physiology to properly assess the cardiorespiratory response to exercise in athletic cohorts.

Background: Ventilatory efficiency (VE/VCO2) is a strong predictor of cardiovascular diseases and defines individuals' responses to exercise. Its characteristics among endurance athletes (EA) remain understudied. In a cohort of EA, we aimed to (1) investigate the relationship between different methods of calculation of VE/VCO2 and (2) externally validate prediction equations for VE/VCO2. Methods: In total, 140 EA (55% males; age = 22.7 ± 4.6 yrs; BMI = 22.6 ± 1.7 kg·m-2; peak oxygen uptake = 3.86 ± 0.82 L·min-1) underwent an effort-limited cycling cardiopulmonary exercise test. VE/VCO2 was first calculated to ventilatory threshold (VE/VCO2-slope), as the lowest 30-s average (VE/VCO2-Nadir) and from whole exercises (VE/VCO2-Total). Twelve prediction equations for VE/VCO2-slope were externally validated. Results: VE/VCO2-slope was higher in females than males (27.7 ± 2.6 vs. 26.1 ± 2.0, p < 0.001). Measuring methods for VE/VCO2 differed significantly in males and females. VE/VCO2 increased in EA with age independently from its type or sex (ß = 0.066-0.127). Eleven equations underestimated VE/VCO2-slope (from -0.5 to -3.6). One equation overestimated VE/VCO2-slope (+0.2). Predicted and observed measurements differed significantly in nine models. Models explained a low amount of variance in the VE/VCO2-slope (R2 = 0.003-0.031). Conclusions: VE/VCO2-slope, VE/VCO2-Nadir, and VE/VCO2-Total were significantly different in EA. Prediction equations for the VE/VCO2-slope were inaccurate in EA. Physicians should be acknowledged to properly assess cardiorespiratory fitness in EA.

POLAR-2, a plastic scintillator based Compton polarimeter, is currently under development and planned for a launch to the China Space Station in 2025. It is intended to shed a new light on our understanding of Gamma-Ray Bursts by performing high precision polarization measurements of their prompt emission. The instrument will be orbiting at an average altitude of 383 km with an inclination of 42° and will be subject to background radiation from cosmic rays and solar events. In this work, we tested the performance of plastic scintillation bars, EJ-200 and EJ-248M from Eljen Technology, under space-like conditions, that were chosen as possible candidates for POLAR-2. Both scintillator types were irradiated with 58 MeV protons at several doses from 1.89 Gy(corresponding to about 13 years in space for POLAR-2) up to 18.7 Gy, that goes far beyond the expected POLAR-2 life time. Their respective properties, expressed in terms of light yield, emission and absorption spectra, and activation analysis due to proton irradiation are discussed. Scintillators activation analyses showed a dominant contribution of ß + decay with a typical for this process gamma-ray energy line of 511 keV.

The radioluminescence (RL) emitted by LiMgPO4 detector under proton beam irradiation was investigated in real time at the radiotherapy facility in the Institute of Nuclear Physics Polish Academy of Sciences in Krakow. The facility uses protons accelerated by the AIC-144 isochronous cyclotron up to the energy of 60 MeV. The measurements of RL were carried out using a remote optical fiber device with a luminophore detector and photomultiplier located at opposite ends of the optical fiber. A thin slice of LiMgPO4 doped with Tm (1.2 mol%) crystal was exposed to the proton beam. The tested detector allowed for the measurement of proton beam current, flux fluence and determination of proton beam time structure parameters. The investigation of LiMgPO4 crystal showed its high sensitivity, fast reaction time to irradiation and possibility of application as the detector for control of proton beam parameters.

Background: Respiratory muscle training (RMT) has been investigated in the context of improved athletic performance and pulmonary function. However, psychophysiological costs of RMT remain understudied. Voluntary isocapnic hyperpnoea (VIH) and inspiratory pressure threshold loading (IPTL) are widely applied RMT methods. The main purposes of this study were to assess whether RMT induces additional load on well-trained triathletes and determine differences in RMT-induced load between sexes and applied methods. Materials and Methods: 16 well-trained triathletes (n = 16, 56% males) underwent 6 weeks of VIH or IPTL program with progressive overload. Blood markers, subjective measures, cardiac indices, near-infrared spectroscopy indices, inspiratory muscle fatigue, and RMT-induced training load were monitored pre-, in and post-sessions. We used multiple ANOVA to investigate effects of sex, training method, and time on measured parameters. Results: There were significant interactions for acid-base balance (p = 0.04 for sex, p < 0.001 for method), partial carbon dioxide pressure (p = 0.03 for sex, p < 0.001 for method), bicarbonate (p = 0.01 for method), lactate (p < 0.001 for method), RMT-induced training load (p = 0.001 for method for single session, p = 0.03 for method per week), average heart rate (p = 0.03 for sex), maximum heart rate (p = 0.02 for sex), intercostales muscle oxygenation (p = 0.007 for testing week), and intercostales muscle oxygenation recovery (p = 0.003 for testing week and p = 0.007 for method). Conclusion: We found that RMT induced additional load in well-trained triathletes. Elicited changes in monitored variables depend on sex and training method. VIH significantly increased subjective training load measures. IPTL was associated with disbalance in blood gasometry, increase in lactate, and reports of headaches and dizziness. Both methods should be applied with consideration in high-performance settings.

Background: Oxygen uptake (VO2) is one of the most important measures of fitness and critical vital sign. Cardiopulmonary exercise testing (CPET) is a valuable method of assessing fitness in sport and clinical settings. There is a lack of large studies on athletic populations to predict VO2max using somatic or submaximal CPET variables. Thus, this study aimed to: (1) derive prediction models for maximal VO2 (VO2max) based on submaximal exercise variables at anaerobic threshold (AT) or respiratory compensation point (RCP) or only somatic and (2) internally validate provided equations. Methods: Four thousand four hundred twenty-four male endurance athletes (EA) underwent maximal symptom-limited CPET on a treadmill (n=3330) or cycle ergometer (n=1094). The cohort was randomly divided between: variables selection (nrunners = 1998; ncyclist = 656), model building (nrunners = 666; ncyclist = 219), and validation (nrunners = 666; ncyclist = 219). Random forest was used to select the most significant variables. Models were derived and internally validated with multiple linear regression. Results: Runners were 36.24±8.45 years; BMI = 23.94 ± 2.43 kg·m-2; VO2max=53.81±6.67 mL·min-1·kg-1. Cyclists were 37.33±9.13 years; BMI = 24.34 ± 2.63 kg·m-2; VO2max=51.74±7.99 mL·min-1·kg-1. VO2 at AT and RCP were the most contributing variables to exercise equations. Body mass and body fat had the highest impact on the somatic equation. Model performance for VO2max based on variables at AT was R2=0.81, at RCP was R2=0.91, at AT and RCP was R2=0.91 and for somatic-only was R2=0.43. Conclusions: Derived prediction models were highly accurate and fairly replicable. Formulae allow for precise estimation of VO2max based on submaximal exercise performance or somatic variables. Presented models are applicable for sport and clinical settling. They are a valuable supplementary method for fitness practitioners to adjust individualised training recommendations. POLAR-2 is a space-borne polarimeter, built to investigate the polarization of Gamma-Ray Bursts and help elucidate their mechanisms. The instrument is targeted for launch in 2024 or 2025 aboard the China Space Station and is being developed by a collaboration between institutes from Switzerland, Germany, Poland and China. The instrument will orbit at altitudes between 340km and 450km with an inclination of 42 ∘ and will be subjected to background radiation from cosmic rays and solar events. It is therefore pertinent to better understand the performance of sensitive devices under space-like conditions. In this paper we focus on the radiation damage of the silicon photomultiplier arrays S13361-6075NE-04 and S14161-6050HS-04 from Hamamatsu. The S13361 are irradiated with 58MeV protons at several doses up to 4.96Gy, whereas the newer series S14161 are irradiated at doses of 0.254Gy and 2.31Gy. Their respective performance degradation due to radiation damage are discussed. The equivalent exposure time in space for silicon photomultipliers inside POLAR-2 with a dose of 4.96Gy is 62.9years (or 1.78years when disregarding the shielding from the instrument). Primary characteristics of the I-V curves are an increase in the dark current and dark counts, mostly through cross-talk events. Annealing processes at 25 ∘ C were observed but not studied in further detail. Biasing channels while being irradiated have not resulted in any significant impact. Activation analyses showed a dominant contribution of ß + particles around 511 keV. These resulted primarily from copper and carbon, mostly with decay times shorter than the orbital period.

Sample-based methods are a useful tool in analyzing the global behavior of multi-stable systems originating from various branches of science. Classical methods, such as bifurcation diagrams, Lyapunov exponents, and basins of attraction, often fail to analyze complex systems with many coexisting attractors. Thus, we have to apply a different strategy to understand the dynamics of such systems. We can distinguish basin stability, extended basin stability, constrained basin stability, basin entropy, time dependent stability margin, and survivability among sample-based methods. Each method has specific properties and gives us important data about the behavior of the analyzed system. However, none of the methods provides complete information. Hence, to have a full overview of the dynamics, one has to collect data from two or more approaches. This study describes the sample-based methods and presents their advantages and disadvantages for the archetypal nonlinear oscillator with multiple coexisting attractors. Hence, we give helpful information in selecting the best method or methods for analyzing the dynamical system.

BACKGROUND: In addition to vocational education and experience, the pharmaceutical profession's proper pursuit requires acquiring and continuously improving professional competencies. In recent years, the need has been increasingly highlighted for developing a medical education system based on helping students develop their competencies. It is necessary to adapt the tools and methods for assessing competencies during formal education. It will enable students to know the directions of further personal or professional development. OBJECTIVE: The study aimed to compare pharmacy students' self-assessment outcomes before and after the Objective Structured Practical Examination (OSPE), which finished the Pharmaceutical Care course (PCc). The study's purpose was also to compare the outcomes of the self-assessment of competencies between the students of two academic years for whom classes on the PCc were provided by different methods. METHODS: The study was conducted over two academic years (2018/2019 and 2019/2020) among 5th-year students enrolled at the Faculty of Pharmacy of the Jagiellonian University Medical College (JUMC) at the end of the Pharmaceutical Care course. Different teaching methods were used in the delivery of the course in these academic years. The students self-assessed their competencies using a questionnaire consisting of a list of personal and patient care competencies. The students completed the questionnaire before and after the OSPE, which followed the completion of the PCc. RESULTS: Students' professional competencies as self-assessed after the exam were higher than those assessed before the exam. Differences were observed in both personal and patient care competencies. Students taking the course in the 2019/2020 academic year set their pre-OSPE competencies higher than students taking the PCc in 2018/2019. CONCLUSION: The self-assessment scores increased for most competencies included in the study following the OSPE. This may suggest that taking part in the exam, involvement in patient's case simulations, and self-assessment of performance at individual stages of the exam contributed to increased subjective assessment of professional competencies.

Cardiopulmonary exercise testing (CPET) is the method of choice to assess aerobic fitness. Previous research was ambiguous as to whether treadmill (TE) and cycle ergometry (CE) results are transferrable or different between testing modalities in triathletes. The aim of this paper was to investigate the differences in HR and VO2 at maximum exertion between TE and CE, at anaerobic threshold (AT) and respiratory compensation point (RCP) and evaluate their association with body fat (BF), fat-free mass (FFM) and body mass index (BMI). In total, 143 adult (n = 18 female), Caucasian triathletes had both Tr and CE CPET performed. The male group was divided into <40 years (n = 80) and >40 years (n = 45). Females were aged between 18 and 46 years. Body composition was measured with bioelectrical impedance before tests. Differences were evaluated using paired t-tests, and associations were evaluated in males using multiple linear regression (MLR). Significant differences were found in VO2 and HR at maximum exertion, at AT and at RCP between CE and TE testing, in both males and females. VO2AT was 38.8 (±4.6) mL/kg/min in TE vs. 32.8 (±5.4) in CE in males and 36.0 (±3.6) vs. 32.1 (±3.8) in females (p < 0.001). HRAT was 149 (±10) bpm in TE vs. 136 (±11) in CE in males and 156 (±7) vs. 146 (±11) in females (p < 0.001). VO2max was 52 (±6) mL/kg/min vs. 49 (±7) in CE in males and 45.3 (±4.9) in Tr vs. 43.9 (±5.2) in females (p < 0.001). HRmax was 183 (±10) bpm in TE vs. 177 (±10) in CE in males and 183 (±9) vs. 179 (±10) in females (p < 0.001). MLR showed that BMI, BF and FFM are significantly associated with differences in HR and VO2 at maximum, AT and RCP in males aged >40. Both tests should be used independently to achieve optimal fitness assessments and further training planning.

BACKGROUND: Genomes within the same species reveal large similarity, exploited by specialized multiple genome compressors. The existing algorithms and tools are however targeted at large, e.g., mammalian, genomes, and their performance on bacteria strains is rather moderate. RESULTS: In this work, we propose MBGC, a specialized genome compressor making use of specific redundancy of bacterial genomes. Its characteristic features are finding both direct and reverse-complemented LZ-matches, as well as a careful management of a reference buffer in a multi-threaded implementation. Our tool is not only compression efficient but also fast. On a collection of 168,311 bacterial genomes, totalling 587 GB, we achieve a compression ratio of approximately a factor of 1,265 and compression (respectively decompression) speed of ∼1,580 MB/s (respectively 780 MB/s) using 8 hardware threads, on a computer with a 14-core/28-thread CPU and a fast SSD, being almost 3 times more succinct and >6 times faster in the compression than the next best competitor.

The low-level radio frequency (LLRF) control system is one of the fundamental parts of a particle accelerator, ensuring the stability of the electro-magnetic (EM) field inside the resonant cavities. It leverages on the precise measurement of the field by in-phase/quadrature (IQ) detection of an RF probe signal from the cavities, usually performed using analogue downconversion. This approach requires a local oscillator (LO) and is subject to hardware non-idealities like mixer nonlinearity and long-term temperature drifts. In this work, we experimentally evaluate IQ detection by direct sampling for the LLRF system of the Polish free electron laser (PolFEL) now under development at the National Centre for Nuclear Research (NCBJ) in Poland. We study the impact of the sampling scheme and of the clock phase noise for a 1.3-GHz input sub-sampled by a 400-MSa/s analogue-to-digital converter (ADC), estimating amplitude and phase stability below 0.01% and nearly 0.01°, respectively. The results are in line with state-of-the-art implementations, and demonstrate the feasibility of direct sampling for GHz-range LLRF systems.

MOTIVATION: The amount of sequencing data from high-throughput sequencing technologies grows at a pace exceeding the one predicted by Moore's law. One of the basic requirements is to efficiently store and transmit such huge collections of data. Despite significant interest in designing FASTQ compressors, they are still imperfect in terms of compression ratio or decompression resources. RESULTS: We present Pseudogenome-based Read Compressor (PgRC), an in-memory algorithm for compressing the DNA stream, based on the idea of building an approximation of the shortest common superstring over high-quality reads. Experiments show that PgRC wins in compression ratio over its main competitors, SPRING and Minicom, by up to 15 and 20% on average, respectively, while being comparably fast in decompression. PURPOSE: To investigate the effect of delayed application of leukocyte-rich platelet-rich plasma (PRP) on rotator cuff repair results as assessed by outcome scores and imaging at one year. METHODS: Patients with a symptomatic rotator cuff tear awaiting arthroscopic repair were approached to take part in the study. Final eligibility for the study was confirmed at the time of surgery. A total of 97 patients were randomized to an ultrasound guided injection of leukocyte-rich PRP or normal saline between 10 and 14 days postsurgery. A total of 87 patients completed clinical evaluation and underwent magnetic resonance imaging imaging at 1 year. Outcome scores included the American Shoulder and Elbow Score, Constant score, Western Ontario Rotator Cuff Index, and the Disabilities of the Arm, Shoulder and Hand Score. Structural integrity of the repair was assessed according to the Sugaya grading. Muscle fatty infiltration was assessed on magnetic resonance imaging using the Goutallier classification. RESULTS: At 1 year postsurgery, there was no significant difference between the treatment groups on any of the patient-reported outcome measures or Constant score. On postoperative imaging analysis, there was no difference in the retear rates (Sugaya 4 and 5) between the groups (21% in control group vs 15.3% in PRP group). Fatty infiltration on postoperative imaging was found to be significantly higher in the normal saline group compared with the PRP group (Kendall's tau-b P = .032). CONCLUSIONS: The delayed application of PRP postrotator cuff repair did not improve function as measured by patient-reported outcome measures and Constant score at 1 year postoperatively. OBJECTIVES: This paper assesses the effect of neurophysiological rehabilitation in children with postural defects on the depth of thoracic kyphosis, lateral spinal deviation and rotation of spinal motor segments. MATERIAL AND METHODS: A total of 201 patients aged 8-15 years old with a postural defect diagnosed by medical examination were enrolled. The analyzed parameters were determined using the DIERS system before the first therapeutic session and after 4 weeks of therapy. The angle of thoracic kyphosis, lateral deviation of the spine and spinal rotation were assessed. The therapy employed techniques associated with the proprioceptive neuromuscular facilitation (PNF) and Vojta's approaches. The results were analyzed separately for both sexes and for patients rehabilitated solely with Vojta's techniques vs. patients rehabilitated according to combined Vojta's and PNF techniques. The χ2 test was used for statistical analyses, at p < 0.05. RESULTS: There was improvement in the angle of thoracic kyphosis, ranging from 0.14 (among boys with kyphosis < 42°) to 5.47 (among girls with kyphosis ≥ 42°), spinal rotation, from 0.37 (among boys with kyphosis ≥ 42°) to 4.33 (among patients with kyphosis ≥ 42° rehabilitated solely according to Vojta's method), and lateral deviations, ranging from 1.32 mm (among boys with kyphosis < 42°) to 2.99 mm (among patients with kyphosis ≥ 42° rehabilitated solely according to Vojta's method). CONCLUSIONS: Neurophysiological rehabilitation of patients with postural defects produced positive effects by improving the angle of thoracic kyphosis, spinal rotation and lateral deviation of the spine. Children with reduced thoracic kyphosis achieved less improvement in the kyphosis angle, lateral spinal deviation and spinal rotation than children with kyphosis ≥ 42°. OBJECTIVES: This paper evaluates the efficacy of using the McKenzie and Vojta methods for patients with low back pain and the use of the DIERS Formetric 4D system as an objective diagnostic tool. MATERIAL AND METHODS: The study enrolled 28 patients aged 15-17 years old. The patients were hospitalized at the Department of Orthopedics and Traumatology of the Swietokrzyskie Center for Pediatrics in Kielce with a diagnosis of back pain associated with a discopathy. The patients were rehabilitated according to the McKenzie and Vojta methods. Assessment by means of the DIERS Formetric system had taken place before the first therapy session and on the day that pain was eliminated to evaluate trunk inclination, angle of thoracic kyphosis, angle of lumbar lordosis, lateral deviation, trunk torsion, surface rotation and pelvic obliquity. Pain intensity and change in pain intensity over time were assessed by means of a numerical rating scale. RESULTS: Pain intensity was reduced to 0 over 3-12 days. The study participants demonstrated reduction in anterior trunk inclination of the mean value at 1.83°. The angle of thoracic kyphosis was also reduced by 7.95°. The angle of lordosis increased by 7.6°. The lateral spinal curvature was reduced by 8.92 mm. There was a reduction of 4.64° in trunk torsion. Surface rotation was reduced by 1.61° and pelvic obliquity was reduced by 3.78°. CONCLUSIONS: In discopathic patients, postural parameters comprising trunk inclination, angle of thoracic kyphosis, angle of lumbar lordosis, lateral deviation, trunk torsion, vertebral rotation and pelvic obliquity fail to reach Hartzmann's physiological reference ranges. A therapeutic intervention based on the Vojta and McKenzie methods may normalize the posture to physiological reference ranges and is effective in the treatment of patients with back pain. BACKGROUND: Achilles tendon rupture is a common injury and the best treatment option remains uncertain between surgical and nonoperative methods. Biologic approaches using multipotent stem cells such as perivascular stem cells pose a possible treatment option, although there is currently a paucity of evidence regarding their clinical therapeutic use. QUESTIONS/PURPOSES: The purpose of this study was to determine whether injected perivascular stem cells (PSCs) would (1) improve histologic signs of tendon healing (such as percent area of collagen); and (2) improve biomechanical properties (peak load or stiffness) in a rat model of Achilles tendon transection. METHODS: Two subtypes of PSCs were derived from human adipose tissue: pericytes (CD146CD34CD45CD31) and adventitial cells (CD146CD34CD45CD31). Thirty-two athymic rats underwent right Achilles transection and were randomized to receive injection with saline (eight tendons), hydrogel (four tendons), pericytes in hydrogel (four tendons), or adventitial cells in hydrogel (eight tendons) 3 days postoperatively with the left serving as an uninjured control. Additionally, a subset of pericytes was labeled with CM-diI to track cell viability and localization. At 3 weeks, the rats were euthanized, and investigators blinded to treatment group allocation evaluated tendon healing by peak load and stiffness using biomechanical testing and percent area of collagen using histologic analysis with picrosirius red staining. RESULTS: Histologic analysis showed a higher mean percent area collagen for pericytes (30%) and adventitial cells (28%) than hydrogel (21%) or saline (26%). However, a nonparametric statistical analysis yielded no statistical difference. Mechanical testing demonstrated that the pericyte group had a higher peak load than the saline group (41 ± 7 N versus 26 ± 9 N; mean difference 15 N; 95% confidence interval [CI], 4-27 N; p = 0.003) and a higher peak load than the hydrogel group (41 ± 7 N versus 25 ± 3 N; mean difference 16; 95% CI, 8-24 N; p = 0.001). The pericyte group demonstrated higher stiffness than the hydrogel group (36 ± 12 N/mm versus 17 ± 6 N/mm; mean difference 19 N/mm; 95% CI, 5-34 N/mm; p = 0.005). CONCLUSIONS: Our results suggest that injection of PSCs improves mechanical but not the histologic properties of early Achilles tendon healing. CLINICAL RELEVANCE: This is a preliminary study that provides more insight into the use of adipose-derived PSCs as a percutaneous therapy in the setting of Achilles tendon rupture. Further experiments to characterize the function of these cells may serve as a pathway to development of minimally invasive intervention aimed at improving nonoperative management while avoiding the complications associated with surgical treatment down the line.

The importance of [11C]-choline as a PET/CT marker has been extensively described, although its production presents considerable technical difficulties. The main ones are short half-lives and the occurrence of dimethylformamide (DMF) as a residual solvent. While the losses resulting from the radionuclide decay can be minimised by shortening the duration of the process, the best solution for reducing the content of DMF is its elimination from the reaction environment. In the current work two methods are compared for [11C]-choline synthesis - a green chemistry approach (with ethanol as a green solvent) and a dry synthesis. The results were compared with each other and with those of the method based on DMF. The solid phase synthesis proved to be the most effective in total elimination of DMF, its final release was the highest, and the synthesis time was the shortest. The optimised synthesis led to the formation of the desired radiotracer with a high radiochemical yield (65% ±3%) in a short production time (12 min) and the residual precursor in the final product at the level of 1 µg/ml. 27% increase of the saturation yield was possible, which resulted in 9 GBq higher activity from 40 minutes of beaming. Each test batch passed all standard quality control requirements, and the levels of residual DMEA were below the limits that have been published in the last Pharmacopoeia monograph.
