RESUMO

Advances in biomedicine and development of modern technologies in the last century have fostered the improvement in human longevity and well-being. This progress simultaneously initiated the need for novel biomaterials. Recently, degradable metallic biomaterials have attracted serious attention in scientific and clinical research owing to their utilization in some specific applications. This work investigates the effect of the polyethylene glycol (PEG) coating of open-cell iron and phosphorus/iron foams on their microstructure and corrosion properties. The addition of phosphorus causes a slight increase in pore size and the deposition of a polymer coating results in a smoothened surface and a moderate decrease in pore diameter. The PEG coating leads to an increase in corrosion rates in both foams and potentially a more desirable product.

Research in the field of biodegradable metallic scaffolds has advanced during the last decades. Resorbable implants based on iron have become an attractive alternative to the temporary devices made of inert metals. Overcoming an insufficient corrosion rate of pure iron, though, still remains a problem. In our work, we have prepared iron foams and coated them with three different concentrations of polyethyleneimine (PEI) to increase their corrosion rates. Scanning electron microscopy (SEM) coupled with energy dispersive X-ray analysis (EDX), Fourier-transform infrared spectroscopy (FT-IR), and Raman spectroscopy were used for characterization of the polymer coating. The corrosion behavior of the powder-metallurgically prepared samples was evaluated electrochemically using an anodic polarization method. A 12 weeks long in vitro degradation study in Hanks' solution at 37 °C was also performed. Surface morphology, corrosion behavior, and degradation rates of the open-cell foams were studied and discussed. The use of PEI coating led to an increase in the corrosion rates of the cellular material. The sample with the highest concentration of PEI film showed the most rapid corrosion in the environment of simulated body fluids.

Surface-enhanced Raman spectroscopy (SERS) of blood plasma on an electrochemically prepared silver surface has been studied as a label-free, noninvasive diagnostic test for colorectal cancer. Indium tin oxide glass substrates were modified with 0.01 mol dm - 3 silver nitrate using the pulsed double-potentiostatic method. The prepared silver substrates were tested with Rhodamine 6G as a model analyte and the surface with the highest signal enhancement was selected. This silver dendritic surface was used as a diagnostic substrate for SERS measurements of human blood plasma. A group of oncological patients with declared colorectal carcinoma (n = 15) and the control group of healthy volunteers (n = 15) were compared. The biomolecular changes in chemical composition in the cancer samples were detected by statistical processing of the resulting SERS spectra. About 94% specificity and 100% sensitivity were achieved for the analysis by the ratio of the SERS peak intensity at 725 cm - 1 for adenine to the peak intensity at 638 cm - 1 for tyrosine and 100% specificity and sensitivity by using principal component analysis. This method of SERS diagnostics of colorectal cancer, which does not require the nanoparticle preparation, mixing, and incubation of plasma with a colloidal solution as in conventional tests, is a rapid, inexpensive method, which could be introduced as a primary diagnostic test.

OBJECTIVES: The aim of this study was to assess the contamination level of aquatic ecosystems of the Svitava River, situated in the South Moravian Region (Czech Republic), by residues of selected brominated flame retardants. We isolated and determined seven indicator PBDE congeners in samples of surface water and bottom sediments using optimized analytical methods. DESIGN: Samples were collected from eight locations along the river basin, particularly near the larger cities in order to assess their possible impact on the aquatic ecosystems. Isolation of selected analytes was performed using the methods of liquid-liquid extraction and cold extraction. Column chromatography and rotary vacuum evaporation were used for the purification and pre-concentration of extracts. Final identification and quantification were carried out by gas chromatography coupled with electron capture detector (GC/ECD). RESULTS: The sum of PBDEs was calculated for individual BDE congeners. Evidence for magnetoreception in mammals remains limited. Magnetic compass orientation or magnetic alignment has been conclusively demonstrated in only a handful of mammalian species. The functional properties and underlying mechanisms have been most thoroughly characterized in Ansell's mole-rat, Fukomys anselli, which is the species of choice due to its spontaneous drive to construct nests in the southeastern sector of a circular arena using the magnetic field azimuth as the primary orientation cue. Because of the remarkable consistency between experiments, it is generally believed that this directional preference is innate. To test the hypothesis that spontaneous southeastern directional preference is a shared, ancestral feature of all African mole-rats (Bathyergidae, Rodentia), we employed the same arena assay to study magnetic orientation in two other mole-rat species, the social giant mole-rat, Fukomys mechowii, and the solitary silvery mole-rat, Heliophobius argenteocinereus. Both species exhibited spontaneous western directional preference and deflected their directional preference according to shifts in the direction of magnetic north, clearly indicating that they were deriving directional information from the magnetic field. Because all of the experiments were performed in total darkness, our results strongly suggest that all African mole-rats use a light-independent magnetic compass for near-space orientation. However, the spontaneous directional preference is not common and may be either innate (but species-specific) or learned. We propose an experiment that should be performed to distinguish between these two alternatives.
