RESUMO

The article deals with indicators of cell composition of umbilical blood (fetus blood) in case of normal pregnancy (n = 38) and hypoferric anemia of light degree (n = 14)10 the hematologic analyzer of medium class was applied. The parameters of erythrocytes of fetus blood in case of hypoferric anemia of light degree were sufficiently stable and had no differences with normal values both in mean values and variability. The variety of adaptive index of fetus blood (Garkavi index) was significantly higher in case of hypoferric anemia as compared with normal values. This occurrence testifies higher stability of indicators of adaptation in case of moderate ferric deficiency. The study results testify the informativity of applied approach related to assessment of stability of cell composition of fetus blood.

RESUMO

The new highly effective method of alcoholism diagnosis was proposed. It included gas-chromatography analysis of breathed out air. The alcoholism was diagnosed if ethane and paraldehyde in this air to be less than 0.47 nM 0.20 nM respectively, pentane level was more that 0.47 nM while ethanol content was found in the range of 0.73-1.96 nM. If ethane concentration proved to be higher that 0.47 nM alcoholism diagnosis was established when ethylene level was found between 0.01-0.19 nM. The assay's diagnostic importance was confirmed by observation of 44 alcoholic patients as well as 26 healthy male individuals which did not consume alcohol.

The study was performed of mitotic cycle stage distribution (G1/0, S, G2 + M) by means of flow cytometry in rats irradiated locally and totally and in 25 cancer patients receiving radiotherapy. Radiation effects manifested themselves in diminished percentage of cells in S-stage, in the block G2 + M reflecting the direct affection of myelokaryocytes. Logarithmic dependence of the quantity, of cells synthetizing DNA on radiation dose occurred in the animals in the range 0.5-6 Gy 24 hours after the exposure. Investigation of the patients' sternal puncture biopsies indicated that the proportion of S-stage cells and the value S/(G2 + M) are significant diagnostic markers of more than 1 Gy irradiation 24-48 h after the exposure. For establishing radiation overdosage these parameters proved much more informative than hemocytological indices (myelogram).

The oxygen-binding properties of Hb molecule are unchanged in the third trimester of normal pregnancy and in pregnancy complicated by slight iron-deficiency anemia. Hb affinity to oxygen in whole blood is formed under the effect of the three main ligands H+, CO2, and 2,3-DPH. The development of hypocapnia in pregnant women results in elevation of pH values, reduction of pCO2, increase of intraerythrocytic 2,3-DPH concentration by 15-18%. Variously directed effects of H+, CO2, and 2,3-DPH on Hb affinity to O2 result in normal P50 values and therefore the Hb-O2 dissociation curve is not shifted to the right. Thus, in pregnant women with alkalosis 2,3-DPH effect on Hb affinity to oxygen completely eliminates H+ and CO2 effects but not compensates for Hb deficiency in the blood.

A new class of drugs is now utilized for in vivo diagnoses and therapy of many widespread diseases. In these pharmacological and diagnostic preparations the active substance is conjugated with a vector which transports the drug to specific biological targets. Monoclonal antibodies are the most commonly used vectors: estimation of their permeability through multilayer and unilayer biomembranes is an important step in the analysis of efficiency of vector drugs. Experiments with Sprague-Dawley rats (mature females weighing 500 to 160 g) have demonstrated the ability of immunoglobulins G to penetrate through the respiratory epithelial-hematic barrier. Using solid phase ELISA, it was found that 5-25% of the total amount of mouse antiinsulin immunoglobulins G1 injected into the trachea under hexenal anesthesia can penetrate into the blood plasma. Accumulation of antibodies in the blood begins 4 hours and ceases 32 hours after the drug application in a dose of 400 micrograms. The kinetics of transmembrane transport is described by an S-like saturation function: C(t) = Cmax/(1+e-(at-b]. Penetration of monoclonal antibodies into the blood is accompanied by their distribution in the organs and tissues as well as by their clearance from the blood plasma. The clearance of monoclonal antibodies is characterized by a 24 hour half-life and is described by an exponential equation: C(t) = C0 x exp-kt. An algorithm for the interaction of these processes which should be taken into account during measurements of the transport of monoclonal antibodies and their complexes through biomembranes is proposed.(ABSTRACT TRUNCATED AT 250 WORDS)

Specific interactions of guanine nucleotides with rat brain membrane preparations were investigated; a two-step scheme of this interaction was proposed. The effect of GTP on the agonist binding to rat brain opioid receptors was studied. Nucleotides were shown to inhibit the ligand interactions with mu- and sigma-receptors. It was found that the GTP-induced inhibition differs from that of the GTP stable analog, GppNp, i.e., GTP does not initiate any noticeable dissociation of the receptor system component and its liberation from the membrane into solution as is the case with GppNp. A kinetic model of the GTP interactions with the opioid receptor system stipulating that the affinity of high affinity centers for mu- and delta-ligands changes after nucleotide hydrolysis on the N-protein, was proposed. The differences in the mechanisms of GTP effect on mu- and sigma-receptors were revealed, i.e., the inhibition of the ligand binding to mu-receptors may occur just after GTP binding to the N-protein. The ligand-receptor interactions in the presence of GTP were modelled according to the proposed scheme.

Individual susceptibility of 10 G6PD- hemizygotes to oxidative hemolytic agents was tested on the basis of multifactorial cluster analysis of biochemical indices of erythrocyte populations; the indices related to G6PD activity and glucose metabolism were analyzed under physiological and oxidative stress conditions in very young, exactly adult and very old red cell suspensions. Biochemical images of G6PD- erythrocytes were obtained and compared with the donor (7 subjects) biochemical image on a IBM-PC computer according to a special "taxon" program. As a result, a stable subdivision of 10 Gd- biochemical images into 5 taxons was formed; each taxon included G6PD subjects with a certain form of clinical appearance of G6PD deficiency. Multifactorial cluster analysis of biochemical data on the erythrocyte population allows a clinical prognosis for G6PD- subjects.

The activity of cAMP-dependent histone kinases (HK) and cAMP-independent casein kinases (CK) as well as cAMP level were assessed in gastric mucosa affected by various forms of gastritis as well as in gastric tumor and adjacent normal tissue. CK activity was higher whereas that of HK and cAMP level were lower in tumor and at a distance of 5-20 cm from its edge as compared to gastritis. CK/HK cAMP ratio in adenocarcinoma was 6 times that in normal tissue and 2-2.5 times that in gastritis. The data obtained and those published earlier suggest the relative increase in cAMP-independent protein phosphorylation with respect to its cAMP-dependent counterpart to be characteristic of human tumors.

Activity of cAMP-dependent and -independent protein kinases as well as content of cAMP were studied in biopsy preparations of gastric mucosal membranes obtained from patients with superficial gastritis, erosion, ulcer, adenomatous polyps and ulcer after treatment with laser irradiation. All the patterns studied were similar in preparations of adenomatous polyps and normal mucosal membrane. Activity of cAMP-independent protein kinases was increased under conditions of erosion, ulcer, gastroduodenal reflux and, especially, in ulcer tissue treated with laser. However, the ratio between cAMP-independent and -dependent phosphorylation was increased only in 50% of ulcerous diseases and in gastroduodenal reflux. The equilibrium of cAMP-dependent and -independent phosphorylation was not altered in the other impairments studied even though benign proliferation was stimulated using laser irradiation.

RESUMO

Phytohemagglutinin (PHA)-effected lymphocyte blastogenesis test (LBGT) in culture has been assessed by flow cytofluorometry in 63 normal subjects and in 160 patients suffering from various pulmonary diseases. An algorithm for the estimation of cellular distribution in accordance with the mitotic cycle stages is suggested, and a computer program for the analysis of the results, based on the mitotic cycle stages, has been compiled. The lymphocyte proliferative activities have been estimated with various PHA doses. The findings have been in good correlation with the results of radionuclide studies (3H thymidine label). The described approach to LBGT assessment yields reliable results and may be used in diagnostic studies.

RESUMO

Blood oxygen balance and acid-base equilibrium were studied in 22 Chuvash patients with hereditary erythrocytosis. Despite the lack of the anomalous types of hemoglobin in the given type of erythrocytosis a number of the patients showed secondary tissue hypoxia associated with hyperoxia of the peripheral venous blood. It is shown that tissues are capable of extracting oxygen from the blood. It is assumed that the development of tissue hypoxia in the patients is determined by a fall in the rate of oxygen diffussion from the capillaries to the tissues or by blood shunting via the arteriolo-venular anastomoses because of microcirculatory disorders. The lowering of hematocrit to the level close to normal with the aid of bloodlettings makes oxygen balance return to normal and reduces tissue hypoxia.
