RESUMO

This article is concerned with the management of the disputable situations arising in the course of establishment of the kinship based on the analysis of autosomal STR loci. It is proposed to enhance the accuracy of determining thekinsip relations in the parent-child pairs (in the absence of one of the parents) by using additional sets of genetic markers localized for example on sex chromosomes, mitochondrial DNA (mtDNA) and bi-allele markers.

RESUMO

We compared the effectiveness of DNA isolation from nail plates using three commercial kits from different manufacturers. The results of the study give reason to recommend the use of PrepFiler BTA Forensic DNA Extraction Kits and QIAamp DNA Investigator Kit for the work with nail microfragments.

RESUMO

The nucleotide sequences of the Verticillium wilt resistance locus of resistant and susceptible tomato genotypes were cloned and analyzed. The nucleotide sequences displaying high degree of homology to the earlier cloned Ve1 and Ve2 genes were detected in the tomato forms susceptible to Verticillium wilt. The polymorphism at this locus between the resistant and susceptible plants was detected and used to elaborate a highly efficient CAPS marker.

RESUMO

Using a combination of degenerate primers designed from the NBS domains of the resistance genes, amplification and subsequent cloning of the resistance gene fragments from sunflower (Helianthus agrophyllus) was conducted. Sequences of cloned PCR products differed from one another and displayed homology to NBS domain fragments of the already known plant resistance genes, as well as to the analogous genes from different classes. The highest homology was shown to the NBS domain regions of cultivated sunflower and the other members of the family Compositae. Two cloned fragments had open reading frames, while the other sequences carried stop codons and seemed to belong to pseudogenes. Amino acid sequences of Helianthus agrophyllus analyzed contained conservative regions typical of NBS domains of the resistance gene products.
