RESUMO

Frequency data for the STR system FGA (HumFibra) were obtained from a Caucasoid German population sample (Düsseldorf area) of 424 unrelated individuals. PCR products were detected by horizontal polyacrylamid gel electrophoresis and a total of 16 alleles was identified by side-by-side comparison with a commercially available sequenced ladder. The observed genotype distribution showed no significant deviation from the Hardy-Weinberg equilibrium. The high information content (pooled German data: rate of heterozygosity = 0.8626; probability of match = 0.0344; mean exclusion chance = 0.7240) render this system a useful tool not only in forensic casework (criminal and paternity cases) but in population genetics too.

Hypervariable loci within the human genome are useful tools in several disciplines: for example in forensic medicine (paternity testing and forensic identification). One of these genetical markers is located at chromosome 1 and is called D1S80 (MCT118). The genotype distribution and the allele frequency of the VNTR locus D1S80 have been studied in a population of 378 unrelated Germans living at Düsseldorf. The determination of genotypes has been carried out by using the polymerase chain reaction and subsequent analysis of the amplified products by polyacrylamide electrophoresis followed by silver staining. The data demonstrate that the locus is highly polymorphic with an observed heterozygosity of 75.66%. The frequency distribution found does not meet Hardy-Weinberg expectations. We think that this is not astonishing because we found only 80 of the possible 231 phenotypes (alleles 16-34, 36-37, without anodal and cathodal variants). So before using the D1S80 data in forensic analyses and paternity tests a larger data base has to be established. The data of the Düsseldorf sample are compared with data of studies on other populations.

RESUMO

This paper reports on the allele and genotype frequencies for five polymerase chain reaction-based loci determined in a sample (n = 295) from Düsseldorf (Germany). The loci are Low-Density-Lipoprotein Receptor, Glycophorin A, Hemoglobin gamma G, D7S8 and Group-Specific Component. Typing was carried out by using the AmpliType PM PCR amplification and typing kit. The loci were tested for possible divergences from Hardy-Weinberg expectations (HWE). The gene frequencies found can be used in forensic analyses and paternity tests. The data from Düsseldorf were compared with six other studies having been published up today.
