RESUMO

We have studied amino acid up-take from incubation media by colchicine-resistant and colchicine-sensitive Djungarian hamster fibroblast cells. It has been shown that arginine and asparagine amino acid contents in the medium are different for colchicine-sensitive and colchicine-resistant cells. Amino acids concentration is not reduced in the medium after incubation of resistant cells, while it is decreased after incubation of sensitive cells. We can suggest that penetration of low-weight sources of nitrogen into sensitive cells is hampered. It can also be suggested that protein macromolecules are the main source of nitrogen for these cells. The protein up-take levels from the incubation medium, as assessed by the ammonia and amino acid contents of cell counts don't exceed 5% of their initial concentrations in the medium.

The experiments on 60 mongrel rats with hepatic failure due to small intestinal fistulas of high and medium location were carried out for the examination of amino acid spectrum in the animal plasma. 16 free amino acids were studied by ion-exchange chromatography. It has been established that hepatic failure due to small intestinal fistulas is characterized by the reduction of the total amino acid content by 20-30%. These changes were brought about mainly by the reduction of glucoblastic and essential amino acids. A dramatic (400%) increase in His and a slight (20%) increase in Asp were noted in high fistulas. The changes were caused by profound losses through fistulas which markedly activated gluconeogenesis and led to full exhaustion of amino acid reserves. It was concluded that the question of diagnostic amino acid value should be considered taking into account the amount of losses and the activity of gluconeogenesis in the pathogenesis of hepatic failure.

Content of free amino acids was altered in blood plasma and erythrocytes of patients with Becker-Kiner progressive muscular dystrophy; these alterations were more distinct in blood plasma as compared with erythrocytes. Spectra of individual free amino acids were distinctly altered both in blood plasma and erythrocytes. Development of the myodystrophia process correlated with alterations in glycogen and branched-chain amino acids content. The data obtained suggest the important functions of erythrocytes in intertissue transport of free amino acids. Similarity between the alterations, observed in progressive muscular dystrophy and in other pathological states accompanied by accelerated metabolism of proteins, suggests that these impairments are among the components in the adaptation reactions to metabolic stresses.

Under examination there were 29 patients (14 females and 15 males aged 20 to 40 years) with various forms of progressing myodystrophies (PMD). The control group consisted of 9 patients (2 females and 7 males) of the same age group. The content of free amino acids in the blood plasma and erythrocytes was determined by the method of column ion-exchange chromatography. The changes of the plasma and erythrocyte levels of most amino acids were found to be diverse in all forms of the PMD. Differences between primary PMD forms and denervation amyotrophies were revealed. The results of the studies show that content of free amino acids in the plasma and erythrocytes reflects both the general adaptation changes in the body in the conditions of the myodystrophic process, and the specificity of amino acid metabolism disturbances in various PMD forms.

RESUMO

Acute disturbances of the cerebral circulation occupy the leading place in the structure of neurovascular diseases. Studies on metabolic processes in the patients, body in the acute period of the stroke and, particularly, the metabolism of free amino acids enable one to expand our knowledge of the pathogenetic essence of the stroke and its differential diagnosis, and thus to decide on the therapeutic measures to be taken for correcting the disturbed metabolism. Of great interest in this connection are dynamic determinations of the levels and the spectrum of free amino acids in biological fluids.
