Introduction: Public safety personnel (PSP), including firefighters, paramedics, and police officers, are exposed to traumatic events as part of their day-to-day jobs. These traumatic events often result in significant stress and increase the likelihood of negative mental health outcomes, including post-traumatic stress disorder (PTSD). The present study sought to develop an in-depth understanding of the lived experiences of PSPs as related to the mental health toll of their service. Through a series of targeted focus groups, Canadian PSP were asked to provide their perspectives on the PTSD-related symptoms that resulted as a by-product of their occupational service. The DSM-5-TR PSTD criteria (A-E) provided a thematic lens to map the self-described symptomatic expression of PSP's lived experiences.Methods: The present study employed a phenomenological focus-group approach with a treatment-seeking inpatient population of PSP. Participants included PSP from a variety of occupational backgrounds. Using semi-structured focus groups, fifty-one participants were interviewed. These focus groups were audio recorded, with consent, and transcribed verbatim. Using an interpretive phenomenological approach, emergent themes within the data were inductively developed, examined, and connected across individual cases.Results: Utilizing the primary criteria of PTSD (Criteria A-E) outlined by the DSM-5-TR, we identified qualitative themes that included exposure to a traumatic event, intrusion symptoms, avoidance symptoms, negative alterations in mood and cognition, and marked alterations in arousal and reactivity.Conclusion: PSP are exposed to extreme stressors as a daily part of their occupation and are at elevated risk of developing mental health difficulties, including PTSD. In the present study, focus groups were conducted with PSP about the mental health toll of their occupations. Their experiences mapped onto the five primary criteria of PTSD, as outlined by the DSM-5-TR. This study provides crucial descriptive information to guide mental health research aims and treatment goals for PSTD in PSP populations.


Repeated exposure to stressful and traumatic events is often a daily occurrence for public safety personnel, actively contributing to an increased risk of development of mental health disorders, including Post-Traumatic Stress Disorder, in this population.Through a series of interviews, the present study examined the subjective experiences of traumatic events in a treatment-seeking population of public safety personnel. Participants' narrative descriptions of their experiences were examined and analysed using the criteria of Post-Traumatic Stress Disorder, as outlined by the Diagnostic and Statistical Manual of Mental Illness-Version V ­ Text Revision, as a thematic lens. Analyses yielded rich descriptive information of the symptomatic expression of criterion-specific themes.The present study offers valuable insights into how a treatment-seeking population of public safety personnel experience their trauma-related symptoms. It also offers an opportunity for both researchers and practitioners to better understand the way public safety personnel may differ from other populations in how they express and understand their experience of Post-Traumatic Stress symptoms.

The effective management of grapevine trunk diseases (GTDs) is an ongoing challenge. Hot water treatment (HWT) is an environmentally friendly and economically viable option; however, the short-term effects of HWT on grapevine (Vitis vinifera L.) health and production are not fully understood. The aim of this study was to compare the effects of HWT on plant growth and fungal community structure in nursery stock until plants were completely established in the field. We assessed eleven graft and three rootstock varieties from four local nurseries in a region of Catalonia (NE Spain) where GTDs are a serious threat. After treatment, the plants were left to grow under field conditions for two growing seasons. Metabarcoding of the ITS region was used to study the mycobiomes of plant graft unions and root collars. We also assessed the influence of plant physiological indicators in community composition. Hot water treatment caused lasting changes in GTD communities in both the root collar and graft union that were not always characterized as a reduction of GTD-related fungi. However, HWT reduced the relative abundance of some serious GTD-associated pathogens such as Cadophora luteo-olivacea in graft tissues, and Phaeomoniella chlamydospora and Neofusicoccum parvum in the root collar. Treatment had the greatest influence on the total and GTD-related fungal communities of Chardonnay and Xarel·lo, respectively. Total community variation was driven by treatment and nursery in rootstocks, whereas HWT most significantly affected the GTD community composition in R-110 rootstock. In conclusion, changes in fungal abundance were species-specific and mostly dependent on the plant tissue type; however, HWT did reduce plant biomass accumulation in the short-term.

Grapevine trunk diseases (GTDs) are caused by cryptic complexes of fungal pathogens and have become a growing problem for new grapevine (Vitis vinifera) plantations. We studied the role of the nursery, variety, and rootstock in the composition of the fungal communities in root collars and graft unions of planting material in Catalonia (NE Spain). We compared necrosis and fungal communities in graft unions and root collars at harvest, and then after three months of cold storage. We evaluated combinations of eleven red and five white varieties with four common rootstocks coming from six nurseries. Fungal communities were characterized by isolation and metabarcoding of the ITS2 region. Our data suggests that nursery followed by rootstock and variety had significant effects on necrosis and fungal community structure in graft and root tissues. Within the plant, we found large differences in terms fungal community distribution between graft and root tissues. Graft unions housed a significantly higher relative abundance of GTD-related Operational Taxonomic Units (OTUs) than root collars. More severe necrosis was correlated with a lower relative abundance of GTD-related OTUs based on isolation and metabarcoding analyses. Our results suggest that nurseries and therefore their plant production practices play a major role in determining the fungal and GTD-related fungal community in grapevine plants sold for planting. GTD variation across rootstocks and varieties could be explored as a venue for minimizing pathogen load in young plantations.

Azospirillum brasilense Sp7 (Sp7) is a diazotrophic, free-living plant growth-promoting rhizobacterium (PGPR) that is increasingly used for its ability to reduce stress and improve nutrient uptake by plants. To test the hypothesis that Sp7 interacts differently with the primary metabolism in C3 and C4 plants, differential proteomics were employed to study weekly protein expression in Sp7-treated maize (Zea mays cv. B73) and tomato (Solanum lycopersicum cv. Boludo) seedlings. Plant and root growth parameters were also monitored. Protein changes were most striking at the four-leaf stage (T1) for both species. Proteins related to metabolism and redox homeostasis were most abundant in tomato at T1, but later, plants experienced inhibited Calvin-Benson (CB) cycle and chloroplast development, indicating that photosynthetic proteins were damaged by reactive oxygen species (ROS). In maize, Sp7 first increased ROS-scavenging enzymes and decreased those related to metabolism, which ultimately reduced photoinhibition at later sampling times. Overall, the early interaction with maize is more complex and beneficial because the photosynthetic aparatus is protected by the C4 mechanism, thereby improving the interaction of the PGPR with maize. Better seedling emergence and vigor were observed in inoculated maize compared to tomato. This study provides an integrated perspective on the Sp7 strain-specific interactions with young C3 and C4 plants to modulate primary metabolism and photosynthesis.

The CRISPR/Cas9 system and related RNA-guided endonucleases can introduce double-strand breaks (DSBs) at specific sites in the genome, allowing the generation of targeted mutations in one or more genes as well as more complex genomic rearrangements. Modifications of the canonical CRISPR/Cas9 system from Streptococcus pyogenes and the introduction of related systems from other bacteria have increased the diversity of genomic sites that can be targeted, providing greater control over the resolution of DSBs, the targeting efficiency (frequency of on-target mutations), the targeting accuracy (likelihood of off-target mutations) and the type of mutations that are induced. Although much is now known about the principles of CRISPR/Cas9 genome editing, the likelihood of different outcomes is species-dependent and there have been few comparative studies looking at the basis of such diversity. Here we critically analyse the activity of CRISPR/Cas9 and related systems in different plant species and compare the outcomes in animals and microbes to draw broad conclusions about the design principles required for effective genome editing in different organisms. These principles will be important for the commercial development of crops, farm animals, animal disease models and novel microbial strains using CRISPR/Cas9 and other genome-editing tools.
