RESUMO

A natural appearing microalgae-bacteria consortium was used to process urban wastewater. The process was done in an 80 m2 raceway reactor and the results were compared to an identical reactor operated using freshwater supplemented with commercial fertilisers. The biomass harvesting was done using commercial ultrafiltration membranes to reduce the volume of culture centrifuged. The membrane allowed achieving a biomass concentration of ∼9-10 g L-1. The process proposed avoids the use of centrifuges and the drying of the biomass, two of the most energy consuming steps of conventional processes. The specific growth rate in freshwater and the wastewater-based media was estimated as 0.30 ± 0.05 and 0.24 ± 0.02 days-1, respectively (p < 0.05). The maximum concentration reached at the end of the batch phase was 0.96 ± 0.03 and 0.83 ± 0.07 g L-1 when the biomass was produced using freshwater and wastewater, respectively (p < 0.05). The total nitrogen removal capacity of the system was on average 1.35 g m-2·day-1; nitrogen assimilation into biomass represented 60%-95% of this value. Furthermore, the P-PO4 3- removal capacity of the system varied from 0.15 to 0.68 g m-2·day-1. The outlet effluent of the reactor was used as a nutrient source in the hydroponic production of zucchini seedlings, leading to an increase in the root dry weight and the stem diameter compared to the water alone. The produced biomass showed potential for use as feedstock to produce plant biostimulants with positive effects on root development and chlorophyll retention.

The microalga Arthrospira platensis BEA 005B was produced in 11.4 m3 raceway photobioreactors and a culture medium based on commercial fertilisers and either freshwater or seawater. The biomass productivity of the reactors operated at a fixed dilution rate of 0.3 day-1 decreased from 22.9 g·m-2·day-1 when operated using freshwater to 16.3 g·m-2·day-1 when the biomass was produced using seawater. The protein content of the biomass produced in seawater was lower; however, the content of essential amino acids including valine, leucine and isoleucine was higher. Seawater also triggered the production of carotenoids and altered the synthesis and accumulation of fatty acids. For example, the biomass produced using seawater showed a 319% and 210% higher content of oleic and eicosenoic acid, respectively. The results demonstrate that it is possible to produce the selected microalga using seawater after an adaptation period and that the composition of the produced biomass is suitable for food applications.

The intensification of livestock activities lead to an increase in waste generation with high content of nutrients, as is the case of piggery wastewater. However, this type of residue can be used as culture media for algae cultivation in thin-layer cascade photobioreactors to reduce its environment impact and produce a valorizable algal biomass. Biostimulants were produced by enzymatic hydrolysis and ultrasonication of microalgal biomass, using membranes (Scenario 1) or centrifugation (Scenario 2) as harvesting methods. The co-production of biopesticides by solvent extraction was also evaluated using membranes (Scenario 3) or centrifugation (Scenario 4). The four scenarios were analyzed by a technoeconomic assessment estimating the total annualized equivalent cost and the production cost, i.e., the minimum selling price. Centrifugation provided biostimulants approximately 4 times more concentrated than membranes, but with higher expense due to the cost of the centrifuge (contribution of 62.2 % in scenario 2) and the electricity requirements. The biopesticide production resulted the highest contribution to investment cost in scenarios 3 and 4 (34 % and 43 % respectively). The use of membranes was also more advantageous to produce biopesticides, although it was 5 times more diluted than using centrifuge. The biostimulant production cost was 65.5 €/m3 with membranes and 342.6 €/m3 by centrifugation and the biopesticide production cost was 353.7 €/m3 in scenario 3 and 2,122.1 €/m3 in scenario 4. Comparing the treatment of 1 ha of land, the cost of the biostimulant produced in the four scenarios was lower than the commercial one (48.1 %, 22.1 %, 45.1 % and 24.2 % respectively). Finally, using membranes for biomass harvesting allowed economically viable plants with lower capacity and longer distance for biostimulant distribution (up to 300 km) than centrifuge (188 km). The algal biomass valorization for agricultural products production is an environmentally and economically feasible process with the adequate capacity of the plant and distribution distance.

The microalga Arthrospira platensis BEA 005B was produced using 80 m2 (9 m3) raceway photobioreactors achieving a biomass productivity of 28.2 g·m-2·day-1 when operating the reactors in semi-continuous mode (0.33 day-1). The produced biomass was rich in proteins (58.1 g·100 g-1) and carbohydrates (25.6 g·100 g-1); the content of phycocyanins and allophycocyanins was 115.4 and 36.9 mg·g-1, respectively. Ultrasounds and high-pressure homogenisation allowed recovering approximately 90% of the initial protein content of the biomass; however, the energetic requirements of the former (∼100 kJ·kg-1) were significantly lower than those of high-pressure homogenisation (∼200 kJ·kg-1). An in silico analysis revealed that papain and ficin would allow releasing a large number of bioactive peptides with antioxidant, antihypertensive (ACE-I and renin), and antidiabetic (DPP-IV, α-amylase, and α-glucosidase) properties. Both were assessed in vitro together with Alcalase and pepsin leading to the generation of enzymatic hydrolysates with in vitro bioactivity.

The present work aims to assess the treatment of unprocessed urban wastewater using the microalga Scenedesmus almeriensis. Two 12 m3 raceway reactors, one supplemented by wastewater and the second by chemical fertilizer, operating outdoors in a semi-continuous mode, were used for eight months. Results suggested that S. almeriensis can be produced in wastewater without affecting the photosynthetic apparatus reaching a productivity of 13 g·m-2·day-1 on average in both the systems. Furthermore, the nutrient content in terms of nitrogen, phosphorous and chemical oxygen demand of the wastewater was reduced under the European limitations during most of the period, with an average removal rate of 2.2, 0.2 and 3.0 g·m-2·day-1 respectively. Therefore, raceways demonstrated a high potential for microalgal production and successful biotreatment, proving robust and reliable. Finally, the effect of environmental conditions on biomass productivity of the clean system was evaluated in a model with high accuracy (R2 = 0.9, p = 0.0002).

The aim of the present study was to assess the potential of producing four microalgal strains using secondary-treated urban wastewater supplemented with centrate, and to evaluate the biostimulant effects of several microalgal extracts obtained using water and sonication. Four strains were studied: Chlorella vulgaris UAL-1, Chlorella sp. UAL-2, Chlorella vulgaris UAL-3, and Chlamydopodium fusiforme UAL-4. The highest biomass productivity was found for C. fusiforme, with a value of 0.38 ± 0.01 g·L-1·day-1. C. vulgaris UAL-1 achieved a biomass productivity of 0.31 ± 0.03 g·L-1·day-1 (the highest for the Chlorella genus), while the N-NH4+, N-NO3-, and P-PO43- removal capacities of this strain were 51.9 ± 2.4, 0.8 ± 0.1, and 5.7 ± 0.3 mg·L-1·day-1, respectively. C. vulgaris UAL-1 showed the greatest potential for use as a biostimulant-when used at a concentration of 0.1 g·L-1, it increased the germination index of watercress seeds by 3.5%. At concentrations of 0.5 and 2.0 g·L-1, the biomass from this microalga promoted adventitious root formation in soybean seeds by 220% and 493%, respectively. The cucumber expansion test suggested a cytokinin-like effect from C. vulgaris UAL-1; it was also the only strain that promoted the formation of chlorophylls in wheat leaves. Overall, the results of the present study suggest the potential of producing C. vulgaris UAL-1 using centrate and wastewater as well as the potential utilisation of its biomass to develop high-value biostimulants.

In this study, a biorefinery based on Oscillatoria sp. is developed to produce high-value compounds such as C-phycocyanin, used in food colourant applications, and biostimulants, used in agriculture-related applications. First, the Oscillatoria biomass production was optimized at a pilot scale in an open raceway reactor, with biomass productivities equivalent to 52 t/ha·year being achieved using regular fertilizers as the nutrient source. The biomass produced contained 0.5% C-phycocyanins, 95% of which were obtained after freeze-thawing and extraction at pH 6.5 and ionic strength (FI) 100 mM, with a purity ratio of 0.71 achieved in the final extract. This purity ratio allows for use of the extract directly as a food colourant. Then, the extract's colourant capacity on different beverages was evaluated. The results confirm that C-phycocyanin concentrations ranging from 22 to 106 mg/L produce colours similar to commercial products, thus avoiding the need for synthetic colourants. The colour remained stable for up to 12 days. Moreover, the safety of the extracted C-phycocyanin was confirmed through toxicity tests. The waste biomass was evaluated for use as a biostimulant, with the results confirming a relevant auxin-like positive effect. Finally, an economic analysis was conducted to evaluate different scenarios. The results confirm that the production of both C-phycocyanin and biostimulants is the best scenario from an economic standpoint. Therefore, the developed biomass processing scheme provides an opportunity to expand the range of commercial applications for microalgae-related processes.

The aim of the present study was to identify the optimum combination of dilution rate and depth of the culture to maximise the Arthrospira platensis BEA005B (Spirulina) productivity using 80 m2 raceway reactors. By varying these two main operational conditions, the areal biomass productivity of the reactors varied by over 55%. The optimum combination, optimised using a surface response methodology, was a depth of 0.10 m and a dilution rate of 0.33 day-1, which led to a biomass productivity of 30.2 g·m-2·day-1 on a dry weight basis when operating the reactors in semi-continuous mode. The composition of the produced biomass was 62.2% proteins, 42.5% carbohydrates, 11.6% ashes, and 8.1% lipids. The isolated proteins contained all the essential amino acids (except for tryptophan, which was not determined); highlighting the content of valine (6.8%), histidine (8.3%), and lysine (7.5%). The functional properties of the proteins were also assessed, demonstrating huge potential for their use in the development of innovative and sustainable foods.

Microalgae and microalgae-derived ingredients are one of the top trends in the food industry. However, consumers' acceptance and purchase intention of a product will be largely affected by odour and flavour. Surprisingly, the scientific literature present a very limited number of studies on the volatile composition of microalgae and cyanobacteria. In order to fill the gap, the main objective of the present study was to elucidate the volatile composition of seven microalgal and cyanobacterial strains from marine and freshwaters, with interest for the food industry while establishing its potential impact in odour. Among the seven selected strains, Arthrospira platensis showed the highest abundance and chemical diversity of volatile organic compounds (VOCs). Aldehydes, ketones, and alcohols were the families with the highest diversity of individual compounds, except in Arthrospira platensis and Scenedesmus almeriensis that showed a profile dominated by branched hydrocarbons. Marine strains presented a higher abundance of sulfur compounds than freshwater strains, while the ketones individual profile seemed to be more related to the taxonomical domain. The results of this study indicate that the VOCs composition is mainly driven by the individual strain although some volatile profile characteristics could be influenced by both environmental and taxonomical factors.

RESUMO

One of the main social and economic challenges of the 21st century will be to overcome the worlds' water deficit expected by the end of this decade. Microalgae based wastewater treatment has been suggested as a strategy to recover nutrients from wastewater while simultaneously producing clean water. Consortia of microalgae and bacteria are responsible for recovering nutrients from wastewater. A better understanding of how environmental and operational conditions affect the composition of the microalgae-bacteria consortia would allow to maximise nutrient recoveries and biomass productivities. Most of the studies reported to date showed promising results, although up-scaling of these processes to reactors larger than 100 m2 is needed to better predict their industrial relevance. The main advantage of microalgae based wastewater treatment is that valuable biomass with unlimited applications is produced as a co-product. The aim of the current paper was to review microalgae based wastewater treatment processes focusing on strategies that allow increasing both biomass productivities and nutrient recoveries. Moreover, the benefits of microalgae based agricultural products were also discussed.

The depth of the culture and the dilution rate have a striking effect on the biomass productivity and the nutrient recovery capacity of microalgal cultures. The combination of culture depth and dilution rate that allows to maximise the performance of the system depends on environmental conditions. In the current study, a response surface methodology was used to explore the relationship between the two most relevant operational conditions and the biomass productivity achieved in 8.3 m2 pilot-scale raceways operated using urban wastewater. Four polynomial models were developed, one for each season of the year. The software predicted biomass productivities of 12.3, 25.6, 32.7, and 18.9 g·m-2·day-1 in winter, spring, summer, and autumn, respectively. The models were further validated at pilot-scale with R2 values ranging within 0.81 and 0.91, depending on the season. Lower culture depths had the advantage of minimising nitrification and stripping but allow to process a lower volume of wastewater per surface area. Biomass productivity was higher at culture depths of 0.05 m, when compared to 0.12 and 0.20 m, while the optimal dilution rate was season-dependent. Results reported herein are useful for optimising the biomass productivity of raceway reactors located outdoors throughout the year.

Astaxanthin is a high-value carotenoid currently being produced by chemical synthesis and by extraction from the biomass of the microalga Haematococcus pluvialis. Other microalgae, such as Chlorella zofingiensis, have the potential for being used as sources of astaxanthin. The differences between the synthetic and the microalgae derived astaxanthin are notorious: not only their production and price but also their uses and bioactivity. Microalgae derived astaxanthin is being used as a pigment in food and feed or aquafeed production and also in cosmetic and pharmaceutical products. Several health-promoting properties have been attributed to astaxanthin, and these were summarized in the current review paper. Most of these properties are attributed to the high antioxidant capacity of this molecule, much higher than that of other known natural compounds. The aim of this review is to consider the main challenges and opportunities of microalgae derived products, such as astaxanthin as food. Moreover, the current study includes a bibliometric analysis that summarizes the current research trends related to astaxanthin. Moreover, the potential utilization of microalgae other than H. pluvialis as sources of astaxanthin as well as the health-promoting properties of this valuable compound will be discussed.

Our aim was to develop a novel medium for the large-scale production of protein-rich Arthrospira with potential applications as a biofertilizer. The novel culture medium, termed as FM-II, was formulated using low-cost commercial chemicals and specifically designed to improve protein production. Both Arthrospira platensis and Arthrospira maxima were produced using FM-II and Arnon medium, which was used as a control. Photosynthetic status of the cells, which was checked by measuring chlorophyll fluorescence, biomass dry weight and protein content, was assessed daily. Arthrospira platensis had higher biomass and protein productivities than A. maxima when cultured in both control and FM-II media. Incorporation of varied micronutrients into FM-II formulation did not improve biomass productivity. Maximum biomass dry weight in FM-II and control medium was 2.9 and 2.5 g · L-1 , respectively. Total protein content of the biomass ranged between 55% and 65%, suggesting potential for being used in the development of high-value agricultural products. As some nutrients were discarded unused, the initial content of phosphates and bicarbonates was reduced by 75% and 50%, respectively, without affecting the process productivity. Results reported herein could promote the production and utilization of Arthrospira platensis by significantly reducing productions costs and therefore increasing the feasibility of the process.

Humans are no strangers to the consumption of microalgae as already in the sixteenth century Spirulina was harvested from Lake Texcoco and consumed in markets in Tenochtitlan (today Mexico City). Nowadays, microalgae are being incorporated into many food formulations. Most of these use microalgae as a marketing strategy or as a colouring agent. However, Spirulina (and compounds derived thereof) show potential for being used as ingredients in the development of novel functional foods, which are one of the top trends in the food industry. Several human intervention studies demonstrated the potential of Spirulina for being used in the prevention or treatment of disorders related to metabolic syndrome. The aim of the current paper was to review current and potential applications of this microalga in the food and functional food industries. Health benefits associated with consuming Spirulina and/or some of the most important compounds derived from Spirulina were also discussed.

Disinfection of fruits is one of the most important steps since they are going to be eaten fresh-or minimally-processed. This step affects quality, safety, and shelf-life of the product. Despite being a common sanitizer in the fruit industry, chlorine may react with organic matter leading to the formation of toxic by-products. Alternative sustainable disinfection strategies to chlorine are under study to minimize environmental and human health impact. Water-assisted UV-C light (WUV-C) is proposed here as an alternative sanitizing method for strawberries. In this study, strawberries were washed for 1 or 5 min in a tank with 2 or 4 lamps on, each emitting UV-C light at 17.2 W/cm2, or in a chlorine solution (200 ppm, pH 6.5). Moreover, trials with 4 lamps on, together with a washing solution consisting on peracetic acid at 40 or 80 ppm, were carried out. Overall, quality and nutritional parameters of strawberries after treatments were maintained. Changes in color were not noticeable and fruits did not lose firmness. No major changes were observed in antioxidant activity, organic acid, anthocyanin, vitamin C, and total phenolic content. Yeasts and molds were not affected by the WUV-C treatment, and 5 min were needed to significantly reduce total aerobic mesophylls population. However, reductions of artificially inoculated Listeria innocua and Salmonella Typhimurium after WUV-C treatments were comparable to those obtained with chlorine-wash, which were 3.0 log CFU / g. Moreover, WUV-C light was effective to minimize microorganisms remaining in washing water, avoiding cross-contamination and thus, allowing water recirculation. This effect was improved when combining the action of UV-C light with peracetic acid, showing the suitability of this combined treatment, understood as an alternative to chlorine sanitation, for sanitizing strawberries and keeping the populations of pathogenic bacteria in washing water lower than 0.6 ±â€¯0.1 log CFU / mL.

Modern city lifestyle is characterized by an increased demand for fresh or minimally processed foods. Lettuce (Lactuca sativa L.), mainly iceberg lettuce, is the main vegetable used during the manufacture of fresh-cut salads. The current study evaluated the phenolic content and antioxidant activity of ten fresh and minimally processed lettuce varieties. The phenolic content of selected lettuce samples varied significantly among varieties. Although a higher phenolic content was observed in modern lettuce varieties, when compared to the traditional ones (except for the landrace Francès 219/855), the antioxidant capacity of modern and traditional lettuce varieties was similar. Minimal processing followed by storage for a 7-day period led to an increased phenolic content in varieties Rutilaï RZ, Abago RZ, Maravilla LS044, Francès 219/855, Negre borratger 386/935, and D'hivern LS008, supporting the hypothesis that wounding can induce the accumulation of phenolic compounds in lettuce leaves. For example, the total phenolic content of Francès 219/855 after processing and storage increased from 8.3 to 11.3 mg/100 g (p < 0.05). Accumulation of phenolic compounds after minimal processing was not observed in all the studied samples, suggesting that this effect could be matrix-dependant. The amount of bioaccessible polyphenols was higher after minimal processing and storage. Indeed, the amount of bioaccessible polyphenols after a simulated gastrointestinal digestion of fresh or minimally processed Pelikan lettuce was calculated as 32.6 or 43.3 mg/100 g respectively (p < 0.05), suggesting that the increased amount of polyphenols caused by processing and storage can also lead to a higher amount of bioaccessible phenolic compounds.

Calçots are the immature floral stems of the second-year onion (Allium cepa L.) resprouts. Modified atmosphere packaging or vacuum packaging are suitable alternatives to preserve fresh-cut vegetables. The objective of this study was to evaluate the effect of postharvest storage time of raw vegetable stored under controlled atmosphere and used packaging system after minimal processing on the quality of fresh-cut calçots. Calçots used for minimal processing were stored under 1.0 kPa O2 + 2.0 kPa CO2 at 1 ℃ for 30 and 60 days. Fresh-cut calçots were packaged using passive modified atmosphere packaging or vacuum packaging and were stored at 4 ℃ for 15 days. Calçots stored under controlled atmosphere for 30 days presented better retention of quality and in turn, being more suitable for minimally processing. Vacuum packaging preserved the physicochemical quality of fresh-cut calçots better after 15 days. Mesophilic aerobic counts were also higher in fresh-cut calçots stored under modified atmosphere packaging, but all counts were below the recommended limits during and at the end of their shelf-life (15 days). The most suitable conservation strategy might be to store whole calçots under controlled atmosphere for 30 days and after minimally processing, packaged under vacuum in order to extend the shelf-life of fresh-cut calçots.

Ultrasound processing can result in cell wall disruption, facilitating the release of the cellular content. Therefore, we hypothesized that sonication of vegetables could be used as a pre-treatment to increase the bioaccessibility of phenolic and antioxidant compounds. Overall, sonication (40 kHz, 250 W, 4 °C, 20 min) did not affect the main physicochemical parameters of tomato, lettuce, zucchini, and green and red pepper (p < 0.05). The polyphenolic content and antioxidant activity of digestive enzymatic extracts was higher than that of water:methanol extracts (p < 0.05). In addition, sonication resulted in increased bioaccessibility of phenolic compounds in lettuce and green pepper (p < 0.05), while no effect was observed for tomato, red pepper, and zucchini samples suggesting a matrix-dependent effect. The amount of phenolic compounds and antioxidants released by vegetables during a simulated gastrointestinal digestion may be higher than the one that can be expected from measurements in usual aqueous-organic extracts.

The risk posed by outbreaks associated with strawberries together with the safety issues of by-products from chlorine disinfection in the fruit industry has led to a search for alternative sanitizers. The disinfection capacity of peracetic acid (PA) at three concentrations (20, 40 and 80 ppm) and washing times (1 and 2 min) was compared to sodium hypochlorite (200 ppm) (NaClO) treatments and a water control, and its influence on the physico-chemical, biochemical and nutritional quality of strawberries was also studied. Counts on total aerobic mesophilic microorganisms were comparable between NaClO and PA. For yeasts and molds, only NaClO and 80 ppm PA reduced contamination in washing water, but no differences wereobserved in strawberries. Artificially inoculated L.innocua was reduced by at least 4 log cfu/g in strawberry by all the PA treatments, except at 20 ppm PA for 1 min. Total soluble solids, pH, titratable acidity, antioxidant activity and total phenolic content values were maintained after all treatments. Only anthocyanin content was affected. Treatments of 20 and 40 ppm PA did not significantly affect fruit color, and there were no losses on strawberry firmness. PA, as a GRAS substance that has shown potential to reduce microorganisms present in strawberries without any major physicochemical or sensorial alteration, could be a suitable alternative to chlorine disinfection.
