The litter trait is one of the most important economic traits, and increasing litter size is of great economic value in the pig industry. However, the molecular mechanisms underlying pig litter traits remain elusive. To identify molecular markers and candidate genes for pig litter traits, a genome-wide association study (GWAS) and selection signature analysis were conducted in a Yorkshire pig population. A total of 518 producing sows were genotyped with Illumina Porcine SNP 50 BeadChip, and 1969 farrowing records for the total number born (TNB), the number born alive (NBA), piglets born dead (PBD), and litter weight born alive (LWB) were collected. Then, a GWAS was performed for the four litter traits using a repeatability model. Based on the estimated breeding values (EBVs) of TNB, 15 high- and 15 low-prolificacy individuals were selected from the 518 sows to implement selection signature analysis. Subsequently, the selection signatures affecting the litter traits of sows were detected by using two methods including the fixation index (FST) and θπ. Combining the results of the GWAS and selection signature analysis, 20 promising candidate genes (NKAIN2, IGF1R, KISS1R, TYRO3, SPINT1, ADGRF5, APC2, PTBP1, CLCN3, CBR4, HPF1, FAM174A, SCP2, CLIC1, ZFYVE9, SPATA33, KIF5C, EPC2, GABRA2, and GABRA4) were identified. These findings provide novel insights into the genetic basis of pig litter traits and will be helpful for improving the reproductive performances of sows in pig breeding.

Kaempferol, a flavonol component of plants, is well-known to exhibit multiple bioactivities, such as anti-oxidative and anti-apoptotic effects. However, the underlying mechanisms responsible for the beneficial effects remain elusive. This study was conducted to test the hypothesis that kaempferol attenuated diquat-induced oxidative damage and intestinal barrier dysfunction by ameliorating oxidative damage and apoptosis in intestinal porcine epithelial cells. Compared with the control group, diquat treatment led to enhanced intracellular ROS production, increased mitochondrial depolarization, and apoptosis, which were accompanied by cell cycle arrest at the G1 phase, reduced cell migration, and disrupted intestinal epithelial barrier function. These effects triggered by diquat were reversed by kaempferol. Further study showed that the protective effect of kaempferol was associated with an enhanced mRNA level of genes related to cell cycle progression (cyclin D1, CDK4, and E2F1) and genes implicated in the anti-oxidant system (GSR, GSTA4, and HO-1), up-regulated abundance of tight junctions (ZO-1, ZO-2, occludin, and claudin-4), as well as enhanced Nrf2, an anti-oxidant transcription factor. In conclusion, we revealed a functional role of kaempferol in the intestinal barrier. Ingestion of kaempferol-rich foods might be a potential strategy to improve the integrity and function of enterocytes.

Vanadium phosphate oxide (VPO) heterogeneous catalysts with different V/P molar ratios were prepared and used for selective oxidation of biomass-derived 5-hydroxymethylfurfural (HMF) to produce 2,5-diformylfuran (DFF) in the liquid phase. It was found that the VPO catalyst with V/P molar ratio 0.25 exhibited the best catalytic performance. Then the VPO catalyst was utilized to catalyze the oxidation of HMF in a batch reactor under different conditions, in terms of type of solvent (water and organic), reaction time and temperature. A high DFF yield of 83.6% with HMF conversion of 100% was obtained under atmospheric pressure.
