In this contribution, ethanolic extracts of Cuminum cyminum (C. cyminum) seeds were evaluated in terms of phytochemical content, total phenol and flavonoid contents. As far as the analytical techniques are concerned, UV-Vis, FTIR, HPLC, NMR (1H and 13C) and ESI-MS were performed. The binding capacity of five different antidiabetic enzymes was tested by in silico molecular docking studies. The HPLC, UV-Vis, FTIR, NMR and ESI-MS data highlighted the presence of seven biologically active molecules e.g. α-pinene, ß-pinene, Δ3-carene, ρ-cymene, α-terpineol, cuminaldehyde and linalool. The results coming from the in silico molecular docking studies showed that such phytochemicals present in the cumin seed extracts play an important role in the activity of key enzymes involved in carbohydrate metabolism. Therefore, C. cyminum is proven to be useful for the treatment of diabetes mellitus and its major secondary complications.

Utilization of carbon dioxide (CO2) in thermochemical treatment of waste plastics may significantly help to improve CO2 recycling, thus simultaneously curtailing dioxins/furans and CO2 emissions. Although CO2 is not such an effective gasifying agent as steam, a few investigations have explored the utilization of CO2 in conjunction with steam to achieve somewhat higher carbon conversion. This work presents a comparative evaluation study of CO2 and steam gasification of a typical post-consumer waste plastics mixture using an Aspen Plus equilibrium model. The effect of flow rate of gasifying medium (CO2 and/or steam) and gasification temperature on product gas composition, carbon conversion, and cold gas efficiency has been analyzed. Simulation results demonstrate that CO2 can serve as a potential gasifying agent for waste plastics gasification. The resulting product gas was rich in CO whereas CO2-steam blends yield a wider H2/CO ratio, thus extending the applications of the product gas.

RESUMO

Probiotics are live microbes which when administered in adequate amounts as functional food ingredients confer a health benefit on the host. Their versatility is in terms of their usage which ranges from the humans to the ruminants, pigs and poultry, and also in aquaculture practices. In this review, the microorganisms frequently used as probiotics in human and animal welfare has been described, and also highlighted are the necessary criteria required to be fulfilled for their use in humans on the one hand and on the other as microbial feed additives in animal husbandry. Further elaborated in this article are the sources from where probiotics can be derived, the possible mechanisms by which they act, and their future potential role as antioxidants is also discussed.

In the present work, we have analysed the changes in the reactive oxygen metabolism of Catharanthus roseus (L.) G. Don. plants in terms of H(2)O(2) content, lipid peroxidation and the free radical quenching systems (non-enzymatic and enzymatic antioxidants) under drought stress. In addition to this, the root alkaloid ajmalicine was extracted and quantified from both control and drought stressed plants. The H(2)O(2) content was analysed from both stressed and unstressed control plants. Lipid peroxidation was estimated as thiobarbituric acid reactive substances. The non-enzymatic antioxidants viz., ascorbic acid, alpha-tocopherol and reduced glutathione contents, antioxidant enzymes like superoxide dismutase, ascorbate peroxidase, and catalase were extracted and estimated from the samples. The alkaloid ajmalicine was extracted and quantified from shade dried root samples and found significantly increased over control. From the results of this investigation, it can be concluded that the water deficit areas may be well used for the cultivation of medicinal plants like C. roseus and the economically important alkaloid production can be enhanced in the plant level.

An experiment was conducted in two varieties, rosea and alba, of Catharanthus roseus plants with two watering treatments viz., 100 and 60% of field capacity, to understand the effects of water deficit on early growth, biomass allocation and photosynthetic pigment responses. We found that there were significant differences in early growth, dry matter accumulation and pigment variations between the two varieties. The root length, shoot length, total leaf area, fresh and dry weights were significantly reduced under water stress treatments. There was a significant reduction in the photosynthetic pigment contents in both the varieties. The rosea variety was more affected due to water deficit when compared to alba variety.

A preliminary study was made on four medicinal plants viz., Ocimum sanctum L., Coleus forskholii Briq, Catharanthus roseus (L.) G. Don. and Aloe vera in order to identify and enumerate the rhizosphere, non-rhizosphere and diazotrophic microorganisms in soil. The diazotrophic bacterial population studied includes Azospirillum, Azotobacter and Pseudomonas. The rhizosphere bacterial populations were 23.33 x 10(6)g(-1) in O. sanctum followed by C. roseus (20.46 x 10(6)g(-1)), A. vera (18.44 x 10(6)g(-1)) and C. forskholii (16.64 x 10(6)g(-1)). The fungi populations were 19.44 x 10(4)g(-1) in C. roseus, 18.66 x 10(4)g(-1) in O. sanctum, 16.44 x 10(4)g(-1) in A. vera and 14.22 x 10(4)g(-1) in C. forskholii. The actinomycetes population was 12.22 x 10(5)g(-1) in O. sanctum, 10.44 x 10(5)g(-1) in C. roseus, 8.44 x 10(5)g(-1) in A. vera and 6.22 x 10(5)g(-1) in C. forskholii. The diazotrophic bacterial population of Azospirillum, Azotobacter and Pseudomonas is 8.2 x 10(4)g(-1), 12 x 10(4)g(-1), 6 x 10(4)g(-1) in the rhizosphere soil. In all the four medicinal plants the microbial population is more in the rhizosphere soil, when compared to non-rhizosphere soil. These results are helpful in developing a biofertilizer consortium for these commercially grown medicinal plants.

We have evaluated the ability of two triazole growth regulators, viz. triadimefon (TDM) and hexaconazole (HEX), in the enhancement of tuber production and quality in cassava (Manihot esculenta Crantz) through their effects on carbohydrate metabolism. One litre of 20 mg(-1) TDM and 15 mg(-1) HEX solution per plant were used for the treatments and groundwater was given to control plants. Triazole treatments reduced plant height and leaf area, but increased fresh and dry weights. Plants treated with TDM showed an increased net assimilation rate, which is followed by HEX and control plants. Triazole compounds increased the relative growth rate of cassava after 200 DAP, i.e. in the phase of tuber enlargement. Triazole compounds increased the starch and other carbohydrate contents and carbohydrate metabolising enzyme activities. From the results of this study, it can be concluded that these triazoles can significantly enhance the tuber production and quality by affecting the starch metabolism, apart from their fungicidal properties.

The application of triazole fungicides is a common practice in the cultivation of carrot (Daucus carota L.) plants. It is there for seems important to test the changes that are occurring in this food crop under triazoles, the non-traditional plant growth regulators, treatments in order to identify the extent to which it tolerate the fungicide application and thereby make it an economical food crop. A field experiment was conducted to find out the effects of two triazole fungicides (hexaconazole (HEX) and paclobutrazol (PBZ) at 20mg l(-1) plant(-1)) on the biomass, yield, electrolyte leakage, lipid peroxidation and antioxidant potential of carrot. The treatments were given to plants on 15, 30 and 45 days after sowing (DAS). The plants were uprooted for analyses of growth and biochemical parameters on 60 DAS. It was found that both HEX and PBZ have significant effects on the growth and biochemical parameters of this plant. Among the triazoles used, PBZ performed best in terms of anthocyanin, protein, amino acid, proline, starch and sugar, contents whereas HEX enhanced carotenoids, fresh weight, dry weight and biomass. There was no significant variation in chlorophyll ('a' and 'b') contents between the two triazole treated plants, but HEX and PBZ proved best when compared to untreated control plants. HEX and PBZ increased alpha- and beta-amylases enzymes activities to a significant level. Out of these two triazoles, PBZ performed best in increasing the starch hydrolyzing enzymes activities. The non-enzymatic antioxidant, reduced glutathione (GSH) and antioxidant enzyme ascorbate peroxidase (APX) were increased under fungicide applications. The data suggests that, the application of triazole fungicides may be a useful tool to increase the tuber quality as well as quantity in carrot plants, apart from their fungicidal properties.

In the present investigation, two watering treatments, viz., 100% and 60% field capacity (FC) were used to understand the effects of water deficit on early growth, biomass allocation, pigment and biochemical constituents and proline metabolism of five varieties of Sunflower (Helianthus annuus L.) plants. We found that there was a significant difference in early growth, dry matter accumulation, pigment, biochemical constituents and proline metabolism among the five varieties. The root length, shoot length, total leaf area, fresh and dry weight, chlorophyll a, b, total chlorophyll and carotenoid were significantly reduced under water stress treatments. Water stress increased the proline, free amino acid and glycinebetaine contents along with increased activity of gamma-glutamyl kinase but the activity of proline oxidase reduced as a consequence of water stress.

Mouse embryos deficient in Gata3 die by 11 days post coitum (d.p.c.) from pathology of undetermined origin. We recently showed that Gata3-directed lacZ expression of a 625-kb Gata3 YAC transgene in mice mimics endogenous Gata3 expression, except in thymus and the sympathoadrenal system. As this transgene failed to overcome embryonic lethality (unpublished data and ref. 3) in Gata3-/- mice, we hypothesized that a neuroendocrine deficiency in the sympathetic nervous system (SNS) might cause embryonic lethality in these mutants. We find here that null mutation of Gata3 leads to reduced accumulation of Th (encoding tyrosine hydroxylase, Th) and Dbh (dopamine beta-hydroxylase, Dbh) mRNA, whereas several other SNS genes are unaffected. We show that Th and Dbh deficiencies lead to reduced noradrenaline in the SNS, and that noradrenaline deficiency is a proximal cause of death in mutants by feeding catechol intermediates to pregnant dams, thereby partially averting Gata3 mutation-induced lethality. These older, pharmacologically rescued mutants revealed abnormalities that previously could not be detected in untreated mutants. These late embryonic defects include renal hypoplasia and developmental defects in structures derived from cephalic neural crest cells. Thus we have shown that Gata3 has a role in the differentiation of multiple cell lineages during embryogenesis.

We found previously that neither a 6-kbp promoter fragment nor even a 120-kbp yeast artificial chromosome (YAC) containing the whole GATA-3 gene was sufficient to recapitulate its full transcription pattern during embryonic development in transgenic mice. In an attempt to further identify tissue-specific regulatory elements modulating the dynamic embryonic pattern of the GATA-3 gene, we have examined the expression of two much larger (540- and 625-kbp) GATA-3 YACs in transgenic animals. A lacZ reporter gene was first inserted into both large GATA-3 YACs. The transgenic YAC patterns were then compared to those of embryos bearing the identical lacZ insertion in the chromosomal GATA-3 locus (creating GATA-3/lacZ "knock-ins"). We found that most of the YAC expression sites and tissues are directly reflective of the endogenous pattern, and detailed examination of the integrated YAC transgenes allowed the general localization of a number of very distant transcriptional regulatory elements (putative central nervous system-, endocardium-, and urogenital system-specific enhancers). Remarkably, even the 625-kbp GATA-3 YAC, containing approximately 450 kbp and 150 kbp of 5' and 3' flanking sequences, respectively, does not contain the full transcriptional regulatory potential of the endogenous locus and is clearly missing regulatory elements that confer tissue-specific expression to GATA-3 in a subset of neural crest-derived cell lineages.

GATA-3 is essential for murine embryonic development, but elucidating the genetic controls over the complex temporal and tissue-specific transcriptional regulatory pattern of this transcription factor gene has been problematic. Here we report the isolation and characterization of two yeast artificial chromosomes (YACs) bearing the murine GATA-3 gene. Ordered deletions of both YACs show that they define a 1-megabase pair contig spanning the GATA-3 locus. We found that a 120-kb YAC transgene, including 35 kb of 5' as well as 60 kb of 3' flanking sequence, confers normal GATA-3 expression at sites not revealed previously through analysis of plasmid transgenic lines. However, even this 120-kb YAC does not contain sufficient information to recapitulate the complete GATA-3 expression program during embryogenesis. While not complete in its regulatory capacity, the YAC transgene is nonetheless able to complement several homozygous GATA-3 mutant phenotypes and thereby prolong embryonic life.
