RESUMO

The objective of this study was to evaluate, characterize and quantify the learning experiences and subsequent application of research-based technologies by beef producers upon conclusion of an online extension certification program (44 Farms International Beef Cattle Academy, IBCA). Upon conclusion of the program, paricipants were invited to complete a structured interview. Interview transcripts (n = 19) were coded, categorized, and merged into four overarching themes: Strengths, Struggles, Courses, and Geographical origin. Within Strengths, the most frequent codes were Connections, Application, and Instructor Experience, with 61, 53, and 50 coded segments respectively. Within Struggles, the most frequent codes were Time Management, Level of Knowledge, and Language issues, with 27, 18, and 15 coded segments, respectively. For Courses in the program, the most frequently mentioned were Nutrition, Reproduction, and Genetics, with 35, 28, and 24 coded segments respectively. Correlation between codes was evaluated using Pearson and only statistically significant (P ≤ 0.05) correlations were included in the matrices for network analysis. Interpretation of the generated network analysis map (P ≤ 0.05; Q = 0.468) including all four categories of codes revealed close relationships between Application and the Strengths of Time management, Instructor Experience, and Connections. Application was also directly related to the Courses of Reproduction and Genetics, and the Struggle of Student Engagement and Guidance. Geographical origin was an important factor mediating different correlations. Developing countries (Brazil, Panama, Dominican Republic, and South Africa) were more closely related to the Struggle of Tuition cost, which, in turn was related to the perceived Prestige of the program. In Europe (Romania, Germany, and Kazakhstan), a stronger correlation to the Struggles of Material Relevance and Language Issues was described. Collectively, these results support the positive impact of a comprehensive and interactive extension initiative to leverage application of research-based principles by beef stakeholders around the world. Further, these outcomes indicate that the most valued aspects of the program regarding application are related to interpersonal experience with faculty and peers of the industry (Instructor Experience and Connections) and that perception of struggles and strengths is greatly influenced by socio-cultural aspects of the learning community.

To determine the effect of CaO-treated Pensacola bahiagrass (Paspalum notatum) hay on intake, ruminal fermentation parameters, and apparent total-tract digestibility of nutrients, nine ruminally cannulated Angus-crossbred steers were used in a triplicated 3 × 3 Latin square design. Steers had ad libitum access to either 1) untreated dry hay (DH; n = 8); 2) hay at 50% DM treated with 8.9% CaCO3 (dry matter [DM] basis; CC; n = 9); or 3) hay at 50% DM treated with 5% CaO (DM basis; CO; n = 8). Water was added to reach 50% DM in the CC and CO diets. Ruminal fluid and blood samples were collected every 3 h for 24 h. Ruminal fluid was analyzed for pH, volatile fatty acids (VFA), and ammonia-nitrogen (NH3-N). Blood was analyzed for plasma urea nitrogen (PUN). Hay and fecal samples were collected for 4 d, four times daily for hay and twice daily for feces, to determine apparent total-tract digestibility of nutrients. The hay provided to steers during the digestibility period was analyzed for in vitro organic matter digestibility (IVOMD) for 48 h. Data were analyzed as repeated measures for blood and ruminal fermentation parameters. Total DM intake was not affected (P ≥ 0.674) by treatment. A treatment effect (P < 0.001) was observed for average ruminal pH, where steers consuming CO had the greatest pH (P < 0.001). Ruminal concentration of NH3-N tended (P = 0.059) to be reduced in steers consuming CO. There was a treatment × time interaction (P = 0.023) on concentrations of PUN, where at 3 h DH and CO were lesser than CC (P ≤ 0.050) and at 21 h DH was lesser than CC (P = 0.020). Total VFA, acetate, propionate, butyrate, branched-chain VFA, and valerate concentrations were affected by treatment (P ≤ 0.035), where a reduction (P ≤ 0.034) occurred in steers consuming CO. No treatment differences were observed for total-tract digestibility of DM (P = 0.186), organic matter (P = 0.169), or crude protein (P = 0.152); however, steers consuming DH had greater neutral detergent fiber (P = 0.038) than CC and tended to be greater than CO (P = 0.082). The CO hay had greater (P = 0.005) IVOMD compared with DH and tended (P = 0.100) to be greater than CC. Bahiagrass hay treated with CaO may reduce ruminal fermentation, as indicated by decreased total VFA concentration without altering DM intake. The addition of CaO did not improve the digestibility of bahiagrass hay in vivo; however, in vitro results are contradictory and warrant further elucidation.


With the ever-growing desire to increase efficiency in beef cattle production, researchers have developed strategies such as treating poor-quality forages with chemicals to increase the digestibility of fiber fractions, consequently increasing their energy value for cattle feeding. Calcium oxide has been proposed as a replacement to more caustic chemicals used in the past (e.g., NaOH) and data indicate that it can promote similar and effective outcomes. The current study evaluated the effects of bahiagrass hay treated with calcium oxide on ruminal fermentation parameters, apparent total-tract digestibility of nutrients, and intake by beef steers consuming hay ad libitum as the sole ingredient in their diet. Additionally, in vitro organic matter digestibility was evaluated on the hay provided to steers to assess treatment effectiveness. Results indicated that steers consuming bahiagrass hay treated with calcium oxide had 1) increased pH and reduced volatile fatty acids concentrations in the rumen; 2) reduced or tendency for reduction on total-tract digestibility of fiber fractions; and 3) no effect on intake, all when compared with steers consuming untreated hay. In contrast, in vitro results indicated that organic matter digestibility was increased when the forage was treated with calcium oxide.

As a novel oilseed crop in Florida, Brassica carinata has the capacity of producing high-quality jet biofuel, with a protein-dense meal (~40% crude protein; CP) obtained as a by-product of oil extraction. Characterization of the meal protein is limited, yet necessary for formulation of beef cattle diets; therefore, the objective of this experiment was to determine ruminal and postruminal digestibility of protein from B. carinata. Eight ruminally cannulated Angus crossbred steers (473 ± 119 kg) were used in a duplicated 4 × 4 Latin square design, in which in situ ruminal and postruminal degradability of nutrients were evaluated. The three-step in vitro procedure was used to compare CP and amino acid (AA) degradation in B. carinata meal pellets (BCM) with that of cottonseed meal (CSM), dry distillers grains with solubles (DDGS), and soybean meal (SBM). In situ bags were incubated in the rumen for 0 to 96 hr, with the undegraded supplement remaining after 16 hr subjected to serial in vitro enzymatic solutions. Data were analyzed using the MIXED procedure of SAS. Ruminal rate of degradation of dry matter, organic matter, and CP was greatest (P ˂ 0.01; 10.9, 11.3, and 11.5 %/h, respectively) for SBM. Rumen degradable protein (RDP) content did not differ (P = 0.20; 47.8% and 55.1%, respectively) between CSM and DDGS, but was decreased (P ˂ 0.01) compared with SBM and BCM, which did not differ (P = 0.99; 72.3% and 71.8% RDP, respectively). Compared with DDGS, SBM had greater (P < 0.01) intestinal digestibility of rumen undegradable protein (RUP). Intestinally absorbable digestible protein (IADP) was greatest (P < 0.01) for CSM, with SBM and BCM having the least IADP. Total tract digestibility of CP (TTDP) was greater (P < 0.01) for SBM compared with CSM and DDGS. The contribution of RUP to intestinally absorbable AA was 7.2 and 3.1 g of lysine and methionine per kilogram of CP in BCM, respectively. The evaluation of B. carinata meal as protein supplemented for cattle consuming a forage-based diet resulted in 71.8% RDP and 97.1% TTDP, thus indicating its viability as a high-quality protein supplement for beef cattle.

The objective of this experiment was to investigate the relationship between residual feed intake (RFI) and parameters associated with reproductive efficiency in growing beef heifers. One hundred and seventy-nine replacement beef heifers (Bos taurus and Bos indicus-influenced) were enrolled in a retrospective cohort design. Heifers were assigned to a 70-d feed efficiency test, and results were utilized to retrospectively classify heifers into low (n = 51), medium (n = 66), or high (n = 62) RFI groups based on their individual RFI values. Blood samples were collected weekly throughout the feed efficiency test and breeding season, which were analyzed for plasma concentration of progesterone to determine age at puberty (PUB). By design, residual feed intake differed among RFI groups where high (1.21 ± 0.08 kg/d) had greater (P < 0.01) RFI than medium (0.00 ± 0.08 kg/d) and low RFI groups (-1.49 ± 0.09 kg/d), and medium had greater (P < 0.01) RFI than low RFI heifers. Yet, no differences were observed between breed types for RFI (P = 0.53). Average daily gain (ADG) differed between breed types where Bos taurus heifers had greater ADG (P = 0.02) than B. indicus-influenced heifers. Furthermore, ADG tended (P = 0.08) to differ among RFI groups, where medium RFI heifers tended to have reduced ADG compared with low and high RFI heifers. No differences were determined between breed types for gain-to-feed ratio (G:F; P = 0.20; however, G:F differed among RFI groups and was greater (P < 0.01) in low vs. high, and low vs. medium RFI heifers. The percentage of cycling heifers by the start of the breeding season differed (P = 0.03) by RFI group, where a greater percentage of low RFI heifers were cycling compared with high RFI heifers. A difference was determined on PUB between breed types (P = 0.03), where Bos taurus (393.40 ± 4.64 d) heifers had a reduced PUB compared with B. indicus-influenced (406.90 ± 5.07 d) heifers. In addition, PUB differed by RFI group where low (385.96 ± 6.20 d; P < 0.01) and medium (398.47 ± 5.47 d; P = 0.02) RFI heifers had a reduced PUB compared with High RFI heifers (416.03 ± 5.58 d). In conclusion, more feed efficient heifers attained PUB earlier than less feed efficient heifers. Therefore, selection of heifers for feed efficiency may positively impact reproductive performance of replacement beef heifers.

This 2-yr study evaluated the growth and puberty attainment of Bos indicus-influenced beef heifers offered 2 different postweaning concentrate supplementation amounts and delivery frequencies. On day 0 of each year, 64 Brangus crossbred heifers were stratified by initial body weight (BW) and age (mean = 244 ± 22 kg; 314 ± 17 d) and assigned into 1 of 16 bahiagrass pastures (4 heifers/pasture/yr). Treatments were randomly assigned to pastures in a 2 × 2 factorial design (4 pastures/treatment/yr) and consisted of concentrate dry matter (DM) supplementation at 1.25% or 1.75% of BW which were offered either daily (7×) or 3 times weekly (3×) for 168 d. On day 56 of each year, heifers were assigned to an estrus synchronization protocol consisting of intravaginal controlled internal drug release (CIDR) insertion on day 56, CIDR removal on day 70, i.m. injection of 25 mg of prostaglandin F2α (PGF2α) on day 86, and i.m. injection of 100 µg of gonadotropin-releasing hormone and timed-AI at 66 h after PGF2α injection (day 89). Heifers were exposed to Angus bulls from day 89 to 168 (1 bull/pasture). Pregnancy diagnosis was assessed on day 213 of each year. Supplementation amount × frequency effects were not detected (P ≥ 0.12) for any variable, except for plasma concentrations of glucose (P = 0.10) and urea nitrogen (PUN; P = 0.01). Herbage mass, herbage allowance, and nutritive value did not differ (P ≥ 0.12) among treatments. Increasing supplementation DM amount from 1.25% to 1.75% of BW increased (P ≤ 0.05) plasma concentrations of insulin-like growth factor 1 (IGF-1), overall average daily gain (ADG), final BW, percentage of pubertal heifers on day 89, pregnancy and calving percentages, and percentage of heifers calving within the first 21 d of the calving season. However, reducing the supplementation frequency from daily to 3× weekly, regardless of supplementation amount, did not impact overall pregnancy and calving percentages (P ≥ 0.42), but caused (P ≤ 0.05) fluctuations in plasma concentrations of insulin and IGF-1 and decreased (P ≤ 0.03) overall ADG, final BW, puberty attainment on days 56, 89, and 168, and percentage of heifers calving during the first 21 d of the calving season. Hence, increasing the supplement DM amount did not prevent the negative effects of reducing the frequency of supplementation (3× vs. 7× weekly) on growth and reproduction of replacement Bos indicus-influenced beef heifers.

A replicated 5 × 5 Latin square design with a 2 × 2 + 1 factorial arrangement of treatments was used to determine the effects of bismuth subsalicylate (BSS) and encapsulated calcium ammonium nitrate (eCAN) on ruminal fermentation of beef cattle consuming bahiagrass hay (Paspalum notatum) and sugarcane molasses. Ten ruminally cannulated steers (n = 8; 461 ± 148 kg of body weight [BW]; average BW ± SD) and heifers (n = 2; 337 ± 74 kg of BW) were randomly assigned to one of five treatments as follows: 1) 2.7 g/kg of BW of molasses (NCTRL), 2) NCTRL + 182 mg/kg of BW of urea (U), 3) U + 58.4 mg/kg of BW of BSS (UB), 4) NCTRL + 538 mg/kg of BW of eCAN (NIT), and 5) NIT + 58.4 mg/kg of BW of BSS (NITB). With the exception of NCTRL, all treatments were isonitrogenous. Beginning on day 14 of each period, ruminal fluid was collected and rectal temperature was recorded 4× per day for 3 d to determine ruminal changes every 2 h from 0 to 22 h post-feeding. Ruminal gas cap samples were collected at 0, 3, 6, 9, and 12 h on day 0 of each period followed by 0 h on days 1, 2, 3, and 14. Microbial N flow was determined using Cr-Ethylenediaminetetraacetic acid, YbCl3, and indigestible neutral detergent fiber for liquid, small particle, and large particle phases, respectively. Data were analyzed using the MIXED procedure of SAS. Orthogonal contrasts were used to evaluate the effects of nonprotein nitrogen (NPN) inclusion, NPN source, BSS, and NPN source × BSS. There was no treatment effect (P > 0.05) on concentrations of H2S on day 0, 1, 2, or 14; however, on day 3, concentrations of H2S were reduced (P = 0.018) when NPN was provided. No effect of treatment (P = 0.864) occurred for ruminal pH. There was an effect of NPN source on total concentrations of VFA (P = 0.011), where a 6% reduction occurred when eCAN was provided. There were effects of NPN (P = 0.001) and NPN source (P = 0.009) on the concentration of NH3-N, where cattle consuming NPN had a greater concentration than those not consuming NPN, and eCAN reduced the concentration compared with urea. Total concentrations of VFA and NH3-N were not affected (P > 0.05) by BSS. There was an effect of BSS (P = 0.009) on rectal temperature, where cattle not consuming BSS had greater temperatures than those receiving BSS. No differences for NPN, NPN source, nor BSS (P > 0.05) were observed for microbial N flow. In conclusion, eCAN does not appear to deliver equivalent ruminal fermentation parameters compared with urea, and BSS has limited effects on fermentation.

This experiment was conducted to evaluate effects of estrous expression and intensity of behavioral estrous expression, assessed by evaluating physical activity, on variables associated with fertility in beef cows. The following treatment regimen was imposed on lactating, multiparous cows (n = 273): 100-µg injection of GnRH and an intravaginal progesterone (P4) releasing device (CIDR) administered on d -10, 25-mg injection of prostaglandin F2α at CIDR removal on d -3, and another GnRH injection + fixed-time artificial insemination (TAI) 60-66 h following CIDR removal (d 0). Cows were fitted with a pedometer on d -10, and an estrous detection patch on the tail-head on d -3. Cows were classified as not expressing estrus (NOESTR) or expressing estrus with net physical activity greater (HIESTR) or less than the median (LWESTR). Only data from cows responsive to the estrous synchronization treatment regimen were utilized (NOESTR, n = 119; LWESTR, n = 50; HIESTR, n = 50). Diameter of dominant follicles on d 0 and corpus luteum volume on d 7 were greater in HIESTR compared with LWESTR and NOESTR groups (P < 0.01). Plasma P4 concentrations on d 7 and pregnancy rates to TAI were greater in HIESTR and LWESTR than the NOESTR group (P < 0.01). Hence, if there was estrous expression during the period of the GnRH-based TAI treatment regimen, there were greater pregnancy rates to TAI, whereas when there was greater intensity of behavioral estrous symptoms, there were larger dominant follicles and corpora lutea dimensions but not greater pregnancy rates.

Efforts to understand biological functions and develop management schemes specific to Bos indicus-influenced cattle raised in tropical and subtropical environments are critical to meet the increasing global demand for protein. In the United States, B. indicus breeds are mostly used to generate B. indicus × B. taurus crosses with increased thermal and parasite tolerance, while retaining some productive characteristics of B. taurus cattle. Although crossbreeding represents a proven strategy to improve cattle adaptation almost immediately, research has also attempted to identify B. taurus genetics that can withstand subtropical and tropical climates. Reduced milk production and delayed reproductive maturation appear to be related with tropical adaptation of B. taurus breeds, as a means to conserve energy under stressful conditions and limited nutrition. Moreover, longevity may be the ultimate adaptation response to unfavorable environments, and retention of bulls and heifers from proven cows is the recommended strategy to improve longevity in B. indicus-influenced herds. Besides selection for longevity, other aspects should be considered when planning reproductive management in tropical and subtropical regions. Bos indicus and B. taurus breeds have multiple differences pertaining to reproductive function, including age at puberty, ovarian dynamics, and pregnancy development. Nutritional strategies such as the stair-step regimen, and use of exogenous progesterone (P4) inserts are options to hasten puberty attainment of late-maturing B. indicus-influenced heifers. Yet, limited pharmacological alternatives are available for reproductive management of B. indicus-influenced females in the United States, which rely on GnRH-based protocols not specifically designed to the reproductive function of B. indicus breeds. In contrast, hormonal protocols based on exogenous P4, estradiol esters, and equine chorionic gonadotropin are available for use in B. indicus females in South America. These include protocols tailored to prepubertal heifers, anestrous cows, and cycling nulliparous or parous females, which often yield pregnancy rates of 50% to fixed-time artificial insemination. The global dairy industry also faces similar challenges in increasing demand and production as the beef industry. Selection of cows capable of sustaining optimal milk yield, reproductive success, and health status in hot and humid conditions is essential for optimal dairy production in subtropical and tropical regions.

To evaluate how the inclusion of Bos indicus genotype influences early fetal development in cattle, a reciprocal embryo transfer approach was used in a completely randomized design with a 2 × 2 × 2 factorial arrangement of treatments to generate 55 pregnancies over 2 consecutive years (n = 55). Recipient cows were randomly assigned to (i) a diet that met daily energy maintenance requirements (MAINT) or (ii) a diet that restricted intake to 70% of the energy maintenance requirements (RESTR). Angus (AN) and Brangus (BN) embryo donors were superovulated and artificially inseminated with female sexed-sorted semen from the same breed. Embryos were then randomly transferred to either AN or BN recipients fed their respective diets for 28 d. Recipients remained on the dietary scheme until day 91 of gestation and were then comingled and fed a common diet that met their energy requirements until calving. Measurements included pregnancy establishment at day 28 of gestation, interferon-stimulated genes (ISG) expression in peripheral blood leukocytes, pregnancy-associated glycoproteins (PAG; using two commercial [A1 and A2] and one in-house assay), and fetal crown-to-rump length (CRL). Recipients in the RESTR diet had lower BWs and BCS (diet × day; P < 0.01) than MAINT recipients. Energy-restricted AN recipients experienced greater (recipient breed × diet, P < 0.01) pregnancy failure by day 28 than the other recipient breed × diet combinations. Restricted recipients that received AN embryos experienced greater pregnancy failure than RESTR recipients receiving BN embryos (embryo breed × diet; P = 0.03). No relevant differences were observed in ISG expression (P > 0.10). Recipients that received BN embryos had greater plasma concentrations of PAG in both A1 (embryo breed × day, P < 0.01) and A2 (embryo breed; P < 0.01). Alternatively, recipients that received AN embryos had greater plasma concentrations of PAG for the in-house assay (embryo breed × day; P < 0.01). In addition, fetuses from AN recipients had greater CRL on day 91 (breed × day, P < 0.01). In summary, Bos taurus cows experienced greater pregnancy failure when nutrient restricted. Furthermore, fetal size and the profile of PAG production during early gestation differed between B. indicus-influenced and B. taurus cattle.

Seminal plasma has conventionally been viewed as a transport and survival medium for mammalian sperm; however, its role now extends beyond this process to actively targeting female tissues. Studies in rodents, swine, and humans demonstrate that seminal plasma induces molecular and cellular changes within the endometrium or cervix following insemination. Seminal-plasma-induced alterations to the maternal environment have been theorized to facilitate embryo development, modulate maternal immunity toward the conceptus, and potentially improve pregnancy success. It is unknown if bovine seminal plasma modulates the uterine environment following insemination in the cow, where routine use of artificial insemination reduces maternal exposure to seminal plasma. We hypothesize that seminal plasma modulates the expression of inflammatory mediators in the endometrium, altering the maternal environment of early pregnancy. In vitro, seminal plasma altered intact endometrial explant expression of CSF2, IL1B, IL6, IL17A, TGFB1, IFNE, PTGS2, and AKR1C4. Furthermore, endometrial epithelial cell CSF2, CXCL8, TGFB1, PTGS2, and AKR1C4 expression were increased after seminal plasma exposure, while endometrial stromal cell CSF2, IL1B, IL6, CXCL8, IL17A, TGFB1, PTGS2, and AKR1C4 expression were increased following seminal plasma exposure. Endometrial expression of IL1B was increased in the cow 24 h after uterine infusion of seminal plasma, while other evaluated inflammatory mediators remained unchanged. These data indicate that seminal plasma may induce changes in the bovine endometrium in a temporal manner. Understanding the role of seminal plasma in modulating the maternal environment may aid in improving pregnancy success in cattle.

Our objectives were to determine the effects of the administration of recombinant bovine somatotropin (bST) at the initiation of a fixed-time AI (TAI) protocol on concentrations of plasma IGF-1, follicle diameter, embryo/fetal size, and pregnancy rates in replacement beef heifers. Four hundred and fourteen Angus-based beef heifers were enrolled in a completely randomized design at 4 locations from January to July of 2016. All heifers were exposed to the 7-d CO-Synch + controlled internal drug release (CIDR) protocol where they received a 100-µg injection of GnRH and a CIDR insert on day -9, 25 mg of PGF2α at CIDR removal on day -2, followed by a 100-µg injection of GnRH and TAI 54 ± 2 h later on day 0. Within location, all heifers were randomly assigned to 1 of 2 treatments: 1) heifers that received 650 mg of bST on day -9 (BST; n = 191); or 2) heifers that did not receive bST on day -9 (CONTROL; n = 223). Blood samples were collected on day -9, 0, 28, and 60 to determine the plasma concentrations of IGF-1. Follicle diameter was determined on day -2 and 0 by transrectal ultrasonography. Pregnancy was diagnosed via transrectal ultrasonography on day 28 or 35, and again at least 30 d after the end of the breeding season. Embryo morphometry was assessed by measuring crown-to-rump length (CRL) on day 28, and fetal size was assessed by measuring crown-to-nose-length (CNL) on day 60. Concentrations of plasma IGF-1 did not differ between treatments on day -9 (P = 0.924), 28 (P = 0.075), and 60 (P = 0.792); however, concentrations of plasma IGF-1 were greater (P < 0.001) in BST-treated heifers at TAI (372.4 ± 16.6 vs. 193.7 ± 16.6 ng/ml). No differences (P = 0.191) were detected for follicle diameter between CONTROL and BST treatments on day -2 or 0. Pregnancy rates to TAI (PR/AI) were greater (P = 0.028) for CONTROL compared to BST heifers (42.5 ± 4.0 vs. 29.9 ± 4.1%). No differences (P = 0.536) in CRL were observed on day 28 between CONTROL and BST heifers. In addition, no difference (P = 0.890) was observed for CNL between CONTROL and BST treatments. Final pregnancy rates did not differ (P = 0.699) between treatments. The administration of bST to beef heifers at the initiation of a TAI protocol increased plasma concentrations of IGF-1 at TAI; however, failed to enhance follicle diameter, embryo/fetal size, and reduced PR/AI.

Two experiments were conducted to evaluate the effects of a high concentrate, s.c. PGF2α compared with a conventionally concentrated, i.m. PGF2α in estrus synchronization protocols for heifers. In Exp. 1, 869 Angus-based beef heifers were enrolled at 8 locations. All heifers were exposed to the 7-d CO-Synch + controlled internal drug release (CIDR) estrus synchronization protocol. On day 7 of the protocol heifers received 100 µg of GnRH i.m., and a CIDR insert for 7 d. On day 0, at CIDR removal, estrous detection patches were applied to heifers and, within location, heifers randomly received 1 of 2 PGF2α treatments: 5 mL of Lutalyse i.m. (CONTROL; n = 434) or a 2 mL of Lutalyse HighCon s.c. (HiCON; n = 435). A second GnRH injection was administered at 54 ± 2 h and heifers were fixed-time AI (TAI). Heifers were evaluated for estrous activity at TAI by determining the activation of estrous detection patches. Pregnancy rates to AI (PR/AI) were diagnosed by transrectal ultrasonography between 35 and 55 d after TAI. The percentage of heifers exhibiting estrus between day 0 and TAI did not differ (P = 0.68) between CONTROL and HiCON treatments (47 vs. 46 ± 4%, respectively). Additionally, PR/AI were similar (P = 0.65) between CONTROL and HiCON treatments (46 vs. 45 ± 3%). In Exp. 2, 190 Angus-based beef heifers were enrolled at 2 locations. Heifers were exposed to the melengestrol acetate (MGA)-PGF2α protocol where they were offered 0.5 mg MGA per day from days 1 to 14. On day 33, heifers were randomly assigned to receive CONTROL (n = 95) or HiCON (n = 95) treatment, and estrous detection aids were applied. Heifers were exposed to AI 12 h after detection of estrus. Heifers not detected in estrus at location 1 received a second PGF2α injection 6 d after the initial PGF2α injection and were placed with fertile bulls. Heifers at location 2 that did not express estrus were administered 100 µg of GnRH i.m. and exposed to TAI 96 h after the initial PGF2α injection. Transrectal ultrasonography was used to diagnose PR/AI between 51 and 57 d after the initial PGF2α injection. The percentage of heifers exhibiting estrus during the estrus detection period was similar (P = 0.40) between CONTROL and HiCON treatments (82 vs. 87 ± 4%). Furthermore, PR/AI were similar (P = 0.62) between CONTROL and HiCON treatments (60 vs. 65 ± 5%). In summary, the 2 concentrations and corresponding routes of administration of PGF2α were similar in efficacy at synchronizing estrus in beef heifers.

To determine the effects of administration of 25 mg of PGF2α 7 d prior to the initiation of the 7-d CO-Synch + controlled internal drug release (CIDR) fixed-time AI (TAI) protocol, 985 Bos taurus beef heifers were enrolled in a completely randomized design at 9 locations from April to July of 2016. Within location, all heifers were randomly assigned to 1 of 2 treatments: 1) CONTROL (n = 496); 100 µg injection of GnRH and a CIDR insert for 7 d [day 7], administration of 25 mg of PGF2α at CIDR removal [day 0], followed by a second injection of GnRH and TAI 54 ± 2 h later; or 2) PRESYNCH (n = 489); same as CONTROL but heifers received an additional injection of 25 mg of PGF2α 7 d prior [day 14] to CIDR insertion. Estrous detection patches were applied to all heifers on day 14 and were evaluated for estrual activity on day 7. Similarly, estrus alert patches were placed on all heifers on day 0 and evaluated for estrual activity at the time of TAI. Pregnancy was diagnosed via transrectal ultrasonography between 35 and 55 d after TAI. The percentage of heifers exhibiting estrus between days 14 and 7 was greater (P < 0.001) for the PRESYNCH (70.1 ± 2.4%) than the CONTROL (41.1 ± 2.3%) treatment, whereas the percentage of heifers exhibiting estrus between day 0 and TAI was greater (P < 0.001) for the CONTROL (55.6 ± 2.4%) than the PRESYNCH (39.7 ± 2.5%) treatment. Estrus response rates differed (P < 0.001) among locations. Pregnancy rates to TAI differed (P = 0.023) among locations; however, they did not differ (P = 0.739) between CONTROL and PRESYNCH treatments (45.4 ± 2.5 vs. 43.2 ± 2.5%, respectively). Final breeding season pregnancy rates did not differ (P = 0.811) between treatments. Therefore, an injection of PGF2α 7 d prior to initiation of the 7-d CO-Synch + CIDR protocol failed to improve pregnancy rates to TAI in replacement beef heifers.

Even with advancements in pre- and post-harvest food safety, Shiga toxin-producing Escherichia coli (STEC) still present challenges to human health. Since cattle are the primary reservoir for STEC, lowering the prevalence of this pathogen in farm animals may reduce STEC outbreaks in humans. However, because many of the factors that modulate the colonization and persistence of STEC in cattle remain unknown, reducing STEC in this host is challenging. In this study, we evaluated a cohort of beef cattle one to eleven years of age to determine the effect of animal age on the prevalence of STEC. During the first year of sample collection, heifers had significantly lower STEC prevalence than cows (37.5% vs. 70%). In the second year of sample collection, STEC prevalence peaked in cows that were two years of age and tended to decrease as animals became older. In addition, by studying a subset of the animals in both years, we observed an increase in STEC prevalence from 40.6% to 53.1% in heifers, whereas cows had a net decrease in STEC prevalence from 71.4% to 61.9%. The results from this study indicate that animal age is a significant factor that influences the prevalence of STEC in cattle. These findings have implications for the development of on-farm mitigation strategies by targeting animals with the highest risk of shedding; it could be possible to reduce pathogen transmission among cattle and prevent zoonotic or foodborne transmission to humans.

Estimations of world population growth indicate that by the year 2050 we will reach nine billion habitants on earth. These estimates impose a tremendous challenge in the current agricultural systems as food supply will need to increase by 100 % in the next 40 years (Food and Agriculture Organization of the United Nations 2009). Beef will be a primary protein source that will assist in meeting the requirements for a portion of the protein in diets of this expanding global populace. Beef is a high-quality protein that contains all essential amino acids for the human body and also contains additional essential nutrients such as iron, zinc, B vitamins, riboflavin, selenium, choline, and conjugated linoleic acid (CLA). Adopting reproductive technologies at greater rates than currently used is a viable method to dramatically enhance production efficiency of beef cattle enterprises.Artificial insemination (AI), estrous synchronization and fixed-time AI (TAI), semen and embryo cryopreservation, multiple ovulation and embryo transfer (MOET), in vitro fertilization, sex determination of sperm or embryos, and nuclear transfer are technologies that are used to enhance the production efficiency of beef operations. In many cases, the development of these technologies is responsible for significant changes to traditional livestock production practices. However, adoption of these technologies appears to has not grown at the same rate in the United States as other formidable beef producing nations. For example, sales of beef semen for AI increased from 3.3 to 11.9 million units between 1993 and 2011 in Brazil, whereas that in the United States has increased from 2.9 to 3.8 million units during the same period. The significant increases in adoption of reproductive technologies in developing countries is likely as a result of the development of practical estrous synchronization and TAI systems that have allowed beef producers the opportunity to eliminate detection of estrus in their AI programs with a high degree of success. In the United States, slow adoption rates of these technologies may result in a future loss of international market share of beef products as other nations take advantage not only of the additional kilogram of beef that can be produced but also the improved quality of beef that can be realized through incorporation of reproductive technologies and resultant genetic improvement. However, current difficulties the US producers have with the incorporation of applied reproductive technologies, such as TAI, MOET, and sex semen, must not be the reason to overlook and incorporate more traditional reproductive technologies such as castration, breeding season management, or weaning. In many cases, beef producers in the United States fail to incorporate these more traditional technologies, which results in a reduction in production efficiency of the US beef industry. This chapter will focus on both traditional and more developed reproductive technologies that will play a role in enhancing future production efficiencies of the US beef cattle production system.

At weaning, heifers should be considered for replacements based on their dam's previous performance; heifer calving date, age, and weight; and previous exposure to implants. Before breeding, heifers should be selected as replacements based on whether they have attained puberty (determined by a prebreeding examination), do not have abnormal pelvic areas, or fail to meet temperament standards. After breeding, heifers should be selected as replacements if they conceive early in the breeding season, are capable of achieving 85% of their mature weight by calving, and calve at a body condition of 5.5 to 6.0.

Controlling the prevalence of Escherichia coli O157 in cattle at the pre-harvest level is critical to reduce outbreaks of this pathogen in humans. Multilayers of factors including the environmental and bacterial factors modulate the colonization and persistence of E. coli O157 in cattle that serve as a reservoir of this pathogen. Here, we report animal factors contributing to the prevalence of E. coli O157 in cattle. We observe the lowest number of E. coli O157 in Brahman breed when compared with other crosses in an Angus-Brahman multibreed herd, and bulls excrete more E. coli O157 than steers in the pens where cattle were housed together. The presence of super-shedders, cattle excreting >10(5) CFU/rectal anal swab, increases the concentration of E. coli O157 in the pens; thereby super-shedders enhance transmission of this pathogen among cattle. Molecular subtyping analysis reveal only one subtype of E. coli O157 in the multibreed herd, indicating the variance in the levels of E. coli O157 in cattle is influenced by animal factors. Furthermore, strain tracking after relocation of the cattle to a commercial feedlot reveals farm-to-farm transmission of E. coli O157, likely via super-shedders. Our results reveal high risk factors in the prevalence of E. coli O157 in cattle whereby animal genetic and physiological factors influence whether this pathogen can persist in cattle at high concentration, providing insights to intervene this pathogen at the pre-harvest level.

The objectives of the present experiment were to determine whether supplementation with progesterone (LO, 1 ng mL(-1) or HI, 100 ng mL(-1)) during either the first (Culture-1, Day 1 to 3) or second (Culture-2, Day 4 to 7) phase of culture of in vitro-produced embryos alters embryo development, embryo metabolism or blastocyst cell number. The percentage of oocytes that cleaved, the percentage of cleaved embryos that developed to the morula stage or greater, the blastocyst stage or greater or the hatched blastocyst stage were similar among treatments. Quantities of glucose metabolised per blastocyst per hour were similar, but when metabolic data was normalised for numbers of cells in each blastocyst, the LO treatment during Culture-2 metabolised more glucose (P=0.03) compared with all other treatments. Embryos receiving LO progesterone tended to have greater (P=0.085) metabolism of glucose compared with embryos receiving HI progesterone. Quantities of pyruvate oxidised per blastocyst per hour, and per cell, were similar among treatments. The number of cells per blastocyst in the control group was increased (P=0.039) compared with cells in progesterone-treated groups. In conclusion, supplementation with progesterone during the culture of in vitro-produced embryos does not appear to improve embryo characteristics.

We performed a series of comparative studies of bull and stallion seminal plasma (SP) and its role on sperm-neutrophil binding as well as the interaction between semen extender and seminal DNase. Because of contrasting roles of SP on sperm-neutrophil binding between horses and cattle, it was suspected there were some species-specific differences on sperm interaction with SP proteins due to the variations in the natural location of semen deposition (uterus compared to vagina). Bull frozen-thawed sperm removed from egg yolk extender showed similar results to fresh sperm, but this also caused extensive sperm agglutination unless SP or egg yolk was included. If similar agglutination occurs after AI with frozen bull semen, it could interfere with sperm transport or sperm functions. Commonly used bull semen extenders were poor media for seminal DNase activity on plasmid DNA degradation, raising the prospect that the same may be true with other SP factors important to fertility. DNase activity per mg SP protein of bulls was less than that of horses (P<0.05), but DNase activity associated with bull sperm was greater (P<0.05) indicating a different affinity of DNase to spermatozoa. This could be related to the fact that bull sperm naturally migrate from the vagina to the uterus leaving the bulk of SP behind. In such migration, sperm cells needed to carry DNase and other SP factors along. Incorporation of egg yolk in bull semen and introducing SP into the uterus of cattle with current AI protocols may contribute to reduced fertility. Modifications of semen extender and/or semen processing should be examined to allow sperm cells a maximum potential for fertilization.
