RESUMO

A previously unpublished reaction of precipitation in agarose between histone and non-histone proteins extracted in saline buffer from nuclei of human skin tumors, is reported. Two bands of precipitation similar to those in an immunodiffusion reaction between NHP and specific antisera, were observed. The reaction described is very similar to the affinodiffusion reaction of glycoproteins and lectins in agarose.

RESUMO

The object of the research is to classify the nuclear proteins of the cellular nucleus of some types of human skin tumours (melanotic and amelanotic melanomas, squamous epitheliomas). Nuclear proteins were extracted from the nuclei with a saline buffer. Nuclear proteins were extracted from a melanoma step by step. After prolonged centrifugation to sediment the DNA and the large ribonucleoprotein complexes, the nuclear proteins were chromatographed on columns of Biorex 70+ to remove proteins were chromatographed on columns of Biorex 70+ to remove the histones. Nuclear proteins from a squamous epithelioma were injected into a rabbit to induce specific antibodies.

RESUMO

In this communication we present the immunoelectrophoretic profiles of non-histone proteins of the cellular nucleus of some types of human skin tumours. The results show that the nuclear non-histone proteins of different types of tumours are quantitatively different from one another. The immunoelectrophoretic profiles of the samples show that an antigenic affinity between the skin tumours concerns most of the nuclear proteins of each sample.

RESUMO

The purpose of this study is to classify the membrane proteins from some cutaneous tumours (basal cell epitheliomas, squamous epitheliomas, melanomas) and possibly detect the ones which are characteristic of each type of tumour. The microsomal, mitochondrial and nuclear fractions were purified following the usual techniques. The membrane proteins were solubilized with sodium dodecyl sulfate, treated with iodoacetamide and chromatographed in SDS-acrylamide slab-gel.

RESUMO

The microsomal, mitochondrial, and nuclear fractions of cutaneous tumors have been investigated. Cutaneous tumors were homogenized and microsomal, mitochondrial, and nuclear fractions were purified. The membrane proteins were solubilized with sodium duodecyl sulfate (SDS). The membrane lipids were removed and membrane proteins were solubilized again in a small volume of SDS solution and chromatographed in SDS-acrylamide slab-gel. The plates were stained with Coomassie Brilliant Blue to show protein bands. The preliminary results show that the electrophoretic profiles of microsomal proteins are characteristic of some tumors.
