Fecal glucocorticoid metabolites (FGMs) have been used as a non-invasive and indirect measurement of the complex stress response in a variety of species. Animals in facilities under managed care allow for the longitudinal study of FGMs in a controlled environment. Animal histories often include environmental, husbandry, and medical notes that can be matched to FGM concentrations to aid in the physiological validation of adrenal stimulation and response. The goal of this study was to demonstrate analytical validations using two enzyme-linked immunosorbent assays (EIA) to measure FGMs from northern sea otters (Enhydra lutris kenyoni) under human care (Seattle Aquarium, Seattle, WA, USA) and to determine baseline and stress response spike levels for individual sea otters. Individual variation was found among the four subjects in the study with fecal baseline levels ranging from 20.2 to 83.7 ng/g for cortisol-immunoreactive metabolites and 52.3 to 102 ng/g for corticosterone-immunoreactive metabolites. As a retrospective study, 39 percent of hormone peaks were associated with notes and most FGM spikes were associated with veterinary procedures or days in which enrichment items were provided and produced an excitatory response. Monitoring baseline FGMs levels and events associated with hormone peak values may provide insight into effective husbandry management to improve the overall welfare of sea otters and other marine mammals.

Human use of marinescapes is rapidly increasing, especially in populated nearshore regions where recreational vessel traffic can be dense. Marine animals can have a physiological response to such elevated human activity that can impact individual health and population dynamics. To understand the physiological impacts of vessel traffic on baleen whales, we investigated the adrenal stress response of gray whales (Eschrichtius robustus) to variable vessel traffic levels through an assessment of fecal glucocorticoid metabolite (fGC) concentrations. This analysis was conducted at the individual level, at multiple temporal scales (1-7 days), and accounted for factors that may confound fGC: sex, age, nutritional status, and reproductive state. Data were collected in Oregon, USA, from June to October of 2016-2018. Results indicate significant correlations between fGC, month, and vessel counts from the day prior to fecal sample collection. Furthermore, we show a significant positive correlation between vessel traffic and underwater ambient noise levels, which indicates that noise produced by vessel traffic may be a causal factor for the increased fGC. This study increases knowledge of gray whale physiological response to vessel traffic and may inform management decisions regarding regulations of vessel traffic activities and thresholds near critical whale habitats.

Mexico is a country that makes heavy use of the shark populations that inhabit the southern portion of the Northeast Pacific Ocean (NEP). Shark meat has become an essential food source in this country, while shark fins are used to supply traditional Asian markets. In addition to consumptive utilization, charismatic shark species support an ecotourism industry that has gained significance in several tourist resorts across the country. In this concluding chapter, we recap the contents of chapters included in volumes 83 and 85 in the Advances in Marine Biology series. The chapters in these volumes address biodiversity, conservation genetics, trophic ecology, migratory movements, fisheries, and shark ecotourism, allowing us to understand the state of knowledge relevant to human: shark interactions in the Mexican Pacific. We discuss the challenges for the sustainable use and conservation of sharks in the southern NEP and highlight the need for a more holistic management approach that includes economic and social factors. To meet these challenges, we recommend updating the Mexican National Plan of Action for Sharks published in, 2004, such that it may continue serving as a roadmap for the conservation and management of sharks in the southern NEP during the years to come.

Sharks are iconic, sometimes apex, predators found in every ocean. Because of their ecological role as predators and concern over the stability of their populations, there has been an increasing amount of work focused on shark conservation around the world in recent decades. The populations of sharks that reside in the Northeast Pacific (NEP) Ocean bordering the west coast of the United States reside in one of the most economically and ecologically important oceanic regions in the world. Volume 78 of Advances in Marine Biology (AMB) is a companion to Volume 77, which focused primarily on NEP shark biodiversity, organismal biology, and ecology. Volume 78 highlights fisheries and the conservation implications of fisheries management; shark population modelling and the conservation impacts of these models given that many life history metrics of NEP sharks necessary to accurately run these models are still unknown; the value of captive sharks to the biology, outreach, and conservation of NEP sharks; and the conservation value of citizen science and shark ecotourism. Together these volumes encapsulate the current state of the knowledge for sharks in the NEP and lay the foundation for protecting, managing, and learning from these species in the face evolving natural conditions and societal opinions.

Because of their regional significance to fisheries and ecological role as predators, and as a result of concern over long-term stability of their populations, there has been an increasing amount of work focused on shark conservation in recent decades. This volume highlights the biodiversity and biological attributes of, and conservation efforts targeted at, populations of sharks that reside in the Northeast Pacific Ocean bordering the west coast of the United States and Canada, one of the most economically and ecologically important oceanic regions in the world. A companion volume addresses details of fisheries and ecotourism in the same region, as well as delving into the relationship between captive husbandry of sharks and education/outreach efforts aimed at fostering a conservation mindset in the public at large. Together, these volumes provide readers a detailed backdrop against which to consider their own actions, and those of resource managers, academics, and educators, as they relate to the long-term conservation of sharks and their relatives.

Conservation genetics is an applied science that utilizes molecular tools to help solve problems in species conservation and management. It is an interdisciplinary specialty in which scientists apply the study of genetics in conjunction with traditional ecological fieldwork and other techniques to explore molecular variation, population boundaries, and evolutionary relationships with the goal of enabling resource managers to better protect biodiversity and identify unique populations. Several shark species in the northeast Pacific (NEP) have been studied using conservation genetics techniques, which are discussed here. The primary methods employed to study population genetics of sharks have historically been nuclear microsatellites and mitochondrial (mt) DNA. These markers have been used to assess genetic diversity, mating systems, parentage, relatedness, and genetically distinct populations to inform management decisions. Novel approaches in conservation genetics, including next-generation DNA and RNA sequencing, environmental DNA (eDNA), and epigenetics are just beginning to be applied to elasmobranch evolution, physiology, and ecology. Here, we review the methods and results of past studies, explore future directions for shark conservation genetics, and discuss the implications of molecular research and techniques for the long-term management of shark populations in the NEP.

Using simple substrates, this methodology explores the scope and reactivity of a new magnesium catalyst for an aziridination reaction capable of building chirality and complexity simultaneously.

The captive husbandry of giant Pacific octopuses Enteroctopus dofleini is well understood, but their endocrine signatures are not well documented. The major vertebrate reproductive hormones--estrogen, progesterone, and testosterone--and the stress-related hormone corticosterone are relatively well known for many vertebrate species. However, few studies on these hormones within invertebrates have been conducted. Our hypothesis was that endocrine signatures within octopuses are similar to those found within vertebrates in response to reproductive activity and stress. Using standard immunoassay techniques, we measured fecal steroids within fecal samples collected from five female and three male giant Pacific octopuses housed at the Seattle Aquarium. The mean estrogen level ranged from 3.67 to 99.39 ng/g of feces, progesterone ranged from 44.35 to 231.71 ng/g feces, testosterone ranged from 9.30 to 18.18 ng/g feces, and corticosterone ranged from 10.91 to 22.14 ng/g feces. The results suggest that octopus fecal hormones are similar to those in vertebrates and may be useful in measuring ovarian activity and stress within captive female giant Pacific octopuses.

Conditions for the phosphoric acid-catalyzed highly enantioselective ring-opening of meso-aziridines with a series of functionalized aromatic thiol nucleophiles are described. The procedure utilizes commercially available aromatic thiols, a series of meso-aziridines, and a catalytic amount of VAPOL phosphoric acid to explore the substrate scope of this highly enantioselective reaction.
