RESUMO

A radioimmunoassay (RIA) has been developed for the determination of picogram amounts of aescine, an anti-inflammatory and anti-oedematous glycoside mixture from unpurified extracts of Aesculus hippocastanum. Practically no interference is observed for various potentially crossreacting compounds. This RIA covers the range 100 pg-50 ng, within which acceptable accuracy and precision are obtained.

RESUMO

A radioimmunological method was developed for determining total and free corticosterone in rat plasma. This method was used to determine the dose-response curve of corticosterone and to measure the elimination and study the half-lives of total and free corticosterone in rat plasma with a dose of 5 mg/kg. The elimination with a dose of 5 mg/kg, when drawn on the half-logarithmic scale, formed a straight line. The half-lives for total and free corticosterone were 25 and 15 min, respectively.

RESUMO

A specific radioimmunoassay (RIA) has been developed for the determination of picogram amounts of trospiumchloride, an anticholinergic agent, from unpurified serum and urine samples. Practically no interference was observed for various potential crossreacting compounds tested. The method is sensitive and excellent values for accuracy, precision and correlation were obtained.

RESUMO

An enzyme-linked immunosorbent assay for vincristine was developed, based on a new procedure for synthesizing the hapten-protein conjugate. In both the immunogen and the enzyme tracer a spacer group was introduced between the hapten and protein, and the vincristine was coupled at a site far from its functional groups. The antibody produced proved to be exceptionally specific as compared with previous immunoassays for bis-indole alkaloids. Thousandfold antibody dilutions could be used and samples at the femtomole range were assayable. Applications of the method to patient plasma samples and to plant material are described.

RESUMO

A radioimmunoassay has been developed for the analysis of atropine ( DL-hyoscyamine) from unpurified ethanolic extracts of Atropa belladonna L. The antiserum used was highly specific for D-hyoscyamine, the unnatural enantiomer. Consequently, a racemization step had to be included in the assay procedure in order to obtain the analysis of atropine. The method is rapid and sensitive, giving excellent values for accuracy, precision and correlation. There was practically no interference in the assay from other compounds present in the extracts.

RESUMO

Radioimmunoassay (RIA) has been used in the analysis of digoxigenin glycoside contents of crude ethanolic extracts prepared from fresh leaf samples of Digitalis lanata Ehrh. The commercial antidigoxin antiserum used in the assay was highly specific against the digoxigenin aglycone. Steroid derivatives with only slight structural differences from digoxigenin remained practically unassayed. The method is highly sensitive, and excellent values for accuracy, precision and correlation were obtained. Digoxigenin glycoside levels of second-year D. lanata leaves were found to be considerably higher than those of frist-year leaves.
