ABSTRACT: Background : It is reported that circVMA21 has an inhibition effect on sepsis-induced acute kidney injury (AKI). Therefore, the underlying molecular mechanisms of circVMA21 in AKI are worthy of further investigation. Material and Methods : Lipopolysaccharide (LPS) was used to induce HK2 cell injury. CircVMA21, miR-337-3p and ZEB2 expression was tested by qRT-PCR. Cell growth was detected by CCK8 assay, EdU assay, and flow cytometry. Protein levels were examined by western blot. The levels of inflammatory factors and oxidative stress markers were measured to evaluate cell inflammatory response and oxidative stress. RNA relationship as verified by dual-luciferase reporter assay, RIP assay, and RNA pull-down assay. Results : CircVMA21 had decreased expression in AKI patients. Overexpressed circVMA21 alleviated LPS-induced HK2 cell inflammation, apoptosis, and oxidative stress. Moreover, circVMA21 sponged miR-337-3p, and miR-337-3p targeted ZEB2. The inhibitory effect of circVMA21 on LPS-induced HK2 cell injury was reversed by miR-337-3p overexpression, and ZEB2 overexpression abolished the promotion effect of miR-337-3p on LPS-induced HK2 cell injury. Conclusions : CircVMA21 could inhibit LPS-induced HK2 cell injury via miR-337-3p/ZEB2 axis.

This study is aimed to explore the value of lymphocyte subsets in evaluating the severity and prognosis of sepsis. The counts of lymphocytes, CD3+ T cells, CD4+ T cells, CD8+ T cells, CD19+ B cells, and NK cells significantly decreased between day 1 and day 3 in both the survivor and the non-survivor groups. The peripheral lymphocyte subsets (PLS) at day 1 were not significantly different between the survivor and the non-survivor groups. However, at day 3, the counts of lymphocytes, CD3+ T cells, CD4+ T cells, and NK cells were remarkably lower in the non-survivor group. No significant differences in CD8+ T cells, or CD19+ B cells were observed. The PLS index was independently and significantly associated with the 28-day mortality risk in septic patients (OR: 3.08, 95% CI: 1.18-9.67). Based on these clinical parameters and the PLS index, we developed a nomograph for evaluating the individual mortality risk in sepsis. The area under the curve of prediction with the PLS index was significantly higher than that from the model with only clinical parameters (0.912 vs. 0.817). Our study suggests that the decline of PLS occurred in the early stage of sepsis. The new novel PLS index can be an independent predictor of 28-day mortality in septic patients. The prediction model based on clinical parameters and the PLS index has relatively high predicting ability.

Some typical defects like delamination, fiber fracture, non-uniform resin distribution and springback are inevitable as titanium-based thermoplastic FMLs are formed by general stamping process at elevated temperature due to the significant differences of various constituent materials in mechanical and thermal properties as well as deformation mechanism. Thus, a novel ultrasonic impact sizing/shaping process method was proposed in the present work, in which a stepped horn and impact tools with a cylindrical tip end was designed by means of vibration modal and harmonic response analysis, and the corresponding experiment setup was established to verify the process mentioned above. Moreover, ultrasonic impact tests were carried out for titanium-based FMLs stacked by titanium sheet, thermoplastic resin film and carbon fiber reinforced fabric to reveal their sizing/shaping mechanism and the effect of key process parameters such as ultrasonic amplitude, scanning speed of impact tool and its tip end diameter on the surface and interface characteristics of titanium-based FMLs. The research results show that the ultrasonic impact scheme with the ultrasonic amplitude of 12 µm, the scanning speed of impact tool within 2 mm/s âˆ¼ 3 mm/s, the tip end diameter of impact tool within Ø4mm∼Ø6mm are proper for the ultrasonic impact sizing/shaping process of titanium-based FMLs.

Boron nitride nanosheets (BNNSs) have been widely used in many fields due to their excellent properties. However, low preparation rates and difficulty in functionalization hinder their further development. This study proposes a novel glucose-assisted ultrasonic cavitation exfoliation (GAUCE) method with glucose as an auxiliary solution to prepare BNNSs. Results show that the method has a high preparation yield of 55.58%, which is higher than the average preparation yield of 33.86%. The mechanism of preparing BNNSs by GAUCE was also investigated. The exfoliation of BNNSs was achieved using the energy of ultrasonic cavitation bubble collapse, which will break the interlayer forces in h-BN. The grafting of hydroxyl groups decomposed by glucose on the edge and surface of BNNSs during cavitation prevented the re-aggregation of the nanosheets, thereby increasing the exfoliation yield of BNNSs. In addition, the contact angle of BNNSs prepared by GAUCE was reduced, and the hydrophilicity was greatly improved.
