RESUMO

Crop diseases caused by fungi threaten food security and exacerbate the food crisis. Inspired by the application of fungicide candidates from natural products in agrochemical discovery, a series of luotonin A derivatives were designed, synthesized, and evaluated for their antifungal activities against five plant fungi. Most of these compounds exhibited significant fungicidal activity against Botrytis cinerea in vitro with EC50 values less than 1 µg/mL. Among them, compounds w7, w8, w12, and w15 showed superior antifungal activity against B. cinerea with EC50 values of 0.036, 0.050, 0.042, and 0.048 µg/mL, respectively, which were more potent than boscalid (EC50 = 1.790 µg/mL). Preliminary mechanism studies revealed that compound w7 might pursue its antifungal activity by disrupting the fungal cell membrane and cell wall. Moreover, in vivo bioassay also indicated that compound w7 could be effective for the control of B. cinerea. The above results evidenced the potential of luotonin A derivatives as novel and promising candidate fungicides.

To study the in vivo inhibition effect of ginsenoside Rh2 on humanleukemia cells, and explore its mechanism from autophagy and apoptosis aspects, human leukemia K562 cells allograft tumor models were applied, and after administration of ginsenosides Rh2 by gavage, the tumor diameter, volume and inhibitory rate were measured, and the anti-tumor activity of ginsenosides Rh2 was observed. The levels of HAT and HDAC in tumor tissues were detected by chemical colorimetry assay, and expressions of HDAC1, HDAC2, HDAC3, HDAC4, HDAC5 and HDAC6 were detected by Western blotting assay. The expression levels of vital genes closely associated with autophagy and mRNA expressions of HDAC6 and Hsp90 were detected by Real time-PCR. HE staining was used to observe apoptosis, and immunohistochemistry was used to detect the protein expressions of HDAC6, Hsp90 and activated caspases 3. The results showed that ginsenoside Rh2 could inhibit the growth of k562 cells allograft tumor, with a tumor inhibition rate up to 53.10%. Ginsenoside Rh2 could significantly decrease HDAC activity and decrease the expressions of HDAC1, HDAC2 and HDAC6, and inhibit the expressions of HDAC6 and HSP90, increase the expressions of vital autophagy genes (beclin-1, LC3A and LC3B). Histopathological results showed that ginsenosides Rh2 could significantly increase the tumor apoptosis. Therefore, ginsenoside Rh2 had good anti-tumor effect in vivo, and the mechanism maybe associated with regulating autophagy and apoptosis through HDAC6 and Hsp90 pathways and inhibiting the in vivo proliferation of tumor cells.

Snow grain size is a key parameter not only to affect the energy budget of the global or local region but also characterizing the status of snow vapor transport and temperature gradient. It is significant to monitor and estimate the snow grain size in large area for global or local climate change and water resource management. Recently, remote sensing technology has become a useful tool for snow grain size monitoring and estimating. In the present paper, the estimate models were built based on simulating the snow surface spectral reflectance curve in visible-infrared region and the sensitive bands and snow indices for snow grain size were selected. These models help estimate snow grain size by hyperspectral remote sensing. Through validating with ground true data, the results show that these models have higher explorative accuracy using 1 030, 1 260 nm and normalized difference snow index (460 and 1 030 nm). In addition, the correlation slopes of estimated and observed valves are 1.37, 0.61 and 0.62, respectively. R2 are 0.82, 0.86 and 0.93 and RMSE are 55.65, 50.83 and 35.91 microm, respectively. The result can provide a scientific basis for snow grain size monitoring and estimating.
