RESUMO

Neuroinflammation is associated with poor outcomes in patients with spinal cord injury (SCI). Recent studies have demonstrated that stimulator of interferon genes (Sting) plays a key role in inflammatory diseases. However, the role of Sting in SCI remains unclear. In the present study, it is found that increased Sting expression is mainly derived from activated microglia after SCI. Interestingly, knockout of Sting in microglia can improve the recovery of neurological function after SCI. Microglial Sting knockout restrains the polarization of microglia toward the M1 phenotype and alleviates neuronal death. Furthermore, it is found that the downregulation of mitofusin 2 (Mfn2) expression in microglial cells leads to an imbalance in mitochondrial fusion and division, inducing the release of mitochondrial DNA (mtDNA), which mediates the activation of the cGas-Sting signaling pathway and aggravates inflammatory response damage after SCI. A biomimetic microglial nanoparticle strategy to deliver MASM7 (named MSNs-MASM7@MI) is established. In vitro, MSNs-MASM7@MI showed no biological toxicity and effectively delivered MASM7. In vivo, MSNs-MASM7@MI improves nerve function after SCI. The study provides evidence that cGas-Sting signaling senses Mfn2-dependent mtDNA release and that its activation may play a key role in SCI. These findings provide new perspectives and potential therapeutic targets for SCI treatment.

Drastic changes in the random load of an electromechanical system bring about a reliability problem for the proportional solenoid valve based on a thermal effect. In order to accurately and effectively express the thermal load of a proportional solenoid valve under random load conditions and to meet the requirements of online acquisition, adaptive anomaly detection, and the missing substitution of thermal load data, a thermal load prediction model based on the Kalman filter algorithm is proposed. Taking the compound operation process of an excavator as the object and based on the field testing of an excavator and the independent testing experiment of a proportional solenoid valve in a non-installed state, a method of obtaining historical samples of the proportional solenoid valve's power and thermal load is given. The k-means clustering algorithm is used to cluster the historical samples of the power and thermal load corresponding to the working posture of a multi-tool excavator. The Grubbs criterion is used to eliminate the outliers in the clustering samples, and unbiased estimation is performed on the clustering samples to obtain the prediction model. The results show that the cross-validation of the sample data under the specific sample characteristics of the thermal load model was carried out. Compared with other methods, the prediction accuracy of the thermal load model based on the Kalman filter is higher, the adaptability is strong, and the maximum prediction deviation percentage is stable within 5%. This study has value as a reference for random cycle thermal load analyses of low-frequency electromechanical products.

The accumulation of arsenic (As) in rice poses a significant threat to food safety and human health. Breeding rice varieties with low As accumulation is an effective strategy for mitigating the health risks associated with arsenic-contaminated rice. However, the genetic mechanisms underlying As accumulation in rice grains remain incompletely understood. We evaluated the As accumulation capacity of 313 diverse rice accessions grown in As-contaminated soils with varying As concentrations. Six rice lines with low As accumulation were identified. Additionally, a genome-wide association studies (GWAS) analysis identified 5 QTLs significantly associated with As accumulation, with qAs4 being detected in both of the experimental years. Expression analysis demonstrated that the expression of LOC_Os04g50680, which encodes an MYB transcription factor, was up-regulated in the low-As-accumulation accessions compared to the high-As-accumulation accessions after As treatment. Therefore, LOC_Os04g50680 was selected as a candidate gene for qAs4. These findings provide insights for exploiting new functional genes associated with As accumulation and facilitating the development of low-As-accumulation rice varieties through marker-assisted breeding.

Background: In China, the number of preterm infants is the second largest globally. Compared with those in developed countries, the mortality rate and proportion of treatment abandonment for extremely preterm infants (EPIs) are higher in China. It would be valuable to conduct a multicenter study and develop predictive models for the mortality risk. This study aimed to identify a predictive model among EPIs who received complete care in northern China in recent years. Methods: This study included EPIs admitted to eighteen neonatal intensive care units (NICUs) within 72 hours of birth for receiving complete care in northern China between January 1, 2015, and December 31, 2018. Infants were randomly assigned into a training dataset and validation dataset with a ratio of 7:3. Univariate Cox regression analysis and multiple regression analysis were used to select the predictive factors and to construct the best-fitting model for predicting in-hospital mortality. A nomogram was plotted and the discrimination ability was tested by an area under the receiver operating characteristic curve (AUROC). The calibration ability was tested by a calibration curve along with the Hosmer-Lemeshow (HL) test. In addition, the clinical effectiveness was examined by decision curve analysis (DCA). Results: A total of 568 EPIs were included and divided into the training dataset and validation dataset. Seven variables [birth weight (BW), being inborn, chest compression in the delivery room (DR), severe respiratory distress syndrome, pulmonary hemorrhage, invasive mechanical ventilation, and shock] were selected to establish a predictive nomogram. The AUROC values for the training and validation datasets were 0.863 [95% confidence interval (CI): 0.813-0.914] and 0.886 (95% CI: 0.827-0.945), respectively. The calibration plots and HL test indicated satisfactory accuracy. The DCA demonstrated that positive net benefits were shown when the threshold was >0.6. Conclusions: A nomogram based on seven risk factors is developed in this study and might help clinicians identify EPIs with risk of poor prognoses early.

N6-methyl-adenosine (m6a) is involved in the occurrence and development of various diseases such as autogenic immune disease and tumors. Methyltransferases regulate primary (pri)-microRNA (miRNA/miR) processing by mediating m6a modifications, consequently affecting pathological processes including immune-related diseases by regulating both innate and adaptive immune cells. However, the roles of m6a on the biological functions of bone marrow mesenchymal stem cells (BMSCs) remain to be elucidated. The relative expression levels of methyltransferase-like 14 (METTL14) and other methyltransferases, demethylases, and miR-873 in bone samples from patients with osteoporosis and from normal individuals were measured by reverse transcription-quantitative PCR. Cell Counting Kit-8 assay was used to examine the proliferation of BMSCs. Co-immunoprecipitation (Co-IP) was used to investigate the binding of METTL14 to DiGeorge syndrome critical region 8 (DGCR8). RNA immunoprecipitation (RIP) was used to examine the binding of METTL14 to pri-miR-873. METTL14 and m6a modifications were highly detected in patients with osteoporosis compared with the controls. Co-IP results indicated that silencing of METTL14 reduced METTL14 and m6a modification levels in BMSCs. Downregulation of METTL14 significantly promoted the proliferation of BMSCs. RIP results suggested that METTL14/m6a methylation modification promoted the processing of pri-miR-873 by binding to DGCR8 in BMSCs. Furthermore, overexpression of miR-873 inhibited the proliferation of BMSCs. The results also showed that miR-873 mimics significantly inhibited the proliferation in small interfering (si)-METTL14 transfected BMSCs; however, miR-873 inhibitors markedly promoted the proliferation of si-METTL14 transfected BMSCs. METTL14 and m6a modifications were upregulated in osteoporosis samples. METTL14 promoted the processing of pri-miR-873 into mature miR-873 by regulating m6a modification. Furthermore, overexpression of miR-873 significantly inhibited the proliferation of BMSCs. Therefore, the METTL14/m6a/miR-873 axis may be a potential target for the treatment of osteoporosis.

Tea plants (Camellia sinensis) typically contain high-flavonoid phytochemicals like catechins. Recently, new tea cultivars with unique purple-colored leaves have gained attention. These purple tea cultivars are enriched with anthocyanin, which provides an interesting perspective for studying the metabolic flux of the flavonoid pathway. An increasing number of studies are focusing on the leaf color formation of purple tea and this review aims to summarize the latest progress made on the composition and accumulation of anthocyanins in tea plants. In addition, the regulation mechanism in its synthesis will be discussed and a hypothetical regulation model for leaf color transformation during growth will be proposed. Some novel insights are presented to facilitate future in-depth studies of purple tea to provide a theoretical basis for targeted breeding programs in leaf color.

The light-sensitive albino tea plant can produce pale-yellow shoots with high levels of amino acids which are suitable to process high-quality tea. In order to understand the mechanism of the albino phenotype formation, the changes in the physio-chemical characteristics, chloroplast ultrastructure, chlorophyll-binding proteins, and the relevant gene expression were comprehensively investigated in the leaves of the light-sensitive albino cultivar 'Huangjinya' ('HJY') during short-term shading treatment. In the content of photosynthetic pigments, the ultrastructure of the chloroplast, and parameters of the photosynthesis in the leaves of 'HJY' could be gradually normalized along with the extension of the shading time, resulting in the leaf color transformed from pale yellow to green. BN-PAGE and SDS-PAGE revealed that function restoration of the photosynthetic apparatus was attributed to the proper formation of the pigment-protein complexes on the thylakoid membrane that benefited from the increased levels of the LHCII subunits in the shaded leaves of 'HJY', indicating the low level of LHCII subunits, especially the lack of the Lhcb1 might be responsible for the albino phenotype of the 'HJY' under natural light condition. The deficiency of the Lhcb1 was mainly subject to the strongly suppressed expression of the Lhcb1.x which might be modulated by the chloroplast retrograde signaling pathway GUN1 (GENOMES UNCOUPLED 1)-PTM (PHD type transcription factor with transmembrane domains)-ABI4 (ABSCISIC ACID INSENSITIVE 4).

Direct absorption of sunlight and conversion into heat by uniformly dispersed photothermal nanofluids has emerged as a facile way to efficiently harness abundant renewable solar-thermal energy for a variety of heating-related applications. As the key component of the direct absorption solar collectors, solar-thermal nanofluids, however, generally suffer from poor dispersion and tend to aggregate, and the aggregation and precipitation tendency becomes even stronger at elevated temperatures. In this review, we overview recent research efforts and progresses in preparing solar-thermal nanofluids that can be stably and homogeneously dispersed under medium temperatures. We provide detailed description on the dispersion challenges and the governing dispersion mechanisms, and introduce representative dispersion strategies that are applicable to ethylene glycol, oil, ionic liquid, and molten salt-based medium-temperature solar-thermal nanofluids. The applicability and advantages of four categories of stabilization strategies including hydrogen bonding, electrostatic stabilization, steric stabilization, and self-dispersion stabilization in improving the dispersion stability of different type of thermal storage fluids are discussed. Among them, recently emerged self-dispersible nanofluids hold the potential for practical medium-temperature direct absorption solar-thermal energy harvesting. In the end, the exciting research opportunities, on-going research need and possible future research directions are also discussed. It is anticipated that the overview of recent progress in improving dispersion stability of medium-temperature solar-thermal nanofluids can not only stimulate exploration of direct absorption solar-thermal energy harvesting applications, but also provide a promising means to solve the fundamental limiting issue for general nanofluid technologies.

STUDY DESIGN: Retrospective cohort study. OBJECTIVES: To evaluate the effect of zoledronic acid, an anti-osteoporosis treatment, during the perioperative period on vertebral body bone mineral density (BMD) after spinal fusion surgery in postmenopausal women with osteoporosis. METHODS: The medical records of postmenopausal patients with osteoporosis who underwent instrumented intervertebral fusion for lumbar degenerative disease between July 2016 and May 2018 were reviewed. Patients with comorbidities or condition which might affect bone metabolism were excluded. Forty-six patients did not receive anti-osteoporosis treatment before surgery and during the postoperative follow-up (untreated group). Another 46 patients who was treated with zoledronic acid perioperatively were matched for age and body mass index to patients in the untreated group. Preoperative and postoperative dual-energy X-ray absorptiometry (DEXA) records and lumbar BMD values of the involved spinal segments and of the cephalad levels, as well as of the femoral neck were recorded. RESULTS: A significant decrease of cephalad vertebral BMD values was observed in the untreated group (-11.47%, P < 0.001), with a slight decrease of the femoral neck (-1.28%, P > 0.05). Zoledronic acid prevented rapid bone loss after instrumented intervertebral fusion surgery, with a bone loss in the cephalad levels of -0.76 ± 4.71% compared to -11.47 ± 16.45% in the untreated group (P < 0.001). while the change in BMD of the femoral neck in the treated group was 1.52 ± 5.88% compared to -1.28 ± 6.58% in the untreated group (P = 0.036). CONCLUSIONS: Perioperative zoledronic acid treatment may offer protection against a significant decrease in BMD of cephalad vertebrae after spinal fusion surgery among postmenopausal women with osteoporosis.

The performance of peat biomethanation was investigated in bioelectrochemical anaerobic digestion at different applied voltages, and compared to conventional anaerobic digestion. The methane yield was stabilized at 16 mL/g peat in the conventional anaerobic digestion. However, in the bioelectrochemical anaerobic digestion, the methane yield was significantly increased to 264 mL/g peat at the applied voltage of 4 V, followed by 1 V, 2 V, 0.5 V and 0 V. The bioelectrochemical system could enrich more electroactive microorganisms on the electrode, as well as in the bulk solution, and further improve the direct interspecies electron transfer for methane production. The 16S rRNA analysis showed a significant increase in the abundance of specific microorganisms in the bulk solution, including Firmicutes phylum and Proteobacteria phylum, in addition to a gradual increase in acetoclastic methanogenesis with an increase in applied voltage. These results provide a solution to turn low-rank coal into a new alternative energy.

Catechins are a cluster of polyphenolic bioactive components in green tea. Anticarcinogenic effects of tea catechins have been reported since the 1980s, but it has been controversial. The present paper reviews the advances in studies on the anticarcinogenic activities of tea and catechins, including epidemiological evidence and anticarcinogenic mechanism. Tea catechins showed antagonistic effects on many cancers, such as gynecological cancers, digestive tract cancers, incident glioma, liver and gallbladder cancers, lung cancer, etc. The mechanism underlying the anticarcinogenic effects of catechins involves in inhibiting the proliferation and growth of cancer cells, scavenging free radicals, suppressing metastasis of cancer cells, improving immunity, interacting with other anticancer drugs, and regulating signaling pathways. The inconsistent results and their causes are also discussed in this paper.

We evaluated the effect of dietary supplementation with Moringa oleifera leaf extract on the resistance to Aeromonas hydrophila infection in crucian carp. The fish were randomly divided into five groups: the basal diet, the basal diet supplied with 0.25% (0.25 M), 0.5% (0.5 M), 0.75% (0.75 M) and 1.0% M. oleifera leaf extract (1.0 M) for 8 weeks. The growth, antioxidant capabilities, related immune genes as well as resistance to A. hydrophila infection were determined. The results showed that compared with the control group, the weight gain, specific growth rate in the group of 0.5% M. oleifera leaf extract, serum superoxide dismutase (SOD), albumin (ALB) and glutathione peroxidase (GSH-Px), the gene expression of hepatopancreas BTB and CNC homolog 1 (Bach1), NF-E2-related factor 2 (Nrf2), peroxidases (PRX) and NADPH oxidase (NOX) in the group of 0.5%-1.0% M. oleifera leaf extract increased, while feed conversion ratio, serum cortisol, red blood cell (RBC), alanine aminotransferase (ALT), malonaldehyde (MDA) decreased in the group of 0.5%-1.0% M. oleifera leaf extract before the stress. After the infection, the group of 0.5% or 0.75% M. oleifera leaf extract also could improve the serum ALB, hepatopancreas Kelch-like-ECH-associated protein 1 (Keap1), Bach1, Nrf2, TOR, PRX and NOX and reduce cortisol compared with the control group. In summary, this study suggested that 0.5% M. oleifera leaf extract inclusion increased the growth performance, even had positive effects on physiological and immune function, and enhanced resistance against pathogenic infections in crucian carp. The optimum level of M. oleifera leaf extract for crucian carp was estimated to be 0.35%-0.48% based on polynomial comparison with FCR and SGR.

Rabbit Hemorrhagic Disease (RHD) is an economically significant infectious disease of rabbits, and its infection causes severe losses in the meat and fur industry. RHD Virus (RHDV) is difficult to proliferate in cell lines in vitro, which has greatly impeded the progress of investigating its replication mechanism and production of inactivated virus vaccines. RHDV VP60 protein is a major antigen for developing RHD subunit vaccines. Herein, we constructed a TK-deactivated recombinant Swinepox virus (rSWPV) expressing VP60 protein and VP60 protein coupled with His-tag respectively, and the expression of foreign proteins was confirmed using immunofluorescence assay and western blotting. Transmission electron microscopy showed that the recombinant VP60, with or without His-tag, self-assembled into virus-like particles (VLPs). Its efficacy was evaluated by comparison with available commercial vaccines in rabbits. ELISA and HI titer assays showed that high levels of neutralizing antibodies were induced at the first week after immunization with the recombinant strain and were maintained during the ongoing monitoring for the following 13 weeks. Challenge experiments showed that a single immunization with 106 PFU of the recombinant strain protected rabbits from lethal RHDV infection, and no histopathological changes or antigenic staining was found in the vaccine and rSWPV groups. These results suggest that rSWPV expressing RHDV VP60 could be an efficient candidate vaccine against RHDV in rabbits.

The contribution analysis of different electron transfer pathways to CH4 production was investigated in bioelectrochemical anaerobic digestion (BEAD). It demonstrates that the indirect interspecies electron transfer (IIET) pathway and the direct interspecies electron transfer (DIET) pathways contributed to 41.7 % and 58.3 % of the CH4 production in the BEAD reactor, respectively. The DIET pathway was further divided into DIET via electrode (eDIET) and biological DIET (bDIET) in the bulk solution, and contributed 11.1 % and 47.2 % of CH4 production, respectively. This indicates that the dominant electron transfer pathway for CH4 production is from the bulk solution, rather than on the polarized electrode. The electroactive microorganisms were well enriched in the bulk solution by the electric field generated between anode and cathode. The enriched electroactive microorganisms significantly improved the CH4 production in the bulk solution through the bDIET pathway.

Following the publication of this paper, it was drawn to the Editors' attention by a concerned reader that certain of the data panels showing results from flow cytometric experiments in Figs. 2D, 4D and 5D, and tumor images shown in Fig. 7A, were strikingly similar to data that had appeared in different form in other articles by different authors. Owing to the fact that the contentious data in the above article had already been published elsewhere, or were already under consideration for publication, prior to its submission to International Journal of Oncology, the Editor has decided that this paper should be retracted from the Journal. The performance of the bioelectrochemical anaerobic digestion (BEAD) reactor was investigated with different carbon material-modified electrodes for the methanation of maize straw. The carbon material-modified electrodes used titanium (Ti) mesh modified with carbon nanotube (CNT), carbon black (CB), and activated carbon (AC). The maximum cumulative methane production obtained in the Ti-CNT reactor was (616.4 ± 9.3) mL/g VS, while the maximum methane production rate in the Ti-AC reactor was (61.9 ± 1.0) mL/g VS.d.The electroactive bacteria were well enriched by the different electrodes, and the enriched electroactive bacteria further facilitate the direct interspecies electron transfer (DIET) for methane production. Additionally, we found the phylum Firmicutes showed a linear relationship to methanogenic performance, as well as the Genus Proteiniborus. The Ti-CNT electrode shows better performance by the electrochemical analysis. These findings provide critical knowledge for the large-scale use of the BEAD process and the treatment of maize straw.

QUESTIONS/PURPOSES: The aim of this study is to (1) describe the epidemiological characteristics of spine injuries in electric bicycles related collisions, (2) develop clinical guideline of spine injuries in electric bicycles related collisions and (3) support the new road safety policy for electric bicycle riders. METHODS: A retrospective review of spine injuries in electric bicycle related collisions was performed from 86 patients in an urban trauma center between 2018 and 2020. The variables including gender, age, radiographic findings, associated injuries, neurologic injuries, treatment, average length of stay were fully collected. Chi-square test and paired sample mean t-test were used to test for statistically significant differences. All statistical analyses were performed using Statistical Product and Service Solutions 20.0.0 software. A P<0.001 was considered as significant. RESULTS: A total of 86 cases were involved in electric bicycles related collisions lead to spine injuries. The spine injuries victims were predominantly male (79.07%) and middle-aged (41-60years, 44.19%). The most common spine injuries were L1 fractures (10.48%). The most common fracture type was AO Fracture Classification type A (71.40%). Age and multivertebral fractures had significant difference between patients who suffered from an ASOI and from those who did not (P-value: 0.005, 0.005). There was significant difference between patients who suffered from neurologic injuries and from those who did not in AO Classification, multivertebral fractures and ASOI (P-value: 0.0001, 0.001, 0.032). Age, AO Classification, multivertebral fractures, ASOI and neurologic injuries had significant difference to influence patients' ISS and ALOS. CONCLUSIONS: Spine injuries in electric bicycles related collisions may make patients suffer from severe consequences including their potential permanent disability, economic cost, or even life safety. Patients with spine injuries in electric bicycles related collisions should be paid close attention to avoid missed diagnosis and misdiagnosis. Helmet, protective clothing or other safety gear should be required to use to provide greater protection for electric bicycle riders.

Adverse medical events (AMEs) often occur in the healthcare workplace, and studies have shown that a positive atmosphere can reduce their incidence by increasing peer report intention. However, few studies have investigated the effect and action mechanism therein. We aimed to extend upon these studies by probing into the relationship between hospital climate and peer report intention, along with the mediating effect of attribution tendency and moderating effects of rewards. For this purpose, a cross-sectional survey was administered in a hospital among health professionals. We collected 503 valid questionnaires from health professionals in China and verified the hypothesis after sorting the questionnaires. The results of empirical analysis show that a positive hospital climate significantly induces individual internal attribution tendency, which in turn exerts a positive effect on peer report intention. Contract reward also helps to increase peer report intention, especially for health professionals with an internal attribution tendency. The findings contribute to the literature regarding AME management in hospitals by providing empirical evidence of the necessity for hospital climate and contract reward, and by providing insights to improve their integrated application.

BACKGROUND: Rice (Oryza sativa L.) is an important crop and a staple food for half of the population around the world. The recent water and labor shortages are encouraging farmers to shift from traditional transplanting to direct-seeding. However, poor germination and slow elongation of the coleoptile constrains large-scale application of direct-seeding. OBJECTIVE: This study was aimed to investigate the genetic basis of the anaerobic germination (AG) potential using a set of Oryza nivara (O. nivara) introgression lines (ILs). METHODS: In this study, a total of 131 ILs were developed by introducing O. nivara chromosome segments into the elite indica rice variety 93-11 through advanced backcrossing and repeated selfing. A high-density genetic map has been previously constructed with 1,070 bin-markers. The seeds of ILs were germinated and used to measure coleoptile length under normal and anaerobic conditions. QTLs associated with AG potential were determined in rice. RESULTS: Based on the high-density genetic map of the IL population, two QTLs, qAGP1 and qAGP3 associated with AG tolerance were characterized and located on chromosomes 1 and 3, respectively. Each QTL explained 15% of the phenotypic variance. Specifically, the O. nivara-derived chromosome segments of the two QTLs were positively tolerance to anaerobic condition by increasing coleoptile length. In a further analysis of public transcriptome data, a total of 26 and 36 genes within qAGP1 and qAGP3 were transcriptionally induced by anaerobic stress, respectively. CONCLUSIONS: Utilization of O. nivara-derived alleles at qAGP1 and qAGP3 can potentially enhance tolerance to anaerobic stress at the germination stage in rice, thereby accelerating breeding of rice varieties to be more adaptative for direct-seeding.

BACKGROUND: The frequencies that can evoke strong steady state visual evoked potentials (SSVEP) are limited, which leads to brain-computer interface (BCI) instruction limitation in the current SSVEP-BCI. To solve this problem, the visual stimulus signal modulated by trinary frequency shift keying was introduced. OBJECTIVE: The main purpose of this paper is to find a more reliable recognition algorithm for SSVEP-BCI based on trinary frequency shift keying modulated stimuli. METHODS: First, the signal modulated by trinary frequency shift keying is simulated by MATLAB. At different noise levels, the empirical mode decomposition, singular value decomposition, and synchrosqueezing with the short-time Fourier transform are used to extract the characteristic frequency and reconstruct the signal. Then, the coherent method is used to demodulate the reconstructed signal. Second, in the paradigm of BCI using trinary frequency shift keying modulated stimuli, the three methods mentioned above are used to reconstruct EEG signals, and canonical correlation analysis and coherent demodulation are used to recognize the BCI instructions. RESULTS: For simulated signals, it is found that synchrosqueezing with short-time Fourier transform has a better effect on extracting the characteristic frequencies. For the EEG signal, it is found that the method combining synchrosqueezing with short-time Fourier transform and coherent demodulation has a higher accuracy and information translate rate than other methods. CONCLUSION: The method combining synchrosqueezing with short-time Fourier transform and coherent demodulation proposed in this paper can be applied in the SSVEP system based on trinary frequency shift keying modulated stimuli.
