RESUMO

In current study, curcumin-loaded bioactive nanocomplexes (Cur NCs) (2 %, 5 %, 8 %, and 11 %) were used to prepare corn starch (CS)-based composite films (CS-Cur NCs). Fourier-transform infrared spectroscopy, X-ray diffraction and scanning electron microscopy revealed that Cur NCs were uniformly dispersed in the polymer matrix via physical interaction. Moreover, the mechanical, gas barrier, hydrophobicity, optical, and thermal properties and the antioxidant activity of composite films were potentially improved with the addition of Cur NCs. Subsequently, CS-based film with 11 % Cur NCs exhibited high antioxidant activity (the scavenging rates of DPPH and ABTS are 50.07 % ± 0.82 % and 65.26 % ± 1.60 %, respectively) and was used for packaging blueberries. Compared with the control, the CS-Cur NCs packaging treatment effectively improved the appearance and nutrition of blueberries, and maintained the high activity of several antioxidant enzymes. Furthermore, CS-Cur NCs packaging treatment significantly improved the ascorbic acid (AsA) and glutathione (GSH) levels, thus regulating the AsA-GSH cycle system and suppressing the accumulation of reactive oxygen species (ROS). In summary, the CS-Cur NCs packaging could effectively conserve the postharvest quality of blueberries by improving antioxidant enzyme activity and suppressing excessive accumulation of ROS, which contributes to the development of bioactive packaging and provides novel insights into the preservation of blueberries. This work demonstrates that the development of active packaging is promising to absorb the oxidative radicals from food, and protect the food from inherent and external factors, thus enhancing the quality, security, and shelf-life of the food during storage.

Soil environmental safety has received much attention during the past few decades due to its significance in agricultural production and human health. Special attention is required for soil pesticide residues and ecological risks. This study examined 197 soil samples from industrial, residential and agricultural areas for the presence of 12 organophosphorus pesticides (OPPs) and 8 synthetic pyrethroids (SYPs) in the 16 cities in Henan Province, and the center of CPUA, based on the Central Plains Urban Agglomeration (CPUA) concept proposed by China. The total average concentrations of ∑12OPPs in industrial, residential and agricultural soils were 194, 217, 267 ng/g dry weight, and those of ∑8SYPs were 26.8, 35.7, 25.5 ng/g dry weight, respectively. The two pollutants with the greatest concentrations in the soils were malathion and fenpropathrin, respectively, the dominant components of OPPs and SYPs. The soil environmental carrying capacity (SECC) analysis, representing the maximum residual load that can be supported, shows that acephate and cyhalothrin were overloaded, with a predicted period of over 500 years. Among the 16 cities of CPUA, a higher frequency of high ecological risk could be observed only in Shangqiu. The OPPs in children had total non-carcinogenic risk values of more than 1.0. Similarly, the non-carcinogenic risks of SYPs in adults and children in the residential areas were more than 1.0. The study provides knowledge on how to effectively manage soil safety in Henan Province, which is the center of the CPUA, with a large population and grain province to protect ecosystems and reduce the risks of soil pesticide residues in humans.

Scutellaria barbata D. Don 1825 is an important medicinal plant distributed in wetlands about 2000 m above sea level and used to treat various diseases. The complete chloroplast genome of S. barbata is 152,050 bp with four subregions consisting of a large single-copy region (84,053 bp), a small single-copy region (17,517 bp), and a pair of inverted repeats (25,240 bp). In the chloroplast genome of S. barbata, 131 genes were detected, comprising 87 protein-encoding genes, eight ribosomal RNA (rRNA) genes, and 36 transfer RNA (tRNA) genes. Phylogenetic analysis based on the complete chloroplast genome and protein-coding DNA sequences of 27 related taxa of the genus (out group included Holmskioldia sanguinea and Tinnea aethiopica) indicates that S. barbata was made a clade with S. orthocalyx, and S. meehanioides was a sister to them. The first chloroplast genome of S. barbata was reported in this work, serving as a potential reference for important medicinal plants within the Scutellaria genus.

BACKGROUND: Tobramycin (TOB), an essential aminoglycoside antibiotic in human life, poses potential threats due to its residues in the environment. The primary concern is the adverse impact of excessive TOB on human kidneys, hearing, and other organs, significantly affecting human health. Constructing a sensitive electrochemical platform for simple and rapid trace detection is crucial. Herein, to enhance the sensitivity of TOB detection in the environment and mitigate the risks associated with residual antibiotics, an ultrasensitive electrochemical aptasensor was developed. RESULTS: The sensor employs a dual-cycle amplification strategy involving catalytic hairpin assembly (CHA) and exonuclease III (Exo III) for efficient signal amplification. Simultaneously, the electrode performance was optimized by incorporating gold nanowires (AuNWs) onto the surface of reduced graphene oxide (PDA-rGO). Specifically, in the presence of TOB, which binds to the aptamer (Apt), dsDNA dissociates, releasing cDNA to open hairpin 1 (HP1) and initiate the CHA cycle with the participation of hairpin 2 (HP2). Exo III shears HP1 in the HP1/HP2 complex, freeing HP2 to participate in the CHA cycle again. Ultimately, a significant amount of signal label is retained on the electrode by hybridizing with sheared HP1, generating a robust electrical signal. SIGNIFICANCE: Through the signal amplification strategy, the aptasensor design provides a broad linear range of 0.005-500 nM, with a low detection limit of 0.112 pM for TOB. It is worth mentioning that the aptasensor displayed favorable stability, specificity, and reproducibility, and has been successfully applied to practical samples, demonstrating its utility in practical applications.

Isolation and analysis of double-stranded RNA (dsRNA) from the phytopathogenic fungus Setosphaeria turcica f. sp. zeae revealed the presence of a new double-stranded RNA (dsRNA) virus, tentatively named "Setosphaeria turcica polymycovirus 2" (StPmV2). The genome of StPmV2 consists of five segments (dsRNA1-5), ranging in size from 965 bp to 2462 bp. Each dsRNA contains one open reading frame (ORF) flanked by 5' and 3' untranslated regions (UTRs) with conserved terminal sequences. The putative protein encoded by dsRNA1 shows 64.52% amino acid sequence identity to the RNA-dependent RNA polymerase (RdRp) of the most closely related virus, Cladosporium cladosporioides virus 1, which belongs to the family Polymycoviridae. dsRNAs 2-4 encode the putative coat protein, methyltransferase (MTR), and proline-alanine-serine-rich protein (PASrp), respectively, and dsRNA5 encodes a protein of unknown function. Phylogenetic analysis based on the RdRp protein indicated that StPmV2 clustered with members of the family Polymycoviridae and is therefore a new mycovirus belonging to the genus Polymycovirus in the family Polymycoviridae. In addition, three other distinct isolates of StPmV2 were identified: one isolated from S. turcica f. sp. zeae and two from S. turcica f. sp. sorghi. To our knowledge, this is the first report of a polymycovirus infecting both S. turcica f. sp. zeae and S. turcica f. sp. sorghi.

In this research, novel citrus-pectin-based Ag@AgCl/graphite carbon nitride nanocomposites (CACNs) were formed by assembling citrus-pectin-based Ag@AgCl (CP-Ag@AgCl) prepared by daylight-driven reduction onto a graphite carbon nitride (g-C3N4) surface. The as-prepared samples were characterized by various techniques to study their morphology, structure, optical and electrochemical properties. The CACNs were found to have enhanced photoelectrochemical properties, which provided excellent photocatalytic activity to degrade the food colorant new coccine (NC) under visible light. For the NC degradation, the recommended photocatalyst offered approximately 95.7% of the degradation efficiency under visible-light irradiation for 60 min, which has superior photostability and reusability. Its degradation rate constant was approximately 5.16 and 37.6 times those of pure g-C3N4 and CP-Ag@AgCl, respectively. The NC degradation conformed to pseudo-first-order kinetics. Thus, the developed nanocomposites provide a useful reference for the removal of hazardous food colorants and resource utilization of peel waste.

Colorectal cancer (CRC) is the third most common cancer worldwide. Long non-coding RNA (lncRNA) small nucleolar RNA host gene 8 (SNHG8) acts as an oncogene in different types of cancer, including prostate, breast and ovarian cancer. SNHG8 promotes the tumorigenesis of CRC; however, its underlying molecular mechanism remains unclear. The present study aimed to explore the mechanism of SNHG8 on CRC development via various assays, including western blot, pull-down, PCR and immunofluorescence assays. The results of the present study demonstrated that SNHG8 expression was substantially upregulated in primary tumor tissues from The Cancer Genome Atlas dataset. Western blot and immunofluorescence analyses demonstrated that SNHG8 facilitated cell proliferation and autophagy in CRC cells. Notably, the function of SNHG8 in enhancing autophagy was dependent on autophagy-related gene 7 (ATG7). In addition, western blot analysis indicated that the effect of SNHG8 on autophagy in CRC cells was dependent on the miR-588/ATG7 axis. Taken together, the results of the present study suggest that SNHG8 promotes autophagy in CRC cells.

In our research, 16 sets of primers were used to amplify contiguous, overlapping segments of the complete mitochondrial DNA (mtDNA) of Odontobutis obscura. The aim of our research was to provide some important genetic background information for conservational practice of these valuable fish species. The total length of the mitochondrial genome is 16 864 bp and deposited in the GenBank with accession numbers KT438552. The gene arrangement and transcriptional direction were similar to other bony fishes which contained 37 genes (13 protein-coding genes, 2 ribosomal RNA, and 22 transfer RNAs) and a major non-coding control region. The nucleotide skewness for the coding strands of O. obscura (GC-skew = -0.26) is biased toward G and the negative GC-skew ranges from -0.51(ND2) to -0.24(CO1). The phylogenetic analysis showed that the Gobiiformes could be divided into three groups, Eleotridae, Odontobutidae and Rhyacichthyidae. The O. Obscura and O. Potamophila are sister species and clustered together.

By using time domain reflectometry (TDR), a fixed-position monitoring research on the dynamics of soil water under Eucommia ulmoides plantation was conducted in a hilly red soil region of southern China. The results showed that there was a significant difference in the soil water storage among different months, and the dynamics of soil water could be divided into the phases of reduction, increase, and more reduction. Soil water varied significantly in its vertical distribution, and the variation pattern also differed with seasons. The vertical distribution of soil water could be divided into two phases, i.e., accumulation and depletion based on the seasonal variation of soil water, or rainy season and dry season based on the monthly variation of rainfall. Soil water was correlated significantly (P < 0.05) with relative humidity (RH), air temperature (t), vapor pressure deficit (VPD) and rainfall (R), and regulated by these meteorological factors synthetically, among which, rainfall was the most important factor, followed by air temperature. The soil water loss rate after rain had a significant hyperbolic relationship with durative droughty days, while soil water storage had a significant linear negative relationship with this duration (P < 0.05). With the extension of drought duration after rain, soil water loss tended to vary gently with increasing soil depth.
