RESUMO

Colorectal cancer (CRC) is a prevalent tumour with high morbidity rates worldwide, and its incidence among younger populations is rising. Early diagnosis of CRC can help control the associated mortality. Fungi are common microorganisms in nature. Recent studies have shown that fungi may have a similar association with tumours as bacteria do. As an increasing number of tumour-associated fungi are discovered, this provides new ideas for the diagnosis and prognosis of tumours. The relationship between fungi and colorectal tumours has also been recently identified by scientists. Therefore, this paper describes the limitations and prospects of the application of fungi in diagnosing CRC and predicting CRC prognosis.

This article presents a comprehensive review of the factors influencing the efficacy of mesenchymal stem cells (MSCs) transplantation and its association with platelet concentrates (PCs). It focuses on investigating the impact of PCs' composition, the age and health status of platelet donors, application methods, and environmental factors on the outcomes of relevant treatments. In addition, it delves into the strategies and mechanisms for optimizing MSCs transplantation with PCs, encompassing preconditioning and combined therapies. Furthermore, it provides an in-depth exploration of the signaling pathways and proteomic characteristics associated with preconditioning and emphasizes the efficacy and specific effects of combined therapy. The article also introduces the latest advancements in the application of biomaterials for optimizing regenerative medical strategies, stimulating scholarly discourse on this subject. Through this comprehensive review, the primary goal is to facilitate a more profound comprehension of the factors influencing treatment outcomes, as well as the strategies and mechanisms for optimizing MSCs transplantation and the application of biomaterials in regenerative medicine, offering theoretical guidance and practical references for related research and clinical practice.

Background: Copper (Cu) is a vital trace element involved in numerous physiological processes, including glycolysis and lipid metabolism. Imbalances in Cu homeostasis can contribute to various diseases. However, current research on the impact of Cu on lipid metabolism has yielded inconsistent findings. Moreover, studies investigating the effects of dietary Cu intake on blood lipids among women of childbearing age are rare. Understanding of this relationship could enhance lipid management, given that most women obtain Cu through their diet. Additionally, the gut microbiota may play a role in this process. This study aims to investigate the effects of dietary Cu intake on blood lipids in women of childbearing age and to analyze the role of gut microbiota in this process. Methods: This study utilized data from the National Health and Nutrition Examination Survey (NHANES) to conduct a preliminary analysis of the correlation between dietary Cu levels and blood lipid indicators in women of childbearing age. Subsequently, an on-site research was conducted to further investigate this relationship, followed by animal experiments to verify the effect of different Cu doses on blood lipid levels. Multiple linear regression models, ANOVA, XGBOOST were employed to analyze the impact of Cu on blood lipids and the role of intestinal microbiota in this process. Results: In the population study, the NHANES results were consistent with on-site findings. The TG, and TC levels in women with childbearing were increased with higher dietary Cu intake. Animal experiments have shown that as Cu intake increases, TC levels increase. Furthermore, when the Cu intake reached 8 mg/day (the recommended dietary Cu intake limit of China, RDI), the TG levels in the research animals decrease, alongside a reduction in the abundance of Weissella cibaria (probiotics related to lipid metabolism), and the levels of LPS and IL-6 increase. Conclusion: The blood lipid levels of women of childbearing age increase with higher dietary Cu intake. RDI of 8 mg/day for women of childbearing age in China may need to be appropriately reduced. Regulating the gut microbiota, especially by increasing the abundance of Weissella cibaria may be an effective intervention for blood lipids.

BACKGROUND: Cerebral palsy (CP) is a condition resulting from perinatal brain injury and can lead to physical disabilities. Exosomes derived from human amniotic mesenchymal stromal cells (hAMSC-Exos) hold promise as potential therapeutic options. OBJECTIVE: This study aimed to investigate the impact of hAMSC-Exos on neuronal cells and their role in regulating apoptosis both in vitro and in vivo. METHODS: hAMSC-Exos were isolated via ultracentrifugation and characterized via transmission electron microscopy, particle size analysis, and flow cytometry. In vitro, neuronal damage was induced by lipopolysaccharide (LPS). CP rat models were established via left common carotid artery ligation. Apoptosis levels in cells and CP rats were assessed using flow cytometry, quantitative reverse transcription polymerase chain reaction (RT-qPCR), Western blotting, and TUNEL analysis. RESULTS: The results demonstrated successful isolation of hAMSC-Exos via ultracentrifugation, as the isolated cells were positive for CD9 (79.7%) and CD63 (80.2%). Treatment with hAMSC-Exos significantly mitigated the reduction in cell viability induced by LPS. Flow cytometry revealed that LPS-induced damage promoted apoptosis, but this effect was attenuated by treatment with hAMSC-Exos. Additionally, the expression of caspase-3 and caspase-9 and the Bcl-2/Bax ratio indicated that excessive apoptosis could be attenuated by treatment with hAMSC-Exos. Furthermore, tail vein injection of hAMSC-Exos improved the neurobehavioral function of CP rats. Histological analysis via HE and TUNEL staining showed that apoptosis-related damage was attenuated following hAMSC-Exo treatment. CONCLUSIONS: In conclusion, hAMSC-Exos effectively promote neuronal cell survival by regulating apoptosis, indicating their potential as a promising therapeutic option for CP that merits further investigation.

Introduction: Depression symptoms are prevalent globally, including China, with a notable impact on college students. This study aims to not only estimate the prevalence of depressive tendencies and attributional styles among college students in the post-pandemic era but also explore the relationship between the two factors. The findings of this study can provide new insights into early intervention and support services for individuals exhibiting tendencies toward depression. Methods: The survey was administered to college students from various academic backgrounds at a specific university in southern Jiangxi Province by employing two scales to investigate depressive tendencies and attributional styles. Depressive tendency was evaluated using the Patient Health Questionnaire-9 (PHQ-9), while the attributional styles were assessed using the Multidimensional-Multiattributional Causality Scale (MMCS). Univariate analysis and multiple logistic regressions were conducted to identify the related factors of depressive tendency. Results: A relatively high (48.9%) prevalence of depression among college students was found in this study. College students with higher grades (OR = 1.574, 95%CI: 1.369-1.810), profession of medicine and allied health sciences (OR = 1.779, 95%CI: 1.203-2.629), experiencing higher study stress (OR = 2.006, 95%CI: 1.601-2.514), and having poor physical condition (OR = 1.527, 95%CI: 1.247-1.869) were identified as risk factors for depressive tendency. The correlation between higher grades and increased learning pressure, coupled with poorer physical condition, heightens the vulnerability of college students to depression. Moreover, the more they attribute these experiences to achievement effort (OR = 0.897, 95%CI: 0.828-0.972), achievement ability (OR = 0.903, 95%CI: 0.838-0.972), and affiliation context (OR = 0.919, 95%CI: 0.860-0.982), the less likely they are to develop depression. Conclusion: In the group of college students, especially those in higher grades, profession of medicine and allied health sciences or experiencing high learning pressure and poor physical condition, emphasizing the significance of their mental well-being becomes crucial. Offering suitable support and assistance is essential. Additionally, fostering the cultivation of positive attributional and coping strategies by attributing difficulties to controllable factors and instilling a belief in their ability to overcome challenges can help reduce the risk of depression.

Microbial volatile organic compounds (mVOCs) can serve as a communication channel among microorganisms, insects and plants, making them important in ecosystem. In order to understand the possible role of mVOCs in Arctic ecology, the microbes in Arctic flowers and their mVOCs and effects on plants were investigated. This study aims to isolate different yeast species from the flowers of five Arctic plant species and further to explore the function of mVOCs emitted by these microbes to plant. It was found that the composition and amount of mVOCs produced by the isolated yeasts were considerably affected by changes in incubation temperature. When the incubation temperature rose, the species of alcohols, aldehydes, esters, organic acids, and ketones increased, but substances specific to low temperature decreased or disappeared. When yeasts were co-cultured with Arabidopsis thaliana without any direct contact, mVOCs produced by the isolated yeasts inhibited the seed germination of A. thaliana at low temperatures; however, the mVOCs promoted the chlorophyll content, fresh weight, root weight and flowering rate of Arabidopsis plants. Although the overall growth-promoting effect of yeast mVOCs was higher at 20°C than at 10°C, the growth-promoting effect on roots, flowers and chlorophyll was highest at 10°C. When cultured at 10°C, the mVOCs produced by Cystofilobasidium capitatum A37, Cryptococcus sp. D41, and Sporidiobolus salmonicolor D27 had the highest growth-promoting effects on the root, flowering rate and chlorophyll content of Arabidopsis, respectively. In the co-culture system, some new mVOCs were detected, such as hendecane, tetradecane, and 1-hexanol that have been proven to promote plant growth. In addition, mVOCs of the isolated Arctic yeasts could inhibit the growth of several microorganisms, especially filamentous fungi. It was the first time to prove that mVOCs produced by the isolated yeasts had varying effects on plant growth at different incubating temperatures, providing a reference for the interactions between microorganisms and plants and their possible responses to climate change in the Arctic area. Moreover, the characteristics of promoting plant growth and inhibiting microbial growth by mVOCs of Arctic yeasts would lay a foundation for potential applications in the future.

A pale pink, Gram-reaction-negative, non-motile, aerobic bacterium, designated MC 3624T, was isolated from a tundra soil near Ny-Ålesund, Svalbard Archipelago, Norway (78° N). Growth occurred at 10-37 °C (optimum 25-30 °C) and at pH 6.0-9.0 (optimum pH 8.0). The predominant fatty acids were C16 : 0 (17.7 %), C18 : 1ω7c 11-methyl (13.4 %), summed feature 3 (C16 : 1ω7c and/or C16 : 1ω6c) (10.1 %) and summed feature 8 (C18 : 1ω6c and/or C18 : 1ω7c) (38.3 %). The major respiratory quinone was ubiquinone-10, and the main polar lipids were phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, phosphatidylcholine and an unidentified aminolipids. The DNA G+C content was 68.9 mol%. Carotenoids of the spirilloxanthin series were produced. The nearest neighbour to the novel strain was Roseomonas wooponensis WW53T (94.36 % 16S rRNA gene sequence similarity). On the basis of phenotypic, chemotaxonomic and phylogenetic data, strain MC 3624T represents a novel species of the genus Roseomonas, for which the name Roseomonas arcticisoli sp. nov. is proposed. The type strain is MC 3624T (=CCTCC AB 2014278T=LMG 28637T).

A Gram-stain-negative, aerobic, motile rod-shaped bacterium, designated 6-67T, was isolated from a soil sample collected from Longyearbyen, Svalbard. Its taxonomic position was investigated by genotypic, phenotypic and chemotaxonomic analyses. This isolate grew at 4-28 °C (optimum, 20 °C), at pH 5.0-9.0 (optimum, pH 7.0) and with 0-0.2 % (w/v) NaCl. Strain 6-67T contained Q-8 as a major respiratory quinone and MK-7 as a minor component; the major fatty acids were summed feature 3 (C16 : 1ω6c and/or C16 : 1ω7c) and C16 : 0. The major polar lipids were phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, two unidentified phospholipids and one unidentified aminophospholipid. The polyamines were putrescine and 2-hydroxyputrescine. 16S rRNA gene sequence analysis indicated that the novel strain 6-67T belonged to the family Oxalobacteraceae within the class Betaproteobacteria. The DNA G+C content of strain 6-67T was 56.21 mol%. On the basis of phylogenetic, physiological and chemotaxonomic data, strain 6-67T is considered to represent a novel species of the genus Undibacterium, for which the name Undibacterium arcticum sp. nov. is proposed. The type strain is 6-67T (=CCTCC AB 2015162T=KCTC 42986T).
