•Four newly recorded species of Podostemaceae from southern China were identified by molecular and morphological evidence.•17 plastomes of Podostemaceae were newly sequenced and two novel polymorphic barcodes (ccsA and ndhA) detected.•Our findings reveal greater species richness (15 species from five genera) of Podostemaceae in China and supply molecular resources for research on taxonomy and phylogenomics of this enigmatic aquatic family.

Plant terrestrialization (i.e. the transition to a terrestrial environment) is a significant evolutionary event that has been intensively studied. While certain plant lineages, particularly in angiosperms, have re-adapted to freshwater habitats after colonizing terrene, however, the molecular mechanism of the terrestrial-freshwater (T-F) transition remains limited. Here, the basal monocot Araceae was selected as the study object to explore the T-F transition adaptation mechanism by comparative genomic analysis. Our findings revealed that the substitution rates significantly increased in the lineage of freshwater Araceae, which may promote their adaptation to the freshwater habitat. Additionally, 20 gene sets across all four freshwater species displayed signs of positive selection contributing to tissue development and defense responses in freshwater plants. Comparative synteny analysis showed that genes specific to submerged plants were enriched in cellular respiration and photosynthesis. In contrast, floating plants were involved in regulating gene expression, suggesting that gene and genome duplications may provide the original material for plants to adapt to the freshwater environment. Our study provides valuable insights into the genomic aspects of the transition from terrestrial to aquatic environments in Araceae, laying the groundwork for future research in the angiosperm.

Background: Acute myocardial infarction (AMI) can lead to sudden cardiac death after prolonged ischemia or heart failure (HF) and impaired left ventricular pump function. However, the underlying mechanism remains largely unknown. The purpose of this study was to investigate the role of mitofilin in alleviating AMI. Methods: Recombinant adenoviral vectors for mitofilin overexpression or mitofilin knockdown were constructed, respectively. A mouse AMI model was established and the effect of mitofilin on myocardial pyroptosis was examined by detecting the lactate dehydrogenase (LDH) level and inflammatory factors. Moreover, a cellular model of AMI was established by treating cardiomyocytes with hypoxia/reoxygenation (H/R). An enzyme-linked immunosorbent assay (ELISA) and a western blot analysis were used to detect the effect of mitofilin knockdown on the expression of pyroptosis-related factors. Furthermore, the regulatory role of mitofilin in PI3K/AKT pathway was evaluated by the western blot and PI3K inhibitor. Results: Mitofilin was downregulated in the heart tissue of the AMI mice and H/R induced cardiomyocytes. The overexpression of mitofilin significantly alleviated AMI and reduced pyroptosis-related factors. Meanwhile, in cardiomyocytes, mitofilin knockdown aggravated cellular damages by promoting pyroptosis. Further analysis showed that the anti-pyroptotic effect of mitofilin was dependent on the activation of the PI3K/AKT signaling pathway. Conclusions: Our study suggests that mitofilin regulates pyroptosis through the PI3K/AKT signaling pathway in cardiomyocytes to ameliorate AMI, which may serve as a therapeutic strategy for the management of AMI.

BACKGROUND: Primary percutaneous coronary intervention (PPCI) is the standard procedure for reperfusion for ST-segment elevation myocardial infarction (STEMI), but the occurrence of the no-reflow phenomenon remains common and is associated with adverse outcomes. AIMS: This study aimed to evaluate whether prolonged balloon inflation in stent deployment would lessen the occurrence of the no-reflow phenomenon in PPCI compared with conventional rapid inflation/deflation strategy. METHODS: Patients were randomly assigned to either the prolonged balloon inflation in stent deployment group (PBSG) or conventional deployment strategy group (CDSG) in a 1:1 ratio. A subset of patients was included in the cardiac magnetic resonance (CMR) assessment. RESULTS: Thrombolysis in MI (TIMI) flow grade 3 was found in 96.7% and 63.3% of the patients of the PBSG and CDSG, respectively (P = 0.005). The results of the PBSG and CDSG are respectively shown as follows: 0% versus 30% no-reflow or slow flow (P = 0.002); 90% versus 66.7% ST-segment resolution ≥ 50% (P = 0.028); 35.6 ± 14.5 frames versus 49.18 ± 25.2 frames on corrected TIMI frame count (P = 0.014); and 60% versus 20% myocardial blush grade 3 (P = 0.001). At 1 month, the major cardiovascular adverse event (cardiovascular mortality) rate was 3.3% in both groups; at 1 year, the rate was 3.3% and 6.7% for the PBSG and CDSG, respectively (P = 1.00). In the CMR subset of cases, the presence of microvascular obstruction (MVO) was detected in 6.7% and 50% of the patients in the PBSG and CDSG, respectively (P = 0.023). CONCLUSION: In our pilot trial, prolonged balloon inflation during stent deployment strategy in PPCI reduces the occurrence of the no-reflow phenomenon in patients with STEMI and improved the myocardial microcirculation perfusion (ClinicalTrials.gov number: NCT03199014; registered: 26/June/2017).

The spatial distribution and association of populations can reflect succession patterns and its adaptation strategies to the change of environmental factors, with important significance for vegetation restoration, reconstruction, and biodiversity conservation. We used the point-pattern analytical method to examine the spatial distribution and its association of the Loropetalum chinense population in karst hills of Guilin, China, based on field surveys. On the basis of Ripley K function, we used the pair-correlation function statistic derived to explore difference in the distribution patterns.We used Ripley L function to examine the spatial associations among the three diameter classes. The results showed that the diameter class structure of the population exhibited an irregular inverted "J" type, with the small diameter class occupying a large proportion, indicating an increased population structure with good regeneration capability. The individuals of the three diameter classes had a clumped distribution at the small scale. With the increases of spatial scales, the aggregation intensity gradually weakened and tended to be randomly distributed. There was an independent spatial association between individuals among the three diameter classes at small scale. As the scale increased, the spatial association between individuals with different diameter classes became positive or negative association. The greater the difference in diameter class of L. chinense population, the weaker their spatial correlation was, which might gradually turn into negative association. Our findings contributed to a clearer understanding of the ecological strategies and scale-dependent cha-racteristics of species coexistence and underlying mechanisms during the growth and development of L. chinense population in karst hills of Guilin, providing reference for the restoration, reconstruction, protection and management of forest in karst hills.

Analyzing the interspecific association of major tree populations of Cyclobalanopsis glauca community and revealing the coexistence among the populations would provide theoretical basis for the restoration and reconstruction, management and biodiversity conservation of Karst hills in Gui-lin. Based on field survey, the χ2 test, association coefficient (AC), co-occurrence percentage (PC), Ochiai index (OI), Dice index (DI), and principal component analysis (PCA) were used to investigate the interspecific association and eco-taxonomic classification of the top 22 tree species in Cyclobalanopsis glauca community in Guilin Karst Rocky Mountains. The results showed that there was a significant positive correlation between the interspecific associations of the 22 main tree populations, indicating that this community was in a stable climax stage. The results of χ2 test showed that 108 pairs of 231 species pairs had positive associations, 115 species pairs had negative associations, 8 species pairs were unrelated, with the positive and negative linkage ratio being 93.9%. The vast majority of species pairs showed no significant association. The strong indepen-dence between species might be mainly caused by the higher habitat heterogeneity of the Karst hills in Guilin which led to the differentiation of niches between species. The results of AC, PC, OI and DI were basically consistent with that of χ2 test, indicating that the analysis of interspecific association should be based on the χ2 test and supplemented by AC, PC, OI and DI indices. According to the PCA and evaluation of interspecific relationships, the 22 main tree species were divided into three ecological groups. The species within each group had the same adaptability to habitat but different ecological requirements.

Heavy metal pollution is a major environmental problem of mine wasteland. Finding out the tolerant plants, which can adapt to the local climate and the soil conditions, is the premise of vegetation restoration. An extensive vegetation survey and sampling were conducted in Huayuan Mn and Pb/Zn mineland, 76 species belonging to 69 genera and 39 families were recorded. The main dominant species and their associated soils were determined for heavy metal concentrations. The results showed that soil Pb, Zn and Cd levels exceeded the threshold levels of Class II of China Environmental Quality Standard for Soils, which suggested minesoils might be polluted by the three elements. The main dominant plants can adapt to the unfavorable edaphic conditions of mineland and were tolerant to heavy metals. There were great variations of metal uptake and accumulation among different plant species. They were classified into three types according to the metal concentrations in the plant shoots and roots: the accumulator, e. g. Camellia oleifera and Dicranopteris dichotoma, absorbed a large amount of heavy metals by the roots and transported to the shoots, which can be used to clean up the soils containing light to moderate toxic metal concentration and with high-value; the root compartment, e. g. Rubus tephrodes, R. corchorifolius, R. chroosepalus, Artemisia princeps and Pteridium aquilinum also absorbed a large amount of heavy metals but held in the roots; and the excluder, e. g. Miscanthus sinensis, Imperata cylindrica, Indocalamus tessellatus and Toddalia asiatica, absorbed less heavy metals than the accumulators. The root compartment and the excluder were more suitable for remediation of the mine wastelands with high heavy metal concentration, low-value and extensive area.
