Cancer is a highly heterogeneous disease, and different individuals of the same cancer type can display different therapeutic effects and prognosis. Genetic variation of long non-coding RNA is the key factor driving tumor development, and plays an important role in genetic and biological heterogeneity. Therefore, it is of great significance to identify lncRNA as a driving factor in the non-coding region and explain its function in tumors for revealing the pathogenesis of cancer. In this study, we developed an integrated method to identify Personalized Functional Driver lncRNAs (PFD-lncRNAs) by integrating the DNA copy number data, gene expression data, and the biological subpathways information. Then, we applied the method to identify 2695 PFD-lncRNAs in 5334 samples across 19 cancer types. We performed an analysis of the association between PFD-lncRNAs and drug sensitivity, which provides medication guidance in disease therapy and drug discovery in the individual. Our research is of great significance for elucidating the biological roles of lncRNA genetic variation in cancer, revealing the related mechanism of cancer, and providing novel insights for individualized medicine.

Traffic activities release large amounts of trace metal(loid)s in urban environments. However, the impact of vehicle operation-associated emissions on trace metal(loid) enrichment in road dust and the potential migration of these trace metal(loid)s to the surrounding environment remain unclear. We evaluated the contamination, sequential fraction, and bioaccessibility of trace metal(loid)s in urban environments by assessing their presence in road dust, garden vegetables, and tree tissues, including bark and aerial roots, at a traffic-training venue impacted by vehicle operation emissions and, finally, calculated the bioaccessibility-based health risk. The results indicated a significant accumulation of trace metal(loid)s in road dust, with the highest lead (Pb), cadmium (Cd), and antimony (Sb) concentrations in the garage entrance area due to higher vehicle volumes, frequent vehicle starts and stops, and lower speeds. Aerial roots exposed to hill start conditions exhibited the highest Pb, Zn, and Sb levels, potentially caused by high road dust resuspension, confirming that this tree tissue is an appropriate bioindicator. Sequential extraction revealed high percentages of carbonate-, Fe/Mn oxide-, and organic/sulphide-associated fractions of Pb, copper (Cu), and zinc (Zn) in road dust, while most Cd, Cr, Ni, and Sb occurred as residual fractions. According to the potential mobilizable fractions in sequential extraction, the in vitro gastrointestinal method could be more suitable than the physiologically based extraction test to evaluate the bioaccessibility-related risk of traffic-impacted road dust. The bioaccessibility-based health risk assessment of the road dust or soil confirmed no concern about noncarcinogenic risk, while the major risk originated from Pb although leaded gasoline was prohibited before the venue establishment. Furthermore, the cancer risks (CRs) analysis showed the probable occurrence of carcinogenic health effects from Cd and Ni to adults and from Cd, Cr, and Ni to children. Furthermore, the Cd and Pb concentrations in the edible leaves of cabbage and radish growing in gardens were higher than the recommended maximum value. This study focused on the health risks of road dust directly impacted by vehicle emissions and provides accurate predictions of trace metal(loid) contamination sources in the urban environment.

This article systematically investigates the technology licensing by Stanford University. We analyzed all the inventions marketed by Stanford's Office of Technology Licensing (OTL) between 1970 to 2020, with 4,512 inventions from 6,557 inventors. We quantified how the innovation landscape at Stanford changed over time and examined factors that correlate with commercial success. We found that the most profitable inventions are predominantly licensed by inventors' own startups, inventions have involved larger teams over time, and the proportion of female inventors has tripled over the past 25 years. We also identified linguistic features in how the inventors and OTL describe the inventions that significantly correlate with the invention's future revenue. Interestingly, inventions with more adjectives in their abstracts have worse net income. Our study opens up a new perspective for analyzing the translation of research into practice and commercialization using large-scale computational and linguistics analysis.

An estimated 3 billion people lack access to dermatological care globally. Artificial intelligence (AI) may aid in triaging skin diseases and identifying malignancies. However, most AI models have not been assessed on images of diverse skin tones or uncommon diseases. Thus, we created the Diverse Dermatology Images (DDI) dataset-the first publicly available, expertly curated, and pathologically confirmed image dataset with diverse skin tones. We show that state-of-the-art dermatology AI models exhibit substantial limitations on the DDI dataset, particularly on dark skin tones and uncommon diseases. We find that dermatologists, who often label AI datasets, also perform worse on images of dark skin tones and uncommon diseases. Fine-tuning AI models on the DDI images closes the performance gap between light and dark skin tones. These findings identify important weaknesses and biases in dermatology AI that should be addressed for reliable application to diverse patients and diseases.

The nucleolus is a nuclear structure composed of ribosomal DNA (rDNA), and functions as a site for rRNA synthesis and processing. The rDNA is guanine-rich and prone to form G-quadruplex (G4), a secondary structure of DNA. We have recently found that HERC2, an HECT ubiquitin ligase, promotes BLM and WRN RecQ DNA helicases to resolve the G4 structure. Here, we report the role of HERC2 in the regulation of nucleolar localization of the helicases. Furthermore, HERC2 inactivation enhances the effects of CX-5461, an inhibitor of RNA polymerase I (Pol I)-mediated transcription of rRNA with an intrinsic G4-stabilizing activity. HERC2 depletion or homozygous deletion of the C-terminal HECT domain of HERC2 prevented the nucleolar localization of BLM and WRN, and inhibited relocalization of BLM to replication stress-induced nuclear RPA foci. HERC2 colocalized with fibrillarin and Pol I subunit RPA194, both of which are required for rRNA transcription. The HERC2 dysfunction enhanced the suppression of pre-rRNA transcription by CX-5461. These results suggest the effect of HERC2 status on the functions of BLM and WRN on rRNA transcription in the nucleolus. Since HERC2 is downregulated in numerous cancers, this effect may be clinically relevant considering the beneficial effects of CX-5461 in cancer treatments.

To assess the impact of antibiotic pollution to the ecosystem in urban water, the occurrence, seasonal, and spatial distributions, potential sources, and ecological risks of 18 targeted antibiotics in urban river, Pearl River located in Guangzhou city, were investigated. Surface water samples were sampled from 24 sites in Guangzhou center of Pearl River during dry and wet seasons. The results indicated that the concentrations of antibiotic residues were at the nanogram per liter level, except sulfamethazine (SMD) (µg/L). Sulfonamides (SAs) were the dominant antibiotics, contributing 60.4-65.0% to the total antibiotics. The concentrations of SAs, fluoroquinolones (QUs), macrolides (MLs), tetracyclines (TCs), and lincosamides (LCs) were higher in dry season than those in wet season at most sampling sites, which possibly resulted from the dilution effect of heavy rainfall. The concentrations of the antibiotic residues in Guangzhou were comparable or higher than other urban rivers. The calculation on risk quotients indicated that erythromycin-H2O (ETM-H2O) and tetracycline (TC) were of high risks. The source identification by the Pearson correlation analysis and principal component analysis-multiple linear regression (PCA-MLR) method suggested that municipal wastewater treatment plants were primary sources of antibiotics. These results would provide important information for the environmental protect.

Replication protein A (RPA) binds to and stabilizes single-stranded DNA and is essential for the genome stability. We reported that an E3 ubiquitin ligase, HERC2, suppresses G-quadruplex (G4) DNA by regulating RPA-helicase complexes. However, the precise mechanism of HERC2 on RPA is as yet largely unknown. Here, we show essential roles for HERC2 on RPA2 status: induction of phosphorylation and degradation of the modified form. HERC2 interacted with RPA through the C-terminal HECT domain. Ubiquitination of RPA2 was inhibited by HERC2 depletion and rescued by reintroduction of the C-terminal fragment of HERC2. ATR-mediated phosphorylation of RPA2 at Ser33 induced by low-level replication stress was inhibited by depletion of HERC2. Contrary, cells lacking HERC2 catalytic residues constitutively expressed an increased level of Ser33-phosphorylated RPA2. HERC2-mediated ubiquitination of RPA2 was abolished by an ATR inhibitor, supporting a hypothesis that the ubiquitinated RPA2 is a phosphorylated subset. Functionally, HERC2 E3 activity has an epistatic relationship with RPA in the suppression of G4 when judged with siRNA knockdown experiments. Together, these results suggest that HERC2 fine-tunes ATR-phosphorylated RPA2 levels through induction and degradation, a mechanism that could be critical for the suppression of secondary DNA structures during cell proliferation.

The proliferation of pollution in aquatic environments has become a growing concernand calls for the development of novel adsorbents capable of selectively removing notorious andrecalcitrant pollutants from these ecosystems. Herein, a general strategy was developed for thesynthesis and functionalization of molecularly imprinted polymer microspheres (MIPs) that couldbe optimized to possess a significant adsorption selectivity to an organic pollutant in aqueousmedia, in addition to a high adsorption capacity. Considering that the molecular imprinting alonewas far from satisfactory to produce a high-performance MIPs-based adsorbent, further structuralengineering and surface functionalization were performed in this study. Although the more carboxylgroups on the surfaces of the MIPs enhanced the adsorption rate and capacity toward an organicpollutant through electrostatic interactions, they did not strengthen the adsorption selectivity in aproportional manner. Through a systematic study, the optimized sample exhibiting both impressiveselectivity and capacity for the adsorption of the organic pollutant was found to possess a smallparticle size, a high specific surface area, a large total pore volume, and an appropriate amount ofsurface carboxyl groups. While the pseudo-second-order kinetic model was found to better describethe process of the adsorption onto the surface of MIPs as compared to the pseudo-first-order kineticmodel, neither Langmuir nor Freundlich isothermal model could be used to well fit the isothermaladsorption data. Increased temperature facilitated the adsorption of the organic pollutant onto theMIPs, as an endothermic process. Furthermore, the optimized MIPs were also successfully employedas a stationary phase for the fabrication of a molecularly imprinted solid phase extraction column,with which purchased food-grade fish samples were effectively examined.

Malachite green (MG) is currently posing a carcinogenic threat to the safety of human lives; therefore, it is highly desirable to develop an effective method for fast trace detection of MG. Herein, for the first time, this paper presents a systematic study on polymer microspheres, being prepared by combined Pickering emulsion polymerization and molecular imprinting, to detect and purify MG. The microspheres, molecularly imprinted with MG, show enhanced adsorption selectivity to MG, despite a somewhat lowered adsorption capacity, as compared to the counterpart without molecular imprinting. Structural features and adsorption performance of these microspheres are elucidated by different characterizations and kinetic and thermodynamic analyses. The surface of the molecularly imprinted polymer microspheres (M-PMs) exhibits regular pores of uniform pore size distribution, endowing M-PMs with impressive adsorption selectivity to MG. In contrast, the microspheres without molecular imprinting show a larger average particle diameter and an uneven porous surface (with roughness and a large pore size), causing a lower adsorption selectivity to MG despite a higher adsorption capacity. Various adsorption conditions are investigated, such as pH and initial concentration of the solution with MG, for optimizing the adsorption performance of M-PMs in selectively tackling MG. The adsorption kinetics and thermodynamics are deeply discussed and analyzed, so as to provide a full picture of the adsorption behaviors of the polymer microspheres with and without the molecular imprinting. Significantly, M-PMs show promising solid-phase extraction column applications for recovering MG in a continuous extraction manner.

BACKGROUND Combretastatin A4 (CA4) is a potential therapeutic candidate for a variety of human cancer treatments. However, the inhibitive effects of CA4 on thyroid cancer cells are still not well-clarified. This study aimed to investigate the potential effect of CA4 on thyroid cancer cells, as well as underlying mechanism. MATERIAL AND METHODS Human thyroid papillary carcinoma cell line TPC1 was pre-treated with 5 concentrations of CA4 (0, 1, 2, 5, or 10 µM) for 2 h. Cell proliferation was determined by 3-(4, 5-dimethyl-2- thiazolyl)-2, 5-diphenyl -2-H-tetrazolium bromide (MTT) assay. Cell migration and invasion were detected by a modified Boyden chamber assay. Moreover, cell apoptosis was detected by terminal deoxynucleotidyl (TUNEL) staining assay and flow cytometry method. Western blot analysis was performed to determine the expression changes of epithelial-mesenchymal transition (EMT)-related proteins and phosphatidylinositol-3-kinase/serine/threonine kinase (PI3K/Akt) signaling pathway proteins. RESULTS CA4 significantly inhibited the cell proliferation, migration, and invasion, and significantly promoted cell apoptosis in a dose-dependent manner compared with the control group. The EMT-related protein levels of N-Cadherin, Vimentin, Snail1, Slug, Twist1, and ZEB1 were significantly decreased by CA4, while E-cadherin had no significant difference compared with the control group. Moreover, PI3K/Akt signaling pathway protein levels of p-PI3K and p-Akt were significantly decreased, whereas PI3K and Akt had no significant differences compared with the control group. CONCLUSIONS CA4 can inhibit proliferation, migration, and invasion and promote apoptosis of TPC1 cells. These effects might be through the PI3K/Akt signaling pathway. CA4 may be a potential therapeutic target for the treatment of thyroid cancer.

Oil spills and industrial organic pollutants have induced severe water pollution and threatened every species in the ecological system. To deal with oily water, special wettability stimulated materials have been developed over the past decade to separate oil-and-water mixtures. Basically, synergy between the surface chemical composition and surface topography are commonly known as the key factors to realize the opposite wettability to oils and water and dominate the selective wetting or absorption of oils/water. In this review, we mainly focus on the development of materials with either super-lyophobicity or super-lyophilicity properties in oil/water separation applications where they can be classified into four kinds as follows (in terms of the surface wettability of water and oils): (i) superhydrophobic and superoleophilic materials, (ii) superhydrophilic and under water superoleophobic materials, (iii) superhydrophilic and superoleophobic materials, and (iv) smart oil/water separation materials with switchable wettability. These materials have already been applied to the separation of oil-and-water mixtures: from simple oil/water layered mixtures to oil/water emulsions (including oil-in-water emulsions and water-in-oil emulsions), and from non-intelligent materials to intelligent materials. Moreover, they also exhibit high absorption capacity or separation efficiency and selectivity, simple and fast separation/absorption ability, excellent recyclability, economical efficiency and outstanding durability under harsh conditions. Then, related theories are proposed to understand the physical mechanisms that occur during the oil/water separation process. Finally, some challenges and promising breakthroughs in this field are also discussed. It is expected that special wettability stimulated oil/water separation materials can achieve industrial scale production and be put into use for oil spills and industrial oily wastewater treatment in the near future.

Solid surfaces possessing both superhydrophobic and superoleophilic properties have attracted significant interest in fundamental investigations and potential applications in the fields of self-cleaning surfaces, oil/water separation, and microfluidic channels. In this paper, a general methodology for robust superhydrophobic fabrics and sponges was proposed via the in situ growth of both transition-metal oxides and metallic nanocrystals, including the simple neutralization reaction and oxidation-reduction reaction. The porous surfaces coated with Group VIII and IB nanocrystals (such as Fe, Co, Ni, Cu, and Ag) can not only present multiscale surface roughness, but also readily coordinate with thiols, leading to special wettability. In our previous work, it has been confirmed that the interaction between the nanocrystals and thiols plays a significant role in the introduction of hydrophobic ingredients. In this work, it has been demonstrated that the efficient control of the nucleation and growth of Group VIII and IB nanocrystals on the porous surfaces becomes the key factor in the formation of multiscale surface roughness, resulting in the achievement of controllable special wettability. In addition, these as-prepared superhydrophobic and superoleophilic fabrics and sponges were successfully used for application in oil/water separation.
