The species Leopardus pardalis is one of the species of neotropical felines more studied in its ecological aspects, however, information of morphophysiological questions of the digestive tract are not found in the literature. Aiming to contribute with such information, the objective of this work was to characterize the morphology of the stomach of this species. Five adult specimens, collected after death by trampling, or donated by IBAMA to the Laboratory of Zoology and Animal Morphology of the University of the State of Mato Grosso - Alta Floresta, were used in this research. The animals were fixed with aqueous solution of formaldehyde 10%, dissected through the basic techniques and instruments of anatomy, and later the material was collected for microscopy. The macroscopic analysis revealed that the stomach was of the unicavitary type, with small and great curvature, located in the left antimer and constituted by the characteristic regions of the organ. In its microscopic aspects it was formed by the four usual tunics of the digestive tube: tunica mucosa, submucosa, muscular and serosa, each one with the peculiarities characteristic of each region, mainly in the tunica mucosa. The stomach of L. pardalis revealed similar morphology to the domestic and wild species, with small histomorphological peculiarities in the pyloric region, which may be related to evolutionary adaptations in the digestive process of this species.(AU)

RESUMO

Abstract With the growing volume of research involving Thysanoptera in Brazil, studies were carried out to improve our understanding of the diversity of thrips in areas where the fauna has historically been neglected. Accordingly, we recorded the diversity of thrips (Insecta: Thysanoptera) associated with a restinga fragment located on the campus of the State University of Ceará (UECE), Fortaleza, Ceará state, and computed the estimated richness and diversity indices. Samples were collected from 2011 through 2013 from flowers of 86 plant species. The material was taken to the Laboratory of Insect-Plant Interaction, where thrips were screened under stereomicroscope. We collected 456 adults and 58 immatures, representing 14 species, in addition to one unidentified species of Treherniella. Microcephalothrips abdominalis was found on a large number of host plants, and Frankliniella insularis was the most common species. About two-thirds of the total richness of thrips species was associated with three plant families (Amaranthaceae, Caesalpiniaceae and Poaceae); six thrips species were each associated with only one plant species. The richness of the species collected was close to that estimated by Bootstrap and Jackknife 1 analysis. The Shannon-Wiener (H') and Simpson (D) diversity indexes were 1,7607 and 0.7769, respectively. Although the species are common, 46 new associations between plant species and thrips were established, 13 of which are true host associations, which demonstrates the importance of coastal vegetation in maintaining populations of thrips.

We report for the first time the detection of Scirtothrips dorsalis Hood (Thysanoptera: Thripidae) in Brazil and describe the occurrence of the thrips on leaves of ungrafted dwarf-cashew Anacardium occidentale Linnaeus 1753 (Anacardiaceae), maintained into a greenhouse, in the northeastern state of Ceará. This exotic polyphagous species listed as absent quarantine pest in the country is originated in Asia, but since the beginning of this century, it has readily dispersed despite the strict quarantine regulations currently in several countries. Individuals of S. dorsalis identified in Brazil are similar to specimens found in South Africa rather than Asia by virtue of the absence of conspicuous reticulation on the posterior half of the metanotum and the presence of complete lines of microtrichia restricted to the posterior part of the abdominal sternites. Scirtothrips dorsalis is a particularly invasive pest and its introduction represents a potential threat to various crops in Brazil, especially mango.

The present study aimed to evaluate the profile of immunoglobins profile, clinical and bacteriological cure after different treatment routes of clinical bovine mastitis. Here, 30 Holstein cows with clinical mastitis in one quarter were uniformly divided into: 10 dairy cows that received 8.5mg of cefquinome sulphate administrated intramammarily during three consecutive milkings and 2.5mg/kg of enrofloxacin administrated parenterally during three consecutive days (Group 1); 10 dairy cows that received 8.5mg of cefquinome sulphate administrated intramammarily during three consecutive milkings (Group 2); and 10 dairy cows that received 2.5mg/kg of enrofloxacin administrated parenterally during three consecutive days (Group 3). Milk samples for somatic cell count, California Mastitis Test (CMT), bacteriological culture and quantification of IgG1, IgG2, IgM and IgA were collected before antimicrobial treatment, and after two, five and 12 days after the antimicrobial treatment. The combined treatment was more effective in the clinical cure rate, reduction of somatic cell count and CMT scores. Furthermore, the results demonstrated that milk concentration of different Igs classes were not related to prognosis of bovine clinical mastitis, and then cannot be considered as robust markers associated with clinical and bacteriological cures.(AU)

RESUMO

The present study evaluated the impact that orchiectomy, a routine but painful intervention in bovine husbandry, can cause on pulmonary immunity. To identify whether orchiectomy can impair lung defense, analyses of serum cortisol concentration and of alveolar macrophage and their function (phagocytosis and respiratory burst) were evaluated. Sixteen Holstein bulls (7 mo old, 250±50kg of body weight BW) were divided in two homogeneous groups - the castrated group and the sham group - and the sample were collected on Days -7, 1 and 7 relative to the day of the procedure. Serum cortisol concentration statistically increased on Days 1 and 7 (D-7: 4,97±1,28ng/ml; D1: 6,35 ±1,10ng/ml; D7: 8,28±1,94ng/ml) after castration and these variables seem to impact the alveolar macrophage percentage on D1 (D-7: 76,86±3,44%; D1: 60,92±2,44%; D7: 74,17±2,56%) and their respective function of phagocytosis (P) and the oxidative burst (OB) on Days 1 and 7 for the castrated group (P D-7: 56,25±15,63 arbitrary values; D1: 54,75±14,07 arbitrary values; D7: 31,77±8,44 arbitrary values; and OB D-7: 222,34±39,52 arbitrary values; D1: 135,25±37,68 arbitrary values; D7: 117,73±18,17 arbitrary values). These results indicate that surgical castration affected lung defense until seven days after the practice, so the pulmonary cell function was impaired for a period higher than that reported in the literature.(AU)

Several predisposing factors are described for mammary diseases or secondary disorders that compromise the quality and productivity of milk from females in different species. The phenotypic characteristics of the udder are considered in the economic evaluation of a female goat, whether for production potential, or as the genealogical record for these females. The limitation of studies correlating these features with the udder health raises doubt about the meaning of the conformation to the health and productivity of the mammary gland. Therefore, this study aimed to list the parameters of the udder conformation with the cellularity of the mammary gland checked by California Mastitis Test (CMT) and somatic cell count (SCC) in 80 Saanen goats without alterations in the clinical examination of the mammary gland or the Tamis test. It was observed that most of the phenotypic parameters of the udder did not influence the CCS, being considered purely esthetic. The circumference and udder depth showed negative correlation with the cellularity and moderate to high heritability traits can be considered parameters for genetic selection of goats.

RESUMO

Peritoneal dialysis (PD) is a renal substitutive therapy based on the infusion of a dialysate in the peritoneum, which induces through an osmotic gradient the ultrafiltration of water and the clearance of blood stream impurities by the peritoneal membrane. The colonization of Tenckhoff catheters (TCs) used in PD by pathogenic microorganisms can lead to peritonitis, and probably catheter removal. Here, optical microscopy and scanning electron microscopy were applied to study biofilm formation in 11 TCs. Biofilms varied in their morphology and thickness. Short-term catheters (6 months) presented thinner deposits (3 µm) with granular or flat morphologies, either on the intraluminal or external surfaces. Bacterial colonies were found on catheters from infected patients. A tendency was observed for long-term catheters (6-8 years) to present thicker biofilms (30-35 µm). Surprisingly, patients' cells colonized the deep layers of the thicker biofilms, forming a complex multicelullar community. It was concluded that the presence of a biofilm is not necessarily related with peritonitis, and biofilm features may correlate to the therapy time.

RESUMO

The effects of chloroquine intake on the retinal function in a Brazilian population of patients were assessed by multifocal electroretinography. Twenty-four randomly chosen eyes of patients treated with chloroquine for rheumatoid arthritis and systemic lupus erythematosus were examined using multifocal electroretinography (mfERG). Control measurements were acquired from 21 randomly chosen eyes of age-matched healthy subjects. None of the study participants had an inherited retinal disease or a Snellen visual acuity reduced to less than 20/40. In patients and control subjects, cumulative chloroquine dose, total daily dose, duration of treatment, retinal examination, visual field defects, visual acuity, and the mfERG were assessed. The average amplitudes and implicit times of the N1, P1, and P2 components of the mfERGs were measured in the central hexagon (R1) and in five rings (R2-R6). The values measured in patients and normal subjects were compared. The P1 amplitudes in R2 were significantly decreased in the patients. In addition, the amplitudes of N1 and N2 in R1 were significantly smaller in the patients. The implicit times of none of the components were significantly different between patients and controls. The response amplitude was not significantly correlated with cumulative dose and duration of intake. There was no correlation with retinal appearance, visual field, and visual acuity. In agreement with earlier data, the central mfERG amplitudes were decreased in chloroquine patients indicating functional alterations in the retina. These changes are also present in a Brazilian population suggesting that the effects of chloroquine are general and that genetic background and life circumstances probably have, if at all, only little effect.

RESUMO

AIMS: The purpose of this study was to demonstrate the usefulness of lectin obtained from Talisia esculenta (TEL) seeds as a tool to recognize and study Microsporum canis. For this purpose, we investigated the antifungal and marker action of this lectin and the relationship of these effects with the presence of carbohydrates on the structure of this fungus. METHODS AND RESULTS: The in vitro antifungal activity of TEL was analysed by broth microdilution assay. In addition, TEL was assessed against the arthroconidia present on hairs obtained from infected dogs and cats. The affinity of fluorescein isothiocyanate (FITC)-labelled TEL for macroconidia and arthroconidia of M. canis was also tested. The effects of TEL on the growth of the M. canis strains began with 0.125 mg ml(-1), and 100% inhibition was obtained with a concentration of 2 mg ml(-1). The addition of carbohydrates, especially N-acetyl-glucosamine and d-mannose, inhibited these antifungal effects. TEL was able to inhibit the growth of arthroconidial chitin-rich forms of M. canis obtained from hairs of infected animals and strains cultured in Sabouraud agar. FITC-labelled TEL efficiently marked macroconidial and arthroconidial forms of M. canis, as shown by fluorescent microscopy. CONCLUSIONS: These results show that the inhibitory effects of TEL on M. canis growth may be related to the interaction of lectin with the carbohydrates present at the micro-organism's surface, mainly D-mannose and N-acetyl-glucosamine. SIGNIFICANCE AND IMPACT OF THE STUDY: Talisia esculenta can be used as an important tool in the biochemical study of M. canis or as a molecule to recognize this dermatophyte in infected tissue.

RESUMO

There is increasing interest in the immune response induced by plant viruses since these could be used as antigen-expressing systems in vaccination procedures. Cowpea severe mosaic virus (CPSMV), as a purified preparation (300 g of leaves, 2 weeks post-inoculation), or crude extract from cowpea (Vigna unguiculata) leaves infected with CPSMV both administered by gavage to Swiss mice induced a humoral immune response. Groups of 10 Swiss mice (2-month-old females) were immunized orally with 10 daily doses of either 50 microg viral capsid protein (boosters of 50 microg at days 21 and 35 after immunization) or 0.6 mg protein of the crude extract (boosters of 0.6 mg at days 21 and 35 after immunization). Anti-CPSMV antibodies were quantified by ELISA in pooled sera diluted at least 1:400 at days 7, 14, 21, 28, 35 and 42 after the 10th dose. IgG and IgA against CPSMV were produced systemically, but IgE was not detected. No synthesis of specific antibodies against the proteins of leaf extracts from V. unguiculata, infected or not with CPSMV, was detected. The use of CPSMV, a plant-infecting virus that apparently does not induce a pathogenic response in animals, induced a humoral and persistent (at least 6 months) immune response through the administration of low antigen doses by gavage. These results raise the possibility of using CPSMV either as a vector for the production of vaccines against animal pathogens or in quick and easy methods to produce specific antisera for viral diagnosis.

RESUMO

The celebration of the Brazilian Nursing Association seventieth anniversary, former Brazilian Graduated Nurses Association, represents, in fact, seventy years of the history of Nursing in Brazil. We have certified, in a historical retrospective that the entity's creation, its organization which has been through several changes, adapting itself to the demands imposed by the conjunctures; the cultural landmarks, where the Brazilian Nursing Magazine, Brazilian Nursing Week, National Seminar of Research in Nursing, the Nursing Regional Meetings and the Brazilian Congress of Nursing are highlighted and are a demonstration of power and guts of this entity that survives from the struggle, conquests and achievements for the category. We also highlight the political advance these last two decades for the political and structural organization, the more democratic decisory process both internal and externally (ABEn's National Council); an autonomous social and political expression while civil society, better relationship with other national and international entities. Finally, we relate the challenges which appear to the entity's present management.

RESUMO

INTRODUCTION: An inhibitory serum factor of mixed lymphocyte culture (MLC) has been associated with successful pregnancy after lymphocyte transfusion in women with unexplained recurrent spontaneous abortions (RSA). OBJECTIVE: Investigate whether the inhibitory serum factor of MLC is essential for a successful pregnancy. METHOD: Sera from 33 healthy pregnant women and from 40 women with RSA were assessed by a one-way MLC in which the woman's lymphocytes were stimulated with her partner's lymphocytes or with third party lymphocytes. RESULTS: An inhibitory serum effect (inhibition > 50% as compared to normal serum) was detected in 45% of the pregnant women who had at least 1 previous parity, in 8% of the primigravidea, in 29% of those with one abortion and in 58% of those with more than one abortion. CONCLUSION: MLC inhibitory serum factor does not seem to be an essential factor for pregnancy development. Therefore, it should not be considered as a parameter for the assessment of RSA patients.

RESUMO

IL-4 is an essential mediator of IgE synthesis; it also upregulates the expression of the low affinity receptor for IgE (CD23) and its release in soluble form (sCD23). The involvement of IL-4 and sCD23 on the IgE synthesis control has been observed in experimental studies. IL-4 and sCD23 serum levels in asthmatic atopic children were determined in order to investigate a possible correlation between these factors and IgE levels. IL-4, total sCD23 and Dermatophagoides pteronyssinus specific IgE were determined in the serum of 19 asthmatic atopic children (aged 7 to 14) and in 13 age-matched controls. Since sCD23 may present an age-dependent variation, sCD23 and IL-4 serum levels were evaluated in 20 "wheezy babies." IL-4 serum levels were significantly higher in patients as compared to controls, whereas there was no significant difference between them concerning sCD23 levels. sCD23 levels were, however, significantly higher among "wheezy babies" as compared to atopic and nonatopic children. There was no correlation between IL-4 and sCD23 serum levels, nor between any of these factors and IgE levels in all groups. In conclusion, the enhanced IL-4 levels suggest that atopic patients have a preferential activation of Th2 subset. CD23 expression is markedly influenced by age.
